Purpose To investigate the expression of Apaf-1 and Caspase-9 in prostate cancer ( PCa) and benign prostatic hyperplasia ( BPH) , and to analyze their correlation with clinicopathological parameters of PCa. Methods Immunohistochemistry was used to de-tect Apaf-1 and Caspase-9 protein in 45 cases of PCa and 60 cases of BPH. Results The positive rate of Apaf-1 and Caspase-9 in PCa tissues was significantly lower than that in BPH (P<0. 05). The expression of Apaf-1 and Caspase-9 was not correlated with the age of patients and distant metastasis (P>0. 05), but it was correlated with the pathological grade and clinical stage of PCa (P<0. 05). Conclusion Apaf-1 and Caspase-9 are lowly expressed in PCa. There is positive correlation between the expression of Apaf-1 and Caspase-9 (rs =0. 645, P<0. 01). Apaf-1 and Caspase-9 might be correlated with the carcinogenesis and development of PCa.

Objective To investigate the value of CT scans in differential diagnosis of thyroid neoplasm.Methods CT findings ofthyroid neoplasm in 29 cases including 8 cases thyroid adenoma,4 cases nodular goiter and 17 cases thyroid carcinoma identified by operation and pathology were analyzed retrospectively.Results The contour of thyroid adenoma was distinct,a completely enhanced ring in 5 cases and enhanced nodules in 3 cases were showed on contrast-enhanced scan.4 cases of nodular goiter showed homogenous cystic low density the lesions were localized and distinct in contour,the cystic wall could be seen.Thyroid carcinoma appeared unhomogenous density,the edges contour of lesion was not distinct and infiltration on adjacent tissue might be seen.Conclusion CT scan might play an important role in diagnosis and differential diagnosis of thyroid neoplasm.

ObjectiveTo explore the expression of hemopoietin(EPO) mRNA on cerebral ischemia-reperfusion injury in rats brain tissue and the effect of Tongxinluo on it.Methods The model of rat (MCAO) were perfused with Tongxinluo,the changes of neural stem cell proliferation and differentiation related cell factors of EPO mRNA were detected after ischemia-reperfusion injury 3、5、7、14 d by means of reverse transcriptase polymerase chain reaction(RT-PCR).ResultsEPO mRNA of ischemia-reperfusion models showed expression in different period,the expression enhanced in the third day,reached the highest in the fifth day; the ischemia side EPO mRNA expression enhanced in the third day after give Tongxinluo,in the 5,7 and 14 day,PCR expression gray values were higher than the model group.ConclusionEPO mRNA expression enhanced after cerebral ischemia,this expression can be strengthened by Tongxinluo,and may further induce neural stem cell proliferation and differentiation.

Objective To investigate the correlations between the hippocampal acetylcholine (ACh) content and neuronal apoptosis in the hippocampal CA1 region as well as the spatial memory impairment in a rat model of vascular dementia (VaD).Methods Forty male healthy Sprague-Dawley rats were randomly divided into either a VaD group or a sham operation group (n =20 in each group).A VaD model was induced by intermittently clipping common carotid artery.Microdialysis was used to collect dialysis solutions in rat hippocampus.High-performance liquid chromatographic analysis was used to detect the ACh content in the dialysis fluid.Morris water maze test was used to test their learning and memory abilities.TUNEL staining was used to detect neuronal apoptosis in the hippocampal CA1 region.Results Microdialysis analysis showed that the ACh content in the hippocampus in the VaD group was significantly lower than that in the sham operation group (0.442 ± 0.028 μmmol/L vs.1.560 ± 0.092 μ mmol/L; t =51.697,P =0.000).TUNEL staining showed that the apoptosis rate in the hippocampal CA1 region in the VaD group was significantly higher than that in the sham operation group (55.652％ ±2.051％ vs.6.530％ ± 1.872％ ; t =79.114,P=0.000).The escape latencies at different detection time points were prolonged significantly (At day 3:49.713 ± 18.161 s vs.13.322 ± 2.454 s; t =-8.881,P =0.000; at day 4:34.368 ± 7.424 s vs.10.503±1.415 s; t=-14.121,P=0.000; at day 5:30.676± 6.669s vs.7.311± 1.534 s; t=-15.270,P =0.000),and the numbers of cross platform were reduced significantly (3.768 ± 1.072 vs.10.218 ± 1.165; t =18.224,P=0.000).Pearson correlation analysis showed that the ACh contents in the VaD group were negatively correlated with the escape latencies (at day 3:r =-0.476,P =0.034; at day 4:r=-0.700,P=0.001; at day 5:r=-0.693,P=0.001).They were positively correlated with the numbers of cross platform (r =0.689,P =0.001),and negatively correlated with the neuronal apoptosis rates in the hippocampal CA1 region (r =-0.271,P =0.031).Conclusions The decreased ACh content,the increased neuronal apoptosis rate in the rat hippocampal CA1 region in the VaD model may be one of the mechanisms of cognitive impairment in VaD rats.

