1.Effects of leisure satisfaction on stress and depression of undergraduates: a path analysis

Objective To explore the relationship among different dimensions of leisure satisfaction,stress and depression of Chinese undergraduates.Methods A stratified sample of 1 000 undergraduates completed the Leisure Satisfaction Questionnaire of College Students (LSQ),Adolescent Self-Rating Life Check List (ASLEC),Self-Rating Depression Scale (SDS),and the basic information questionnaire.The data was analyzed by methods of Pearson' s product-moment correlation and structural equation modeling.Results Firstly,total score of LSQ (3.63 ±0.78) had significant effect on depression(β=-0.30,t=-8.81>2),and it' s indirect effect on depression buffered by stress was also significant(β=-0.05,t 1 =-5.69,t2 =7.54).Secondly,in the three dimensions of LSQ,satisfaction of leisure relaxation(3.80±0.89) had both significant main effect(β=-0.18,t=-3.28) and indirect effect buffered by stress (β=-0.06,t1 =-6.53,t 2 =7.28) on depression,while satisfaction of leisure contact (3.51 ±0.82) had neither main effect nor indirect effect buffered by stress on depression significantly(t <2).And satisfaction of leisure transcendence (3.56 ± 0.85) had significant main effect (β =-0.15,t =-2.75) but no indirect effect buffered by stress on depression.Conclusion The impact paths to stress and depression were distinguished between the three dimensions of leisure satisfaction of Chinese undergraduates.
2.Development of leisure satisfaction questionnaire for university students

Objective To develop a questionnaire which measures leisure satisfaction for university students. Methods Initial items of the questionnaire were compiled according to theoretical analysis and interviews. 400 college students were recruited to respond to the items. The explorative factor analysis was used to decide the final items and dimensions. Another 350 college students were recruited to respond to the decided questionnaire and a criterion questionnaire. Results The leisure satisfaction questionnaire consisted of three dimensions : Relaxing Satisfaction (RS) , Contacting Satisfaction (CS), and Transcend Satisfaction (TS). Internal consistency (Cronbach' sα) of the three dimensions were 0.86,085 and 0. 84 and the Cuttmann Split-half Coefficient were 0.76,0.83 and 0.86. The cumulative explanation of the variance was 58.956%. A confirmative factor analysis revealed that the three-dimension model was proper(x2/df = 1. 74, NFI = 0.88, NNFI = 0.93, CFI = 0.94, GFI =0.93,AGFI=0.91,IFI=0.94,RMSEA =0.048). The leisure satisfaction Questionnaire was positively related to freedom satisfaction( r = 0.20 ~ 0. 30, P < 0.01). Conclusion The leisure satisfaction questionnaire for university students has satisfactory psychometric characteristics.
3.Comparison of long-term oncological outcomes of laparoscopic and open resection of rectal cancer.

OBJECTIVETo compare the long-term oncological outcomes of laparoscopic and open resection of rectal cancers.
METHODSBetween January, 2003 and December, 2008, 514 patients with rectal cancer were admitted in our hospital, among whom 186 underwent laparoscopic tumor resection and 238 received open radical resection of the tumors. The long-term survival of the patients and the recurrence pattern were compared between the two groups.
RESULTSThe median follow-up of the patients was 48.54∓28.76 months. No significant differences were found between the two groups in the local recurrence rate (3.9% vs 5.5%, P=0.284), 5-year overall survival (69.4% vs 61.3%, P=0.067), or the 5-year disease-free survival rates (67.7% vs 60.7%, P=0.110). Both the 5-year overall survival and progression-free survival of the patients in stage IV were better in the laparoscopic group than in the open surgery group (P<0.05).
CONCLUSIONLaparoscopic resection of rectal cancer can achieve long-term oncological outcomes comparable to those of conventional open surgery.

