Objective To study the effects of A?25-35 on the expression of gene P21,CDK4,E2F1 and BAX in cultured PC12 cells.MethodsPC12 cells were treated with 25 ?mol/L A?25-35,the relation between cell cycle redistribution and apoptosis was analyzed by flow cytometry(FCM).Protein and mRNA expression of P21,CDK4,E2F1 and BAX were detected by RT-PCR and Western-blot respectively.Results About 90% PC12 cells were found arrest on G0/G1 by FCM being deprived serum.Treated with 25 ?mol/L A?25-35 for 8,16,24 h,the percent of S phase cells was raised remarkably(P

Aim To study the effective mechanism of curcumin on abnormal cell cycle and apoptosis of serum-deprived PC12 cells induced by ?-amyloid peptide 25-35(A?25-35).Methods Synchronized PC12 cells were pretreated with 5 ?mol?L-1 Cur for 1 h,and then 25 ?mol?L-1 A?25-35 for 24 h.Protein and mRNA expression of p21,CDK4,E2F1 and bax were detected by RT-PCR and Western blot respectively.Results After synchronized PC12 cells being pretreated with 5 ?mol?L-1 Cur for 1 h,the mRNA and protein expression of p21 gene were increased gradually,while CDK4,E2F1 and bax gene were decreased.Conclusion Cur maybe effects the redistribution of cell cycle and apoptosis of serum-deprived PC12 cells induced by A?25-35,through increasing the mRNA and protein expression of p21,and decreasing the mRNA and protein expression of CDK4,E2F1 and bax gene.
3.Changes of serum soluble apoptosis factors and their relations with cognition disorders in the patients with vascular dementia

Objective To investigate the changes of serum soluble factor-related apoptosis (sFas) and soluble Fas ligand (sFasL) and their relations with cognition disorders in the patients with vascular dementia (VaD). Methods Serum concentrations of sFas and sFasl were detected by enzyme-linked immunosorbent assay (ELISA) and compared between 70 patients with VaD aged (72.5± 7.5)years and 50 healthy elderly people aged(72.5 ± 7.5)years.The VaD patient's cognitive functions were evaluated by activity of daily living scale (ADL),mini mental state examination (MMSE) and hachinski ischemia score (Hachinski). Results The serum levels of sFas and sFasL in VaD patients were (228.0±60.7)μg/L and (146.8±30.1)μg/L,and in the healthy elderly were (62.4±22.6)μg/L and (82.3 ± 18.7)μg/L,respectively.The serum levels of apoptosis factors in VaD patients were significantly higher than in the healthy controls (t=20.883,14.453,P<0.01).sFas level was negatively correlated with age,the scores of ADL and Hachiuski while positively with the scores of MMSE (r=-0.956,-0.943,-0.950 and 0.904,all P<0.01). sFasL level was negatively correlated with the scores of MMSE while positively with age,the scores of ADL and Hachinski (r=-0.899,0.963,0.948 and 0.939,a11 P<0.01). Conclusions Apoptosis may be involved in the pathological change during VaD and the serum levels of sFas and sFasL might be related with cognition disorders.
4.Correlation between polymorphism of cytochrome P450 1A2 gene and antidepressant clinical efficacy of duloxetine

Objective To investigate the relationship between C734A and G-2964A polymorphism of cytochrome P450 1A2 gene and clinical efficacy of duloxetine.Methods 223 patients with depression were treated with duloxetine for six weeks.The clinical efficacy was evaluated with the Hamilton rating scale for depression (HAMD) ;single nucleotide polymorphism (SNP) at position C734A and G-2964A of CYP1A2 gene were identified with restriction fragment length polymorphism(RFLPs) ;then one-way ANOVA was adopted to analyze the relationship between SNP and clinical efficacy.Results (1) In 223 patients,the frequency of allele A at locus 734 was 63.64%,while that of allele A at locus-2964 was 26.82%.(2) 220 patient underwent the whole treating course.The conjoint analysis of two locuses indicated that the decreasing ratio of HAMD score of high-activity group,middle-activity group and low-activity after treatment was (56.05± 10.13) %,(66.36± 8.66) % and (73.82± 7.10) % respectively,the differences obtained by pairwise comparison of the three groups were of statistical significance(P<0.01).Conclusion There is close relationship between C734A and G-2964A polymorphism of CYP1A2 gene and clinical efficacy of duloxetine in the treatment for depression.
5.Current status of illness perception and analysis of its correlation with quality of life in patients undergoing radiotherapy for lung cancer

