1.Maxillomandibular advancement and contrarotation for the treatment of severe complicated multilevel ob-structive sleep apnea/hypopnoea syndrome

Objective:To evaluate the effects of maxillomandibular advancement and contrarotation in the treatment of severe com-plicated multilevel obstructive sleep apnea/hypopnea syndrome (OSAHS).Methods:1 2 patients (all males,aged 26 to 56 years) with severe complicated multilevel OSAHS were treated by orthognathic surgery methods of maxillomandibular advancement and cont-rarotation and followed up for 6 -1 2 months.Results:After surgery the symptoms of all patients,such as snoring,waking up be-cause of stoke and streaming with sweat,disappeared or were ameliorated remarkably.The AHI index declined from 51 -70 before operation to 0 -3 after operation.While sleeping,their blood oxygen saturation augmented from 30% -70.1 % presurgically to 93%-99% postsurgically,and their average blood oxygen saturation increased from 60% -72% presurgically to 95% -99% postsurgi-cally in all 1 2 patients.Conclusion:Orthognathic surgery method of maxillomandibular advancement and contrarotation is effective in the treatment of severe complicated multilevel OSAHS.

3.Selection High-Yield PHB Depolymerase Producing Strain with UV

Conidial suspension was obtained from the Penicillium sp. DS9713a which could decompose PHB. Through screening a lot of mutants with the methods of transparent zones and flask culture, five strains were selected from starting strain treated with UV and their activity were higher than the original .The best mutant strain named DS9713a-CS01, it's enzyme activity was as 197.42% high as that of the original ,furthermore, the characters of crude enzyme were studied.
5.Expression Pattern of Apolipoprotein J in Rat's Model of Carotid Artery Restenosis

Objective: To study the expression pattern of apolipoprotein J (Apo J) in rat's model of vascular restenosis after cartid balloon dilation. Methods: A total of 40 Wistar rats were randomly divided into 2 groups: Experimental group, the rat's model of carotid artery injury was established by 2 F Fogarty balloon catheter scratching in right carotid artery and Control group, the rats received sham operation without catheter scratching. n=20 in each group. Right carotid artery tissue was taken at 1, 2, 3, 4 weeks after the operation respectively and 5 rats were used for each time point. The morphological changes were measured by HE staining, protein and gene expressions of Apo J were examined by immunohistochemistry and qRT-PCR. Results: Compared with Control group, at each time point Experimental group had obvious intimal hyperplasia and up-regulated protein and gene expressions of Apo J, P<0.05. In Experimental group, with prolonged time of injury, consistent endometrial hyperplasia was observed and it reached the maximum at 4 weeks after operation; the peak protein and gene expressions of Apo J was found at 3 weeks after operation, then decreased at certain point at 4 weeks after operation, P<0.05. Conclusion: Apo J might be closely related to intimal hyperplasia after vascular injury, high expression of vascular Apo J was mainly derived from vascular smooth muscle cell synthesis, it could be a kind of compensation with protective role and might be used as a new biological target for treating vascular retenosis after percutaneous coronary intervention.
6.High density fed-batch culture of Escherichia coli DH5 alpha/pDH-B2m with DO feed-back control of nutrient feeding.

Optimization of cultivation condition of recombinant E. coli DH5 alpha/pDH-B2m and the condition suitable for expression of recombinant mature peptide of human bone morphogenetic protein-2 was carried out in 500 mL shaking flasks and then transferred to NBS Bioflo IV, a 20 L DO feed-back fed-batch culture system, to obtain rhBMP-2. The results indicate that keeping dissolved oxygen at 40% and controlling nutrient feeding rate with DO feed back strategy can obtain theoretically 3.59 g recombinant mature peptide of hBMP-2 per liter of broth, the final cell density OD600 reaches 57(22.8 g dry cell weight/L), and the expression of rhBMP-2 amounts to 30% of the total protein in E. coli.

7.Thinking and Methods for Demonstrating the Effectiveness of Chinese Herbal Compounds from the Perspective of Pharmacokinetics.

8.Clinical analysis of 1 423 cases of root canal therapy.

