1.The influence of different doses of rhEPO to the expression of new BPD rat lung eNOS and the Bcl-2 protein

Objective To observe changes of the expression of Bcl‐2 and endothelial nitric oxide synthase (eNOS) in lung tissue ,which was used the indifferent treatment doses of recombinant human erythropoietin (rhEPO) of newborn SD model of a new type of bronchial pulmonary dysplasia .To explore its effect on the protection of the new type of bronchial pulmonary dysplasia (BPD) ,and find out the appropriate drug delivery time and dosage .Methods We used the things lipopolysaccharide (LPS) and continuous high oxygen preparation to make the new model of the BPD .We made random normal newborn mice 18 as normal air group ,and then 90 newborn baby mice model of random points normal saline group (group A) ,rhEPO800 group (group B);rhEPO 1000 group (group C) ,rhEPO1200 group (group D) ,rhEPO1400 group (group E) ,in experiment 1 ,7 ,and 14 days ,we were ran‐domly selected 6 executed only lung tissue under liquid nitrogen .Using RT‐PCR and Western blot method to detect the Bcl‐2 and eNOS protein and mRNA expression in lung tissue .Results Compared with the normal air group ,the expression of protein and mRNA of Bcl‐2 in group A was decreased ,while the expression of eNOS was increased .Compared with group B ,C ,D ,E ,with the increase of rhEPO concentration ,the expression of Bcl‐2 protein and mRNA in group A gradually increased ,while the expression of eNOS gradually decreased ,and the expression of the group E was the most obvious ,and the difference was statistically significant (P<0 .05) .Conclusion It is prompted that rhEPO has some treatment function to the new BPD ,while the dose of 1 400 IU/kg of 14 days have the best function .
2.Analysis of 75 cases of suspected type A influenza results by real-time fluorescent quantitative PCR test

Objective To analyze the detection results of 75 suspected type A influenza cases and to understand the distribution situation of influenza patients to provide the guidance for its early prevention and control .Methods The nasopharyngeal swabs specimens from 75 patients with suspected type A influenza were detected for understanding the viral infection situation by real‐time fluorescent quantitative PCR .Results In the specimens of 75 suspected patients ,13 cases with type A influenza virus were detected out ,in which 9 cases were the H1N1 subtype ,mean age > 50 years old .The temporal distribution was 3 cases in March 2013 ,1 case in April ,7 cases in January 2014 and 2 cases in February .No H7N9 subtype was detected out .Conclusion Real‐time fluores‐cent quantitative PCR has the characteristics of rapidness and high accuracy .The winter is the high onset season of flu ,elderly pa‐tients are susceptible to influenza virus .The monitoring and early treatment should be strengthened .
3.Effect of compound glycyrrhizin on airway inflammation in asthmatic rats

Objective To investigate the effect and mechanism of compound glycyrrhizin on airway inflammation in asthmatic rats. Methods Forty wistar rats were randomly divided into health group (group Ⅰ), asthma group(group Ⅱ), treating group (group Ⅲ) and treating control group (group Ⅳ) (n=10 in each group). Group Ⅱ, group Ⅲ and group Ⅳ received intraperitoneal injection of ovalbumin (OVA) and aluminum hydroxide gel for sensitization and OVA aerosol by ultrasonic nebulization for challenge, in addition, group Ⅲ re-ceived intraperioneal injection of 200 μg of compound glycyrrhizin 1 hour before OVA aerosol challenging every time, and group Ⅳ were in-jected with normal saline instead of compound glycyrrhizin. Group Ⅰ were injected and nebulizated with normal saline instead of OVA and alu-minlum hydroxide. All rats were sacrificed at the 6th hour after the last challenging, venous blood and bronchoalveolar lavage fluid(BALF) were collected, the IgE in venous blood were determined and the concentration of IL-4, IL-5, IL-13 and IFN-γ in BALF were detected, the cells in BALF were counted and detached. Results The concentration of IgE, IL-4, IL-5, IL-13 and the count of neutrophilic granulocytcs, lymphocytes, cosinophils in group Ⅱ were significantly higher than those in group Ⅰ and group Ⅲ(P<0.01), and the concentration of IFN-γ in group Ⅰ and group Ⅳ were significantly lower than those in group Ⅰ and group Ⅲ(P <0.01). There was no significant difference be-tween group Ⅱ and group Ⅳ(P >0. 05). The concentration of IgE, IL-4, IL-5, IL-13 and the count of neutrophilic granulocytes, lympho-cytes, cosinophils in group Ⅲ were higher than those in group Ⅰ, and the concentration of IFN-γ and the count of mononucleat macroplile cells in group Ⅲ were lower than those in group Ⅰ, but there were no significant difference(P>0.05). Conclusion Compound giycyrrhiz-in alleviated the airway inflammation and reduced the airway hyperresponsiveness in asthmatic rats, which mechanism may be related to regu-lating the balance of Th1/Th2 cytokines, suppressing the immune ability of Th2 and promoting the immune ability of Th1.
4.Efficacy and short-term safety of percutaneous coronary intervention in elderly patients with acute myocar-dial infarction

