1.EFFECT ON ALTRASTRUCTURE OF CHONDROCYTE IN THE EPIPHYSEAL PLATE AFTER CHRONIC COLLISION INJURY

Five young rabbits were divided into 5 groups (2, 4, 6, 8 and 12 week), one rabbit in each group.A special collision unit was used in the experiment by which the distal ends of the right fore legs of rabbits were struck repeatedly. Altrastructure changes of chondrocyte at the epiphyseal plate in the distal endof radius were observed in H-500 electron microscope. In two and four week groups, organaelles swelling was found such as rough endoplasmic reticulum andmitochondria, especially mitochondria swelling which showed different shapes like sausagge and racket.Space around nucleus increased, nucleus shape changed, myeline structure in the mitochondria and chroma- tin margination in the nucleus were found in the six and eight week groups. Altrastructure changes weremore serious in the eight and twelve week groups than other groups. Necrosis of chondrocyte was appearedsuch as part organaelles dissolved or disappeared, nucleus brokeu aud nucleus memberane disappeared. The results showed in altrastructure pathology that chronic collision injury can cause not only degeneration of chondrocyte in the pressure epiphyseal plate but necrosis of chondrocyte as well. It concluses that chondrocyte necrosis is related to local ischemia resulted from chronic collision injury.

3.AN EXPERIMENTAL PATHOLOGICAL STUDY OF CHRONIC COLLISION INJURY OF THE DISTAL RADIAL EPIPHYSIS

A special collision unit was used in the experiment by which the distal ends of the fore legs of young rabbits were struck repeatedly on the right aspect. In another group of young rabbits, deep branches of the artery of the right fore legs were ligated and severed. X-Ray examination was made before the two groups of rabbits were killed.Roentgenogram demonstrated: Growth plate of the right fore leg showed wave-like growth, thickened or thinned in the collision group; no difference showed between 2 fore legs in vessel group.Histological demonstrations1. Collision group: 1) Chondrocytcs of articular surface decreased, degenerated and disappeared. 2) Trabecula of the cpiphysis and metaphysis atrophied; osteoblasts and ostcocytcs decreased and degenerated. 3) Growth plate thickened, some arranged wave-like; chondrocytes increased and degenerated. 4) Fibrous tissues and cartilage increased in the mctaphysis. 5) Perichondral ring thickened.2. Vessel group: Similar to the collision group except fibrous and cartilage proliferation in the epiphysis and no arrangement disorder in the growth plate.
4.Analysis of proteomic spectra in serum from patients with laryngeal carcinoma by SELDI-TOF-MS technology

Objective:To screen the tumor markers of laryngeal carcinoma and to investigate their expression in preoperative and postoperative serum.Method:The distinct protein in serum was detected in 32 cases of laryngeal carcinoma and 38 healthy people by IMAC-Cu proteinchip array and surface-enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS).The distinct proteins in serum were detected in 32 cases of laryngeal carcinoma preoperation and within 10 days 15 cases of laryngeal carcinoma postoperation with the same methods.The discriminatory profiling between preoperative and postoperative patients was analyzed by Biomarker Wizard software and Biomarker Pattern software.Result:The results showed that fifteen differentially expressed proteins in serum were screened by analysis of protemic spectra of preoprative patients and normal subjects.Seventeen kinds of protein differentially expressed in serum were screened by analysis of protemic spectra of preoperative and postoperative patients.Six kinds of protein(2 958.52,3 796.89,5 148.86,6 115.57, 052.18,and7 770.76)were obtained for making up patterns that was able to class the preoperative-team and postoperativeteam.Corresponding correct ratio were 84.38%(27/32)and 73.33%(11/15).Conclusion:The preliminary results suggest that classification system will provide a highly accurate and innovative approach for the arly dioagnosis of laryngeal carcinoma and judgement of prognosis.SELDI-TOF-MS technology is a useful tool for a high throughput screening of large-sized serum samples tO discover potential biomarkers for laryngeal carcinoma.

