1.Effects of impulsivity on executive functions in heroin addicts:an ERP study

Objective To investigate the influence of impulsivity on the functions of the conflict monitoring and the conflict resolution process for heroin addicts.Methods 80 heroin addicts were drawn randomly using simple random sampling method and finished Opioid Addiction Severity Inventory ( OASI ) and Barratt Impulsivity Scale ( BIS-11) .High impulsivity group( 7 females of 22) and low impulsivity group (8 females of 21) were divided according to 27%of the total BIS-11score followed the grouping method in psychometrics.Then the event-related potentials (ERPs) technique with the classical color-word Stroop task was used to reveal the influence of impulsivity on the executive function for heroin addicts.Results ( 1) The scores of motor impulsivity(23.00±1.73) and no-plan impulsivity(27.77±3.22) were higher in high im-pulsivity group than those in low impulsivity group(19.31±2.80,23.38±3.59)(P<0.01);and there was posi-tive correlation between impulsivity and addiction severity( r=0.415, P<0.05).(2) The behavioral data showed significant Stroop interference effects in both groups(P<0.01).(3) ERPs data showed that there were significant incongruent-N450 and SP effects in the low impulsivity group,whereas incongruent N450 and SP effects disappeared in high impulsivity group ( Low impulsivity group incongruent vs congruent condition N450:(2.82±3.09)μV vs (4.51±2.77)μV, P<0.05; SP:(3.54±1.25)μV vs (2.84±1.03)μV, P<0.05;High impulsivity group incongruent vs congruent condition N450:(4.98±4.10)μV vs (3.39±3.31)μV, P<0.05;SP:(3.43±3.84)μV vs (4.66±4.53)μV, P<0.05).Conclusion The brain time-interval change of executive function such as the conflict monitoring and the conflict resolution process is influenced by the im-pulsivity levels of heroin addicts.
2.Semen quality and urogenital Ureaplasma urealyticum infection in 5 306 preconception males in one district

Objective To investigate the semen quality and the influence of urogenital Ureaplasma urealyticum infection in pre‐conception males in Shanghai .Methods From Jan ,2014 to Nov ,2015 ,5 306 preconception males in our andrology department were received semen analysis .The difference of semen index between ages and urogenital Ureaplasma urealyticum infected or not were analyzed .Results The average pH value was(7 .27 ± 0 .14) ,the average volume was(3 .15 ± 1 .42)mL ,the average sperm concen‐trationwas(57.51±40.22)×106/mL,theaverageoftotalspermcountwas(172.83±134.90)×106 ,theaveragevitalityratioof grade(A+B) sperm was(38 .50 ± 17 .54)% ,the average motile sperm ratio was(46 .36 ± 20 .08)% ,the average of total motile sperm count was(89 .86 ± 92 .82) × 106 .Motile sperm ratio of preconception males from twenty to twenty nine was(49 .60 ± 20 .70)% ,total motile sperm count was(93 .40 ± 95 .83) × 106 ,which was greater than other ages .Ureaplasma urealyticum turned to positive in 2 311 cases was (43 .55% ) .There were statistically significant differences in total sperm count ,total motile sperm count ,sperm concentration ,vitality ratio of grade A sperm ,vitality ratio of grade(A+B) sperm ,motile sperm ratio ,seminal acid phosphatase ,seminalα‐glucosidase and seminal plasma fructose between positive and negative samples(P<0 .01) .There was differ‐ence in pH and seminal plasma zinc between positive and negative samples(P<0 .05) .Conclusion The sperm quality of preconcep‐tion males in Shanghai is not encouraging ,urogenital Ureaplasma urealyticum infection in preconception men is very important .
3.Optimizing the establishment of endometriosis model in rats by orthogonal experiment

Aim To probe into the best conditions for the establishment of endometriosis model in rats by the orthogonal experiment method.Methods Autotransplantation was used to establish the endometriosis model in rats by the orthogonal experiment method in which four main factors were considered to optimize the condition of model,according to the volume of heterotopic endometrium.Results The para ligament of uterus was the best place for transplanting.Both dissecting muscular layer and injecting estrin played an important role in the establishment of endometriosis model in rats. The value of four factors decreased progressively according to the injection of estrin,the dissection of muscular layer,the direction of endometrium implantation and oestrus.Conclusions By optimizing the condition for the establishment of endometriosis model in rats by orthogonal experiment,the volume of heterotopic endometrium was improved.A technology platform for pharmacodynamics research was thus offered.
4.Vertebroplasty combined with anti-osteoporosis treatment reduces refracture rate

