Corneal endothelial cell(CEC)is the most critical part for the cornea, of which activity can influence the postoperative vision.It is very important for the clinical cornea preservation considering the function and its self-purification of donor cornea.There are a variety of classical methods, which can significantly prolong the saving time of donor cornea with its good quality of CEC.We reviewed the published papers about present preservation methods of cornea, which can give us many suggestions for the clinical cornea preservation.
2.Evaluation on capability of parasitic disease control and prevention among professional and technical personnel in medical and health institutions in Wuxi City

Objective To understand the capability of parasitic disease control and prevention among professional and tech?nical personnel in medical and health institutions in Wuxi City,so as to provide the evidence for promoting relative capability building. Methods Forty?one professional and technical persons from 22 medical and health institutions received the evalua?tion through the theoretical knowledge exam and laboratory operation skill assessment. Results The average score of theoretical knowledge exam was(76.5±15.6)with the pass rate of 80.5%and excellent rate of 48.9%. The average score,pass rate and ex?cellent rate for Plasmodium blood slide making were(7.3 ± 1.5),87.8%and 41.5%respectively,the average score,pass rate and excellent rate for Plasmodium blood slide reading were(14.0 ± 7.2),31.7% and 12.2% respectively;the average score, pass rate and excellent rate for helminthes microscope examination were(19.4 ± 10.4),24.4%and 0 respectively;the average score,pass rate and excellent rate for Oncomelania hupensis snail identification were(8.6±1.1),95.1%and 73.2%respectively. The average scores of helminthes microscope examination and Oncomelania hupensis snail identification were higher in the par?ticipants with middle?level professional title or above than in the participants with primary level professional title (both P <0.05). The average scores of theoretical knowledge exam,Plasmodium blood slide reading and helminthes microscope examina?tion were higher in the participants from disease control and prevention institutions than in the staff who came from medical insti?tutions(all P<0.05). Conclusions The professional and technical personnel in medical and health institutions in Wuxi do bet?ter in theoretical knowledge,Plasmodium blood slide making and Oncomelania hupensis snail identification. However,the capa?bility of parasite microscope examination is urgently needed to be improved in the future.
3.The impact of respiratory motion for target dose during external-beam partial breast irradiation

Objective To evaluate the impact of respiratory motion for dose of target and organ at risk during external-beam partial breast irradiation (EB-PBI).Methods 4D-CT scan sets were acquired for 20 patients who underwent EB-PBI.The volume of the tumour bed (TB) was determined based on seroma or surgical clips on the ten sets of 4D-CT images.For each patient a conventional 3D conformal plan (3D-CRT) was generated based on the 4D-CT end inhalation phase images,then copied and applied to the other phases.The following parameters were calculated to analyse:mean dose (D),homogeneity index (HI),conformal index (CI),and the volumes that received ≥ x Gy (Vx).Results During free breathing,the TB centroid motion was 0.90,0.75 and 0.80 mm in the lateral,anteroposterior and superior-inferior directions,respectively.The medium spatial motion vector was 0.95 mm.In the superiorinferior direction,TB motion significantly correlated with D HI,and CI of PTV (r =-0.458,-0.451 and 0.462,P < 0.05),as well as D V20 and V30 received by the ipsilateral normal breast (r=0.527,0.488 and0.526,P <0.05).And in the motion vector,the D V5,V10,V20 of the ipsilateral lung all correlated with TB motion (r =0.416,0.503,0.522 and 0.498,P < 0.05).A correlation also existed between dose and percent volume of heart and volume variation of heart (Dmean,V5 and V10) (r =0.727,0.704 and 0.695,P < 0.05).Conclusions Small TB motion caused by respiratory motion during free breathing result in dosimetric variation of the target and potential dosimetric off-target or suboptimal dose coverage for EB-PBI.The doses of lung during free breathing were relatively sensitive to TB motion and thorax expansion,while heart doses were not influenced notably.
4.Studies on transdermal delivery of ferulic acid through rat skin treated by microneedle arrays.

