1.Study on mRNA expression of TNFRSF12 in peripheral blood leukocytes of SLE patients

Objective:To explore whether the mRNA expression of TNFRSF12 in peripheral blood leukocytes of SLE patients is abnormal and to determine whether the TNFRSF12 gene is susceptibility gene to SLE.Methods:The level of mRNA expression of TNFRSF12 in peripheral blood leukocytes was detected by TaqMan one-step RT-PCR method.Results:The mRNA expression pattern in SLE patients is different to that of normal control.The mRNA in SLE consists of 2 types of isoforms of TNFRSF12,encoding membrane bound glycoprotein and potentially secreted molecules respectively,whereas only the latter in normal control.After PHA treatment,the mRNA expression pattern in normal control becomes the same as that of SLE patients.Overall TNFRSF12 mRNA expression was significantly decreased in active SLE group and significantly increased in lupus nephritis group.Conclusion:The results indicated that TNFRSF12 may play a role in the pathogenesis of SLE and was susceptibility gene to SLE.
2.Prevention of myonephropathic metabolic syndrome after operation of acute arterial occlusion by continuous renal replacement therapy

Objective To study the effect of continuous renal replacement therapy(CRRT) in preventing myonephropathic metabolic syndrome(MNMS) after operation of acute arterial occlusion. Methods Twenty-four patients with acute arterial occlusion were divided randomly into 2 groups: CRRT group(n=11) and control group(n=13).The patients were treated with embolectomy or revascularization.In control group,we used conventional therapy such as anti-inflammation,expansion of blood capacity,anticoagulation,and correcting acidosis and electrolyte disorder.In CRRT group,patients were treated by continuous veno-venous hemofiltration(CVVH) with 6 h during operation and 24 h after operation. 3.Study of the expression of Id1 gene to invasion of the pituitary adenoma

Objective To explore the pathogenesis of invasive pituitary adenomas,and to improve the effect of clinical treatment.Methods 15 pairs of invasive pituitary adenoma tissues and noninvasive pituitary adenoma tissues were selected from 114 specimens collected and the Id1 gene expression and Id1 protein expression in invasive pituitary adenoma tissues and noninvasive pituitary adenoma tissues were tested by quantitative RT-PCR,Western blot and immunohistochemical methods;Then small interfering RNA (siRNA) was used to interfer Id1 gene expression in NQ-04 cells.Results The Id1 gene expression in invasive pituitary adenoma tissues was significantly higher than that in noninvasive pituitary adenoma tissues(n =15,t =2.725,P =0.013) ;The migration of pituitary adenoma cells interfered was significantly reduced.Conclusion The invasive pituitary adenoma tissues has high level of Id1 gene expression,which may be correlated with its invasion.And Id1 is expected to become a biological marker of diagnosis and determination in the prognosis of pituitary adenoma.
4.Quality standard study on Entadae Semen.

OBJECTIVETo establish the quality standard of Entadae Semen, and provide scientific basis for its quality control.
METHODTLC and HPLC were used for qualitative identification and quantitative analysis of phaseoloidin and entadamide A-O-β-D-glucopyranoside in Entadae Semen. The test of water content, ash and ethanol-soluble extractives of Entadae Semen was carried out according to the methods recorded in appendix of Chinese Pharmacopeia (2010 edition).
RESULTThe TLC was well separated with clear spots. The linear range of phaseoloidin was between 0.014 to 2.747 g x L(-1) (r = 0.999 6, n = 9) with an average recovery rate of 101.06% (RSD 0.90%, n = 6); the linear range of entadamide A-O-β-D-glucopyranoside was between 0.002 to 0.452 g x L(-1) (r = 0.999 7, n = 9) withan average recovery rate of 101.52% (RSD 1.09%, n = 6). The content of phaseoloidin in sample is between 5.12% to 9.24%, entadamide A-O-β-D-glucopyranoside is between 0.55% to 2.17%, alcohol-soluble extracts is between 30.9% to 45.2%, water is between 6.6% to 8.6%, and total ash is between 2.4% to 2.9%.
CONCLUSIONThe established standard is acceptable for quality control of Entadae Semen.

5.Cyclophosphamide impulsion therapy for child patients with systemic lupus erythematosus (a report of 6 cases)

Objectives To search for the therapeutic methods of child patients with systemic lupus erythematosus (SLE) and the effect of cyclophosphamide (CTX) for SLE.Methods The 14 child patients with SLE were divided into two groups: CTX impulsion therapy group (6 cases) and prednisone therapy group (8 cases). The paticnts in CTX group were treated by CTX 20-30mg/kg, divided into two days for intravenous drip. There was a interval of 15 days between the two course of treatment. The prodnisone (1mg/kg) were used too at the same time. The patients in prednisone group were treated by prednisone, 2mg/kg. d, per os, tolal for 8 weeks, then reduced the dose gradually.Results The 5 cases get the obvious effect and 1 casc get the effect in 8 waeks in CTX group, total offective rate is 100%. one case is complete remission, 5 cases have effect and 2 cases are not effect, total effective rate is 75% (P
6.Prevalence and risk factors of sexual dysfunction in patients with Parkinson's disease

