1.ADVANCES ON THE PHARMACOLOGICAL EFFECTS AND CLINICAL APPLICATIONS OF FISH OIL PREPARATIONS

The pharmacological effects and clinical applications of fish oil preparations were reviewed. Fish oil is a natural material. Its excellent clinical tolerance and multiple pharmacological activities make it a pharmaceutical of broad prospects. Fish oil preparations have been used in the therapy and prevention of hyperlipidemia and atherosclerosis in diabetics. They have also been proved effective in improving symptoms of burns and abscesses. Additionally,fish oil preparations can replace non-steroidal anti-inflammatory drugs in the therapy of immune system disorders and have effects of slowering aging and improving the memory faculty of the aged. Applications of fish oil in organ transplantation have also been reported.
2.Application of Virtual Reality Training System in Rehabilitation Medicine Education

Objective To apply the virtual reality training in clinical education of rehabilitation therapy. Methods 26 undergraduate students majored in rehabilitation therapy were randomly divided into 2 groups during clinical education of physiotherapy. The experimental group (n=14) received virtual reality training first, and then with routine teaching, while the control group (n=12) learnt with routine teaching only. Self-assessment and physiotherapy test were conducted after training. Results The experimental group reported more interested, initiative and positive in training than those of the control group. The experimental group achieved higher scores in practical tests than the control group (P<0.05), while no significant difference in the theoretical examination between the groups. Conclusion Application of virtual reality training may improve the teaching of physiotherapy.
3.Construction of mouse PPAR? adenovirus vector and its expression in INS-1 cells

Objective To construct the recombinant adenovirus vector containing mouse PPAR?,and infect the INS-1 cells.Methods The pCDNA3.0-mPPAR? plasmid was digested with KpnⅠ and EcoRⅤ to obtain full-length coding sequence of mPPAR?,then the mPPAR? cDNA was ligated to the pAdtrack-CMV vector.The plasmid of pAdTrack-mPPAR? was linearized with PmeⅠ,and the fragment was reclaimed and transformed into BJ5183 which contains pAdeasy.After being screened,the extracted plasmid of positive bacteria linearized with PacⅠ was transfected into HEK293 cells with lipofectamine~(TM) 2000 and was identified by the green fluorescence protein(GFP).The harvested virus was amplified in HEK293 cells and infected INS-1 cells.After infection,the expression of PPAR? was proved by GFP expression and Western blotting.Results The recombinant adenovirus vector containing mouse PPAR? was constructed successfully and the titer was 1.5?10~(10) pfu/ml.The PPAR? expression in INS-1 cells infected by the virus was much higher than the controls.Conclusion The constructed recombinant adenovirus vector containing mouse PPAR? provides a potent tool to investigate its biological function in pancreatic islets and other tissues.
4.Data base establishment and data mining on regular pattern of TCM beauty prescription

ObjectiveTo explore the regular pattern of TCM beauty prescription.MethodsBased on the ACCESS database platform,we established TCM beauty prescription database and analyzed the information in detailed.ResultsThe total number of beauty prescription in the database was 1821.We focused on analyzing the frequency of medicine application in ancient beauty prescriptions,medicinal diet,acne,chloasma and freckles prescriptions.Statistics showed that dahurian angelica root,indian bread,medlar,honey,black sesame,baical skullcap root,and Chinese angelica were the most commonly used seven kinds of Chinese herbs.ConclusionThe main parts of Chinese medicines for making beauty were those herbs which can nourish yin and blood,supplement yang and qi; and the main parts of Chinese medicine for treating skin diseases were those herbs which can clear heat and detoxicate.
5.Effects of active component in Cichorii on lipid metabolism of rat with hypertriglyceridemia complicated by hyperuricemia and hyperglycemia

OBJECTIVE: To observe the effect of active component in Cichorii (N3) on lipid metabolism of rat with hypertriglyceridemia complicated by hyperuricemia and hyperglycemia. METHODS: Hypertriglyceridemia complicated by hyperuricemia and hyperglycemia was induced in rats by feeding a kind of feedstuff containing high fructose for 21 days. Then three different doses of N3 were administered continuously to the rats for 35 days. The effects of N3 on lipid metabolism were observed by determining the levels of serum triglyceride (TG), uric acid (UA), glucose (GLU) and free fatty acids (FFA) and the activities of fatty acid synthetase (FAS) and hepatic lipase (HL) from the liver and lipoprotein lipase (LPL) from the heart. RESULTS: Twenty-one days after feeding a high-fructose diet, the levels of serum UA, TG and GLU in the untreated group were significantly higher than those of the normal control group. Thirty-five days after administering N3, there was no statistical difference in serum TG and heart LPL activity between the untreated group and the normal control group, while liver FAS activity, serum FFA level and liver HL activity in the untreated group were significantly higher than those in the normal control group. Different doses of N3 treated rats all showed a decrease of TG, UA and GLU levels. N3 could decrease FAS activity and FFA level, and increase LPL and HL activities significantly. CONCLUSION: N3 in Cichorii may regulate the lipid metabolism of the rats with hypertriglyceridemia complicated by hyperuricemia and hyperglycemia by decreasing FAS activity and FFA level, and increasing LPL and HL activities.
6.Biological characteristics and hematopoietic support of bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus

