2.An event-related functional MRI study on working memory impairment in children with primary nocturnal enuresis

Objective To assess the working memory and explore the activation of brain areas for children with primary nocturnal enuresis (PNE) with fMRI scan. Methods Twenty three right-handed children with PNE and 20 age-matched right-handed healthy children as the controls were recruited.Intelligence tests were performed by means of Wechsler Young Children Scales of Intelligence (C-WISC) in children with PNE and normal controls. The full intelligence quotient (FIQ), verbal IQ (VIQ),performances IQ (PIQ) and the memory/caution (M/C) factor of PNE children and the controls were measured. After Intelligence tests, an enent-related fMRI scan was performed using the categorial N-Back working memory task. Percent of correct responses (PCR) and mean reaction time to correct response (mRT) were recorded and analyzed by the student t test. The fMRI data were analyzed using Statistical Parametric Mapping 2 (SPM2), the differences in activation were compared between two groups. Results The data of 15 PNE children and 15 healthy children were evaluated. The FIQ, VIQ and PIQ in PNE group were in a normal range and no statistical significance with the control group ( P > 0. 05). M/C factor in the PNE group(90.4 ±10.2)was significantly lower than that in the control group (99. 6 ± 11.9) (t =2. 260,P < 0. 05). In the N-Back test, PNE children had significantly less PCR [(72.7±6.3 ) % vs. ( 86. 3 ±6. 7) %, t = 5. 727,P < 0. 01] and longer mRT [ (625. 8 ± 72.5) ms vs. (534. 8 ± 63. 3 ) ms, t = 3. 684,P < 0.01] than the healthy controls. The activation regions of PNE patients and healthy children were mainly in the dorsal right frontal lobe, right parietal lobe, left temporal lobe gyrus fusiformis and bilateral cerebellum posterior lobe. The activation level in left posterior cerebellar lobe in PNE children was significant lower than that in healthy controls (P<0.01). Conclusion The children with PNE have deficits in working memory which might be associated with the dysfunction of the left cerebellum.
3.Correlations study between grip strength, lung functions, six-minute walk test and left ventricular ejection fraction in patients with coronary artery disease

Objective:To investigate the clinical significance of grip strength, three simple lung functions[functional volume capacity(FVC), forced expiratory volume in the first second(FEV1)and peak expiratory flow(PEF)], 6-minute walking distance test(6MWD)and left ventricular ejection fraction(LVEF)for assessing overall integrative function in elderly versus non-elderly patients with coronary artery disease(CAD).Methods:A total of 35 hospitalized CAD patients aged 70.4±10.6 years from January 2017 to December 2018 were retrospectively enrolled.Patients were divided into the non-elderly group(<65 years, n=8)and the elderly group(≥65 years, n=27). FVC, FEV1 and PEF were tested by a portable lung function machine.The grip strength, 6MWD, three simple lung functions(FVC, FEV1, PEF), LVEF and echocardiography were detected after admission.The above indexes were re-examined in outpatient clinic at 3 months after discharge.The results of above indexes were compared at admission versus at three months after discharge.The consistency and correlation between FVC, FEV1, PEF, 6MWD, grip strength and LVEF were analyzed.Results:The detected results of FVC, FEV1, PEF, 6MWD, left-hand grip strength, right-hand grip strength and LVEF showed no significant difference at admission versus at 3 months after discharge( P>0.05)in a total of 35 hospitalized CAD patients.Pearson bivariate correlation analysis showed that FVC, FEV1, PEF, left-hand and right-hand grip strength had pairwise correlation between them( P<0.01).6MWD had no correlation with LVEF.And LVEF was related with FVC and FEV( P<0.01), but not related with PEF, 6MWD and grip strength( P>0.05). The detective values of FVC, FEV1, PEF, 6MWD, left-hand and right-hand grip strength were lower in the elderly group than in the non-elderly group[(2.11±0.66 )L vs.(2.88±0.55) L, (1.74±0.46 )L vs.(2.62±0.49 )L, (5.50±1.79 )m/s vs.(8.22±1.59) m/s, (332.07±115.58)m vs.(446.14±99.81 )m, (25.14±7.87) vs.(35.15±8.30), (27.37±8.39 )kg vs.(38.37±10.20)kg, P<0.01]. LVEF had no significant difference between the two age groups. Conclusions:FVC, FEV1, PEF, 6MWD and grip strength are lower in elderly group than in non-elderly group.A comprehensive set of tests of grip strength, three simple lung function and 6MWT is helpful to evaluate the overall integrative function in elderly CHD patients.
4.The dendritic cell subsets function and the cytokines levels in plasma change in different period in patients with HBV infection.