Objective Percutaneous coronary intervention( PCI) is effective in improving the ischemia and prognosis of pa-tients with acute myocardial infarction ( AMI) to reduce the short-term mortality.However, little research has been done on PCI in eld-erly AMI patients.The study aimed to evaluate the efficacy and the safety of percutaneous coronary intervention in elderly AMI patients (≥75 years old) . Methods 213 AMI patients who underwent emergency PCI in Jingling Hospital from January 2012 to December 2013 were divided into 2 groups:elderly group (≥75 years old, n=57) and non-elderly group (<75years old,n=156).Retrospec-tive analysis were made on the clinical data and the coronary intervention features of the patients. Results There were more patients having dyspnea, fatigue and other heart failure symptoms at the onset of first-break AMI in elderly group than in non-elderly group (21.1%vs 3.2%,P<0.0).More women (47.4% vs 16.7%,P<0.01) and more patients with hypertention or diabetes mellitus were found in elderly group.The procedure success rates with TIMI-3 flow grade of post-PCI in both groups were very high (100%). Compared with non-elderly group, the occurrence of the procedure-related complications (3.5%vs 2.6%,P=NS) and major adverse cardiac event rates (8.8%vs 6.4%,P=NS) and in-hospital mortality (5.3%vs 2.6%,P=NS) showed no significant difference.Conclusion There are more atypical clinical symptoms in elderly AMI patients. The emergency PCI in elderly AMI patients can effectively make artery unimpeded with high successful rate, few com-plications and a favorable short-term prognosis.

6.Effects of aminoguanidine on endotoxin-treated rat aorta and pulmonary artery in vitro

Objective: To investigate the effects of selective NOS inhibitor aminoguanidine (AG) on endotoxintreated rat aorta (AO) and pulmonary artery (PA) in vitro. Method: The reaction to noradrenaline (NE) (10~(-9)-10~(-4)mmol/L) was measured in the vessels with complete endothelium, then the vessels were divided into 8 groups after incubated in LPS(300ng/ml)for 4 hours. Each group was incubated with AG(100?mol/L)or L-NAME (300?mol/L)for 20, or 60min, respectively. Result: Both AO and PA showed hyporeaetivity to NE after incubated in LPS. and the reactivity of AO and PA to L-NAME and AG increased significantly after incubated in them for 60min or 20min. The sen sitivities of AO to AG,AO and PA to L-NAME increased greatly. AO had a higher reactivity to AG in 60min than in 20min. L-NAME caused earlier and greater contraction than AG in both 60 min and 20 min groups. Conclusion: LNAME and AG can both increase the decreased reactivities of AO and PA to NE. AG only increases the AO sensitivity.
7.Biological effects of down-regulating of TIZ gene on ovarian cancer cells in vitro

Objective To construct TIZ gene RNA interference plasmid,as well as study the biological effects of down-regulating of TIZ gene on ovarian cancer cells.Methods According to the mRNA sequence of TIZ on GenBank,3 pairs of small interference RNA (siRNA) for TIZ gene expression were designed and introduced into SKOV3 cells by liposome transfection reagent; the real-time quantitative PCR (QRT-PCR) was used to detect the interference efficiency and selected the most efficiency siRNA segment;pGPU6/GFP/Neo carrier was used to construct pGPU6/GFP/Neo-siRNA-TIZ-573 restructured interference plasmid,introduced into SKOV3 ccll (sh-TIZ-573/SKOV3).The cell growth curves were made by methyl thiazolyl tetrazolium (MTT) method.The clonogenicity efficiency was observed by clony formation test.The cell cycles were measured by flow cytometry (FCM).The ability of invasion,metastasis and adhesion of ovarian cancer cells were detected by the matrigel invasion assay,transwell migration assay and adhesion assay,respectively.Results QRT-PCR results showed that ovarian cancer SKOV3 cells which transfected by recombinant plasmids pGPU6/GFP/Neo-siRNA-TIZ-573 could down-express TIZ gene.The cell growth curves,FCM and clony formation tests showed that the growth and proliferation of sh-TIZ-573/SKOV3 cells were significantly speeded up compared with the control cells (P < 0.01).There was no significant difference in cell invasion,migration and adhesion between sh-TIZ-573/SKOV3 cells [(48.5 ± 1.7) %,(53.6 ±3.4)%,(64.9 ±5.0)%,respectively] and the control cells (P >0.05).Conclusion The growth of ovarian cancer cells could be speeded up by down-expressing of TIZ gene,which TIZ gene may be play a biological role as a tumor suppressor gene.
9.Effect of amplitude of respiratory movements on dose distribution of static intensity-modulated radiotherapy