In order to investigate the characteristics of transdermal delivery of ferulic acid under the treated of microneedle arrays and the influence on permeability of rat skin capillaries, improved Franz-cells were used in the transdermal delivery experiment with the rat skin of abdominal wall and the length of microneedle arrays, different insertion forces, retention time were studied in the influence of characteristics of transdermal delivery of FA. The amount of FA was determined by HPLC system. Intravenous injection Evans blue and FA was added after microneedle arrays treated. Established inflammation model was built by daubing dimethylbenzene. The amount of Evans blue in the rat skin was read at 590 nm wavelength with a Multiskan Go microplate reader. Compared with passive diffusion group the skin pretreated with microneedle arrays had a remarkable enhancement of FA transport (P <0.01). The accumulation of FA increased with the enhancement of insertion force as to as the increase of retention time. Microneedle arrays with different length had a remarkable enhancement of FA transport, but was not related to the increase of the length. The research of FA on the reduce of permeability of rat skin capillaries indicated that the skin pretreated with microneedle arrays could reduce the content of Evans blue in the skins of rat significantly compared with the untreated group. The permeation rate of ferulic acid transdermal delivery had remarkable increase under the treated of microneedle arrays and the length of microneedle arrays ,the retention time so as to the insertion force were important to the transdermal delivery of ferulic acid.

5.Research advances of endoplasmic reticulum stress in high glucose environment and its related ophthalmic diseases

· Endoplasmic reticulum (ER) is a place where it folds and synthesizes the proteins.ER stress was induced when a variety of physiological and pathological factors happened,under which the protein misfolding occurred,the unfolded protein accumulated and the calcium ion imbalanced in the ER.The ER of high-glucose environment can change the protein redox state and produce reactive oxygen species,which affects the ER channel function and chaperone protein buffer;meanwhile changes the balance of calcium ions;finally induces the formation of ER stress.More and more studies have confirmed that ER stress in high glucose environment can cause a variety of ophthalmic diseases.So we review the recent articles about ER stress of high glucose environment and its related ophthalmic diseases.
6.Genetic diagnosis and weight loss surgery of a case of Prader-Willi syndrome

To investigate the clinical features, genetic diagnosis, and treatment of a patient with Prader-Willi syndrome(PWS). For a case with clinically suspected PWS, methylation specific PCR(MSPCR)amplification was applied to CpG islands of SNRPN(exon α)gene locus in the 15q11-q13. Furthermore, the diagnosis was comfirmed by the method of bisulfite sequencing PCR(BSPCR). Metabolic status before and after the operation of sleeve gastrectomy were compared. Absence of amplification of paternal allele on chromosome 15q11-q13 was detected in the case by MSPCR, different from the normal control. Results of BSPCR further proved a full methylation of CpG islands in the SNRPN gene locus. Four months after sleeve gastrectomy, systemic metabolic status and ventricular function were improved. MSPCR and BSPCR were both consistent with genetic diagnosis of PWS. Weight loss surgery is expected to be a major therapy of this disease.
7.Determination of ursolic acid of Liuwei Dihuangwan simulation samples by NIR.

METHODUsing NIR with PLS, PCA-BPANN and WT-BPANN.
RESULTThe predication recovery were 100.7%, 100.6%, 100.1%, and the RSD were 5.42%, 6.49%, 6.52% respectively.
CONCLUSIONNIR can be used in the determination of ursolic acid, which set up the foundation of on-line control of traditional Chinese medicine.

8.Renovascular morphological changes in a rabbit model of hydronephrosis.