OBJECTIVEWe studied the function of dendritic cells subsets and the cytokines levels in plasma in patients with different periods of returning (immune tolerance, hepatitis active and non-replicating period) and aimed to explore the possible reasons for HBV chronic infection.
METHODSThirty HBV infected patients in different stages of infection were enrolled and divided into three groups: the immune tolerance group (10 cases), the hepatitis active group (10 cases), and the non-replicating group (10 cases). Ten healthy people were enrolled and served as controls. Blood from the patients and controls were collected and the dendritic cells subsets function (The cytokine levels in different groups) were analyzed using statistical method.
RESULTSThe total IL-12 output and single nucleus IL-12 output of the total HBV infected patients were lower than that of the healthy control group (P value less than 0.01). The total IFNa output and single nucleus IFNa output of the total HBV infected patients had no significant difference between the total HBV infected patients and the healthy controls (P value more than 0.05). The total IL-12 output of the healthy control group was higher than that in others 3 groups of the HBV infected patients. (Z = -3.039, -2.967 ,-2.949, P value less than 0.01) and the single nucleus IL-12 output of the healthy control group was also higher than that in others 3 groups of the HBV infected patients. (Z =-3.027, -2.671 , -2.863, P value less than 0.01) . The total IFNa output and the single nucleus IFNa output of the healthy control group was higher than that in the hepatitis active group of HBV infected patients (Z = -3.016, -3.176, P value less than 0.01). While the plasma IFNa cytokine levels in the 3 HBV infection groups were higher than in the healthy control group (Z = -2.967, -2.896, -3.054, P value less than 0.01).
CONCLUSIONDifference existed between the function of dendritic cells subsets and the IFNa levels in different returning periods of HBV infected patients. The function of dendritic cells subsets has no significant difference in HBV patients with different periods of returning. The flawed function of dendritic cells subsets and the abnormal IFNa level may be one of the reasons for chronic HBV infection.

5.Investigation of oral health status in 3 516 undergraduate university students.

OBJECTIVETo investigate the oral health status of undergraduate university students and to guide their oral hygiene behaviors.
METHODS3516 college students in Northwest University for Nationalities were investigated in this study according to the criterion issued by World Health Organization (WHO) on the basic methods of oral health investigation in 1997.
RESULTSThe caries prevalence rate among the students investigated was 47.87%. The average decayed teeth were 1.29. The obturation rate of caries was 20.42%. The students came from city showed significant higher caries rate than those from countryside, and so did the female students compared with the male students. Dental calculus rate was 79.78%. The dental segments related were 3.75. The worst situation was among male students and students from countryside. The examined rates were 53.30% of crowded incisor, 42.58% of maximum upper-incisor anomaly, 50.60% of the maximum lower-incisor anomaly and 31.09% of molar malposition, respectively. More rates of dentofacial anomalies showed no differences of significance between different area and gender.
CONCLUSIONIn general, the oral health status of the college students are not ideal. It is necessary to advocate early examination, early prevention and early treatment among the students in university.

6.Comparison on sensitivity and specificity of serum miR-122 as biomarkers of drug-induced liver injury

