1.Progress of nuclear transcription factor Sp1 regulating angiogensis of tumor
Journal of International Oncology 2012;39(1):18-21
Sp1 is one of the nuclear transcription factors which are widely expressed in human body.It plays an important role in human growth and physiological activities, and is highly expressed in many kinds of tumors'tissue.Sp1 participates in regulating growth, invasion, angiogensis actions of tumor by regulating downstream gene(s) transcription. Some grow factors and tumor suppressor genes can regulate the expression of VEGF by Sp1,and then participate in angiogensis of tumor by two main mechanisms:kinases inducing Sp1 phosphorylation and the interact of Sp1 and other proteins.

Objective To explore the effects of Xiefuzhuyu decoction on myocardial cellar apoptosis and expression of ICAM-1 gene protein in rats of experimental acute ischemic myocardium. Methods The left anterior descending coronary artery was ligated to make a model of acute ischemic myocardium. The changes in myocardial cellar apoptosis and the ICAM-1 gene expression were detected. Results The number of myocardium cellar apoptosis in Xiefuzhuyu group and Diaoxinxiekang group were significantly less than those of ischemic model group respectively. The number of myocyte necrotic cell increased in ischemic group. At the same time, the expression of ICAM-1 supported this condition. Conclusion Xiefuzhuyu decoction and Diaoxinxiekang can protect ischemic myocardium.

Acid-sensing ion channels( ASICs )are ubiquitously expressed both in periphery nervous system, where they are involved in nociception, mechanosensation, inflammation ,cardiac angina, and in central nervous system, where they are essential to a variety of physiologic and pathophysiologic processes, such as synaptic plasticity ,learning ,memory and ischemic brain injury. Here in the article, we present a collection of key points about ASICs, ranging from molecular identity and expression, regulatory mechanisms responsible for channel gating to their multiple functions. Finally, a future prospect for the investigation of ASICs is outlined.

Spasticity can influence the outcome of the patients with cerebral injury. Most traditional physical therapies often focused on the spastic muscle. The recent development has showed that stimulating the antagonistic muscle can relieve the spasticity, which based on the reciprocal inhibition theory. This paper reviewed these approaches on its theory and application.

Objective To study the effects of sibutramine on the human-ether-a-go-go-related gene (hERG) current in HEK293 cell line that overexpresses hERG potassium channels. Methods HEK293 cell line that overexpresses hERG potassium channels was constructed. Whole cell patch clamp techniques were employed to investigate the effects of sibutratime on Iherg in the cell line. Results Sibutramine reduced hERG current in a concentration-dependent manner:the inhibition rate was (8.82?2.32)%,(26.7?5.78)%,(44.93?4.60)% and (85.22?3.37)% at the dose of 3,10,30 and 100 ?mol/L. IC50 value was 32.66 ?mol/L (n=8). Ⅰ-Ⅴ relationship was inhibited by sibutramine (3,10,30 and 100 ?mol/L) at membrane potentials from -10 mV to +60 mV (step) and from 0 mV to +60 mV (tail) (n=8). Conclusion Sibutramine significantly inhibited Iherg in HEK293 cell line that overexpresses hERG potassium channels in a concentration-dependent manner, which may be a major mechanism of causing QT interval prolongation.

Aim To investigate the blockade of cadmium on store operated calcium channels and the fluorescence interference of cadmium with Fura-2.Methods PC12 cells were used to determine the intracellular calcium concentration [Ca~(2+)]i indicated by change in Fura-2 fluorescence ratio(F_(340)/F_(380)).Results Introduction of cadmium induced a significant increase in Fura-2 fluorescence ratio following thapsigargin-evoked calcium entry;Besides, cadmium gave rise to a remarkable elevation in Fura-2 fluorescence ratio following breakdown of the plasma membrane by triton,a detergent. The Fura-2 fluorescence was increased in the presence of cadmium and Fura-2/AM,simultaneously in the absence of PC12 cells. Conclusion Cadmium can interfere with the Fura-2 fluorescence.

Objective To compare the cell-killing and sensitization effect of 6-gingerol on human hepatoma carcinoma (HepG-2) cell in normal mode versus hypoxia-hypoglycemia mode in chemotherapy. Methods The HepG-2 cells was cultured to logarithmic phase and treated with adriamyein doxorubicin hydrochloride (ADM) (5,10,15,20,40,60 mg/L) and 6-gingerol(25,50,100,200 μmol/L)in different concentrations. Then the cell counting kit (CCK-8) assay kit was used to determine the proliferation inhibition of HepG-2 cells.Cell apoptosis was detected by combining flow cytometry and AnnexinV-FITC PI double staining after treated with different drugs. The expressions of bcl-2,bax and birc-5 mRNA in HepG-2 cells was detected by real time fluorescence quantitative polymerase chain reaction(RT-PCR)assay.Results 6-gingerol and ADM had a certain degree of growth inhibition on HepG-2 cells. In two modes, the inhibition ratios of the 6-gingerol and ADM were both increased along with the increase of the concentration, which showed a dose-dependent manner.Flow cytometry analysis demonstrated that the apoptosis rate in the control group,6-gingerol group,ADM group and the 6-gingerol+ADM group in the normal mode was (7.98±0.76)%,(9.63 ± 1.00) %,(12.70 ± 2.13) % and (19.92 ± 1.41) % respectively. The apoptosis rate in the control group,6-gingerol group,ADM group and the 6-gingerol+ ADM group in the hypoxia-hypoglycemia mode was (13.92 ± 2.02)%,(19.36 ±-1.22)%,(27.87 ± 0.99)% and (38.63 ± 2.25)% respectively. It demonstrated that the apoptosis rate was increased in the experimental groups as compared to the control group under the two culture conditions(the normal mode and the hypoxiahypoglycemia mode)(t=7.250,5.259,12.185,8.140,15.000,47.576,respectively,all P<0.05,0.01 or 0.001).The combination group had the highest number of apoptosis cells, and the number of apoptosis cells was higher in hypoxia-hypoglycemia group than in normal culture group.Real-time PCR analysis showed that, compared with the control group, the expressions of bcl-2 and birc-5 mRNA were decreased and the expression of bax mRNA had no significant changes in experimental group under the normal culture conditions. The expressions of bcl-2 and birc-5 mRNA were significantly decreased and the expression of bax mRNA was increased in experimental group as compared with the control group under the hypoxia-hypoglycemia conditions.Under the hypoxiahypoglycemia environment, the expression of bcl-2 mRNA was significantly increased, the expressions of bax and birc-5 mRNA was significantly reduced, and the ratio of bcl-2 and bax was significantly increased as compared with the normal culture conditions. Conclusions 6 gingerol may decrease the inhibitory effect of survivin protein on tumor cells apoptosis by reduced the expression of birc-5,which generates the cell-killing and sensitizing effect on HepG-2 cell in chemotherapy. This performance is more obvious in the hypoxia-hypoglycemia environment.
