1.Effect of adenovirus-mediated local tissue factor pathway inhibitor gene transfer on thrombosis formation in rabbit carotid artery injuries

Objective To observe the effects of tissue factor pathway inhibitor(TFPI) on thrombosis formation in rabbit carotid arteries after ballon injury. Methods Fouty rabbits with the weight 2.5-3.0 kg were respectively divided into 4 groups, Ad-TFPI, Ad-LacZ, PBS and normal control groups. The normal control group was not given any treatment and other 3 groups were given 0.2 ml Ad-TFPI, Ad-LacZ or PBS reproduced by the Dispatch catheter respectively after the PTCA balloon iniury on the right carotid arteries. Ten days after gene transfer the repeated balloon injury was performed in the 3 groups, and the first balloon injury was performed in the normal control group by the same method. The carotid blood flow was recovered immediately after the injury. Thirty minutes later all the animals were sacrificed. The injured carotid arteries and one part of contralateral normal artery were cut down, scissored along the long axis, flattened and fixed in the 2% glutaral. The platelet aggregation and thrombosis formation on the luminal surfaces was observed under electron microscope. Results The electron microscope results showed that the vascular endothelial cell structure was integrated and lined up in order in the nomal artery which had no any injury. After the balloon injury in the normal control group, the structure of the endothelial cell was disintegrated, and there was some platelet aggregation but no fibrosis formation. A large amount of platelet aggregated but no fibrosis formed in Ad-TFPI group after the repeated balloon injury. A large amount of fibrosis formed and red cells piled up in the Ad-LacZ and PBS group. The positive rate of thrombosis formation among groups had siginificant differences(χ2=14.95, P<0.01). The positive rate in Ad-TFPI group(20%) was lower than that in Ad-LacZ group(80%, χ2=7.20, P<0.01) and PBS group(70%, χ2=5.05, P<0.05), but was higher than that in the normal control group(10%, χ2=0.39, P>0.05). The positive rate in Ad-LacZ group(80%) was higher than in the normal control group(10%, χ2=9.90, P<0.01) and in the PBS group(70%, χ2=0.27, P> 0.05). The positive rate in PBS group(70%) was higher than that in the normal control group(10%, χ2=7.50, P< 0.01). Conclusions The repeated balloon injury method can cause a large amount of fibrosis formation in the rabbit carotid. TFPI gene inhibits thrombosis formation in balloon-injured rabbit carotid arteries.

3.Establishment of animal model of temporomandibular joint synovitis and its histological investigation.

OBJECTIVETo establish a stable animal model of temporomandibular joint (TMJ) synovitis.
METHODSSixteen 6-week-old male SD rats were classified into four groups, control group, occlusal dimension increase group, masseter resection group, occlusal dimension increase group and masseter resection group. The rats in the occlusal dimension increase group were adhered composite resin to their maxillary molars in order to increase the occlusal vertical dimension when they were 9-week-old. The rats in the masseter resection group were cut off their bilateral masseter muscles when they were 6-week-old. In the occlusal dimension increase group and masseter resection group, rats' bilateral masseter muscles were resected and occlusal vertical dimension was increased. All rats were sacrificed at their 10 weeks old. TMJ samples were prepared for histology to evaluate the animal model.
RESULTSThe control group showed non-inflammatory changes. The occlusal dimension increase group and the masseter resection group showed vascular dilation and synovial lining proliferation, but there were no statistically significant differences between the two groups (P > 0.05). Compared to the two disposed groups, the occlusal dimension increase group and masseter resection group showed significant inflammatory changes (P < 0.05), including synovial lining proliferation, vascular dilation and fibrin deposit.
CONCLUSIONThe animal model of TMJ synovitis created in the present investigation could simulate the real pathological features of synovitis in vivo, and this animal model showed the obvious merits of high stability and reproduction.

4.Effect of Xuebijing injection on TLR4--NF-kappaB--IL-1beta pathway of myocardial hypoxia/reoxygenation in rats.

