1.Characteristics of Behavior and Self-concept in Psychological Abused/Neglected Juveniles with Generalized Anxiety Disorder

Objective: To invesgate the situation of psychological abuse and neglect among juveniles with generalized anxiety disorder(GAD) and explore the characteristics of behavior and self-concept in psychological abused/neglected juveniles with GAD.Methods: A sampling survey was made in 9495 juveniles(5~17) from six regions of Hunan Province.After using the two-stage epidemiologic research method,409 children including 206 normal controls,99 GAD without abuse/neglect and 104 GAD with abuse/neglect,completed CPANS,CSCS and their parents completed CBCL.Results: There were significantly higher rates of psychological abuse or neglect in the GAD group than the normal controls;the factor scores of juveniles with GAD were significantly higher than those with no mental disorders in all six aspects in CPANS(P=0.000),higher scores of internalizing and externalizing behaviors characterized the subgroup of juveniles with both GAD and abuse history compared with the subgroup with GAD without such history,and the scores including total score,behavior,anxiety,gregariousness and happiness factors(P=0.000),of the subgroup of juveniles with GAD and abuse histories were significantly lower than the GAD subgroup without abuse or neglect.Conclusion: Psychological abuse and neglect are more prevalent and severe in juveniles with GAD than in normal ones;when psychological abuse/neglect co-occurs with GAD,juveniles’ behavior problems are even greater and their level of self-concept are much lower.
2.The relationship between impulsive behavior and alexithymia in pupils

ObjectiveTo study the relationship between impulsive behavior and alexithymia in pupils in Changsha.Methods 702 pupils were tested with Barratt impulsivity scale (BIS-11) and Toronto alexithymia scale (TAS-20).Results ( 1 ) Male pupils had significantly higher scores of total BIS and non-planning impulsiveness than female (24.47 ± 5.30 vs 23.40 ± 5.59,P < 0.05 ; 62.25 ± 9.46 vs 60.35 ± 9.91,P < 0.05 ).Pupils in elder age group had only significantly higher total score of BIS than pupils in younger age group ( total:62.36 ± 9.69 vs 60.29 ± 9.68,P< 0.01 ).(2) Pupils with high impulsivity had significantly higher scores of total and factor scores of alexithymia than pupils with low impulsivity (total:57.43 ± 10.16 vs 46.55 ± 8.77,P <0.01 ).The total scores of BIS were positively correlated with total scores of alexithymia ( r =0.40,P < 0.01 ) and factor scores of alexithymia ( r =0.25 ~ 0.33,P < 0.01 ).Stepwise regression analysis revealed that total scores of BIS explained by DIF,DDF and EOT were 18%.ConclusionThere are certain gender and age differences in impulsivity impulsive behavior of 8 to 13 years old pupils.There exist the low to moderate correlations between impulsivity and alexithymia in pupils.
3.The relationship of impulsive personality and parental rearing patterns in junior middle school students

Objective To study the development of impulsive personality in junior middle school students of Changsha, and to explore its relationship with parental rearing patterns. Methods 409 students were investigated by Egna Minnenav Barndoms Uppfostran ( EMBU ) and Barratt Impulsivity Scale ( BIS-11 ). Results Male and female had significant difference in attention impulsiveness scores((20.56 ±3.60) vs ( 19.63 ±3.07) , P< 0.01). Grade 7 and Grade 9 had significant difference in attention and motor impulsiveness scores((20. 58 ± 3.47)vs (19.63 ±3.46) , P<0.05;(20. 31 ±4.37)vs (21.75 ±4.00) , P< 0.01). Parental warmth and understanding was negatively correlated with attention , non-planning and total scores of impulsiveness ( r= - 0. 23 ~ -0.33, P<0.01). Parental refusal and rejection was positively correlated with total and factors of BIS-11 ( r = 0. 23 ~0. 33 , P<0. 01 ) . Parental punishment and rigor was positively correlated with motor and total scores of im-pulsiveness( r = 0.22 -0.26, P<0.01). Parental over-interference and overprotection was positively correlated with motor impulsiveness ( r = 0. 23 ~0. 34, P<0. 01). Mother's punishment and rigor was positively correlated with attention impulsiveness( r = 0.22, P<0. 01 ). Father' s overprotection and Mother' s over-interference and overprotection was positively correlated with total scores of impulsiveness ( r = 0. 23 ;0. 25 , P < 0. 01) . Stepwise regression analysis revealed that factors and total of BIS-11 explained by patterns rearing were 12% ,14% ,15% and 19% . Conclusion Rearing patterns can severely impact the development of impulsive personality. Impulsive personality is positively predicted by parental rearing patterns.
4.A Rho-kinase inhibitor, fasudil, attenuates progressive glomerulosclerosis induced by daunorubicin in rats.