Objective To investigate the prevalence and related factors of sexual dysfunction (SD) in patients with Parkinson's disease.Methods A total of 411 patients with Parkinson' disease in Nanjing Brain Hospital were analyzed retrospectively.The ICD-10 diagnostic criteria for SD was used to evaluate SD.The Unified Parkinson's Disease Rating Scale (UPDRS) part Ⅲ, Hoehn and Yahr (H&Y) staging, Non-motor Symptoms Questionnaire (NMS-Quest), Parkinson's Disease Sleep Scale (PDSS), Hamilton Anxiety Rating Scale (HAMA), Hamilton Rating Scale for Depression (HRSD) and Montreal Cognitive Assessment (MoCA) were used to evaluate patients.Results SD was found in 145 (35.3 %) patients and the prevalence was higher in male than in female.Patients with SD had older age, longer duration, severer motor symptoms, and higher scores of UPDRS-Ⅲ, HAMD, HAMA and NMS-Quest (all P<0.05).In male patients, SD was positively associated with age, duration of disease, UPDRS-Ⅲ , H-Y stage, HAMD, HAMA and NMS (r=0.127, 0.303, 0.240, 0.236, 0.181, 0.221 and 0.302, all P<0.05).In female patients, SD was positively associated with duration of disease, HAMD, HAMA and NMS (r=0.194, 0.163, 0.189 and 0.178, all P<0.05).The forward binary Logistic regression analysis indicated that female gender (OR=0.539, P<0.05), duration of disease (OR=1.133, P<0.05), NMS (OR=1.104, P<0.05) and history of exposure to heavy metals (OR=5.268, P<0.05) were associated with SD.Conclusions SD is a common complication in patients with Parkinson's disease, and a great attention should be paid to its screening and diagnosis.
7.Effects of health education and stove improvement in preventing coal-burning-borne endemic fluorosis in Pingba County of Guizhou Province

Objective To observe the effect of health education and installation of improved stove in control of coal-burning-borne endemic fluorosis in Pingba County of Guizhou Province.Methods Two towns(Shizi and Qibo) of Pingba County that had implemented the health education and installed the improved stoves for control of coal-burning-borne endemic fluorosis were selected as the investigation sites.Two villages in each selected town and 30 householders as well as all students of grade 4-6 were investigated.Questionnaires were made by survey of the fluorosis control knowledge,household investigations were used to know the use of household stoves,corn and peppers drying method,and the dental fluorosis of children aged 8-12 were examined by Dean methods before the intervention(2006) and after implementation(2009) of health education and improvement of stoves.Results The awareness of fluorosis control among students and households after the intervention was 97.36% (405/416) and 86.46%(415/480),respectively,and the difference was statistically significant(x2 =658.86,163.10,all P < 0.01) compared with that before the intervention [32.19%(1083/3364),43.33%(130/300)].Correct using rate of the household stoves was 95.42% (229/240) after the intervention and 45.85% (3976/8672) before the intervention,the difference was statistically significant(x2 =230.25,P < 0.01).The correct rates of drying corn and chili were 91.67% (110/120) and 97.50% (117/120) before the intervention,and 20.00% (12/60),26.67% (16/60) after the intervention,the difference was statistically significant(x2 =94.07,104.02,all P < 0.01).The incidences of dental fluorosis of children aged 8-12 were 31.21%(1305/4182) after the intervention and 87.71%(2856/3256) before the intervention,the difference was statistically significant(x2 =2371.91,P < 0.01).Conclusions The measures of health education and intervention of stove improvement in Pingba County of Guizhou Province for prevention and control of coal-burning-borne endemic fluorosis is effective.
8.Clinical characteristics of patients with ketosis-onset diabetes

A total of 105 patients with ketosis-onset diabetes were divided into four groups (A+B-,A + B +,A - B -,A - B +) based on the results of islet-associated autoantibodies (A +,A - ) and β-cell function (B +,B - ).Nearly 94% patients in the A -B + group and 1/10 patients in the A - B - group were non-insulin dependent.While in the A + B - and A + B + groups,the detection rates of subsequent non-insulin dependency were 20% and 58%,respectively.Among the four groups,patients in the A - B + groups had a lower level of IL-6 and a higher level of TNF-α. Patients in the A + B - and A + B + groups had a higher level of IL-18.Our study indicates that patients with ketosis-onset diabetes may present different clinical characteristics. Evaluate the islet-associated autoantibodies and pancreatic β-cell function may be helpful to the clinical diagnostic and therapeutic strategies in diabetes.
9.Identification of the nanobacteria in prostatic fluid of CPPS

Objective To identify the nanobaeteria in prostate fluid of patients with CPPS.Methods Expressed prostatic secretion(EPS)and urine specimens were collected by Meares-Stamey way from CPPS patients(n=100)and normal controls(n=100).The specimens were cultured and nanobacteria was identified by indirect immunofluoreseenee staining with rnonoelonal antibody.The morphological features were observed by using transmission electron microscopy(TEM). Results The positive rate of nanobaeteria in the EPS cdture of CPPS patients and controls were 43% and 5% respectively,with significant statistical difference(X2=39.58,P<0.01).By TEM,the sizes of NB ranged from 100 to 500 nm and appeared eoccoid-ccccobacillary shape. Conclusion Nanobaeteria infection may exist in EPS of CPPS patients.