OBJECTIVETo investigate the role of Xuebijing injection(XBJI, traditional Chinese medicine), in inhibiting TLR4--NF-kappaB--IL-1beta pathway of myocardial hypoxia/reoxygenation in rats.
METHODSThirty six male SD rats (280 +/- 30) g were randomly divided into six groups (n = 6): normal group (N group), balanced perfusion group (BP group), model group (M group), low dose XBJI group (XBJI(L) group), middle dose XBJI group (XBJI(M) group), high dose XBJI group (XBJI(H) group). By Langendorff isolated heart perfusion device to establish the model of myocardial hypoxia/reoxygenation in rats. ELISA was used to detect the concentration of interleukin-1beta (IL-1beta); Western blot was used to detect the expression of nuclear factor kappa B p65 (NF-kappaB p65) protein and toll like receptor 4 (TLR4) protein; and RT-PCR to determine the expression of NF-kappaB p65 mRNA and TLR4 mRNA;To observe microstructure changes of hypoxia/reoxygenation myocardial by light microscopy.
RESULTSCompared with M group, the IL-1beta concentration, NF-kappaB p65 and TLR4 protein,NF-kappaB p65 and TLR4 mRNA of XBJIL group, XBJI(M) group, XBJI(H) group expression decreased in varying degrees,and decreased most obviously all in XBJI(M) group (P < 0.05, P < 0.01); Myocardical structural damage was serious in M group, and improved after treatment XBJI, the most obvious was the XBJI(M).
CONCLUSIONDifferent dose of XBJI can inhibit TLR4--NF-kappaB--IL-1beta signal transduction pathway and reduce several inflammatory reaction after myocardial hypoxia/reoxygenation injury, the 4 ml/100 ml of XBJI is the best.

Objective To analyze the application of serum 25-hydroxyvitamin D (25-OH-D), bone source alkaline phosphatase (BAP) and ultrasonic bone mineral density (BMD) in the detection of infantile rickets aged 3-12 months. 6.Construction and optimization of a novel plasma membrane model for membrane affinity assay based on polydiacetylene vesicles

Most drugs need to interact with cell membrane to reach the biological target, so that membrane affinity assay is an important early screening step in drug discovery. However, at present, the traditional oil-water distribution method is still used, a new, simple and accurate method for membrane affinity assay is urgently needed. In this study, according to the colorimetric principle, a new assay model based on polydiacetylene vesicles was optimized through a series of experiments including different concentrations of vesicle solution, temperature, or pH reaction environment. On this basis, tetracaine hydrochloride, 2-methylimidazole and histamine were used as model drugs to measure the membrane affinity constants and verify the between-batch precision of the optimized assay model (relative standard deviation less than 5%). In addition, polydiacetylene vesicles were stable for up to 180 days, demonstrating the potential application of the assay model. This strategy is simple, stable, reliable, with high reproducibility, low cost and easy to promote, which provided a new tool and a new direction for the high-throughput assay of membrane affinity.

9.A survey on the periodontal status in type 2 diabetic patients

Objective To investigate the type 2 diabetic patient's periodontal condition and to analyze the influencing factors of periodontitis.Methods A total of 182 type 2 diabetic patients were included in the survey and requested to fill out a questionnaire,and their periodontal status was evaluated by measuring probing depth(PD),attachment level(AL),sulcus bleeding index(SBI),simplified oral hygiene index(OHI-S).Results The prevalence of periodontitis in this group of patients was 96.7%(176/182),including 20 patients with mild periodontitis.156 with moderate to advanced periodontitis.The mean PD and AL of the 182 patients were(2.92±0.67)rain and(2.87-4-1.31)mIIL At least one tooth Was lost in57.1%(104/182)of the patients.The factors related to periodontitis were age,gender,smoking,living in town or country,and 2 h plasma glucose of oral glucose tolerance test(OGTT).There Was no relationship between the severity of periodontitis and education level. The majority of patients did not receive any periodontal therapy.Conclusions Periodontal status was bad in patients with type 2 diabetes.It is important to develop an education program on oral health for tvpe 2 diabetic patients.
10.Analysis of coping strategies of community population in Shenzhen.

OBJECTIVETo investigate the coping strategies of the community-based population in Shenzhen.
METHODSTotally 5,940 subjects from the communities in Shenzhen were sampled and tested with the Simplified Coping Style questionnaire.
RESULTSThe most common coping strategies adopted by the community residents in Shenzhen were "consoling themselves" (33.1%) and "looking at the bright side of things" (31.4%), and the least adopted coping strategy was "relieving distress by smoking, drinking, taking medicine and eating" (57.8%); the strategies left out from their choices were mostly negative strategies. The scores for positive coping strategy factors of community residents in Shenzhen were greater than those of negative ones, with a mean difference of 0.43. The scores of both positive and negative coping strategy factors of these residents were significantly lower than those of general population in other mainland cities (P<0.001), with mean differences of 0.12 and 0.36, respectively.
CONCLUSIONThe community residents in Shenzhen adopt mostly positive strategies for coping with frustration, which benefit their status of self-rated health.
