1.Clinical Effect of Interferon α-2 b on Scar Formation after Glaucoma Filtering Surgery

Objective:To investigate the preventive efficacy of interferon α -2b on scar formation after glaucoma filtering opera-tion. Methods:Totally 62 patients with primary glaucoma(68 eyes)were randomly divided into the observation group(31 cases with 36 eyes)and the control group(31 cases with 32 eyes). The patients in the observation group were given wet compress with a piece of cotton infiltrated by interferonα-2b,while the control group was given wet compress with a piece of cotton infiltrated by mitomycin C. After 4-month postoperative follow-up,the type of filtering bleb,complications and the changes of intraocular pressure were compared between the two groups. Results:The observation group was mainly with type II bleb(58. 33%),while the control group was mainly with type I bleb(53. 13%),and there was significant difference in the filtering bleb type between the two groups(P<0. 05). After the operation,the intraocular pressure at each time point in the two groups was decreased significantly when compared with that before the operation(P<0. 05),while the difference between the two groups was not statistically significant(P>0. 05). The percentage of visual invariableness or improvement in the observation group was significantly higher than that in the control group(P<0. 05),and the occurrence of complications after the operation in the observation group was significantly lower than that in the control group( P<0. 05). Conclusion:Interferon α-2b is an ideal drug for anti-scarring formation after glaucoma filtration surgery.
2.Fluorescence properties of 5-iodoaccetamidofluorescein-labeled rabbit cardiac troponin C

BACKGROUND:Measurement of cardiac troponin plays an important role in diagnosis of myocardial infarction.OBJECTIVE:To label the rabbit cardiac troponin C(cTnC) by a fluorescent probe 5-iodoaccetamidofluorescein(5-IAF),and to observe whether the 5-IAF can be used to study the interaction between cTnC and other contractile regulatory proteins.DESIGN,TIME AND SETTING:A randomized control experiment was performed at Department of Human Anatomy,Guangzhou Medical College,from January 2002 to December 2005.MATERIAL:Adult rabbits were provided by Experimental Animal Center of Guangzhou Medical College.METHODS:The rabbit cTnC DNA fragment was prepared with RT-PCR method.This gene fragment was cloned to pET expression vector by gene recombination technology.The site-directed mutagenesis were used to produce a mutant containing single cysteine at position 84 by replacing Cys35 with Ser,cTnC(C35S).The cTnC(C35S) was labeled by 5-IAF and 2-(4'-(iodoacetamido) anilino) naphthalene-6sulfonic acid(IAANS),Respectively.And then,the fluorescence emission(steady-state and time-resolved) was performed.MAIN OUTCOME MEASURE:The fluorescence properties of 5-IAF-labeled cTnC(C35S) and IAANS-labeled cTnC(C35S).RESULTS:The excitation of apo-cTnC(C35S)IAF was performed at 491 nm,and the emission peak was at 520 nm.Saturation of cTnC(C35S)IAF with Mg led to a 35% decrease in fluorescence intensity.Another 35% decrease with a 3 nm-blue shift was seen as the protein was saturated with Ca.The two-phase transitions of fluorescence emission from IAANS-labeled cTnC in response to Mg and Ca did not appear in fluorescence emission of 5-IAF-labeled cTnC.However,the Ca-induced conformational change in cTnC remained unchanged no matter which probe was used.Ca titration experiments showed that binding parameters derived from the fluorescence emission of the two probes were comparable.CONCLUSION:5-IAF is an appropriate probe that can be used to study the interaction between cardiac troponin C and other contractile regulatory proteins.
3.Application value of combined detection of virus nucleic acid and antibody in HFMD

Objective To investigate the application value of combined detection of enterovirus 71 type(EV71) nucleic acid and antibody in diagnosis of hand-foot-mouth disease(HFMD).Methods From Jan.to Nov.2015,a total of 180 children with suspected HFMD were divided into nucleic acid detection group,antibody detection group and combined detection group,with 36 subjects for each group.Viral nucleic acid was detected by using reverse transcription polymerase chain reaction.Antibody was detected by using enzyme linked immunosorbent assay.All detected results were analyzed.Results The positive rate of nucleic acid detection group was the highest(80.55%),while that of antibody detection group was the lowest(47.22%).Among various symptoms,the positive rates of nucleic acid,antibody and combined detection in subjects with skin rash were the highest.Conclusion Combined detection of EV71 nucleic acid and antibody could avoid the false positive result of nucleic acid detection and false negative result of antibody detection,thus could be helpful for increasing diagnostic accuracy rate of HFMD.
4.Determination of 2-Indole Ketone Derivative and Its Related Substances by High Performance Liquid Chromatography