BACKGROUND:Currently, vertebral compression fractures are the most common osteoporotic fracture in postmenopausal women;however, incidence of refracture has aroused increasing attention due to a lack of standard treatment. OBJECTIVE:To evaluate whether vertebroplasty combined with anti-osteoporosis treatment can reduce refracture rate fol owing osteoporotic vertebral compressive fractures. METHODS:Eighty-nine patients with osteoporotic vertebral compressive fractures undergoing vertebroplasty were divided into control group (n=38) and treatment group (n=51) after making an informed choice about treatment. Chest/lumbar X-ray and bone mineral density determinations were performed through outpatient or inpatient fol ow-up. The spinal stability, bone mineral density and refracture rate of patients in both groups were fol owed up. RESULTS AND CONCLUSION:Seventy-eight patients achieved complete fol ow-up (ranged from 6-39 months, average 26.73 months). There was no significant difference in the spinal stability between both groups (P>0.05), while rare bone trabecula was found in the control group. There was a significant difference in bone mineral density between both groups at postoperative 12, 24, and 36 months (P<0.05). The refracture rate in the treatment group was significantly lower than that in the control group (P<0.05). Our results indicate that anti-osteoporosis treatment can effectively reduce the incidence of refracture after vertebroplasty in patients with osteoporotic vertebral compressive fractures, and this study found satisfactory short-and medium-term clinical outcomes.

6.Human amniotic epithelial cells:culture technology optimization and biological characteristics

BACKGROUND:As current studies on isolation, culture andcryopreservation of human amniotic epithelial cells (hAECs) are relatively scattered, it is difficult to form a comprehensive and effective solution to meet the clinical needs of stem cells for transplantation in future.OBJECTIVE:To establish the technology of isolation, culture and cryopreservation of hAECs, and to study the biological characteristics of hAECs.METHODS:Orthogonal method was used to study the effects of different factors on the separation, culture and cryopreservation, and range method was adopted to analyze the data to optimize the separation, culture and cryopreservation. We performed cell primary and passage cultures, morphology observed by microscope, drawn cell growth curve and flow cytometry assay, immunofluorescence staining, hepatocyte like cell differentiation to study the biological characteristics of hAECs.RESULTS AND CONCLUSION:(1) The optimal hAECs separation conditions were as follows:trypsin digestions were conducted at a concentration of 0.25%, four times, once for 20 minutes digestion; optimal conditions of culture were 4×108/L cell seeding density, 10 μg/L epidermal growth factor, 5% serum; optimal conditions of cryopreservation were 1×1010/L cell cryopreservation density, 10% dimethyl sulfoxide, 80% serum. (2) The primary cells were adhered to the wall in 2-3 days, exhibiting irregular polygon, paving stone-like growth. Cell adherence and growth rate were accelerated after subculture, and the growth and proliferation ability of passage 2 cells were not significantly decreased after cryopreservation and resuscitation. (3) Immunofluorescence staining showed that the primary cells strongly expressed SSEA-4 and CK19, but did not express Vimentin, CD45 and HLA-DR. The immunophenotype statistics of the primary and passage 4 cells showed the epithelial mesenchymal transition of hAECs in culture process. (4) Immunofluorescence staining showed that the liver cell marker expression of ALB, CK18 was significantly increased after hAECs were induced to differentiate into hepatocyte-like cells. Glycogen staining revealed glycogen synthesis in hAECs after 3 weeks of induction. To conclude, hAECs are easy to obtain and have strong proliferation ability in vitro, and express surface markers for undifferentiated embryonic stem cells.
7.Chronic pain and emotional disorders as well as the effect of interventional treatment

OBJECTIVE: To analyze the relationship between chronic pain and painprone meutal disorders and investigate the effect of psychological intervention and medication.DATA SOURCES: Related literatures about the relationship between chronic pain and pain-prone mental disorders were searched on computer in Medline and Embase from January 1989 to May 2004 with the retrieval word of "Chronic pain, emotional disorder, treatment", the language was limited to English. The subjects were patients who suffered from chronic pain for above 6 months.STUDY SELECTION: Related literatures about the relationship between chronic pain and pain-prone mental disorders and its therapeutic interventions were carried on primary trial to delete those obsolete and repetitive literatures, and the others were used to search in the full text. The quality was appraised mainly on the basis of the data authenticity, preciseness of the designation, strictness of the implementation and reasonability of the statistical analysis.DATA EXTRACTION: Altogether 20 literatures were obtained and 11literatures were proved accordant with the inclusion standard. 9 literatures were excluded because 7 were repetitive experiments and two were Meta analyses.DATA SYNTHESIS: 11 literatures include experimental group and control group, patients in the experimental group were treated with analgetic combined with psychological intervention and antidepressant while those in the control group were only given analgetic; the outcomes of the two groups were compared.CONCLUSION: Chronic pain may co-exist with emotional disorders. It can be remarkably alleviated by the administration of analgetic combined with psychological intervention and antidepressant, which proves better than single analgetic medication.
8.Study on the correlation between MTHFR gene polymorphism and unexplained recurrent spontaneous abortion

