1.Acetyl-CoA carboxylase expression in the liver of quail with hyperuricemia and abdominal obesity

The quail model of hyperuricemia combined with abdominal obesity was induced by high purine diet.Body weight in model group showed no change.Serum uric acid level in model group was increased significantly on 7,14,21,and 28 d( P<0.05 or P<0.01 ).Abdominal fat index in model group increased significantly on 28d.On 7 d and 28 d,serum free fatty acid level was increased significantly.Acetyl-CoA carboxylase( ACC ) protein expression in the liver of model quail was increased as shown by ELISA and immunohistochemisty ( P<0.05 or P<0.01 ),suggesting that the alteration of ACC expression contributes to the pathogenesis of hyperuricemia combined with abdominal obesity.
2.Correlation analysis of Sirt1 gene variant and susceptibility to chronic obstructive pulmonary disease

Objective To explore the relationship between Sirt1 single nucleotide polymorphisms ( rs2236319 ) and chronic obstructive pulmonary disease ( COPD ) susceptibility. Methods The subjects included 149 patients with stable COPD in the First People′s Hospital of Jining outpatient or ward from March 2014 to March 2015, and 160 cases of the same period health check. Sirt1?rs 2236319 genotype distribution frequency was detected by real?time PCR,and the levels of inflammatory factors were compared,including IL?6 and CRP. Results The AA,AG,GG genotype frequency of COPD group were 59. 1%( 88/149) ,35. 6%( 53/149),5. 4%(8/149) respectively,of control group were 45. 0%(72/160),48. 8%(78/160),6. 3%(10/160) respectively,the allele A,G frequency of COPD group were 76. 8%(229/298),23. 2%(69/298) respectively,of control group were 69. 4%( 222/320 ) , 30. 6%( 98/320 ) respectively, there were statistically significant differences between the two groups(χ2=6. 23,6. 06,P<0. 05) . IL?6 and CRP levels were significantly increased in COPD group carrying AA genotype as compared to COPD group carrying AG+GG genotype((10. 30±0. 40) mmol/L,(10. 16±0. 36) mmol/L;(2. 56±0. 20) mg/L,(2. 46±0. 22) mg/L;F=4. 52,8. 04,P<0. 05);There were no significant differences in terms of IL?6 and CRP levels in control group carrying AA genotype as compared to control group carrying AG+GG genotype ( P>0. 05 ) . Both the level of IL?6 and CRP were significantly increased in COPD group carrying AA genotype and AG+GG carriers compared to controls((10. 30 ± 0. 40 ) , ( 9. 88 ± 0. 56 ) mmol/L, ( 10. 16 ± 0. 36 ) , ( 9. 86 ± 0. 57 ) mmol/L;( 2. 56 ± 0. 20 ) , ( 2. 28±0. 25) mg/L,(2. 46±0. 22),(2. 26±0. 26) mg/L;F=34. 52,11. 09,73. 06,19. 38;P<0. 01). Conclusion The polymorphisms of Sirt1?rs2236319 may be an additional risk factor for COPD and associate with inflammatory factors. Furthermore the involvement of rs2236319 in the initiation and progression of COPD may relate to inflammatory processes.
3.Study on GC Fingerprint of Lingyang Ganmao Tablets

OBJECTIVE:To establish the GC fingerpriont for Lingyang ganmao tablets.METHODS:The determination was performed on DB-5 ms capillary column.Flame ionization detector was adopted with temperature of 250 ℃ (temperature programming).The temperature of injector was 240 ℃.Carrier gas was nitrogen with flow rate of 1.0 mL/min;the sample size was 1 μL by split sampling with split ratio of 20 ∶ 1.Using peppermint ketone as reference,Similarity Evaluation Software for Chromatographic Fingerprint of Traditional Chinese Medicine (2012 edition) was used for the similarity analysis of 10 batches of Lingyang ganmao tablets.RESULTS:There were 14 common peaks in the batches of Lingyang ganmao tablets,similarity degrees were higher than 0.98.It was proved that the GC profiles and control fingerprint of 10 batches of samples had good consistency.CONCLUSIONS:The established fingerprint can provide reference for the identification and quality evaluation of Lingyang ganmao tablets.
5.Laparoscopic repair of esophageal hiatal hernia.

This study was to appraise safety and feasibility of laparoscopic approach and investigate the clinical effects of laparoscopic tension-free repair of esophageal hiatal hernia using mesh. From August 2006 to July 2009, 24 patients with esophageal hiatal hernia underwent laparoscopic repair. Twenty-three patients received laparoscopic tension-free repair using mesh, at the same time, Toupet or Dor partial fundoplication was performed. One patient was converted to open surgery. The average operating time was 90 min (70-210 min) and the blood loss was between 10-110 mL. There was no death. The mean postoperative hospital stay was 5 days (3-30 days). During a follow-up period of 12-20 months (mean 15 months), there was no recurrence of the hernia, and no complication with use of mesh. The present study suggested that laparoscopic approach was secure and minimally invasive operation for esophageal hiatal hernia and the use of mesh could reduce recurrence rate.
6.Expression of NogoA in 21a area of visual cortex in visual developing normal cat and strabismic amblyopia cat

