China Pharmacy 2015;26(35):4989-4991
OBJECTIVE:To observe therapeutic efficacy and safety of low molecular heparin calcium (fraxiparine) combined with vapour-pressure type blood circulation driver for the prevention and treatment of postoperative lower limb deep vein thrombo-sis(DVT)in elderly fracture patients. METHODS:80 elderly patients receiving fracture surgery were randomly divided into con-trol group and observation group, with 40 cases in each group. Control group only received preventive measures for lower limb DVT under the action of vapour-pressure type blood circulation driver, while observation group was additionally given Low molecu-lar heparin calcium injection on the basis of control group,Preoperative 2 h,postoperative 12 h and 24 h,0.4 ml was administrat-ed;Postoperative d2-d3,0.4 ml was subcutaneously injected;postoperative d4,0.6 ml was administrated and lasted for 10 d. The incidence of lower limb venous thrombosis, therapeutic efficacy, the changes of blood rheology indexes and the cross-section diam-eter difference of uninjured side limb were observed in 2 groups;the incidence of ADR was compared between 2 groups. RE-SULTS:The observation group;total effective rate (95.00%) was higher than control group (72.50%),with statistical signifi-cance (P<0.05). After treatment, the cross-section diameter difference of side and uninjured side limb in observation group de-creased to(1.10±0.87)cm,which was significantly lower than control group(2.59±1.55)cm,with statistical significance(P<0.05). The blood viscosity at both high and low shear rates, plasma viscosity and fibrinogen of observation group were all better than those of control group, with statistical significance(P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:In the prevention of lower limb DVT in elderly fracture patients,Low molecular heparin calcium injection combined with vapour-pres-sure type blood circulation driver show high effective rate, significantly improve blood rheological index and lower the incidence of lower limb DVT with good safety.

By analyzing the mechanism of pronunciation, traditional acoustic parameters, including fundamental frequency, Mel frequency cepstral coefficients (MFCC), linear prediction cepstrum coefficient (LPCC), frequency perturbation, amplitude perturbation, and nonlinear characteristic parameters, including entropy (sample entropy, fuzzy entropy, multi-scale entropy), box-counting dimension, intercept and Hurst, are extracted as feature vectors for identification of pathological voice. Seventy-eight normal voice samples and 73 pathological voice samples for /a/, and 78 normal samples and 80 pathological samples for /i/ are recognized based on support vector machine (SVM). The results showed that compared with traditional acoustic parameters, nonlinear characteristic parameters could be well used to distinguish between healthy and pathological voices, and the recognition rates for /a/ were all higher than those for /i/ except for multi-scale entropy. That is why the /a/ sound data is used widely in related research at home and abroad for obtaining better identification of pathological voices. Adopting multi-scale entropy for /i/ could obtain higher recognition rate than /a/ between healthy and pathological samples, which may provide some useful inspiration for evaluating vocal compensatory function.

Objective To investigate the effect of imatinib on the growth of A549 non-small cell lung cancer transplanted tumors and the expression of PDGFB and PDGFRβ proteins in tumor tissues and stroma in nude mice and to explore the underlying tumor suppression mechanism. Methods A transplantation tumor model of A549 non-small cell lung cancer was established in nude mice. The mice were then randomly divided into four groups: control group (0.9%NaCl), low-dose imatinib group (50 mg/(kg·d)), medium-dose imatinib group (100 mg/(kg·d)), and high-dose imatinib group (200 mg/(kg·d)). The effect of different concentrations of imatinib administered by continuous gavage on tumor growth was observed for 28 days. HE staining was performed to observe the pathological changes of tumor tissues. The expression of PDGF/PDGFR pathway-related proteins and the phosphorylation levels of AKT and ERK1/2 proteins in tumor tissues were detected by Western blot analysis. Double immunofluorescence staining was used to detect the expression of PDGFB and PDGFRβ proteins in the tumor stroma. Results Imatinib inhibited the growth of A549 non-small cell lung cancer cells in nude mice, suppressed the expression of PDGFB in tumor tissues, and decreased the phosphorylation levels of PDGFRβ, AKT, and ERK1/2. The expression of PDGFB and PDGFRβ in tumor stromal fibroblasts of the administered group was significantly lower than that of the control group. Conclusion Imatinib exhibits a pronounced inhibitory effect on A549 xenografts of nude mice with non-small cell lung cancer, and its antitumor mechanism may involve the downregulation of PDGFB and PDGFRβ expression in tumor stromal fibroblasts.

Objective To construct a eukaryotic expression plasmid carrying human full-length Notch ligand Delta-like 3 (DLL3) gene and study the effect of DLL3 knockdown and overexpression on the proliferation of gastric cancer cells in vitro. Methods Human full-length DLL3 gene was amplified by PCR and cloned into the eukaryotic expression vector pCMV-Tag4. After verification by restriction enzymes and sequencing, the recombinant DLL3/pCMV-Tag4 vector was transiently transfected into HEK293T cells, in which the expressions of human DLL3 mRNA and protein were detected using real-time quantitative PCR and Western blotting, respectively. The expression of DLL3 in normal gastric epithelial cells and gastric cancer cell lines was detected by qRT-PCR and Western blotting. DLL3/pCMV-Tag4 was transfected into 3 gastric cancer cell lines, and their proliferation was assessed with MTT assay. Human gastric cancer cells MGC803 and MKN45 were also transfected with a specific human DLL3-siRNA to assess the effect of DLL3 down-expression on the cell proliferation. Results The recombinant eukaryotic expression vector DLL3/pCMV-Tag4 was successfully constructed and human full-length DLL3 was expressed in HEK293T cells. MTT assay showed that DLL3 over-expression obviously promoted the proliferation and down-regulation of DLL3 inhibited the proliferation of the gastric cancer cells. Conclusion DLL3 overexpression can promote the proliferation of gastric cancer cells in vitro, and down-regulation of DLL3 inhibits the proliferation of gastrc cancer cells, which provides a novel strategy for targeted thrapy of gastric cancer.
