Objective To explore the pathogenesis, reason, diagnosis and treatment of cerebral salt wasting syndrome (CSWS) in the patients with serious cerebral injury. Methods The clinical data of 26 cases of CSWS after serious cerebral injury were analyzed retrospectively. Results The diagnosis of CSWS was confirmed by clinical symptoms of the patients and the results of laboratory examinations. After the effective treatment, the low plasma sodium level of 21 patients was corrected, and 5 patients died. Conclusion Hyponatremia,hypernatruria, hypovolemia, the increase of urinary amount and the elevation of plasma ANP and BNP level were main characteristics of CSWS.The supplement of water and salt was safe and effective in the treatment of CSWS.

OBJECTIVE To analyze the caloric test findings of the unilateral primary horizontal and posterior semicircular canal benign paroxysmal positional vertigo(HSC-BPPV and PSC-BPPV).METHODS Caloric test were tested in 152 PSC-BPPV patients and 25 HSC-BPPV patients after Dix-Hallpike test and Roll test.RESULTS Of 152 PSC-BPPV patients 64 canal paresis(CP)were found in the abnormal side and 44 in the normal side;Of 25 HSC-BPPV patients 16 canal paresis were found in the abnormal side and 9 in the normal side.The incidence of CP in PSC-BPPV is 30.3% and the incidence of CP in HSC-BPPV is 52%.13 Patients with PSC-BPPV were found abnormal Directional Preponderance(DP)and 10 Patients with PSC-BPPV were found abnormal Directional Preponderance.CONCLUSION The incidence of CP in the abnormal side of HSC-BPPV and PSC-BPPV is more than that of the normal side.The incidence of CP in abnormal side of the HSC-BPPV is more than that of PSC-BPPV.The incidence of abnormal DP in HSC-BPPV is more than that of PSC-BPPV.

Objective To study the correlation between the dynamic changes of sex hormones and the severity and prognosis of head injury,and to investigate the role of sex hormones in traumatic brain injury.Methods The dynamic changes of sex hormones were quantitatively dynamic measured in 100 patients with head injuries,while the relationships between glasgow coma score(GCS),glasgow outcome score(GOS) and the concentration of sex hormones were also studied.Results The estradioi,progesterone and testosterone levels in serum increased within 12 hours after trauma in all groups.Estradiol increased markedly continuously within 1,3 days in GCS≤12 group,testosterone levels in serum decreased within 3 days after trauma in GCS=3～8 group,with a significant differnce compared with others groups.The concentration of sex hormones correlated positively with GOS.Conclusions The measurement of sex hormones may be useful to judge the severity and prognosis of head injury in earlystage.The increases of estradiol in the early stage after traumatic brain injury may exert powerful protective effects on brain tissues.Decreases of sex hormones are important factors leading to death of patients of traumatic brain injury.

ATP-binding cassette protein E (ABCE1) has been annotated as an Rnase L inhibitor in eukaryotes. Previous study showed that the overexpression of ABCE1 was related with the occurrence and clinical stage of lung adenocarcinoma. As an initial investigation into the novel functions of ABCE1, siRNA-expressing vectors targeting sites of the ABCE1 gene were constructed from RNAi-Ready pSIREN-DNR-DsRed-Express vector. Cultured 95-D and NCI-H446 lung carcinoma cells were transfected with the siRNA-expressing vectors using FuGENE 6 and transfection efficiency was determined by using fluorescence microscopy. The expression level of ABCE1 protein was determined by Western blot and immunofluorescence staining. Cell viability was determined by MTT, cell cycle was analysed by flow cytometry.The apoptotic rate was observed by ELISA. Fluorescence microscopy showed a satisfactory transfection efficiency which was about 42.70%. Cell viability and the growth fraction were markedly suppressed, whereas the apoptosis was significantly increased in SiRNA-95-D and SiRNA-NCI-H446 cells than controls(P< 0.05). It can be concluded that the siRNA targeting ABCE1 gene shows a dramatic inhibitory effect on RNA transcription and protein expression and a promoting effect on the apoptosis in 95-D/NCI-H446 cells, which offers a reliable base for the further in vivo experiment.

Objective To investigate paclitaxel and ezrin-siRNA therapy for metastatic liver cancer.Methods Human metastatic liver cancer cells MHCC97-H were divided into four groups in vitro and in vivo,including control cells,exposure to paclitaxel alone,ezrin-siRNA alone,and the combination of paclitaxel and ezrin-siRNA.Nude mice lung lesions were formed by tail vein injection of tumor cells.The tumor cells'apoptosis rate,wound healing ability,the ability to cross an artificial membrane,and the numbers of metastatic lung lesions were measured.Results The apoptosis rate in normally cultured MHCC97-H tumor cells was 2.52％,compared to the other three groups at 11.66％,2.35％,7.38％ respectively.The wound healing ability was best in the negative control and weakest in cells treated with combination paclitaxel and ezrin-siRNA.The numbers of cells which could cross the artificial membrane in the negative,paclitaxel,ezrin-siRNA,and combination paclitaxel and ezrin-siRNA group were 52.5 ± 3.9,32.2 ± 3.2,26.6 ± 2.4,and 19.4 ± 1.1 respectively.The numbers of metastatic lung lesions in the nude mice (n =40) were 12.5 ±2.4,8.2 ± 1.5,7.0 ± 1.3,and 3.8 ± 0.3 respectively.Conclusions The ezrin-siRNA combined with paclitaxel therapy demonstrated a greater efficacy over single agent therapy for depressing liver cancer growth,motility,aggression,and metastasis in vitro and in vivo.