OBJECTIVETo evaluate the clinical quality of root canal therapy (RCT) in West China Dental Hospital of Sichuan University.
METHODS1 423 RCT teeth were finished from Mar. 2001 to Feb. 2002 in West China Dental Hospital. Root canal filling quality and treatment period of these teeth were evaluated. 695 teeth of the total were revisited 2 years later and 2-year success rate were evaluated.
RESULTSThe ratios of adequate filling, underfilling, and overfilling were 79.97%, 14.62% and 5.41%, receptively. Full canal RCT ratio of molar was 89.44%. Average RCT treatment period was 2.8 weeks. 2-year success rate of RCT was 94.39%.
CONCLUSIONClinical RCT level of West China Dental Hospital was satisfactory from 2001 to 2002.

9.Relationship of cytokine level with cancer cachexia and therapeutic effects of indomethacin for cancer cachexia.

BACKGROUNDTo investigate the relationship between cytokine level and cancer cachexia on murine model, and to observe the effects of regular dose of indomethacin on cancer cachexia and survival of mice.
METHODSC57 mice bearing Lewis lung carcinoma were used to establish murine cancer cachexia model. The serum cytokine levels (IL-1, IL-6, TNF-α and IFN-γ) and body weight of the mice were measured at different time points before and after treatment with daily intraperioneal injection of either indomethacin (1 mg/kg) or saline.
RESULTSThe mice in cancer cachexia group had significantly higher serum levels of IL-1, IL-6 and TNF-α ( P < 0.05) and lower body weight ( P < 0.05) than those in healthy control group. Compared to saline treatment, indomethacin intervention apparently down regulated the levels of IL-1, IL-6 and TNF-α ( P < 0.05 ), and remarkably prolonged the survival of mice ( P < 0.05). No significant difference in IFN-γ level was observed between cancer cachexia and healthy control groups ( P > 0.05), as well as between indomethacin and saline groups ( P > 0.05).
CONCLUSIONSThe results suggest that serum IL-1, IL-6 and TNF-α are possibly associated with cancer cachexia, however, IFN-γ seems to be irrelevant. Indomethacin may potentially ameliorate cancer cachexia through down-regulating levels of cytokines.
10.Sensitivity of two cell lines with acquired resistance to gefitinib to several chemotherapeutic drugs.

OBJECTIVETo explore the sensitivity of tumor cell lines with acquired resistance to gefitinib to several chemotherapeutic drugs and provide preclinical basis of available chemotherapy regimens after failure of molecular targeted therapy.
METHODSHuman lung adenocarcinoma cell lines PC9 and PC9/G with acquired resistance to gefitinib were cultured in vitro. The sensitivity to chemotherapeutic drugs and inhibition rate of cell proliferation was determined by MTT assay. Effects of drugs on apoptosis and expression of P-170 were determined by flow cytometry. Difference of gene expression profile between PC9 and PC9/G cells was analyzed by DNA microarray. Western blot was used to test the expression of Akt, phospho-Akt and integrin beta1.
RESULTSThe resistance index of PC9/G cells to cisplatin was about 5.4-fold compared with that of PC9 cells. LY294002 may significantly elevate the sensitivity of PC9/G cells to cisplatin (P < 0.05). PC9/G cells were more sensitive to docetaxel than PC9 cells. No significant difference of sensitivity to pemetrexed was found between these two cell lines. Expression level of P-170 in PC9/G cells was lower than that in PC9 cells. In PC9/G cells, the expression of integrin beta1 and DNA healing gene was high and expression of gene during mitosis was low. The level of expression of Akt, phospho-Akt and integrin beta1 in PC9/G cells was higher than that in PC9 cells.
CONCLUSIONIn PC9/G cells, a cell line with acquired resistance to gefitinib, over-expression of PI3K, integrin and DNA restoration gene and continuous activation of PI3K is found to be correlated with resistance to cisplatin. Docetaxel or pemetrexed is a more reasonable choice than cisplatin for treatment of NSCLC patients who failed to respond to EGFR-TKI.
