1.Endometrial stromal stem cells from early-pregnancy decidua differentiate into endometrial epithelial cells

BACKGROUND:Decidua tissue is one of the most important sources of endometrial stem cel s, from which stromal stem cel s can differentiate into various kinds of cel s under contain inductions. OBJECTIVE:To observe the differentiation of endometrial stromal stem cel s isolated from early-pregnancy decidua into endometrial epithelial cel s. METHODS:Endometrial stromal stem cel s from early-pregnancy decidua were cultured, identified and directional y induced. Then induced cel s were randomized into test group cultured in conditioned medium and control group cultured in normal culture medium containing fetal bovine serum. Expression of keratin was detected by immunofluorescence assay and western bolt assay in the two groups. RESULTS AND CONCLUSION:The endometrial stromal stem cel s from early-pregnancy decidua were successful y isolated and cultured, and presented with a S-shaped curve. Within 1-48 hours after inoculation, cultured cel s were in the adaptation phase;then, the cel s proliferated rapidly and began to grow logarithmical y at 48 hours after culture;after 60 hours of culture, the cel growth rate decreased, with a steady rise in the cel curve. Results from the flow cytometry detection showed that the endometrial stromal stem cel s could express CD73 and CD90, but not express CD34, CD45 and cytokeratin. Immunofluorescence findings showed the expression of keratin negative in the control group, and mostly positive in the test group. Besides, the relative expression of keratin in the test group was significantly higher than that in the control group (P<0.05). These results suggest that the endometrial stromal stem cel s can be isolated from the human decidua in early pregnancy, and differentiate into endometrial epithelial cel s by directional induction.
2.Rifampicin Injection-Capsule Sequential Therapy for Retreatment Smear Positive Pulmonary Tuberculosis at Intensive Stage:Observation of Curative Effect

OBJECTIVE: To observe the curative effect of Rifampicin Injection-Capsule sequential therapy for retreatment of smear positive pulmonary tuberculosis(TB) at intensive stage.METHODS: 80 patients with smear positive pulmonary tuberculosis at intensive stage were enrolled: 40(treatment group) were randomly assigned to receive HZES plus Rifampicin Injection-Capsule sequential therapy,and another 40(control group) to receive HZES plus Rifampicin capsule.Follow-up of sputum negative conversion rate,pulmonary lesion,and toxcity were scheduled at the end of first and second month during intensified stage.RESULTS: At the end of first and second month during intensified stage,the sputum negative conversion rates were 82.5% and 97.5% respectively in the treatment group versus 65.0% and 85.0% respectively in control gorup;the X-ray imaging test of chests revealed that the responsive rates were 17.5% and 40.0% in the treatment groups versus 10.0%and 25.0% in the control group.At the end of second month,the cavity closure(or diminution) rate was 41.7% in the treatment group versus 22.7% in the control group;the incidence of hepatic lesion was 22.5% verus 25.0% and that of gastrointestinal tract toxcity was 20.0% versus 45.0%.CONCLUSION: Rifampicin Injection-Capsule sequential therapy was proved to be of rapid onset of effect,remarkable efficacy,less toxicity and satisfactory efficacy in controlling symptom,shortening bacterial clearance time,reducing side effects and treating severe TB.

4.Research advances on risk factors for arm lymphedema following breast cancer surgery

Arm lymphedema is a common and persistent complication of breast cancer surgery. Surgery or radiotherapy damages axillary lymphatic vessels and obstructs lymphatic reflux, leading to the accumulation of lymph fluid in the superficial soft tissues of the upper limbs and the subsequent formation of edema. Clinical treatment of breast cancer-related lymphedema remains a worldwide problem. Currently, there is still no cure.

6.Variation of prothrombotic state markers in patients with type 2 diabetes mellitus who have microvascular complication

Objective To investigate the variation of coagulation parameters in type 2 diabetes mellitus(T2DM)patients with microvascular complication. Methods Coagulation and fibrinolysis parameters were measured in 40 healthy controls (control group) and 80 T2DM patients (T2DM group) with (47 patients) and without (33 patients) microvasular complications. Results Compared with those in control group, the serum levels of fibrinogenand D-D dimmersin in T2DM group were found to be increased significantly:(4.29±1.70) mg/L vs. (3.12±0.49) mg/L, 0.395 (0.265-0.910) mg/L vs. 0.215 (0.163-0.300) mg/L, P<0.05;while the activity of antithrombin III and protein S levels were significantly decreased in T2DM group:(94.11±25.04)%vs. (103.90±12.48)%,(70.23±23.22)%vs. (90.35±17.35)%, P<0.05. Amongst the T2DM patients, the serum levels of APTT, fibrinogen,D-D dimmers, and the activity of protein S in patients with microvascular complication were found to be significantly higher than those in patients without microvascular complication:(38.09±5.73) s vs. (34.53±4.13) s,(4.60±1.93) mg/L vs.(3.86±1.21) mg/L, 0.630(0.320-1.200) mg/L vs 0.310(0.240-0.405) mg/L, (79.4± 22.16)%vs. (57.15±18.05)%, P<0.05. Conclusion Hypercoagulable state and decreased anti-coagulation ability may contribute to the risk of development of microvascular complication in T2DM patients.