Objective To quantitatively investigate the effect of low-dose ionizing radiation on the frequencies of chromosome aberrations and micronucleus-containing cells of radiation workers.Methods Nine electronic databases were systematically searched on the basis of the published studies evaluating the effects of low-dose ionizing radiation on the frequencies of chromosome aberrations and micronucleuscontaining cells.Of the 195 studies searched,21 studies were identified with a total of 1 970 626 cells under studying.Cochrane' s Q and I2 statistics were used to evaluate heterogeneity among studies and pooling odds ratio (OR) with 95％ confidence intervals (CI) were calculated using random-effect models or fixed-effect models,and publication bias were also calculated.Meta-analysis was performed using Stata 12.0.Results The pooling OR of chromosome-type aberrations [OR =3.03 (2.59,3.56)],dicentric plus centric rings [OR =4.12 (2.99,5.67)],translocations [OR =2.73 (1.67,4.46)],micronucleuscontaining cells [OR =1.70 (1.40-2.06)] were higher for radiation workers when compared with control group.Conclusions The frequencies of chromosome aberrations and micronucleus cell of peripheral lymphocytes are significantly high in radiation workers who were occupationally exposed to low-dose ionizing radiation.It should be noted that the radiation protection of radiological workers be enhanced.

Objective To analyze the inhibitive effect of Keerkang Oral Liquid on adenovirus (ADV3) , parainfluenza virus (HVJ) , respiratory syncytial virus (RSV) , and herpes simplex virus 1 (HSV-1) , herpes simplex virus 2 ( HSV-2 ) in cell culture. Methods Sensitive cell culture was adopted, and chick embryo kidney (CEK) cells and bit hamster kidney (BHK) cells were used to infect homologous virus. Keerkang Oral Liquid was given after 2 hours, then cytopathie phenomenon (CPE) was observed. Results When the amounts of infected virus are less than or equal to 100TCID50, the group which was given Keerkang Oral Liquid showed CPE, while the virus control group showed 25%～75% CPE. Besideds Keerkang Oral Liquid showed inhibitive effect on maximum non-venom concentration (TD0) , medium effective concentration (IC50), minimum valid concentration (MTC) and therapeutic index (TI) ofADV3, HVJ, RSV, BSV-1, and HSV-2 by 0.5, 0.25, 0A25-0.0625 ml/ml and 4.8 respectively. Conclusion Keerkang Oral Liquid has obvious inhibitive effects on ADV3, HVJ,RSV and HSV-1, HSV-2 in cell culture, which provides experimental basis for treating ADV3, HVJ, RSV and HSV-1, and HSV-2.

Objective To analyze the effect of Kecrkang oral Liquid on extracorporeal bacteriostasis. Methods Tube dilution method was adopted to test the effect of different concentrations of Keerkang oral liquid on bacillus coli, klebsiella pneumoniae, bacillus aeruginosus, staphylococcus aureus, staphylococcus epidermidis, β-Streptococcus hemolyticus, streptococcus pneumoniae, haemophilus influenzae and candida albicans. Results Keerkang Oral Liquid has obvious inhibited effects to all the bacteria that mentioned above. Conclusion Keerkang Oral Liquid has obvious extracorporeal bacteriostasis and resistance to fungi.

Objective To study the changes in miRNAs expression in the exosomes of human umbilical vein endothelial cells(HUVECs) after 60 Co γ-rays expose using microRNA(miRNA) chips and bioinformatics techniques so as to provide new clues to the mechanism of radiation-induced vascular tissue injury and its bystander effects.Methods HUVECs exosomes were collected in the control and 4 Gy irradiated cells by ultra-high-speed centrifugation,and further confirmed using transmission electron microscopy (TEM) and Western blotting of exosomes biomarkers.miRNA microarray was used to analyze miRNA expression profiles of exosomes and cells.Also,real-time quantitative PCR(qRT-PCR) was used to verify differentially expressed miRNAs,and the miRDB and TargetScan were performed to predict the target genes of the differentially expressed miRNAs.Bioinformatics analysis was performed using DAVID,KEGG and other online tools.Results Compared with the control exosomes from non-irradiated HUVECs,miRNA microarray analysis revealed that 5 up-regulated,and 13 down-regulated miRNAs were identified in the exosomes from HUVECs at 0.5 h after 4 Gy-irradiation,and 16 up-regulated and 5 down-regulated miRNAs at 2 h after 4 Gy-irradiation.Moreover,38 and 85 miRNAs were differentially expressed respectively in the HUVECs at 0.5 h and 2 h after radiation.The difference was statistically significant(P<0.01).The results of bioinformatics showed that these miRNAs might exert the radiation-induced bystander effect (RIBE) by regulating MAPK signal pathways,RAS and PI3K-Akt signal pathways.Conclusion The ionizing radiation injury significantly alters the components and expression levels of exosomal miRNAs,which play important roles in regulating the signal pathways in response to radiation.