Accumulating evidence suggests that the small G protein Rho and its downstream effector Rho kinase may play important roles in kidney biology. The present study examined the effects of a Rho-kinase inhibitor, fasudil, on daunorubicin-induced progressive glomerulosclerosis and explored the underlying mechanism by which fasudil ameliorates glomerulosclerosis. Thirty-six male SD rats were randomly allocated into sham-operation group (sham group, n=12), unilateral nephrectomy (UNX)+daunorubicin (DRB) group (model group, n=12), UNX+DRB+Fasudil group (treatment group, n=12). Two to four weeks after the establishment of the animal model, 6 rats in each group were taken randomly for the detection of 24-h urine protein excretion. Kidney sections were examined by HE and PAS staining, immunohistochemistry and transmission electric microscopy (TEM). The expression of Rho-kinase mRNA and P27 mRNA in kidney were detected by RT-PCR. It was found that the 24-h urine protein excretion in model group was increased significantly as compared with sham group (P<0.01). But this increase was significantly suppressed by fasudil (P<0.05). At 4 week, the foot process effacement in podocytes, mesangial proliferation and ECM accumulation were observed in model group, presenting as focal segmental glomerulosclerosis. But in the treatment group, the fasudil alleviated glomerular injury, with proliferating cell nuclear antigen (PCNA)-positive cell infiltration ameliorated and the expression of P27 increased. The expression of Rho-kinase mRNA was significantly enhanced in model group and was suppressed in treatment group. Moreover, fasudil up-regulated the mRNA expression of P27. Our study demonstrated that the glomerulosclerosis was substantially ameliorated by inhibiting the expression of Rho-kinase. It is suggested that Rho-kinase pathway is involved in the renal injury and the inhibition of Rho-kinase may constitute a therapeutic strategy for the treatment of renal injury.

6.Analysis of PMM2 gene variant in an infant with congenital disorders of glycosylation type 1a.

OBJECTIVE:
To identify potential mutation of PMM2 gene in an infant with congenital disorders of glycosylation type 1a (CDG-1a).
METHODS:
Genomic DNA was extracted from peripheral blood sample of the patient. All coding exons (exons 1-8) and splicing sites of the PMM2 gene were amplified with PCR. Potential variants were detected by direct sequencing of the PCR products and comparing the results against the ESP and SNP human gene databases. A protein BLAST system was employed to analyze cross-species conservation of the variants amino acid. A PubMed BLAST CD-search system was employed to identify functional domains damaged by variants of the PMM2 gene. Impact of potential variants was analyzed using software including PolyPhen-2 SIFT and Mutation Taster. Whole exome sequencing was used to identify additional variants of the PMM2 gene which may explain the condition of the patient.
RESULTS:
The child was found to carry compound heterozygous variants (c.458_462delTAAGA and c.395T>C) of the PMM2 gene, which were inherited respectively from his father and mother. The c.458_462delTAAGA has not been reported previously and may result in disruption of 10 functional domains within the PMM2 protein. The c.395T>C mutation has been recorded by a SNP database with frequency unknown. Both mutations were predicted as "probably damaging". Whole exome sequencing has identified no additional disease-causing variant which can explain the patient's condition.
CONCLUSION
The patient's condition may be attributed to the compound heterozygous variants c.458_462delTAAGA and c.395T>C of the PMM2 gene. Above results has facilitated molecular diagnosis for the patient.

7.Pressure-strain loops technique detects the effects of different pacing modes of left bundle branch on left ventricular myocardial work

Objective:To evaluate the effects of different pacing modes (unipolar/bipolar) under left bundle branch pacing(LBBP) on ventricular mechanical synchrony and myocardial work using the pressure-strain loop technique.Methods:Twenty-nine patients with LBBP due to symptomatic bradycardia were collected as LBBP group in Sun Yat-sen Memorial Hospital of Sun Yat-sen University from December 2018 to July 2020. Another 29 matched patients with right ventricular pacing (RVP) during the same period were also included as a RVP group. Each LBBP patient was programmed to different pacing modes (uni-/bio-polar) within 1 week after the operation.Under each pacing mode, the inter- and intra-ventricular mechanical synchronization were evaluated. Meanwhile, the global work index (GWI), global constructive work (GCW), global wasted work (GWW), and global work efficiency (GWE) were obtained by the left ventricular pressure-strain loops technique.Results:Compared with the RVP group, the mechanical synchrony in the LBBP group was significantly improved (all P<0.05). GWI, GCW, and GWE increased, while GWW decreased, and the differences were statistically significant (all P<0.05), there were no significant differences in ventricular mechanical synchronization, GWI, GCW, GWE, and GWW between unipolar and bipolar pacing in the LBBP group (all P>0.05), there were no significant differences in these parameters when increasing output voltage (all P>0.05). Conclusions:LBBP induces better mechanical synchronization and higher myocardial work efficiency than RVP. Different LBBP pacing modes do not affect ventricular mechanical synchronization and myocardial work efficiency.
8.Survey on standardization of implementing enteral nutrition in patients with stroke in Guangdong province, China