OBJECTIVE To make a comparison of sensitivity and specificity between serum miR-122 and traditional biomarkers of drug-induced liver injury. METHODS Acetaminophen (APAP, 1250 mg · kg-1, ig), α-naphthylisothiocyanate (ANIT, 150 mg · kg-1, ig), methionine-choline deficient diet (MCDD, free feeding) and carbon tetrachloride (CCl4, 50%, ip) were used to establish hepatocellular injury, cholestasis, steatosis and fibrosis models, respectively, which were used to evaluate the sensitivity of serum miR-122 as a biomarker of drug-induced liver injury, when compared with the traditional biomarkers. Isoprenaline hydrochloride (IH) and gentamicin (GM) were used to establish myocardial and renal injury models, respectively, which were used to evaluate the specificity of serum miR-122, when compared with the tradi-tional biomarkers. Serum and liver tissues were collected at different time points during the study. Tradi-tional biomarkers such as glutamic-pyruvic transaminase (GPT), glutamic-oxaloacetic transaminase (GOT) and total bilirubin (TBIL) were measured with an automatic biochemistry analyzer. MiR-122 was detected with real- time quantitative PCR. Histopathological examination with HE staining was performed for liver tissues. RESULTS Serum miR-122 increased significantly earlier [miR-122 was signifi-cantly increased (>2 fold) at 1.5 h, 3 h, 2 weeks and 4 weeks after treatment in the four models respec-tively, while no significant increase (<2 fold) was observed for GPT, GOT and TBIL at 6 h, 12 h, 3 weeks and 6 weeks after treatment in the four models respectively] and more signficantly (the maximum fold change for miR-122 was 235.8, 202.7, 73.8 and 600.3 for the four models, respectively. For the GPT, the maximum fold change was 9.5, 3.9, 2.5 and 6.6, respectively; 6.0, 2.4, 1.4 and 3.5 respectively for GOT; 2.6, 2.3, 1.2 and 1.8 respectively for TBIL) than that of traditional biomarkers in hepatocellular injury, cholestasis, steatosis and fibrosis models, when compared with the control group. In the myocar-dial injury model, a significant increase of GOT was observed after IH treatment (2.1 fold), while no change was observed for serum miR-122. In the renal injury model, no false positive results were observed for serum miR-122. CONCLUSION Serum miR-122 can be used as a biomarker of drug-induced liver injury and serum miR-122 is more sensitive and specific than traditional biomarkers (such as GPT, GOT and TBIL).
7.The Effect of Contusion and Exhaustive Exercise on Satellite Cells Activation and Pax7/CBF1/DAPT Contents in Skeletal Muscles of Rats

Objective To study the effect of contusion and exhaustive exercise on satellite cells' activation and Pax7/CBF1/DAPT contents in skeletal muscles of rats,and reveal the repair mechanism of the skeletal muscle injury.Methods Twenty-four seven-week-old male Sprague-Dawley(SD)rats were randomly divided into a control(C)group,an immediately after exhaustive exercise(E0)group,a 24 hours after exhaustive exercise(E24)group and a 48 hours after exhaustive exercise(E48)group,each of 6.Other 18 SD rats were randomly divided into 3 groups,6 rats in each group:an immediately after contusion group(D0),a 24h post-contusion group(D24)and a 48h post-contusion group(D48).All groups were killed at different time points after exhaustive exercise and the contusion respectively while the control group was at resting state,and their serum was extracted.The right gastrocnemius muscles were resected and divided into 2 parts:one was used immediately for culturing satellite cells,while the other was stored in the bridge at-80℃.All the serum and gastrocnemius muscles were tested for Pax7,CBF1 and DAPT contents using the enzyme-linked immunosorbent assay(ELISA).Moreover,three-day-old newborn rats were also slaughtered for the above experiment.Results The results of culture in vitro showed that one day after the culture began a small amount of spindle satellite cells could be seen in the newborn rats,but the satellite cells of other groups were growing slowly.However,on the third day and the fifth day,the spindle satellite cells of each group began to proliferate in quantity and reached the peak.Then all the satellite cells had a good proliferation until the seventh day after serial passage,and some patial fusions to microtubules occurred.The biggest cell number and proliferation rate was in newborn rats,but no skeletal muscle satellite cell was found in the control group.At the same time,the number and the proliferation rate of skeletal muscle satellite cells in contusion groups were significantly higher than the exhaustive exercise groups.The results of Elisa showed that the contents of CBF1 and Pax7 in skeletal muscles in exhaustive groups and contusion groups,were significantly upregulated,compared to the control group(P<0.05).Amomg them,the CBF1 content in group D0,together with the Pax7 content in group D24 and D48,was significantly higher than that of the other groups(P<0.05).On the other hand,the content of DAPT in skeletal muscles of exhaustive groups and contusion groups,compared to the control group,was significantly downregulated (P<0.05).To be more specific,the value of group D0 was significantly lower than that of the other groups (P<0.05).However,there was no significant difference in it among group E0,E24 and E48.Conclusions Contusion and exhaustive exercise can activate the skeletal muscle satellite cells to proliferate and differentiate.The number of activated skeletal muscle satellite cells in the contusion groups is significantly larger than that of exhaustive groups maybe due to the stronger stimulating by contusion.Both contusion and exhaustive exercise increase the contents of Pax7 and CBF1 in skeletal muscles,but decrease that of DAPT.The Pax7 and CBF1 may play a positive regulating role in terms of activating skeletal satellite cells and repairing skeletal muscle injury,while DAPT may play a negative regulating role.
8.Screening bioactive compounds inhibiting influenza virus from isatidis radix by ultrafiltration mass spectrometry.