Objective To retrospectively analyze medical data of patients with colorectal cancer (CRC) so as to provide evidence for clinical use of opportunistic screening.Methods A total of 2450 CRC patients (male 1377,female 1073) who were treated at five hospitals in North China during October 2001 and September 2011 and had complete medical records and pathological results were recruited.The correlations of incidenceofCRCwithage,gender,tumorlocationandhistologicaltypeswere analyzed.Results Of all the CRC patients,those less than 50 years old accounted for 18.14％ ; and the incidence of CRC was substantially increased in those over 50 years old.Seventy-three percent of tumor occurred at the rectum and sigmoid colon,6％ at descending colon,7％ at transverse colon and 14％ at ascending colon.Moderately,well or poorly differentiated adenocarcinoma accounted for 50.33％,40.35％and 9.32％,respectively.Histological differentiation was not correlated with age and gender ( P ＞ 0.05 ).Conclusions Age and gender should not be considered a determination of opportunistic screening for CRC.Colonoscopy is recommended as an alternative CRC screening procedure.

Objective To investigate the effects and mechanism of retinoie acid on B16 marina mel-anoma cells and human melanocytes in vitro. Methods B16F10 murine melanoma cells and human mela-noeytes were cultured in culture medium which contains different concentration of components, including retinoic acid. Using reverse transcription-polymerase chain reaction (RT-PCR) mRNA expression of the tyrosinase was detected. Tyrosinase activity, melanin content and cell proliferation rate were also deter-mined. Results Retinoieacid exhibited an inhibitory effect on the expression of tyrosinase mRNA. As the concentration of retinoic acid was 100 μmol/L, treating for 72 h, the expression of tyrosinase mRNA de-creased 30.13 %, retinoic acid exhibited an inhibitory effect on tyrosinase activity and melanin production at high concentration (>500 μmol/L), and it could promote the cell proliferation. Retinoic acid and hy-droqninone could be cooperative at high concentration (1 000 μmol/L), and enhanced the down regulation of tyrosinase activity and melanin content. Retinoic acid could also mitigate the inhibitory effect of hydro-quinone on cell proliferation, so as to protect the cells from injury. Hydroquinone had no effect on tyrosi-nase gene expression at mRNA level. Conclusion Retinoic acid inhibits the synthesis of melanin by the genetic regulation at mRNA level.

Objective To analyze the inpatient specialist capacity of a hospital during 2013-2014 in Beijing for its specialists development, based on relevant local specialist development.Methods Analyzing the inpatient specialist capacity using case mix index (CMI) and DRG numbers according to BJ-DRG grouping method.Each MDC of the hospital is divided into four types according to the status and development of inpatient specialist capacity.Results Of the 19 MDCs, 2 (such as nervous system disease) fall into type 1, 6 MDCs (such as neck, ears, nose, mouth, pharynx disease and dysfunction) into type 2, while 5 MDCs (such as female reproductive system diseases and dysfunction) into type 3, and 6 MDCs (such as pregnancy, childbirth and postpartum) into type 4.Condusion Most of the MDC inpatient specialist capacities of the hospital in question are at ranking high in Beijing, and such capacities for various MDC can be improved by all means.

CMTM family is silenced and down-regulated in several kinds of tumors.Its aberrant expression has a strong association with the development,progression and metastasis of tumors.Thus,CMTM family is potential tumor suppressor genes.Epigenetics mechanism is the essential mechanism of the aberrant expression in this gene family.The discovery of this research gives a new direction to the clinical treatment of tumor.

Objective To study the effect of osgentide (OST) on proliferation of mouse preosteoblast MC3T3-E1 under simulated microgravity ( SMG ) .Methods Under normal conditions , cell proliferation was evaluated by MTT assay to screen an OST compound of an effective concentration after MC 3T3-E1 cells were treated with series OSTs .Furthermore, cell proliferation and cell cycle distribution of MC 3T3-E1 cells were analyzed after treatment with 1 nmol/L OST5 by MTT assay and by flow cytometry ( FCM) scanning under SMG .Results Under normal conditions , 1 nmol/L OST5 was able to significantly promote the proliferation of MC3T3-E1 cells (P<0.01).Under SMG, proliferation of MC3T3-E1 cells was significantly inhibited and more cells entered G 1 than under normal conditions (CN).The proportion of S phase of MC3T3-E1 cells after treatment with 1 nmol/L OST5 ( OST-SMG) for 3 d was higher than that of untreated MC 3T3-E1 cells under SMG,suggesting that OST5 could promote DNA synthesis ( P<0.05 ) .Conclusion OST5 facilitates the proliferation of MC3T3-E1 cells under SMG, which provides a basis for the use of OST5 in the prevention and treatment of bone loss relat-ed to microgravity .

Objective To study the inhibitory effect of verapamil and nicardipine on human scar fibroblast in serum-free culture and to compare the effectness of the two drugs.Methods We used MTT method to detect the effect of two drugs on human scar fibroblast proliferation:adding verapamil and nicardipine with different concentrations in the culture of fibroblasts which were in logarithmic growth phase (150,100,50,10,0μmol/L).After 24,72,and 120 h,we used MTT method to detect the cell proliferation,and converted the absorbance into growth inhibitory ratio.Results Verapamil and nicardipine showed the definite inhibition on the hypertrophic scar fibroblast (HSFB) and keloid fibroblast (KDFB) which were cultured in vitro.There was some difference in the action feature.In the earlier period,the effect of verapamil was powerful than that of nicardipine.With time,the effect did not reinforce.When fibroblast had been cultured for three to five days,the inhibition became weak.But nicardipine showed lasting inhibition on fibroblast proliferation.Conclusion Combination of verapamil with nicardipine may be a valuable method in the treatment of scar.
