Chinese Journal of Tissue Engineering Research 2005;9(11):204-206
BACKGROUND: Rhodiola rosco L acts on anti-aging,anti-fatigue and improving immunity in human body and improves angina pectoris.OBJECTIVE: To study the effect of rhodiola root on heart hemodynamics and myocardial contractility and probe into its mechanism.DESIGN: Observational and controlled based on the experimental animals.SETTING: Department of physiology in a medical university.MATERIALS: The experiment was performed in Laboratory of Cardiac Vessels of Department of Physiology of Guangxi Medical University, from January to April 2001.Thirty New Zealand white rabbits of either sex, weighted varied from 1.9 to 2.3 kg,were randomized into two groups, named as the control(physiological saline 4 mL/kg) and rhodiola sacra SH Fu (nuodikang) group(0.5 g/4 mL · kg-1),15 rabbits in each group. A hundred common wild frogs were employed, weighted varied from 120 to 160 g,randomized into 4 groups, named as the control group of physiological saline, nuodikang of small dosage group(blood concentration 209 mg/L),nuodikang of moderate dosage group(blood concentration 418 mg/L) and nuodikang of large dosage group(blood concentration 627 mg/L),25 frogs in each group.METHODS: ms2000 multiple medium biological signal analysis system was applied,by Straub method, frog heart perfusion specimen in vitro was prepared and the cardiac activity was assayed. The relevant indexes of ventricle activity in rabbits were determined with hemodynamics.MAIN OUTCOME MEASURES: Comparison of hemodynamics changes in rabbits of two groups and the comparison of effects of nuodikang at various dosages on frog heart work in vitro.RESULTS: Myocardial contractility in every nuodikang group was strengthened remarkably compared with the control(F = 8.939, P<0.01 ), the myocardial contractility in nuodikang groups at the dosages of 209 mg/L,418 mg/L and 627 mg/L was in the tendency of increasing, strengthened by 52. 3%,70. 1% and 86.4% respectively. But the significant difference was not apparent in the comparison among experimental groups(P>0.05). The significant difference in frog heart rates among groups was not apparent(P>0.05). Concerning to hemodynamics in rabbits,in nuodikang groups, the maximum increasing rate of pressure in the left ventricle was increased by 22.1% (F = 6.259,P<0.05),the mean pressure of artery was decreased by 8.2% (F = 5.688, P<0.05) . The significant difference in other indexes was not apparent in the comparison between the control and nuodikang groups.CONCLUSION: Nuodikang strengthens myocardial contractility and speeds up myocardial contraction, but does not present remarkable influence on myocardial dilation; simultaneously it reduces the mean pressure of artery.

Journal of Third Military Medical University 2003;0(20):-
Objective To construct the endothelial cell model with overexpressed human protein kinase C ?2(PKC?2) after high glucose inducement in order to study the function of human PKC?2. Methods The PRKCB1 gene was amplified from pMD18-T-PRKCB1 plasmid and then directly cloned into shuttle plasmid pDC315 to construct shuttle plasmid. Then the recombinant shuttle plasmid and adenovirus genomic plasmid pBHGlox△E1,3Cre were cotransfected into 293 cells to construct recombinant adenovirus Ad-PRKCB1. The virus titer was calculated by TCID50. The Ad-PRKCB1 was verified by immunocytochemistry and RT-PCR. Ad-PRKCB1 was transfected into human umbilical vein endothelial cells (HUVECs) followed by the treatment of high glucose (25 mmol/L) for 96 h, and then the cells were observed by laser scanning confocal microscopy (LSCM). Results Restrictive endonuclease digestion and PCR result showed that the target gene was correctly cloned into shuttle plasmid. Cytopathic effect (CPE) was observed under inverted microscope through homologous recombination. The virus titer was 7.9?109 IU/ml. Immunocytochemical staining and RT-PCR indicated the expression of human PKC?2 in the transfected HUVECs. The translocation was observed under LSCM. Conclusion A recombinant adenovirus vector with human PKC?2 and the endothelial cell model with human PKC?2 overexpression by high glucose are constructed successfully.

Medical Journal of Chinese People's Liberation Army 1981;0(04):-
In order to evaluate the changes in T cell subsets after acute irradiation injury, and investigate the mechanism of deficiency of cellular immunity in acute irradiation syndrome, Balb/c mice were exposed to a single total body irradiation of 5Gy gamma rays from a 60 Co source. On days 10, 28 and 120, the amounts of CD4 + and CD8 + subsets in spleen cells were evaluated by flow cytometry (FCM), and expression of IL 4 gene in ConA stimulated spleen cells was evaluated by reverse transcription linked polymerase chain reaction, in which ?-actin gene was used as an internal control. It was found that after irradiation, the ratio of CD4 + /CD8 + became significantly higher than that of the controls, and then the ratio lowered gradually until day 120. However, it still did not return to the level of the controls. On day 10, expression of IL 4 gene at mRNA level increased significantly. The results indicated that CD8 + subset was more radiosensitive than CD4 + , and the pattern of cytokine seeretion shifted to that of Th2 soon after irradiation.