In vitro neuraminidase inhibition assays and ultrafiltration liquid chromatography with diodearray detector coupled to time of flight mass spectrometer (UPLC-DAD-TOF-MS) were combined to screen bioactive compounds inhibiting neuraminidase from Isatidis Radix. By comparing the compounds from Isatidis Radix before and after ultrafiltration, we found that arginine, goitrin and adenosinea can bind with neuraminidase, and the binding degree of the three compounds were (36.23 +/- 1.12)%, (32.54 +/- 1.02)% and (9.38 +/- 0.47)%, respectively. The IC50 of arginine and goitrin were (1.16 +/- 0.02), (1.20 +/- 0.02) g x L(-1), respectively. While the IC50 of adenosinea was higher than 500 g x L(-1). The results showed that arginine and goitrin might be the main compounds with antiviral activity of Isatidis Radix. This study may provide a useful method for the screening of bioactive compounds and quality control of Isatidis Radix.

9.Effect of different tooth preparation designs on the marginal and internal fit discrepancies of cobalt-chromium crowns produced by computer-aided designing and selective laser melting processes

PURPOSE:
To evaluate the impact of five different tooth preparation designs on the marginal and internal fit discrepancies of cobalt-chromium (CoCr) crowns produced by computer-aided designing (CAD) and selective laser melting (SLM) processes.
MATERIALS AND METHODS:
Five preparation data were constructed, after which design crowns were obtained. Actual crowns were fabricated using an SLM process. After the data of actual crowns were obtained with structural light scanning, intaglio surfaces of the design crown and actual crown were virtually superimposed on the preparation. The fit-discrepancies were displayed with colors, while the root means square was calculated and analyzed with one-way analysis of variance (ANOVA), Tukey’s test or Kruskal-Wallis test (α =.05).
RESULTS:
The marginal or internal color-coded images in the five design groups were not identical. The shoulder-lip and sharp line angle groups in the CAD or SLM process had larger marginal or internal fit discrepancies compared to other groups (P < .05). In the CAD process, the mean marginal and internal fit discrepancies were 10.0 to 24.2 µm and 29.6 to 31.4 µm, respectively. After the CAD and SLM processes, the mean marginal and internal fit discrepancies were 18.4 to 40.9 µm and 39.1 to 47.1 µm, respectively. The SLM process itself resulted in a positive increase of the marginal (6.0 – 16.7 µm) and internal (9.0 – 15.7 µm) fit discrepancies.
CONCLUSION
The CAD and SLM processes affected the fit of CoCr crowns and varied based on the preparation designs. Typically, the shoulder-lip and sharp line angle designs had a more significant effect on crown fit. However, the differences between the design groups were relatively small, especially when compared to fit discrepancies observed clinically.
10.Genetic polymorphisms of MxA protein and eIF-2a-reg2 and their responses to interferon treatment in patients with chronic hepatitis B.

OBJECTIVESTo identify the host single nucleotide polymorphisms (SNP) of myxovirus resistance A (MxA) protein and eukaryote initiation factor 2alfa regulatory region 2(eIF-2a-reg2) and to predict interferon (IFN) treatment responses in patients with chronic hepatitis B.
METHODSTwo hundred sixty-two patients with chronic hepatitis B (CHB) were treated with interferon alfa (IFN ) for 12 months. Six months later the therapeutic effectiveness was evaluated. All the patients had signed a formal consent form. The patients were grouped into a sustained response (SR) group and a non-sustained response (NSR) group according to their responses to the IFNa treatment. Single nucleotide polymorphisms of the antiviral protein MxA promoter -88,-123 and protein kinase(PKR) activated eIF-2a-reg2 sites were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and were compared with the responsiveness to IFN treatment of these CHB patients.
RESULTSAmong the 262 patients, 212 (80.9%) were non-sustained responders to IFNa and 50 (19.1%) were sustained responders. The rate of sustained responders with GT heterozygote at MxA promoter -88 was higher than that of the GG genotype (OR: 5.3, 95% CI: 2.46-11.43, P less than 0.01) and also higher than that of the TT genotype (OR: 4.1, 95% CI: 1.86-9.09, P less than 0.01). There were no statistically significant differences in IFN therapeutic effectiveness among the patients with different genotypes at MxA promoter -123, eIF-2a-reg2 and haplotypes made by MxA promoter -88 G/T, and -123 C/A alleles (P more than 0.05).
CONCLUSIONPatients with GT genotype at MxA promoter -88 responded well to IFN treatment. SNP as a potential marker could be used to predict IFN treatment responses of patients with chronic hepatitis B.