Objective To study the effect of amplitude of respiratory movements on dose distribution of static intensity-modulated radiotherapy (IMRT),and to provide a basis for dose correction against respiratory movements.Methods A two-dimensional matrix driven by the QUASAR program-controlled respiratory movement instrument was used to simulate human respiratory movements in the head/foot direction.The dose distribution was evaluated on the isocenter plane with different amplitudes of respiratory movements.The Verisoft software and absolute dose analysis were used to analyze dose distribution,percentage errors of absolute dose,and passing rates of radiation field for both collected data and planned dose distribution.Results In spite of little effect on dose distribution in target volume,respiratory movements increased the dose outside the marginal target volume along the movement direction.When the respiratory amplitude was lower than 6 mm,the passing rate of γdistribution met the requirement of clinical dose verification;when the respiratory amplitude was larger than 8 mm,the passing rate of γ distribution was reduced with the increasing respiratory amplitude and fell below the clinical standard.Conclusions Respiratory movements have a blurring effect on static IMRT.It is possible to introduce some compensation methods to static IMRT in the treatment of tumor with periodic respiratory movements.The normal tissue at the edge of target volume along the respiratory movement direction is exposed to a higher radiation dose than expected.Therefore,the radiation dose for the normal tissue around target volume should be made as low as possible in the radiotherapy plan.For the patients with relatively large respiratory amplitude,action should be taken to lower respiratory amplitude and elevate the radiation dose in target volume.
10.Analysis on detection results of postoperative mixed infection of multi-drug resistant bacterial strains among traumatic patients in Zaoyang area

Objective To investigate the pathogenic bacterial distribution and drug resistance situation of multi‐drug resistant bacterial strain mixed infection among postoperative traumatic patients in Zaoyang area to provide a reference for clinical treatment . Methods A total of 71 traumatic cases of multi‐drug resistant bacterial strain mixed nosocomial infection were selected and per‐formed the bacterial culture and drug resistance analysis according to the samples taking from different infection sites .The the re‐sults were statistically analyzed .Results Among 71 cases of multi‐drug resistant strains mixed infection ,205 multi‐drug resistant strains of bacteria were isolated .Among them ,119 (58 .05% ) strains were Gram‐negative bacteria ,Gram‐positive bacteria was in 83 strians(40 .49% ) and fungus was in 3 strains(1 .46% ) .The resistance of Escherichia coli ,Klebsiella pneumoniae ,Enterobacter and Serratia to more than 10 kinds of antibacterial drugs was more than 50% ,which to ampicillin ,cephalothin ,cefuroxime ,azlocillin ,ce‐fotaxime cefuroxime ,azlocillin ,cefotaxime and cefoperazone was more than 80% ,while which to imipenem and amikacin was only 5 .88% and 28 .57% respectively .The resistance rates of Gram‐positive bacteria represented by Staphylococcus aureus ,Staphylococ‐cus epidermidis ,Staphylococcus haemolyticus and Streptococcus pneumoniae to penicillin G ,oxacillin and ampicillin generally reached 98 .80% ,97 .59% ,and 95 .18% respectively .No Gram‐positive bacteria was resistant to vancomycin ,in addition the resist‐ance to rifampin and chloramphenicol also was only 32 .53% and 34 .94% respectively .Conclusion In early stage of infection ,it is unable to determine the types of pathogenic bacteria and difficult to select sensitive antibacterial drugs ,under this occasion ,the com‐bination of imipenem and vancomycin is a good choice .
