Objective:To investigate the effects of Yukuiqing (YKQ) on the expression of connective tissue growth factor (CTGF) in cultured human renal mesangial cells (HRMC) incubated with AGEs. Methods:The HRMCs were incubated with AGEs (200 ?g/mL) and 1.25% YKQ which was prepared by Chinese herbal medicine serum pharmacological approach for 0,8,16,24,48 and 72h or different concentrations (0.313%,0.625% and 1.25%) of YKQ for 48h,respectively,which were incubated with DMEM and rat serum (RS) as the control. CTGF mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western Blotting respectively. 2.Association study between the genetic polymorphism of 15 STR loci and the crime of rape

Objective To investigate the relationship between rapists and related allele genes based on the analysis of 15 short tandem repeats (STRs) loci genetic polymorphism. Methods The method of Genome-wide scan was being used. Buccal swab samples of 129 rapists and 156 random populations were collected and PCR compound amplification was carried out with the aid of AmpFISTR Identifiler system. Then the products were subjected to electrophoresis and gene detection with AB13100 type gene analysis system so as to calculate and compare the alleles of 15 STRs gene frequency in the two groups. Results All the 15 STRs loci allele gene frequency in rapists and random population was found to coincide with Hardy-Weinberg law(P>0. 05). Allele 28 of D21S11 (rapists: 1.55% ,control group:5. 13%) ,allele22 of FGA(rapists:24.03% ,control group:16.99%),allele23 of FGA(rapists: 17.05% ,control group:26.28%) ,allele 10 of TH01(rapists:1.16% ,control group:4.17%) ,allele 8 of TPOX(rapists:55.77% ,control group:63.77%),allele 12 of TPOX(rapists:4.26% ,control group: 1.28%) were different between the two groups (P< 0.05) .while it is no differ significantly in other STRs loci allele gene(P >0.05). Conclusion Allele 28 of D21 S11,allele 22 and 23 of FGA, allele 10 of TH01, allele 8 and 12 of TPOX may be associated with the violent crime of rape. It is suggested that there are existing sensitive or resistance genes about the violent crime of rape in chromosome 2,4,11,21.
3.Real-time fluorescence PCR-molecular beacon assay for detecting mutations in rpoB genes of rifampin-resistant Mycobacterium tuberculosis

Objective To establish a rapid method to detect mutations in rpoB genes of rifampin-resistant Mycobacterium tubereulosis in dinical specimens using Real-time fluorescence PCR molecular beacon assay.Methods 174 strains of Mvcobacterium tuberculosis clinical isolates were analyzed using real-time fluorescence PCR molecular beacon assay foilowed with DNA sequencing while 12 strains of NTM and 4 strains of bacteria other than Mycobacterium tuberculosis were used as the contrast.Results Eighty-two 89.1 of 92 rifampin (RIF)-resistant strains and 3 of 82 RIF-sensitive strains were found to harbor mutation in the rpoB gene using real-time fluorescence PCR-molecular beacon assay.The specificity, sensitivity,and accuracy of this assay were 96.3%,89.1%,and 92.5%,respectively-Eithty-three of 92 RIF-resistant strains and 1 of 82 RIF-sensitive strains were found to harbor mutation in the rpoB gene using the direct DNA sequencing.The specificity,sensitivity,and accuracy of the direct DNA sequencing were 98.8,90.2%,and 94.2%,respectively.As compared with real-time PCR molecular beacon assay,171 of 174(98.3%)strains of myeobactefium tuberculosis clinical isolates had the salne results.Conclusion Real-time fluorescence PCR-molecular beacon assay can be used as a rapid screen method to detect RIF-resistant isolates.
4.Significance of HER-2/neu expression in ovarian epithelial tumours.

OBJECTIVETo investigate expression of Her-2/neu (c-erbB-2) and its significance in ovarian epithelial tumors.
METHODS106 specimens and clinical history of ovarian epithelial tumors were collected including 54 cases of malignant, 33 cases of borderline and 19 benign cases. There were 19 cases of stages I and II and 35 cases of stages III and IV in ovarian carcinoma and all borderline malignant cases were stage I according to FIGO's standard. Immunohistochemical staining for Her-2/neu was performed by LSAB method.
RESULTSThe positives rates of Her-2/neu of benign, borderline and malignant tumors were 47.4% (9/19), 84.8% (28/33,), and 85.2% (46/54), respectively. Both borderline and malignant cases were found to have higher expression of Her-2/neu than those of benign cases (P < 0.02 and P < 0.01) respectively. Overexpression of Her-2/neu showed in 14/54 (25.9%) of malignant cases, 3/33 (9.1%) of borderline, and none of benign cases. Overexpression of Her-2/neu in malignant tumors cases with metastasis was higher than those without metastasis (P < 0.001).
CONCLUSIONOverexpression of Her-2/neu is associated with biological aggressiveness of ovarian cancer.

5.Resveratrol improves cognitive function in severely burned rats by inhibiting hippocampal NF-κB/JNK pathway.

Objective To investigate the protective effect of resveratrol (RSV) on improving cognitive function in severely burned rats and its possible mechanism. Methods 18 male SD rats aged 18-20 months were randomly divided into 3 groups: control group, model group and RSV group, with 6 rats in each group. After successful modeling, the rats in RSV group were gavaged once daily with RSV (20 mg/kg). Meanwhile, the rats in control group and model group were gavaged once daily with an equal volume of sodium chloride solution. After 4 weeks, the cognitive function of all rats was estimated by Step-down Test. The concentration of tumor necrosis factor α (TNF-α) and interleukin 6 (IL-6) protein in serum of rats were detected by ELISA. The expression of IL-6, TNF-α mRNA and protein were estimated by real-time PCR and Western blotting. The apoptosis of hippocampal neurons was tested by terminal deoxynuclectidyl transferase-mediated dUTP-biotin nick end labeling assay (TUNEL). The expression of nuclear transcription factor-κB (NF-κB)/c-Jun N-terminal kinase (JNK) pathway-related proteins in hippocampus were assessed by Western blotting. Results Compared with the rats in model group, rats in RSV group exhibited improved cognitive function. Consistently, the rats in RSV group had a reduced concentration of TNF-α and IL-6 in serum, decreased mRNA and protein expressions of TNF-α and IL-6 in hippocampus, and decreased apoptosis rate and relative expression of p-NF-κB p65/NF-κB p65 and p-JNK/JNK in hippocampal neurons. Conclusion RSV alleviates inflammatory response and hippocampal neuronal apoptosis by inhibiting NF-κB/JNK pathway, thereby improving cognitive function in severely burned rats.