Objective To investigate the illness perception and quality of life in patients undergoing radiotherapy for lung cancer and to explore its correlation with quality of life.Methods Totally 118 patients undergoing radiotherapy for lung cancer in a Class Ⅲ Grade A hospital between June and December 2017 were selected by purposive sampling. The Brief Illness Perception Questionnaire (BIPQ) and the Quality of Life Questionnaire-Core 30 (QLQ-C30) were used to investigate their illness perception and quality of life. The correlation between the two was analyzed by Pearson correlation analysis.Results The illness perception score of these patients was (52.22±9.87). In terms of quality of life, the highest score was found in psychical function (62.45±12.56), whereas the lowest was found in nausea and vomiting (20.65±8.78). The scores in different dimensions of the quality of life questionnaire for patients with lung cancer were correlated with illness perception (r=-0.26, -0.13, -0.41, -0.21, 0.06, 0.22, 0.28, -0.38;P<0.05), except for cognitive function, fatigue, pain, loss of appetite, diarrhea and financial difficulty.Conclusions Patients with lung cancer show more negative perception of illness and lower quality of life. Therefore, nursing measures shall be taken to reduce their negative perception and improve their quality of life.
6.Effects of teach-back method on the compliance of functional exercises in postoperative patients with ;breast cancer

Objective To explore the effects of teach-back method on the compliance of functional exercises in postoperative patients with breast cancer. Methods Totally 105 postoperative patients with breast cancer admitted in our hospital from March to September 2015 were selected as the control group, and 114 patients from October 2015 to March 2016 were selected as the intervention group. Patients in the intervention group received the functional exercises based on teach-back method, while patients in the control group received routine health education method. The mastery skill score and compliance of functional exercise one day before discharge in two groups were compared. Results The mastery skill score of functional exercise one day before discharge was (63.73±5.51) in the intervention group, which was significantly higher than (56.45±4.88) in the control group ( P<0.05);the constituent ratio of fully compliance, partial compliance and noncompliance status in the intervention group one day before discharge were 64. 91%, 25. 44%, 9. 65%, and they were obviously superior to the control group (47.62%, 36.19%, 16.19%) (P<0.05). Conclusions Teach back method can effectively improve the skill and compliance of functional exercise in postoperative patients with breast cancer.
7.Effect of intervention model based on empowerment theory on self-efficacy and self-acceptance of patients with permanent enterostomy

Objective? To explore the effect of empowerment-based intervention model on self-efficacy and self-acceptance of patients undergoing permanent enterostomy. Methods? By purposive sampling, a total of 100 colorectal cancer patients undergoing permanent enterostomy operation in the First Affiliated Hospital of Zhengzhou University from January to December of 2018 were selected as the research objects. The 50 patients admitted from January to June of 2018 were taken as the control group while the other 50 patients admitted from July to December of 2018 were taken as the experimental group. During intervention, there were 7 patients lost in the control group and 43 valid questionnaire retrieved; in the experimental group, 2 patients were lost and 48 valid questionnaires were retrieved. The control group was given conventional nursing care while the experimental group was given the nursing intervention formulated based on the empowerment theory. Stoma Self-Efficacy Scale(SSES) and Self-Acceptance Questionnaire(SAQ) were used to assess the patients. Results? After intervention, the total score of SSES, the dimension scores of "stoma care efficacy" and "social self-efficacy"of the experimental group were higher than those of the control group, with statistical significances (P<0.05). After intervention, the total score of SAQ and the score of "self-acceptance" dimension in the experimental group were higher than those in the control group, and the differences were statistically significant (P<0.05). Conclusions? The intervention based on empowerment theory can improve the self-management efficacy and self-acceptance level of patients with permanent enterostomy. It is suggested that it should be popularized in clinical practice.
8.Application of the "Internet +" model in the nursing management of esophageal cancer patients in chemotherapy

Objective:To explore the application value of "Internet +" model in esophageal cancer patients in chemotherapy.Methods:A total of 90 patients with esophageal cancer treated in the First Affiliated Hospital of Zhengzhou University from January 2017 to January 2019 were selected by convenient sampling method, and all received chemotherapy. They were divided into observation group ( n=45) and control group ( n=45) according to the random number table method. The control group adopts routine nursing management, and the observation group adopts the "Internet +" management model. Blood samples were taken before the nursing intervention and 6 months after the intervention to measure the nutritional indicators of the two groups, including serum prealbumin (PA) , albumin (ALB) , and transferrin (TRF) levels. The Connor-Davidson Psychological Resilience Scale was used to evaluate the psychological resilience of the two groups of patients. Results:The levels of serum PA, ALB, and TRF in the two groups after intervention were higher than those before intervention, and the observation group were higher than the control group, the differences were statistically significant ( P<0.05) . After intervention, the mental impact, acceptance changes, tolerance of negative emotions, ability, control scores, and total psychological elasticity scores of the two groups were higher than before the intervention, and the observation group were higher than the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Compared with traditional nursing management, the "Internet +" management model can further improve the nutritional level and mood of esophageal cancer patients in chemotherapy, and the overall application effect is good.