4.WNK2 delays the proliferation and invasion of hepatocellular carcinoma by inhibiting the ERK1/2/ROS/SHP2 signaling pathway

Objective To investigate the effect of WNK2 on the ERK1/2/ROS/SHP2 signaling pathway in hepatocellular carcinoma(HCC)and to explore its role in cell proliferation and migration in HCC.Methods HepG2 cells were transfected with WNK2-mimic,sh-RNA WNK2,and corresponding negative control.The effect of WNK2 on the proliferation of HCC was examined by subcutaneous tumorigenesis assay in BALB/c nude mice.The expressions of WNK2,p40,gp90,p-SHP2,p-AKT,and p-ERK1/2 in tumor tissues were detected by Western Blot.After treatment with SHP2 inhibitor PHPS1,the expressions of WNK2,P40,gp90,p-SHP2,p-AKT,and p-ERK1/2 in HepG2 cells were detected by Western Blot.The migration ability and invasion ability of HepG2 cells were detected by cell scratch assay and Transwell.The proliferation ability of HepG2 cells was detected by monoclonal proliferation assay.Results Compared with the sh-NC group,the tumor volume of nude mice in the sh-RNA WNK2 group was significantly increased(P<0.01);Compared with the NC-mimic group,the tumor volume of nude mice in the WNK2-mimic group was significantly reduced(P<0.01).Western Blot result showed that compared with the sh-NC group,the expression of WNK2 in the sh-RNA WNK2 group was significantly decreased(P<0.01),while the expressions of p40,gp90,p-SHP2,p-AKT and p-ERK1/2 were significantly increased(P<0.01).Compared with the NC-mimic group,the expression of WNK2 was significantly increased in the WNK2-mimic group(P<0.01),and the expressions of p40,gp90,p-SHP2,p-AKT,and p-ERK1/2 were significantly decreased(P<0.01).In vitro experiment,compared with the sh-NC group,the expression of WNK2 was significantly decreased in the sh-RNA WNK2 group(P<0.01),while the expressions of p40,gp90,p-SHP2,p-AKT and p-ERK1/2 were significantly increased in the sh-RNA WNK2 group(P<0.01).Compared with the sh-NC+PHPS1 group,the expression of WNK2 was significantly decreased in the sh-RNA WNK2+PHPS1 group(P<0.01),while the expressions of p40,gp90,p-SHP2,p-AKT,and p-ERK1/2 were reversed and had no significant differences compared with the sh-NC+PHPS1 group(P>0.05).The cell scratch assay and Transwell result showed that the migration and invasion ability of HepG2 cells in the sh-RNA WNK2 group was significantly increased compared with the sh-NC group(P<0.01).The migration and invasion ability of HepG2 cells in the sh-NC+PHPS1 group and sh-RNA WNK2+PHPS1 group were significantly decreased with no significant difference(P>0.05).The result of the monoclonal proliferation experiment showed that the proliferation capacity of HepG2 cells in the sh-RNA WNK2 group was significantly increased compared with the sh-NC group(P<0.01),while the proliferation ability of HepG2 cells in the sh-NC+PHPS1 group and sh-RNA WNK2+PHPS1 group was significantly decreased with no significant difference(P>0.05).Conclusions WNK2 can inhibit the ERK1/2/ROS/SHP2 signaling pathway,thereby inhibiting ERK1/2/Akt signaling and delaying the proliferation and migration of HCC.
5.Synthesis of taurine-fluorescein conjugate and evaluation of its retina-targeted efficiency in vitro.

In this work, retinal penetration of fluorescein was achieved in vitro by covalent attachment of taurine to fluorescein, yielding the F-Tau conjugate. Nuclear magnetic resonance (NMR) and high resolution mass spectrometry (HRMS) were used to confirm the successful synthesis of F-Tau. The cellular uptake of F-Tau in adult retinal pigment epithelial cells (ARPE-19) and human retinal microvascular endothelial cells (hRMECs) was visualized via confocal scanning microscopy. The results indicated an improvement of solubility and a reduction of logP of F-Tau compared with fluorescein. As compared with fluorescein, F-Tau showed little toxicity, and was retained longer by cells in uptake experiments. F-Tau also displayed higher transepithelial permeabilities than fluorescein in ARPE-19 and hRMECs monolayer cells (P<0.05). These results showed that taurine may be a useful ligand for targeting small-molecule hydrophobic pharmaceuticals into the retina.