BACKGROUND: Insulin resistance and impaired insulin secretion are the key complications of polycystic ovary syndrome (PCOS). The incidences of insulin resistance syndrome (IRS), impaired glucose tolerance (IGT) and diabetes mellitus have been clearly shown to be race-related, thus the association between polycystic ovary syndrome and insulin resistance was race-related.OBJECTIVE: To analyze the correlation of insulin resistance with body mass index (BMI), blood lipids and plasma glucose in Tibetan patients with PCOS.DESIGN: A prospective study based on Tibetan population.SETTING: Department of Cardiology, General Hospital of Chinese PLA; Second Department of Internal Medicine,General Hospital of Tibetan Military Area Command of Chinese PLA.PARTICIPANTS: Thirty-six Tibetan PCOS patients, aged 25-42 years with a mean age of (32±5) years were selected from the Second Department of Internal Medicine and Department of Gynecology, General Hospital of Tibetan Military Area Command of Chinese PLA. Another 36 female inpatients were selected as the controls (control group), aged 24-35 years with a mean age of (30±5) years.METHODS: The disease histories were collected and general conditions were recorded. Oral glucose tolerance test (OGTT, 75 g) was conducted for all the subjects. All the subjects werefasted for 12 hours to collect venous blood samples to detect the levels of hormone, fasting insulin, total cholesterol, triglyceride, high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C). Ovary was detected with echocardiography. The insulin resistance index was calculated by the homeostasis model assessment (HOMA-IR) was calculated by the formula:fasting insulin (mlU/L)×FPG (mmol/L)/22.5. The blood pressure, BMI and levels of blood lipids, plasma glucose and insulin were compared between the PCOS group and control group.MAIN OUTCOME MEASURES: Blood pressure, BMI and levels of blood lipids, plasma glucose and insulin were observed in both groups.RESULTS: All the 36 PCOS patients and 36 controls were involved in the analysis of final results. ① The BMI, systolic blood pressure and diastolic blood pressure in the PCOS group were higher than those in the control group (t =3.426 5,2.448 4, 2.212 5, P < 0.05-0.01). ② The levels of total cholesterol, triglyceride and LDL-C in the PCOS group were higher than those in the control group (t=2.104 9, t =2.304 7, 2.400 0, P< 0.05-0.01), whereas the level of HDL-C was lower that in the control group (t =4.800 0, P< 0.01). ③ The levels of fasting insulin and 2-hour plasma glucose and HOM in the PCOS group were higher than those in the control group (t =7.809 5, 12.365 0, 2.789 9, P< 0.01).CONCLUSION: Tibetan PCOS patients present a clustering of atherosclerotic risk factors, including obesity, adverse lipid profile, hypertension, hyperglycemia, etc.

BACKGROUND: Carotid intima-media thickness (IMT) is the substitution index of coronary atherosclerosis, but the precise relation between IMT and coronary artery disease (CAD) is still unknown.OBJECTIVE: To investigate the role of carotid IMT in coronary atherosclerotic heart disease (CAHD).DESIGN: A case analysis.SETTING: Department of Cardiology, Chinese PLA General Hospital.PARTICIPANTS: The study was carried out in the Department of Cardiology, Chinese PLA General Hospital from January 2000 to January 2002. Totally 285 inpatients aged 33-74 years, mean (54.48±9.44)years with suspected CAHD, including 164 males and 121 females, were involved in this study. According to angiographic results, they were divided into 4 groups: 0-vessel group, 1-vessel group, 2-vessel group and 3-vessel group.METHODS: ①The weight and height of patients were measured, and body mass index (BMI) = weight (kg)/height (m)2.②In the supine position, the blood pressure of left arm were measured. ③Venous blood was sampled todetect blood lipid. ④High-resolution B-mode ultrasound and 10 MHz linear transducer was used to evaluate the common carotid artery, internal carotid artery, external carotid artery and carotid bifurcation.Carotid IMT ≥ 1.0 mm was defined as carotid atherosclerosis. The results of coronary angiography by standard Judkins technique showed that, CAHD extent (0, 1, 2 and 3) was determined by the number of diseased vessels, where luminal stenosis was ≥ 50%. Left stem stenosis ≥ 50% was defined as 2-vessel while left stem merging right coronary affection were taken as 3-vessel.MAIN OUTCOME MEASURES: ①IMT values at different detection sites of the patients with different CAHD lesions; ②The predictors of the CAHD severity;③The predictive values of IMT at different carotid sites on CAHD; ④The relationof different risk factors and IMT.RESULTS: Totally 285 patients were involved in the result analysis.①IMT values at different detection sites of CAD patients: The chi square test demonstrated that, left and right carotid bifurcations took the maximal F value among groups (left: 65.64, right: 63.24). And the IMT values of left and right carotid bifurcations were elevated with the increasing CAHD severity. ②The predictors of the CAHD severity: The regression analysis implied that male, high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), and IMT of left carotid bifurcation, right carotid bifurcation, right internal carotid artery and right external carotid artery were the independent predictors of CAHD severity, while left and right carotid bifurcation were the most important (t=3.61, P=0.391; t=2.58,P=0.247). ③The predictive values of IMT at different carotid sites on CAHD: The left and right carotid bifurcations also found the maximal positive predictive value for detecting CAHD (left: 89.9%, right: 88.8%).④Multivariate regression analysis revealed that age, BMI, cholesterol, triglyceride, HDL-C and LDL-C were the risk factors of left carotid bifurcation atherosclerosis.CONCLUSION: IMT in the carotid bifurcation is related significantly with the presence and extent of CAHD. They share common pathological and hemodynamic basis. Carotid bifurcation IMT may be used as a surrogate marker for CAHD.
