Objective To analyze the allergens detection in asthmatic infants and young children. Methods The specific IgE test and allergens screening test were conducted in 243 infants and young children with whooping diseases including asthmatic bronchitis, asthmatic pneumonia and asthma by skin prick test and Pharmacia UniCAP test. Results 1. Food allergen was the allergen detection most frequently in infants(P = 0.03). The proportion of aeroallergens increased significantly with age (positive rate:67. 2%). Milk and egg white was the main food allergen detection in infants and young children, and the main aeroallergen detection was house dust mite. 2. The allergen - specific IgE test using UniCAP test system was more accurate than skin prick test. Conclusions The infants are more susceptible to food hypersensitivity; the increasing opportunity of aeroallergen exposure may lead to a higher prevalence of aeroallergens in young children. UniCAP system is more accurate than other methods.

Objective To survey the current status of job stressor and exhaustion among nursing staff in the catheter room of grade Ⅲ-A general hospitals and to look for regularities. Methods 178 nursing staff coming from catheter room, operating room, and outpatient department of Shenzhou Hospital and Fengtian Hospital accepted questionnaires of Chinese Nursing Stressor Scale and Exhaustion Scale in Spring 2010.Results Chinese Nursing Stressor Scale indicated that scores of subitem and total score of job stressor in the catheter room were significantly higher than those in the outpatient department; however, there were no significant differences in scores of subitem and total score of job stressor between catheter room and operating room.On the other hand,Exhaustion Scale showed that scores of emotional exhaustion and job unconcerned feeling in the catheter room were significantly higher than those in the outpatient department, but scores of job achieved feeling in the catheter room were significantly lower than those in the outpatient department. There were no significant differences in Exhaustion Scale between catheter room and operating room nurses. Conclusions Job stressor and exhaustion were serious among nursing staff in the catheter room of grade Ⅲ-A general hospitals, which should cause the attention of management department.

Chinese Journal of cardiovascular Rehabilitation Medicine 2016;25(2):133-136
Objective:To study expression of complement C1q tumor necrosis factor-related protein 3 (CTRP-3)in patients with hypertension and its significance. Methods:A total of 562 patients with hypertension were enrolled as hypertension group, and 570 normal subjects undergoing physical examination were regarded as normal control group. General data, plasma levels of adiponectin (APN),leptin and CTRP-3 etc. were measured and compared be- tween two groups. Correlation among CTRP-3 level and related factors of hypertension were analyzed. Results:Compared with normal control group, there were significant reductions in plasma levels of APN [(12.1±0.4)μg/ml vs. (7.3±0.5)μg/ml],leptin [(10.1±0.4)ng/ml vs. (6.2±0.8)ng/ml]and CTRP-3 [(429±15)ng/ml vs. (314±13)ng/ml]in hypertension group,P<0.05 all. Spearman correlation analysis indicated that after correcting age and gender, plasma CTRP-3 level was significant inversely correlated with body mass index, systolic blood pres- sure, diastolic blood pressure, waist circumference, waist hip ratio, levels of total cholesterol, low density lipopro- tein cholesterol (LDL-C),triglyceride,fasting blood glucose, insulin, homeostasis model-insulin resistance index (HOMA-IR),glycosylated hemoglobin, high sensitivity C reactive protein (hsCRP)(r=-0.852~-0.011,P<0.05 or <0.01),and significant positively correlated with levels of high density lipoprotein cholesterol (HDL-C), APN and leptin (r=0.654~0.962,P<0.05 all). Multi-factor regression analysis indicated that compared with high tertile CTRP-3 group, the OR=14.17 (95%CI:3.62~28.34),P=0.001 in low tertile CTRP-3 group,sug- gesting that low plasma CTRP-3 level was independent risk factor for hypertension. Conclusion:Complement C1q tumor necrosis factor-related protein 3 level significantly correlated with hypertension, and it's an independent risk factor for hypertension.

[Objective] To explore the feasibility and advantages of blood microdialysis technique for the pharmacokinetic study of sinomenine. [Methods] In this study, rats served as the experimental subjects and sinomenine was administered to the rats by intravenous injection. With microdialysis technique for blood sampling, the blood concentration of sinomenine was determined by high performance liquid chromatography (HPLC) and the associated pharmacokinetic parameters were analyzed by 3p97 program. [Results] The pharmacokinetics process of sinomenine in rats presented as the two-compartment model. The half-life of distribution phase was 10.98min and that of elimination phase was 44.71 min. [Conclusion] It is feasible to study pharmacokinetic parameters of sinomenine by using blood microdialysis technique. In comparison with the traditional technique, blood microdialysis technique is superior to the examination of drug blood concentration .

Journal of Peking University(Health Sciences) 2003;0(05):-
Objective: Cellular proliferate inhibition, senescence or apoptosis are induced by telomere shortening through the activation of P53 pathway, but so far, little is known of the mechanism. This study aimed to clarify the molecular regulation of P53 through telomere pathway by the investigation of molecular interaction between P53 and the main telomere associated protein telomeric repeat binding protein 1(TRBP1) in vitro. Methods: Glutathione S-transferase (GST) alone and 4 different human P53-GST fusion proteins were expressed in E. coli. and purified through glutathione Sepharose TM 4B by affinity chromatography, P53s were wild type P53 (1-393), N terminal truncated form P53 2C (95-393), C terminal truncated form P53 N5 (2-293) and single amino acid mutant P53 R175H (175 arginine to histidine). Glutathione Sepharose TM 4B, purified GST alone and P53 fusions were mixed with human breast cancer cell line MCF-7 cellular protein extracts through in vitro binding assay-pull down, the molecular interaction between P53 and TRBP1 were detected by Western blot. Results: SDS-PAGE and Coomassie brilliant blue staining showed that the molecular weights of all the purified proteins were as expected and purities were over 90%. Western blot of TRBP1 showed that both wild type P53 and P53 R175H could bind to TRBP1 of MCF-7 cells, and their binding capacities are similar, whereas GST alone and Glutathione Sepharose TM 4B beads couldn’t. Compared with both of them, the interaction between P53 2C and TRBP1 enhanced dramatically, but between P53 N5 and TRBP1 reduced significantly. Conclusion: P53 can interact with TRBP1 directly and in vitro, C terminus of P53 (293-393) is the structural domain of their interaction. This C terminus domain dependent interaction between P53 and TRBP1 may be related to the cellular activities induced by telomere dynamic changing.