Liver is the common metastatic site of breast cancer .The process of liver metastasis consists of multiple steps and involves various factors from breast cancer cells and the liver micro -environment .In this re-view,we reviewed the molecular(including the connection of proteins ,protein kinases,miRNAs),signal transduc-tion pathways(including CXCL12-CXCR4 axis,Wnt/beta-catenin signaling pathway,AF1q/TCF7/CD44 regu-latory axis,laminin receptor/Akt/ERK signaling pathway,leptin/ERK/IL-8 signaling pathway)and the influen-cing factors(including hypoxia inducible factors,tumor-associated fibroblasts,adipose tissue matrix stem cells, metalloproteinases,selective ligands,inflammatory cell infiltration)associated with the liver micro -environment in breast cancer .
8.Diagnosis value of combined detection of CA 199 ,CA242 and DKK1 in pancreatic cancer

Objective To investigate the clinical values of CA199 ,CA242 and DKK1 in diagnosing pancreatic cancer (PCA). Methods The serum levels of CA199 ,CA242 and DKK1 were measured with ECLIA and ELISA method in 112 patients with PCA ,58 patients with benign pancreatic disease and 62 individuals undergoing physical examination.The Kruskal-Wallis single fac-tor analysis of variance (ANOVA) and Mann-Whitney rank-sum test were adopted to conduct the statistical comparison.The ROC curve was drawn by using the Logistic regression model and the area under curve (AUCROC ) was calculated.Results The levels of serum CA199 ,CA242 and DKK1 in the patients with pancreatic cancer were significantly higher than those in the benign pancreatic disease group and healthy normal control group ,the difference was statistically significant ( H=76.30 ,61.2 ,47.60 ,P<0.05);the sensitivities of CA199 ,CA242 and DKK1 for diagnosing pancreatic cancer were 76.7% ,69.6% and 85.7% respectively ;the speci-ficities were 95.0% ,96.7% and 92.5% respectively ;the accuracies were 86.2% ,83.6% and 89.2% respectively.The sensitivity , specificity and accuracy of combined detection of CA 199 ,CA242 and DKK1 by the Logistic regression equation P= 1/[1 +e-(-4.163+0.21X1+0.156X2+0.342X3) ] were 94.4% ,90.8% and 92.2% respectively ;AUC of CA199 ,CA242 ,DKK1 and pre-1 were 0.736 , 0.862 ,0.886 and 0.949 ,respectively.Conclusion Serum CA199 ,CA242 and DKK1 levels have an important value in the diagnosis of pancreatic cancer ,and their combined detection could significantly improve the sensitivity and accuracy of pancreatic cancer diag-nosis.
9.hTERT antisense oligonucleotides enhance apoptosis induced by cisplatin in HeLa cell lines

Aim To investigate the effects of human telomerase reverse transcriptase(hTERT)antisense phosphorothioate oligodeoxynucleotide(ASODN)on telomerase activity and apoptosis induced by crisplatin(DDP)in HeLa cell lines.Methods HeLa cells were collected after transfection of 0.05,0.1,0.2 ?mol?L-1of ASODN and the total DNA was extracted.Telomeric repeat amplification protocol(TRAP)-AgNO3 stain was used to detect the activity of telomerase.Apoptosis of HeLa cells was detected by AO staining morphological observation and flow cytometry(FCM)analysis technology.Results The telomerase activity of HeLa cells was expressed in high level,which was significantly reduced dose-dependently and time-dependently after treatment with ASODN of hTERT.Two days after transfection of ASODN of 0.05 ?mol?L-1,0.1 ?mol?L-1,0.2 ?mol?L-1,the telomerase activity decreased by 21.77%,52.42% and 71.10% respectively.After treatment with ASODN for 24,48 h and 72 h,the telomerase activity decreased by 12.48%,38.27% and 71.10% respectively.In morphological observation using acridine orange(AO)staining method,HeLa cells displayed typical apoptotic figure after treated with DDP plus ASODN of hTERT.The percentage of apoptosis of HeLa cells treated with DDP of 1.5 mg?L-1,3.0 mg?L-1,for 48 hours after 24 hours exposure to ASODN of 0.2 ?mol?L-1(50.35%,29.67%)was significantly higher than that treated with DDP alone(19.67%,11.38%,P

Objective To study the effects and mechanisms of major immune nutrients and to introduce the progresses of clinical applications about enteral immunonutrition.Methods The related literatures about the effects and clinical applications of enteral immunonutrition were reviewed.Results Infection rate can be reduced and the hospitalization can be shortened as a result of the improved nutritional status and immune competence of patients which can be enhanced by reasonable enteral immune nutrition.Most of the patients suffering from serious diseases can benefit from enteral immunonutrition,such as gastrointestinal cancers,post-transplantation complications,chronic liver disorders,acute pancreatitis and so on.However,as a new nutrition therapy,the clinical use of enteral immunonutrition in critically ill patients is still controversial.Conclusions Enteral immunonutrition plays an important role in the nutritional support of patients with serious diseases,such as gastrointestinal cancers,organ failures.However,much work remains to be done.