Objective To explore the biological characteristics and karyotype of bone marrow-derived mesenchymal stem cells(MSCs)in patients with systemic lupus erythematosus(SLE)and hematopoietic sup- port of MSCs.Methods MSCs were isolated from bone marrow of 11 SLE patients and 6 healthy controls by density centrifugation and adhesive culture in vitro.The surface markers were detected by flow cytometry (FCM).The morphological changes of MSCs were observed in primary and passage cultures.The growth curves were assayed.The karyotype of MSCs was detected by blocking cellular mitosis with colchicines.The MSCs from SLE patients and healthy controls were infused to ICR mice after high-dose chemotherapy.The changes of peripheral blood counts of the mice were recorded.Results Approximately(6~9)?10~9 MSCs from SLE were obtained after 5 passages and their growth was slower than normal controls(P<0.01).Both groups were positive for CD29,CD44 and CD105,and negative for CD14,CD34,CD45 and HLA-DR.MSCs from SLE had a normal karyotype.MSCs infusions of the two groups were accompanied by no adverse event and the recovery of white blood cell,hemoglobin and platelet count was quicker when compared with the controls(P<0.05).Conclusion MSCs from SLE have demonstrated abnormalities in expansion in vitro.MSCs from SLE have a normal karyotype.Ex vivo MSCs infusion from SLE patients can support hematopoiesis as normal MSCs.
7.Elucidating hypoglycemic mechanism of Dendrobium nobile through auxiliary elucidation system for traditional Chinese medicine mechanism.

To build the Dendrobium nobile -T2DM network, and elucidate the molecular mechanism of D. nobile to type 2 diabetes (T2DM). Collect the chemical composition of D. nobile and the targets on T2DM by retrieving database and documents, build the network of D. nobile to T2DM using the entity grammar systems inference rules. The molecular mechanism of D. nobile to T2DM includes: (1) regulating lipid metabolism by lowering triglyceride; (2) reducing insulin resistance; (3) protecting islet cells; (4) promoting the glucose-dependent insulin tropic peptide (GIP) secretion; (5) inhibiting calcium channel. Under the guidance of network pharmacology, through entity grammar systems inference rules we elucidate the molecular mechanism of D. nobile to T2DM, and provide the basis for the further development of health care products based on D. nobile.

8.Association of polymorphism of V4 and T2 locus alleles in ADAM33 gene with asthma susceptibility

Objective To verify the association of a disintegrin and metalloproteinase domain 33 (ADAM33) polymorphisms in childhood asthma susceptibility and severity in patients with moderate and severe asthma.Methods A total of 144 controls and 110 asthmatic patients were recruited for this hospitalbased case-control study.Two polymorphic sites (V4,T2) were genotyped using the polymerase chain reaction-restriction fragment length polymorphism method.The gene-gene interactions were analyzed with the multifactor dimensionality reduction (MDR) software.Results The allele frequencies of three SNPs (V4,T2) of ADAM33 in childhood asthma group were significantly higher than control group (P < 0.01,OR =2.36 ~ 2.96,95% CI:1.56-1.78 ~ 3.56-4.94).For the SNPs V4,GC genotype frequencies of both moderate and severe groups were significantly higher from the control group (P < 0.05) ; the CC genotype frequencies in severe group were significantly higher than control group (P < 0.05) ; the genotype GA of T2 locus in severe group and AA genotype frequencies in moderate group were significantly higher than control group (P <0.01 ~0.05) ; there were no significant differences for both allele and genotype frequencies of S2 locus between the childhood asthma and control group (P > 0.05).By MDR analysis,the best interaction model was the four-factor model that the V4,T2 genotypes were the subgroup to predict asthma risk.Conclusions Our results highlight the role of ADAM33 as a susceptibility gene for childhood asthma,and the interactions among ADAM33 V4 and T2 are also associated with childhood asthma.
