2.Variation of asthma patient's quality oflife after anti-inflammation treatment
Chinese Journal of Clinical Pharmacology and Therapeutics 2000;0(03):-
Aim To evaluate the influence of asthma and the effect of anti-inflammation therapy on the quality of life (QOL) in allergic asthma patients. Methods The QOL of 45 patients with asthma were investigated by collecting questionnaire before and after three months anti-inflammation treatment , at the same time the serum eosinophil cationic protein(ECP) level was detected in 20 patients. The QOL questionnaire included 4 domains, they were respiratory symptoms, physical activity limitation, emotional function and environmental stimuli. Results The factors which influenced the QOL in asthma patients ranging from high to low were environmental stimuli, respiratory symptom and emotion function and physical activity limitation. After anti-inflammation treatment, the QOL was significantly improved and the serum ECP was decreased (P

A murine closed trauma model was used to study the modulating effects of macrophages on T lymphocyte proliferation and the mechanism. It was found that the T lymphocyte transformation was significantly lower than the control on the 1st,2nd,4th,7th and 10 day posttrauma and the suppression of macrophages on T lymphocytes was augmented especially on the 1st,2nd and 4th day posttrauma.Interleukin 1 production of macrophages was not obviously changed while tumor necrosis factor and prostaglandin E2 synthesis were significantly increased.Indomethacin 1?g/ml could block the suppression of macrophages on T cell transformation and mitomycin-C 25?g/ml could stop the synthesis of cytokine but could not block completely the suppression on T cell transformation. These findings suggest that closed trauma induced suppression on T cell transformation results from macrophages through the release of large amounts of prostaglandin E2 and direct cell to cell contact.

Objective To explore the relationship between serum placental growth factor (PLGF) and carotid atherosclerosis (CAS) in patients with type 2 diabetes mellitus (T2DM ). Methods Serum PLGF were determined by ELISA in 53 patients with T2DM ,who were divided into T2DM without carotid plague (T2DM group ,n= 27) and T2DM with carotid plague (T2DM + CAS group ,n= 26). Fasting plasma glucose (FPG) ,glycosylated hemoglobin A1c (HbA1c) and some other metabolic variables were also measured. The results were compared to those of 27 healthy controls (NC group). Results Serum PLGF levels were significantly higher in T2DM group and T2DM + CAS group than in NC group (P<0.01). T2DM+CAS group showed significantly higher serum levels of PLGF compared to that of T 2DM group(P<0.01).Correlation analysis showed that serum PLGF was positively correlated with TC ,LDL‐C , hsC‐RP and IL‐6 in T2DM patients (P<0.01). Similarly ,multiple regression analysis showed that LDL‐C and IL‐6 were the independent factors of serum PLGF level (P<0.05). Furthermore ,Serum PLGF was positively associated with carotid plague. Conclusion PLGF is positively related with chronic inflammation factors in T2DM patients. Serum PLGF may play an important role in the occurrence and development of carotid atherosclerosis and other macroangiopathy in T 2DM.

Objective To observe the effect of Ixeris sonchifoila,a kind of Chinese traditional medicine on noise-induced hearing loss. Methods Fourty-two guinea pigs were divided into two groups. The animals were exposed under 110 dB(n=21) and 120 dB(n=21) noise, separately. Each group was divided into medicine group(n=7),saline group(n=7) and control group(n=7).Ixeris sonchifoila was used in the medicine group and saline in the saline group immediately after the noise exposure by abdomen injection. The threshold was recorded 7 and 14 d later by ABR.Cochlea patch was performed to find hair cell loss. Results By using Ixeris sonchifoila, the threshold shift was significantly reduced(P

Objective To study the effect of continuous renal replacement therapy(CRRT) in preventing myonephropathic metabolic syndrome(MNMS) after operation of acute arterial occlusion. Methods Twenty-four patients with acute arterial occlusion were divided randomly into 2 groups: CRRT group(n=11) and control group(n=13).The patients were treated with embolectomy or revascularization. In control group,we used conventional therapy such as anti-inflammation,expansion of blood capacity, anticoagulation, and correcting acidosis and electrolyte disorder. In CRRT group, patients were treated by continuous veno-venous hemofiltration(CVVH) with 6 h during operation and 24 h after operation. Results In control group,24 h after operation, the serum potassium, blood urea nitrogen(BUN),serum creatinine(SCr),and myoglobin(Mb) were significantly increased(P

Objective:To study the mechanism of apoptosis in laryngeal carcinoma cell induced by Stat3 antisense oligodeoxynucleotide (ASODN).Method:The designed Stat3 ASODN was transferred into laryngeal cacinoma Hep-2 cell by lipofection. Expression of Bcl-2, Bax and C-Myc were detected by Western blot and PCR.Result:Western blot and PCR results demonstrated that Stat3 ASODN could significantly increased the expression of Bax and decreased the expression of Bcl-2 and C-Myc when the concentration of antisense oligodeoxynucleotide were heightened.Conclusion:Stat3 ASODN participate in apoptosis by enhancing the expression of Bax and reducing the expression of Bcl-2 and C-Myc.

Objective In this study, the effects of moderate treadmill exercise and estrogen replacement treatment on the production of liver nitric oxide(NO)and the expression of endothelial nitrie oxide synthase(eNOS)and inducible nitric oxide synthase(iNOS)in ovariectomized(OVX)rats were observed. Methods Female mature SD rats were randomly divided into six groups according to their body weight:sham group(group S),sham exercise group (group S+EX),group OVX,OVX+exercise group (group OVX+EX),OVX+estrogen group(group OVX+E_2),and OVX+exercise+E_2 group(group OVX+EX+E_2).The rats ran on the treadmill at a speed of 18m/min for 60min,5 times per week.50μg/kg of E_2 was given subcutaneously three times per week to groups OVX+E_2 and OVX+EX+E_2.After 14 weeks,NO production in the liver was determined by nitrate reductase method.Protein expression of eNOS and iNOS was determined by immunohistochemistry. Results The NO content of group S was significantly lower than that of group S+EX,but significantly higher than that of group OVX.The NO content of groups OVX+EX,E_2,and OVX+EX+E_2 were significantly higher than that of group OVX.The positive reaction of eNOS mainly located in the cytoplasm of hepatocytes and endothelial cells of the hepatic sinusoids in all groups. The changing tendency after exercise or/and estrogen replacement was almost similar with that of the content of NO.The positive reaction of iNOS mainly located in the cytoplasm of hepatocytes in groups S,S+EX,E_2,and OVX+EX+E_2,while it located in the nuclei in both groups OVX and OVX+EX.Conclusion Moderate exercise and estrogen supplementation both had the same effect of upregnlated synthesis of NO in liver of ovariectomized rats.
