1.Effect of different formula diets on mouse model with DEN-induced hepatocellular carcinoma

Objective To establish a mouse model of diethylnitrosamine(DEN)-induced hepatocellular carcinoma (HCC),and to explore the effects of two different diet formulas on the establishment of DEN-induced HCC model.Methods SPF C57BL/6 mice (8 males and 8 females) were injected intraperitoneally with 25 mg/kg DEN at day 14 to establish a HCC model.The mice were divided into two groups after weaning.One group was fed with the SPF class rodents cereal-based diet,another group was fed with AIN-93G formula diet.The mice were sacrificed at the age of 9 months.The livers were weighed and the growth of liver cancer was observed and recorded.Results All the mice in the cereal-based diet group developed HCC as expected.The body weight and liver mass of the mice in the AIN-93G diet group were significantly lower than that of the cereal-based diet group.The incidence of HCC,and the number and size of tumor nodules were also significantly lower in the AIN-93G diet group than that in the cereal-based diet group.Conclusions DEN-induced HCC model has been successfully established in mice fed with cereal-based diet,while mice fed with AIN93-G diet prevented the development of DEN-induced HCC,and their body weight was decreased significantly,suggesting that dietary factors play a key role in establishment of animal disease models.
2.Studies on the toxic effects of magnetic targeting adriamycin-carboxymethyldextran magnetic nanoparticles.

In this paper, adriamycin-carboxymethyldextran magnetic nanoparticles (ADR-CMD MNPs) were prepared. After i.v. administration in mice, acute toxicity, cumulative toxicity and the distribution profiles of heart were studied both for free adriamycin(ADR) and ADR-CMD MNPs. The results showed conjugation with CMD MNPs, the acute toxicity of ADR was decreased significantly, the LD50 value of ADR-CMD MNPs was 5.06 times as high as that of the free ADR. Altogether, the cumulative toxicity of conjugate MNPs is significantly decreased as expressed by the mortality, the loss for both weight and leucocyte after repeated injection. Tissue distribution studies show the reduced cardiac uptake of ADR after i.v. which possibly contributes to minimizing the cardiotoxic effect of ADR.

3.The metastatic pattern of thoracic lymph nodes in 306 patients with lung cancer.

BACKGROUNDTo study the metastatic pattern of thoracic lymph nodes in patients with resectable lung cancer.
METHODSFrom January 1992 to December 2000, radical lobectomy or pneumonectomy and systemic lymphadenectomy were performed in 306 patients with lung cancer. Number, size, colour and hardness of lymph nodes in each region were recorded and neoplastic metastasis was examined by pathology.
RESULTSOut of 4 614 resected lymph nodes from 2 456 regions, 954 lymph nodes from 521 regions were confirmed to have metastasis. The metastatic rates of thoracic lymph nodes and mediastinal lymph nodes were 61.8% and 43.5% , respectively. The metastatic frequencies in regions around the hilar or root of lung (11,10,7,5,4) were higher than those of regions far from the root of lung (9,6,3,2,1). There was a remarkably higher metastatic rate of lymph nodes in small cell lung cancer than that in non small cell lung cancer (P < 0.01 ). The metastatic rate was closely related to size, colour and hardness of lymph nodes (P < 0.001, P < 0.001 , P < 0.001 ).
CONCLUSIONSMost of neoplastic metastasis of lymph nodes spreads from proximal to distal areas, lower to upper regions, and from the hilar to the mediastinal. A few mediastinal lymph nodes show a skipping pattern. It is necessary to perform systemic lymphadenectomy during pulmonary resection.
4.Carinal resection and bronchial sleeve lobectomy in the treatment of central lung cancer: A report of 105 cases.

BACKGROUNDTo summarize the experience of carinal resection and bronchial sleeve lobectomy in the treatment of 105 patients with central lung cancer from November, 1991, to November, 2001.
METHODSA total of 105 patients with central lung cancer underwent pulmonary resection. Carinal resection and reconstruction was performed in 19 patients, bronchial sleeve resection in 81, and bronchial sleeve combined with pulmonary artery sleeve lobectomy in 5.
RESULTSThere was no operative mortality. Postoperative complications such as anastomotic leakage, cardiac arrhythmia, asthma, pulmonary atelectasis occurred in 10.5% of total group. The 1-, 3- and 5- year survival rates were 89.9%, 60.0% and 47.2%, respectively.
CONCLUSIONSBronchial sleeve lobectomy and double-sleeve lobectomy are capable of excising pulmonary tumor as much as possible while remaining healthy lung tissues. Carinal resection and reconstruction is helpful to extend the surgical indication.
5.Research progress of radiolabeled bombesin drugs targeting GRPR in the diagnosis and treatment of prostate cancer

Prostate cancer is the most common cancer among men and the fifth leading cause of cancer-related death among men worldwide. Gastrin-releasing peptide receptor (GRPR) is an important complementary target of prostate specific membrane antigen (PSMA) and a key target for prostate cancer diagnosis and treatment. Radionuclide labeled bombesin (BBN) analogues can accurately target overexpressed GRPR in tumor cells, thus providing early diagnosis and treatment of prostate cancer. In this review, we focus on the studies of 99Tc m, 111In, 68Ga, 64Cu, 18F, and 177Lu nuclide-labeled BBN analogues for diagnosis and peptide receptor radionuclide therapy.
