2.Dynamic observation on Th1/Th2 type immune response in mice immunized with recombinant Bb-Eg95-EgA31 vaccine of Echinococcus granulosus

Objective:To dynamically observe changes of Th1/Th2 type immune responses in mice immunized with recombinant Bifidobacteria bifidum(Bb)-Eg95-EgA31 vaccine of Echinococcus granulosus(Eg).Methods:BALB/c mice were vaccinated by 5?108 colony forming unit(CFU) orally and 5?105CFU intranasally respectively.Mice were killed on week 2,4,6,8,10,12,14,16,18 and 20 after immunization respectively,sera were collected to measure the levels of IgG,its subclasses and IgE by enzyme linked immunosorbent assay(ELISA);Spleens were removed to determine the levels of IFN-?,IL-12,TNF-?and IL-10 of splenocyte culture supernatant by ELISA.Results:Oral immunization mice showed higher levels of IgG,IgG2a,IgG2b,IgG1,IgG3 and IgE on the 8~10th week,2~20th week,2~20th week,4~8th week,6~12th week and 10th week respectively,while IFN-?,IL-12,TNF-?and IL-10 on the 2~16th week,2~12th week,2~6th week and 4~12th week post vaccination respectively;Intranasal immunization mice showed higher levels of IgG,IgG2a,IgG2b,IgG1,IgG3 and IgE during the 4~10th week,4~20th week,2~20th week,2~12th week,4~12th week and 10~12th week respectively,while IFN-?,IL-12,TNF-?and IL-10 on the 2~8th week,2~12th week,2~8th week and 6~16th week post vaccination respectively.Conclusion:Mixed Th1/Th2 type immune responses may be induced in mice immunized with the recombinant Bb-Eg95-EgA31 vaccine of Echinococcus granulosus.Oral and intranasal inoculation are two better immune routes,and the former is better than the latter.
3.Influence of Baicalin on TNF-? and Soluble Intercellular Adhesion Molecule-1 in Rats Infected with Pneumocystis carinii

Objective To explore the effect of baicalin on tumour necrosis factor-?(TNF-?) and soluble inter-cellular adhesion molecule-1(sICAM-1) of immunosuppressed rats infected with Pneumocystis carinii.Methods Forty-nine SD rats were randomly divided into 7 groups:normal control(A), immunosuppressed control(B), SMZ/TMP control(C), baicalin prevention(D), low dose(E), moderate dose(F) and high dose(G).Rats of group D received six injections at three-day intervals with 3.5 mg dexamethasone sodium phosphate for 3 weeks, while groups B, C, E, F and G received same immunosuppressor but for 6 weeks.Rats in group D were given 100 mg/kg baicalin daily for 5 weeks, and groups C, E, F and G were given 250 mg/kg SMZ+50 mg/kg TMP, low dose 100 mg/(kg?d), moderate dose 200 mg/(kg?d), and high dose 400 mg/(kg?d) baicalin daily for 2 weeks, respectively.At the end of 8th week after immunosppression, the contents of TNF-? and sICAM-1 in peripheral blood were detected by radioimmunoassay(RIA) and ELISA, respectively.The pathological change of lung was observed by lung imprint smear with gormori methenamine silver(GMS) staining and lung section with hematoxylin-eosin(HE) staining.Results The content of TNF-? in group D(2.14?0.14) ng/ml, group E(2.57?0.15) ng/ml, group F(1.46?0.14) ng/ml, group G(1.12?0.13) ng/ml andgroup C(1.59?0.14) ng/ml were higher than that of group A(0.70?0.21)ng/ml(P
4.Construction and expression of a recombinant BCG-TSOL18 vaccine of Taenia solium

We constructed a recombinant Bacillus Calmette‐Guerin(BCG)‐TSOL18 vaccine of Taenia solium and observed the expression of the TSOL18 gene in BCG .The TSOL18 gene of Taenia solium was obtained from the recombinant plasmid pGEX‐TSOL18 by digestion method and cloned into Escherichia coli (E .coli)‐mycobacterium shuttle plasmid pMV261 to con‐struct the recombinant plasmid pMV261‐TSOL18 of Taenia solium ,and the recombinant plasmid was identified by restriction enzyme digestion ,PCR and DNA sequencing .Then ,the recombinant plasmid was transformed into BCG by electroporation to construct the recombinant BCG‐TSOL18 vaccine of Taenia solium ,and the vaccine was identified by PCR .The expression of the TSOL18 gene in BCG was identified by SDS‐PAGE and Western blot .The 393 bp TSOL18 gene fragment was successfully obtained by restriction enzyme digestion .Restriction enzyme digestion ,PCR and DNA sequencing suggested that the recombi‐nant plasmid pMV261‐TSOL18 of Taenia solium was successfully constructed .PCR confirmed that the recombinant plasmid pMV261‐TSOL18 of Taenia solium was successfully transformed into BCG ,suggesting that the recombinant BCG‐TSOL18 vaccine of Taenia solium was successfully constructed .SDS‐PAGE showed that the relative molecular mass (Mr) of TSOL18 target protein was approximately 14 .7 kD .Results of western blot showed the TSOL18 target protein could be recognized by rabbit antiserum or cysticercosis swine serum .The recombinant BCG‐TSOL18 vaccine of Taenia solium was successfully con‐structed .The TSOL18 gene of Taeniasolium was successfully expressed in BCG and the expressed TSOL18 recombinant pro‐tein had specific antigenicity .This result would lay a foundation for further study of the vaccine .
5.Research on the application of flipped classroom based on MOOC in human parasitology teaching