Objective:To investigate the standardization of implementing enteral nutrition in patients with stroke among nurses in Guangdong province.Methods:The results of 175 valid questionnaires submitted by 175 nurses from 60 hospitals in Guangdong province from June to July 2022 were analyzed. Contents of the questionnaires included status of enteral nutrition management of stroke patients (facility, staffing, risk screening, and formulation, implementation and quality control of the plans), nurses' knowledge of enteral nutrition monitoring and complication prevention, follow-up status of enteral nutrition in stroke patients, and nurses' willingness in participating in special training of enteral nutrition.Results:(1) In terms of enteral nutrition facilities and staffing, only 76% and 36% used enteral infusion pumps and heating infusion devices, and only 3% had multidisciplinary team configuration; in terms of risk screening, only 63% had all-patient-screen for enteral nutrition risk; in terms of plan formulation, the proportion of no enteral nutrition process reached to 44%; in terms of plan implementation, the implementation rate of enteral nutrition≤48 h and gastrointestinal function evaluation using gastrointestinal function evaluation scale was less than 50%; in terms of plan quality control, less than 50% Departments had sensitive indexes of enteral nutrition nursing. (2) Among 14 risk management problems of enteral nutrition monitoring and complication prevention, only 3 had accuracy rate higher than 70%. (3) 78% nurses mentioned that patients had experienced enteral nutrition complications, and 77% nurses mentioned that patients/family members had consulted about enteral nutrition related issues; 81% nurses wanted to participate in training or refresher courses related to enteral nutrition, and 88% nurses wanted to join the enteral nutrition Nursing Alliance for stroke patients to serve patients after referral and discharge.Conclusion:The current situation of enteral nutrition management of stroke patients in Guangdong province is not optimistic; nurses lack knowledge of enteral nutrition monitoring and complication prevention, but are strongly willing to participate in the special training of enteral nutrition; relevant measures to further improve the standardized management of enteral nutrition of stroke patients is suggested.
9.Effect of LAG3 deficiency on natural killer cell function and hepatic fibrosis in mice infected with Echinococcus multilocularis

Objective To investigate the effect of LAG-3 deficiency (LAG3-/-) on natural killer (NK) cell function and hepatic fibrosis in mice infected with Echinococcus multilocularis. Methods C57BL/6 mice, each weighing (20 ± 2) g, were divided into the LAG3-/- and wild type (WT) groups, and each mouse in both groups was inoculated with 3 000 E. multilocularis protoscoleces via the hepatic portal vein. Mouse liver and spleen specimens were collected 12 weeks post-infection, sectioned and stained with sirius red, and the hepatic lesions and fibrosis were observed. Mouse hepatic and splenic lymphocytes were isolated, and flow cytometry was performed to detect the proportions of hepatic and splenic NK cells, the expression of CD44, CD25 and CD69 molecules on NK cell surface, and the secretion of interferon γ (IFN-γ), tumor necrosis factor α (TNF-α), interleukin (IL)-4, IL-10 and IL-17A. Results Sirius red staining showed widening of inflammatory cell bands and hyperplasia of fibrotic connective tissues around mouse hepatic lesions, as well as increased deposition of collagen fibers in the LAG3-/-group relative to the WT group. Flow cytometry revealed lower proportions of mouse hepatic (6.29% ± 1.06% vs. 11.91% ± 1.85%, P < 0.000 1) and splenic NK cells (4.44% ± 1.22% vs. 5.85% ± 1.10%, P > 0.05) in the LAG3-/- group than in the WT group, and the mean fluorescence intensity of CD44 was higher on the surface of mouse hepatic NK cells in the LAG3-/- group than in the WT group (t = −3.234, P < 0.01), while no significant differences were found in the mean fluorescence intensity of CD25 or CD69 on the surface of mouse hepaticNK cells between the LAG3-/- and WT groups (both P values > 0.05). There were significant differences between the LAG3-/- and WT groups in terms of the percentages of IFN-γ (t = −0.723, P > 0.05), TNF-α (t = −0.659, P > 0.05), IL-4 (t = −0.263, P > 0.05), IL-10 (t = −0.455, P > 0.05) or IL-17A secreted by mouse hepatic NK cells (t = 0.091, P > 0.05), and the percentage of IFN-γ secreted by mouse splenic NK cells was higher in the LAG3-/- group than in the WT group (58.40% ± 1.64% vs. 50.40% ± 4.13%; t = −4.042, P < 0.01); however, there were no significant differences between the two groups in terms of the proportions of TNF-α (t = −1.902, P > 0.05), IL-4 (t = −1.333, P > 0.05), IL-10 (t = −1.356, P > 0.05) or IL-17A secreted by mouse splenic NK cells (t = 0.529, P > 0.05). Conclusions During the course of E. multilocularis infections, LAG3-/- promotes high-level secretion of IFN-γ by splenic NK cells, which may participate in the reversal the immune function of NK cells, resulting in aggravation of hepatic fibrosis.