Objective To discuss the correlation between MTHFR gene polymorphism and unexplained recurrent spontaneous abortion.Methods A case control study was used in this study,140 patients with unexplained recurrent spontaneous abortion(UR-SA) (abortion group)and 143 cases of normal women(control group)were recruited.Genomic DNA was obtained and extracted from the oral mucosa cells.Fluorescence quantitative PCR was used to examine the MTHFR gene polymorphisms,and Taqman-MGB technology was conducted to analysis the relationship between single nucleotide polymorphism and disease.Results There was statistically significant difference in the frequencies of C677T genotype and alleles between the two groups(P<0.05).However,no significant difference in the frequencies of A1298C genotype and alleles between the two groups(P>0.05).Conclusion MTHFR gene C677T polymorphism might be one of the genetic risk factors of URSA.
9.Prediction of colon neoplasms by rectal aberrant crypt foci

Objective To evaluate the relationship between endoscopic rectal aberrant crypt foci (ACF) and colon neoplasm, including lesions with high risk of aggressive progression (adenoma with diameter ≥ 1cm, villus adenoma, tubular-villus adenoma, adenoma of high grade dysplasia, or cancer). Methods The rectum of 212 patients who underwent colonoscopy was stained by 0. 4% indigo armine, and the patient was classified into different grade according to the number of rectal ACF, namely Grade 0 as no ACF, Grade Ⅰ as 1-4, Grade Ⅱ as 5-9 and Grade Ⅲ as more than 10. The correlation between rectal ACF grade and colon lesions was analyzed. Results Of 212 patients, 72 were classified as Grade 0, 48 as Grade Ⅰ , 41 as Grade Ⅱ, and 51 as Grade Ⅲ. The detection rate of colon neoplasm and lesions with high risk of aggressive progression in patients with rectal ACF were significantly higher than that in patients without rectal ACF (OR at 95% CI was 22. 352 (6. 716 -74. 395) and 7. 982 ( 1. 838-34. 672), respcetively). Conclusion Rectal ACF may predict the detection of colon lesions, including those with high risk of aggressive progression.
10.Influence of jiangzhuo mixture on glucose and lipid metabolism in MSG obese-induced rats.

OBJECTIVETo study the effect of Jiangzhuo Mixture (JZM) on glucose and lipid metabolism, free fatty acid (FFA) and insulin sensitivity index of sodium glutamate (MSG) induced obese rats.
METHODSSixty-four male MSG rats, 8-10 weeks old, were randomly divided into 4 groups equally, the low dose and high dose JZM group were treated respectively with 10 mL/(kg d) and 20 mL/(kg d) JZM, the rosiglitazone (RGZ) group with water solution of RGE 20 mg/(kg d), while the model group fed only with distilled water 10 mL/(kg d) for control, all by gastrogavage for 7 successive weeks. Levels of fasting blood glucose (FBG), triglyceride (TG), cholesterol (TC), and serum fasting insulin (Ins) were measured before and after treatment. Besides, the level of FFA and the proportion of fat weight to body weight (F/B) were measured and insulin sensitivity index (ISI) was calculated after treatment.
RESULTSAfter being treated for 7 weeks, the indexes including TC, FBG, Ins, F/B and FFA were all lower than those in the model group. Compared with before treatment, TC level lowered, FBG and Ins level raised in the two JZM groups and the RGZ group. Comparisons between the three treated group showed a significant lower level of Ins in the RGZ group. Level of ISI was significantly lowered in the 3 groups after treatment (P <0.05), but still higher in the RGZ group than that in the model control group (P <0.05). As for level of FFA, it was 314.81 +/- 110.25 micromol/L in the high dose JZM group and 305.56 +/- 92.33 micromol/L in the RGZ group, which were lower than that in the low dose JZM group (375.00 +/- 219.95 micromol/L, P <0.05).
CONCLUSIONSJZM could decrease the serum levels of TC, FBG, Ins and FFA in MSG rats, and decrease the fat content of organism in rats' growth process. The FFA decreasing action is dose-dependent. But its effect on ISI in MSG rats in the growth stage is insignificant. JZM can not reverse the forming processes of hyperglycemia, hyperinsulinemia, and insulin resistance in MSG rats.