Objective To establish an high performance liquid chromatography method for determination of 2-indole ketone derivative and its related substances. Methods Agilent Eclipse XDB-C18(250 mm×4.6 mm,5 μm) column was adopted.The mobile phase was acetonitrile-water with gradient elution mode at a flow rate of 1. 0 mL?min-1; the column temperature was 35 ℃;the injection volume was 20 μL and the detection wavelength was set at 257 nm. Results 2-indole ketone derivative ID and related substances could be well separated. The 2-indole ketone derivative had good linear correlation ( r=0.999 4) within the range of 40-300 μg?mL-1 . It had a good precision ( RSD<1%) . The limit of detection was 8 ng. Conclusion The method is accurate,simple,sensitive and selective,which can be used for the quality control of 2-indole ketone derivative and related substances.
5.Nonpharmacological therapy of gastroesophageal reflux in preterm infants

Gastroesophageal reflux (GER) was very common among preterm infants, which could cause several complications and severely affects the prognosis. The standardization of diagnosis and treatment of gastroesophageal reflux was very important for preterm infants. Due to less adverse reactions, nonpharmacological therapy was the most advisable choice in preterm infants with GER. In this paper, nonpharmacological therapies including body positioning, feeding frequency and modality, changing food patterns, gastric tube, and nonnutritive sucking were reviewed. Body positioning was the safest and most effective nonpharmacological therapy. Other nonpharmacological strategies such as changes of feeding frequency and modality might also be useful. The role of some strategies was still controversial, thereby needing to be further investigated. Selection of what kind of specific nonpharmacological therapy should be based on the specific conditions.
6.Determination of Nine Residual Solvents in Grape Seed Extract by Headspace Gas Chromatography

Objective To establish a headspace gas chromatography method for determination of nine residual solvents in grape seed extract. Methods The residual solvents in grape seed extract were separated on DB-WAX column (30 m× 0.45 mm, 0.85 μm) with an FID detector; the injector temperature and the detector temperature was set at 220 ℃ and 250 ℃ , respectively; the chapiter pressure was 27.58 kPa; the containers of headspace injector were in equilibrium for 30 min at 80 ℃ ;the N, N-dimethylformamide was used as the solvent. Results The detected solvents were well separated. Good linear relationship of the benzene and ethanol was obtained within the range of 0.04-0.3,100-750 μg?mL-1 , respectively. Good linear relationships of the hexane, o-xylene, m-xylene, p-xylene, styrene, toluene and 1,2-diethyl benzene solvents were obtained within the range of 0.4-3.0 μg?mL-1 (r≥0.995 8), respectively. The average recoveries (n = 3) of the nine solvents were 96.35%,97.08%,97.31%,89.93%,92.35%,90.65%,88.56%,93.84%,86.51% and the RSDs were 4.38%,2.16%,3.49%, 4.19%,4.80%,4. 83%,4. 70%,5. 00%,4. 39%, respectively. Conclusion The established method is simple, rapid and accurate, which can be used for the determination of residual solvents in grape seed extract and can simultaneously provide scientific basic for quality control.
7.Effect of IgG gene silencing by RNA interference on radiosensitivity of prostate cancer PC3 cells.

OBJECTIVETo investigate the effect of RNA interference of IgG gene on the radiosensitivity of the human prostate cancer PC3 cell line.
METHODSPC3 cells were trasnfected via lipofectamine by the shRNA vector FCGR1AshRNA targeting the Fc segment of IgG, using NCshRNA as the negative control. Q-PCR and Western blotting were used to analyze the expression of IgG in the trasnfected cells. The cells were then exposed to ⁶⁰Co γ ray at 0, 2, 4, 6, 8, 10 Gy, and the cell proliferation was evaluated by MTS and the cells apoptosis estimated by flow cytometry at 12, 24 and 48 h.
RESULTSMTS assay showed that ⁶⁰Co γ ray significantly inhibited the proliferation of PC3 cells transfected with FCGR1AshRNA as compared with NCshRNA-transfected and blank control cells (P<0.05). Flow cytometry showed that the cell apoptosis rate was significantly higher in FCGR1AshRNA group than in NCshRNA and blank control groups at 48 h after γ ray exposure (P<0.05). At 12, 24 and 48 h after 6 Gy radiation, the cells in FCGR1AshRNA group showed a significantly lowered proliferation rate and an increased apoptosis rate (P<0.05).
CONCLUSIONThe shRNA targeting IgG gene can significantly enhance the sensitivity of PC3 cells to radiation. The combination of RNA interference targeting IgG gene with radiotherapy may be more effective in the treatment of prostate cancer.