Obstructive nephropathy ultimately leads to end-stage renal failure. Renovascular lesions are involved in various nephropathies, and most renal diseases have an ischemic component that underlies the resulting renal fibrosis. The aim of this study was to investigate whether morphological changes occur in the renal vasculature in hydronephrosis and the possible mechanisms involved. A model of complete unilateral ureteral obstruction (CUUO) was used. Experimental animals were divided into five groups: a normal control group (N) and groups of animals at 1st week (O1), 2nd week (O2), 4th week (O4) and 8th week (O8) after CUUO. Blood pressure was measured, renal arterial trees and glomeruli were assessed quantitatively, and renovascular three-dimensional reconstruction was performed on all groups. Glomerular ultrastructural changes were examined by transmission electron microscopy. The results showed that the systolic blood pressure was significantly increased in the obstructed groups (O1, O2, O4 and O8). Three-dimensional reconstruction showed sparse arterial trees in the O8 group, and a tortuous and sometimes ruptured glomerular basement membrane was found in the O4 and O8 groups. Furthermore, epithelial media thickness and media/lumen ratio were increased, lumen diameters were decreased, and the cross-sectional area of the media was unaltered in the segmental renal artery, interlobar artery and afferent arterioles, respectively. In conclusion, renal arterial trees and glomeruli were dramatically altered following CUUO and the changes may be partially ascribed to vascular remodeling. Elucidation of the molecular mechanisms of renovascular morphological alterations will enable the development of potential therapeutic approaches for hydronephrosis.
9.Chinese word processing in nonfluent aphasic patients.

BACKGROUNDChinese nonfluent aphasic patients experience apparent speech production deficit, but it remains less known in which part of Chinese speech production this deficit occurs. The present study aimed to examine the ability of nonfluent aphasic patients in Chinese orthography, phonological and semantic processing via two experiments. Experiment I disclosed the general pattern of deficit of Chinese nonfluent aphasic patients in speech production. Experiment II tested whether this deficit occurs in orthography, phonological or semantic processing.
METHODSThe present study adopted neuropsychological testing methods to compare speech production and Chinese word processing between nonfluent aphasic patients (the patient group) and normal individuals (the control group). Character reading and word reading tasks were used to test speech production. Chinese radical decision, rhyme decision and semantic decision tasks were used to examine word processing. Reaction time and the correct answer rate were collected.
RESULTSThe patient group had a longer reaction time and was more prone to errors in both character reading and word reading tasks than was the control group. For the patient group, there was no difference between the reaction time of character reading and word reading, the error rate of the former was higher than the latter. In radical decision task the reaction time and error rate to the radical "mu " were higher in the patient group than in the control group. In the rhyme decision task, the reaction time and error rate to the rhyme "ang" were higher for the aphasic patients. In the semantic decision task the reaction time to characters in the category of animals was higher for the aphasic patients, yet the error rate was not significantly different between the two groups.
CONCLUSIONSNonfluent aphasic patients seemingly have decreased speed of speech production and an increased error rate. There is a deficit in phonological processing of aphasic patients while their semantic processing may remain intact.

10.Study on a pedigree with Leydig cell hypoplasia caused by novel mutation of luteinizing hormone receptor

Objective To investigate a Chinese pedigree suffering from Leydig cell hypoplasia ( LCH) based on clinical data and genetic diagnosis. Methods The patient was diagnosed by means of clinical data, hormone profiles, and human chorionic gonadotropin ( hCC) test. The luteinizing hormone/chorionic gonadotropin receptor(LHCGR) gene of the patient and family members was amplified and sequenced. Results The patient presented with male pseudohermaphroditism, low level of testosterone, which did not respond to hCG. Genetic analysis of the LHCGR revealed two novel mutations: a missense mutation located in exon 5, resulting in Ile replaced by Thr in the extracellular domain; and a splice site mutation in the 3' terminal of intron 6( IVS6-3 C→A). Proband's sister (46, XX) who lacked clinical manifestations showed the identical genotype with the patient. Conclusions A mutation in the consensus sequence of 3' splice site, in addition to a missense mutation (Ile 152Thr)in the extracellular ligand-binding domain is the cause of inactivation of the LHCGR gene in patient with Leydig cell hypoplasia.