In order to improve the quality of teaching, the teaching and research section of parasitology explores the teaching effects of human parasitology based on independent learning. Taking the clinical medicine specialty (excellent doctor class) as an example, in order to overcome the problems that the teaching content brought by traditional infusion teaching is difficult to meet the needs of modern teaching and students have low interest in learning. On the basis of optimizing teaching content and teaching methods, some courses are adopted flipped classroom teaching model based on the MOOC videos learning before class, emphasizing student-centered and self-directed learning, focusing on independent learning ability and critical thinking training, and introducing modern information technology and improving teaching effectiveness. The teaching effect shows that the student-centered teaching thought can be better reflected, students' ability of self-learning has been enhanced, and their sense of learning achievement has been strengthened, in favor of cultivating their ability to analyze and solve problems, as well as lifelong learning ability and teamwork ability.
6.Research progress of immunoregulation of T lymphocytes in cysticercosis

Taenia solium is one of the main human parasites. Both larvae and adults can cause diseases, among which neurocysticercosis caused by Cysticercus is the most serious disease. The immune response induced by Cysticercus infection and its regulatory mechanism have been the key problems in the prevention and treatment of Cysticercus infection. This review reports the progress of T lymphocytes-mediated immune responses and immunoregulation in cysticercosis, which is expected to provide a theoretical basis for the immunopathogenesis and prevention and treatment of cysticercosis.
7.Construction and identification of a recombinant Bifidobacteria (pGEX-TSO45W-4B) vaccine of Taenia solium

Objective To construct and identify a recombinant Bifidobacteria(Bb) (pGEX-TSO45W-4B) vaccine of Taenia solium.Methods The TSO45W-4B gene of Taenia solium was synthesized.It was expected that the fragment length of the gene was 351 bp.The gene was cloned into Escherichia coli-Bifidobacteria(Bb) shutde vector pGEX-1λT to construct a recombinant plasmid pGEX-TSO45W-4B.The recombinant plasmid was electroporated into Bb to construct a recombinant Bb (pGEX-TSO45W-4B) vaccine.The vaccine was identified by restriction analysis,PCR and sequencing,which would demonstrate that the constructed recombinant Bb (pGEX-TSO45W-4B) vaccine of Taenia solium contained the gene of fragment length of 351 bp.Results The TSO45W-4B gene of 351 bp was synthesized.Restriction analysis,PCR and sequencing results showed that the recombinant Bb (pGEX-TSO45W-4B) vaccine of Taenia solium was successfully constructed and contained the gene fragment of 351 bp.Conclusion The recombinant Bb (pGEX-TSO45W-4B) vaccine of Taenia solium is successfully constructed,which lays a foundation for the study of expression and immunogenicity of the vaccine.
8.Research Progress in Antigens for the Diagnosis of Alveolar Echinococcosis

Specific antigens of Echinococcus multilocularis is essential for the diagnosis of alveolar echinococcosis and vaccine development.Because of the limited source of nature antigen,its application is restricted.The development of recombinant antigens can provide large amount of antigen under effective quality control.This review summarizes the recent progress in antigen research,especially the recombinant antigen used for diagnosis of the disease.
9.Human parasitology teaching reform based on culturing students' ability to practice

To improve the quality of tea.ching,investigating the effect of practice-based in the teaching of human parasitology by department of parasitology in our school.We take medical laboratory science professional as an example,the parasite teaching is the traditional model of infusion teaching at present,besides the classical contents,it is difficult for some courses to meet the needs of social development at present,which has deficiency in attracting students' interest.Based on the optimization of teaching content,and teaching methods for the teaching process,part of contents were carryed out on the base of cultivating students' ability to practice.The objective of this teaching model is to solve practical problems as the goal,particularly focus on learning process of solving the problem.These results show that the teaching model based on culturing students' ability to practice could better embody the idea of studentcentered teaching,to some extent,it helps to stimulate students' independent thinking ability,and enhance students' learning achievement,and cultivate students' analysis and problem-solving ability,practical ability and teamwork spirit.
10.Changes of splenocyte subsets in mice immunized with recombinant Bb-Eg95-EgA31 vaccine against Echinococcus granulosus

To investigate the weight reduction of hydatid cyst and level of splenocyte subsets in mice immunized with recombinant Bifidobacteria bifidum (Bb) Bb-Eg95-EgA31 vaccine of Echinococcus granulosus (Eg) and challenged with Eg protoscoleces,BALB/c mice were immunized with the vaccine subcutaneously,intramuscularly,intranasally or orally.Then the mice were challenged with 50 Eg protoscoleces on 8~(th) week after vaccination and killed on 25~(th) week after infection.The weight of hydatid cyst was measured and the reduction of the weight was calculated.The spleens were collected for splenocytes preparation.CD_4~+ and CD_8~+ T cells were determined by flow cytometry(FCM),and the blank vector,Bb or MRS were set as control.The hydatid cyst weight was reduced by 45.33%,41.33%,70.67% and 62.67% respectively for the 4 immunization groups.Number of CD_4~+ and CD_8~+ subsets in the immunization groups was more than that in the control group.CD_4~+ and the ratio of CD_4~+/CD_8~+ in the intranasal or oral immunization group was much higher than that in the subcutaneous or intramuscular immunization group.It's concluded that CD_4~+ and CD_8~+T cells may play an important role in the protection induced by recombinant Eg95-EgA31 vaccine of Eg.In addition,intranasal and oral vaccinations could be the better ways for immunization.