8. Effect of mindfulness trait factors on sleep quality of college students: the mediating role of cognitive emotion regulation strategies and anxiety

Objective:
To explore the effect of mindfulness traits on sleep quality of college students, and to investigate the mediating mechanism of cognitive emotion regulation and anxiety for mindfulness trait factors, and sleep quality in college students.
Methods:
Totally 397 college students from five colleges were investigated using Five Facet Mindfulness Questionnaire, Cognitive Emotion Regulation Questionnaire, Self-Rating Anxiety Scale and Pittsburgh Sleep Quality Index.SPSS 22.0 version and Bootstrap method were used to analysis data and test intermediate effect.
Results:
(1)The total score of mindfulness trait and the score of describe, act with awareness and non-judge factors in mindfulness traits were(118.44±9.38), (24.82±3.87), (27.41±5.53) and (25.07±4.60), respectively.The total score of sleep quality was (5.57±2.62), and it was negatively related to the above three factors (
9.Expression of OCT4 protein in bladder cancer and its clinicopathological implications.

OBJECTIVETo study the expression of OCT4 protein in bladder cancer and its correlation to the clinicopathologic features and prognosis of bladder cancer.
METHODSOCT4 mRNA and protein expression was detected in 5 bladder cancer cell lines (RT-4, Tcc-Sup, KK47, T24, and 5637) and 1 normal bladder cell lines by real-time PCR and Western blotting, respectively. Immunohistochemical analysis was used to detect the expression of OCT4 protein in 46 bladder cancer samples.
RESULTSAll the 5 bladder cancer cell lines expressed detectable levels of OCT4 mRNA and proteins, whereas the normal bladder cell line SV-HUC-1 was negative for OCT4 expression. The clinical bladder cancer tissues showed a high positivity rate of OCT4 expression (76.1%), which was not detected in normal bladder tissues. Specific OCT-4 signals were localized mainly in the nuclei of the cancer cells. The expression rate of OCT4 protein was significantly higher in bladder cancer tissue than in normal bladder epithelium (P<0.05), and showed a positive correlation to the grade of tumor differentiation and metastasis (P<0.05) but not to the patients' age, gender or TNM stage.
CONCLUSIONOCT4 protein expression is associated with tumor differentiation and metastasis in bladder cancer and may play an important role in the early diagnosis and prognostic evaluation of bladder cancer.

10.Qualitative studies on experience of the primary malignant bone tumors caregivers:a Meta-synthesis

Objective To systematically evaluate the qualitative studies on the care experience of caregivers for primary malignant bone tumors patients,in order to provide references for the construction of bone tumor support care system.Methods The Cochrane Library,PubMed,Embase,CNKI,Wanfang Database,VIP database,and China Biomedical Literature Database were searched by computer to collect qualitative studies on the care experience of caregivers of malignant bone tumors patients from the establishment of the databases to November 2022.The quality of the literature was evaluated using the Joanna Briggs Institute(JBI)Quality Evaluation Criteria for Quality Research in Evidence-Based Health Care Centers(2016),and the results were integrated by a pooled integration approach.Results A total of 12 studies were included;48 themes were extracted and summarized into 9 categories,which were combined into 3 integrated results.Integration result 1 is obvious physical and mental disturbance.Integration result 2 is multiple role maladaptation.Integration result 3 is positive growth after adjustment.Conclusion Caregivers of patients with malignant bone tumors have serious physical and mental burden and are eager for multiple support.It is suggested that medical staff pay attention to the multi-dimensional needs of patients,formulate personalized support strategies,help caregivers adapt and transform their roles,and promote the post-traumatic growth of caregivers.