BackgroundThe study of the molecular mechanism of visual plasticity is helpful for the explaining and prevention of strabismus and amblyopia.The effect and significance of NogoA in the strabismus and amblyopia formation are attracting more attention.ObjectiveThe present study was to investigate the expression of NogoA in 21a area of visual cortex in strabismic-induced amblyopia cats and explore the possible molecular mechanism of strabismic-induced amblyopia.MethodsSixteen 4-week old clean cats were randomizedly divided into normal group and strabismic-induced amblyopia group and eight for each group.The strabismic-induced amblyopia models were created by cutting off the external rectus in 8 cats.Pattern visual evoked potentials ( P-VEP ) were recorded 1 week after operation and compared with normal cats,and depression of amplitude and prolongation of implied time of P100 wave were as the successful criterion of model.The 200 ml paraformaldehyde was infused via heart to fax the brain under the deep anesthesia and then the cats were sacrificed and the brain cortex sections were prepared.The morphology of 21a zone of cat visual cortex was examined by haematoxylin and eosin staining,and the expressions of NogoA in 21a area of visual cortex were detected by immunohistochemistry with a monoclonal antiNogoA antibody.The use of the animals complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission.ResultsThe implied time and amplitude ratio of VEP P100 wave were (98.10±7.07)ms and (0.83±0.14) in normal group,and those in strabismic-induced amblyopia group were (108.50±6.95 )ms and (0.35 ±0.09 ),showing significant differences between two groups (t=4.450,P=0.005 ; t =5.970,P =0.005 ).The numbers of neurons were similar in 21a area of visual cortex between the two groups,but the volume of the neurons was lessen in strabismic-induced amblyopia group.The positive cell densities for NogoA were ( 387.37±2.01 ) cells/mm2,( 354.58± 1.85 ) cells/mm2 and ( 289.68± 1.81 ) cells/mm2 in layer Ⅱ/Ⅲ,Ⅳ and V/V[ respectively in strabismic-induced amblyopia group,and those in normal group were ( 161.39± 1.98 ) cells/mm2,( 128.93 ± 1.26 ) cells/mm2 and ( 96.25 ± 1.49 ) cells/mm2,indicating a significant increase in model cats ( t =- 160.400,- 201.890,- 164.740,P =0.000 ).Conclusions NogoA may play an important role in the regulation of the sensitive developmental period of visual sense and its plasticity.
7.Impact of scanning and observing conditions on definitive volume of rebuilt CT images in three dimensional conformal radiotherapy planning system

Objective To measure changes of definitive volume of rebuild CT image in three dimensional conformal radiotherapy (3DCRT) treatment planning system (TPS) under different scanning and observing conditions, which may provide a reference in selecting proper condition of scanning and observing, delineating target volume, and delivering treatment plan. Methods A self-made phantom was scanned under different CT scanning conditions, datas were imported to TPS, CT images were rebuilt in three dimensions under different observing conditions, and then the volumes of large, middle and small simulated tumors on CT images were compared with their real volumes. Results The volume defined by rebuilt CT imags in 3DCRT TPS varied according to different scanning and observing conditions. When scanning axial thickness was set at 10. 000 mm, errors of large, middle and small simulated tumor were - 13.60%, - 17. 68% and - 27. 53%, respectively. When 0. 625 mm was set, errors changed to -0. 50%, -0. 68% and -0. 81% ,respectively. When scanning pitch was set at 0. 562, 0. 938, 1. 375 and 1. 750, the errors were - 1.06%, - 1.09%, 5.52% and 4.75%, respectively. Extreme observing condition, window width and window level markedly increased errors of definitive volume of rebuilt CT images with a maximum of 80. 06%. Conclusions This study may provide clues in selecting scanning and observing condition, delineating target volume, and delivering treatment plan.
8.Determination of Vitamin B_1 and Vitamin B_6 in Gengnianling Capsules by HPLC Simultaneously

OBJECTIVE:To determinate vitamin B1 and vitamin B6 in Gengnianling capsules by HPLC simultaneously .METHODS: The separation was performed on Hypersil-ODS C18 column, methanol - sodium hexanesulfonate solution(20 : 80) was used as mobile phase with a flow rate of 0.8ml/ min and detection wavelength of 280nm.RESULTS: Linear correlations with peak area scores were achieved when the sample size of vitamin B1 and vitamin B6 were with a range of 0.884?g-2.652?g (r = 0.9 999) and 0.714?g-2.142?g(r = 0.9 999) .respectively, the average recovery of which were 95.87%(RSD = 0.82%) and 101.96% (RSD = 0.86%), respectively .CONCLUSION: The method is simple, accurate and it can be used for quality control of Gengnianling Capsule.
9.A study of association rules in three-dimensional property-taste-effect data of Chinese herbal medicines based on Apriori algorithm.

The theory of four properties (Qi) and five tastes (Wei) is the core of the property theory of Chinese materia medica. It is known that Qi and Wei are associated with the pharmacological effects (Xiao) of herbs. This study took records of all 365 Chinese herbs in Shennong's Classic of Materia Medica (Shennong Ben Cao Jing) as the data resource and established a three-dimensional data cube, in the purpose of finding out and analyzing the frequent patterns and valued association rules of Qi, Wei and Xiao based on Apriori algorithm. The results of this study may give rise to innovative ideas and methods in research of traditional Chinese materia medica.