7.Clinical significance of hepatic artery variation in hepatic portal lymphadenectomy

Objective:To investigate the recognition and injury prevention strategies of hepatic artery variations during hepatic portal lymphadenectomy. Methods:A retrospective analysis was performed, and 12 patients of hepatic arterial variation among 62 pa-tients with hepatic portal lymphadenectomy were the subjects. The study was conducted in the First Affiliated Hospital of Bengbu Medi-cal College between January 2013 and July 2014. The intraoperative treatment and postoperative complications were recorded. Results:Among 12 cases of hepatic artery variation, we found the following cases:3 cases (25.0%) of Michels' Type III, 2 cases (16.7%) of Mi-chels' Type VI, 1 case (8.3%) of Michels' Type IX, 1 case (8.3%) of Hiatt's Type 6, 2 cases (16.7%) of spatial location variation between right hepatic artery and hepatic duct, 2 cases (16.7%) of left and right hepatic artery originating from a common hepatic artery, and 1 case (8.3%) of right hepatic artery originating from the gastroduodenal artery. No injury of hepatic artery occurred. Two cases had post-operative complications, including 1 case of pancreatic leakage and 1 case of incision infection;postoperative hemorrhage, bile leakage, hepatic abscess did not occur in these two cases. Patients recovered well in general. Conclusion:Hepatic arterial injury can be signifi-cantly reduced by the following:increased familiarity with the various types of hepatic artery variations;complete imaging examina-tions for inspection and evaluation before surgery;and careful and meticulous operations in surgery.

9.Procedure and clinical assessments of malariotherapy: recent experience in 20 HIV patients.

OBJECTIVETo demonstrate the side effects of malariotherapy and to explore safe procedures in conduct of malariotherapy for human immunodeficiency virus (HIV) infected patients.
METHODSTwenty HIV/acquired immunodeficiency syndrome (AIDS) patients were selected for the study of malariotherapy and were intravenously infected with Plasmodia vivax to induce therapeutic malaria. Malaria was terminated with chloroquine after 10 - 20 malarial febrile episodes. Clinical assessments were made before (baseline), during (malarial phase) and after (post) termination of malaria. The density of Plasmodia in peripheral blood from the HIV/AIDS patients were compared to that from HIV-negative naturally infected malarial patients who donated the blood for the therapeutically induced malaria. CD(4) cell baseline levels were correlated to the severity of malarial symptoms and parasitemia.
RESULTSThere were no significant differences of Plasmodium density between the HIV/AIDS patients injected with P. vivax and the HIV-negative blood donors. However, it was found that the HIV-positive patients had milder malarial symptoms and parasitemia with progressively lower CD(4) cell baseline levels. All patients developed every day or every other day fever episodes with headache and shaking chill. These symptoms were well tolerated with the aid of anti-pyretic medications. Spleen and liver enlargement were seen in 15 of 20 and 4 of 20 patients respectively. Transitory liver effects with increase of serum glutamic-pyruvic transaminase were seen in 2 of 20 during malarial phase. Most patients experienced mild to medium anemia and 6 of 20 patients developed thrombocytopenia during malarial phase. All these side effects disappeared after termination of malaria or within one month thereafter. No complications occurred in these patients.
CONCLUSIONSTherapeutically induced acute vivax malaria was well tolerated in 20 HIV-positive subjects who represented a range of CD(4) cell levels from 1868/ micro l to 15/ micro l. Malariotherapy did not induce complications while increasing CD(4) cell levels in most treated HIV/AIDS patients (results published elsewhere).

10.Application of SELDI-TOF-MS technology in study of laryngeal carcinoma biomarkers.

OBJECTIVE:
In order to establish a primary diagnosis model of laryngeal carcinoma, we explore the changes of proteomic spectra in serum from patients with laryngeal carcinoma and healthy controls.
METHOD:
Proteomic spectra of 32 serum samples from laryngeal carcinoma (18 cases of glottis and 14 of supraglottis) patients and 38 from healthy people whose age and sex was matched were generated by IMAC3-Cu protein chip array and SELDI-TOF-MS (surface was enhanced laser desorption/ionization time-of-flight mass spectrometry). The discriminatory profiling between cancer and normal samples was analyzed by Biomarker Wizard software and Biomarker Pattern software.
RESULT:
Fifteen differentially expressed proteins in serum were screened by analysis of proteomic spectra of preoperative patients and normal subjects. Eight proteins (3795.04, 5068.18, 5339.78, 5908.28, 6629.74, 9266.32, 13879.6 and 14037.1) were obtained for making up patterns that was able to class the patients team and normal team correctly. Corresponding correct ratio were 87.50% (28/32) and 86. 84% (33/38).
CONCLUSION
SEI.DI-TOF-MS Protein Chip technology is a new tool for the early diagnosis of laryngeal carcinoma.
