Objective To observe the effects of sodium arsenite (NaAsO2) on the expression of transcription factor nrf2 (Nrf2) and heme oxygenase 1 (HO-1) in Chang liver cells, and to explore the possible mechanism by using buthionine sulfoximine ( BSO), a GSH synthesis inhibitor. Methods Chang liver cells were treated with NaAsO2 at the doses of 5, 10 and 20 ?mol/L, alone, for 24 h, or pretreated with BSO (3 mmol/L, 12 h). Western blot assays were used to detect the protein expression of Nrf2 and HO-1. Tamoxifen and letrozole are two of the most effective pharmaceuticals mainly used in hormonal dependent) breast cancer therapy. A trace analytical method using ultra performance liquid chromatography/tandem mass spectrometry(UPLC-MS/MS) was developed to simultaneously detect these two drugs in both influent) and effluent of municipal sewage treatment plant. Sewage samples were passed through an Oasis HLB cartridge, then an amino solid phase extraction cartridge was connected with Oasis HLB cartridge. Target compounds) was firstly eluted with 6 mL methanol and then 3 mL methanol was used to elute the amino solid phase extraction cartridge, collected and pooled the elutants. After concentration, target compounds were separated) on a BEH C_(18)) column using a gradient elution profile with a mobile phase consisting of 0.1% formic acid aqueous) solution and acetonitrile and detected by an electro spray ionization tandem mass spectrometry with multiple reactions monitoring(MRM). Satisfactory linearity(R~2>0.997) was obtained over the range of 1.0-100 μg/L) and 0.1-100 μg/L for tamoxifen and letrozole, respectively, with limits of quantification(LOQ) of 1.0 and 0.1 ng/L. Mean recoveries of two target compounds(spiked in sewage samples at three concentration levels) ranged from 68.8% to 103.0%, with relative standard deviations(RSD) less than 15%. This method can be applied for the analysis of target drugs sewage samples.
3.Evaluation of real-time fluorescent quantitative PCR and cefoxitin screening test in detection of methicillin-resistant Staphylococcus aureus

Objective To evaluate the cefoxitin screening test in detecting(MRSA)with the real-time fluorescent quantitative polymerase chain reaction(RT-PCR)for detecting mecA gene as the golden standard.Methods Staphylococcus aureus strains iso-lated from various bacterial infection specimens were collected,which were amplified by RT-PCR and detected by the cefoxitin screening test respectively.The results were compared.Results In 121strains of MRSA,66 strains carrying mecA gene were detec-ted by RT-PCR(P >0.05),accounting for 54.55%(66/121).There was no statsitcal difference in the detection results between the cefoxitin screening test and RT-PCR.The sensitivity of the cefoxitin screening test was 93.93%,the specificity was 81.82%,the positive predictive value was 84.93% and negative predictive value was 91.83%.Conclusion The RT-PCR technique can accurate-ly and rapidly detect MRSA.
4.CPT-11 transarterial chemoembolization prolongs survival of patients with unresectable hepatocellular carcinoma

Objective To study the efficacy of CPT-11 TACE in the treatment of unresectable HCC.Methods A retrospective review was undertaken on unresectable HCC patients receiving doxorubicin transarterial chemoembolization (59 cases) and irinotecan(CPT-11) in 24 cases from May 2003 to November 2011.Survival analysis was performed with Kaplan-Meier statistics.Differences in survival between the two groups were assessed for statistical significance with the log-rank test.Results Overall survival time was significantly longer in patients treated with CPT-11 compared with doxorubicin treated group (21.7 vs 14.5 months,P =0.042).There was no significant difference in time to progression between the two groups,but time to progression was longer in CPT-11 group than doxorubicin treated group (11.42 vs 9.46 months,P =0.091).Subgroup analysis showed that for intermediate-stage HCC,CPT-11 resulted in a significantly longer time to progression and overall survival compared with doxorubicin treated group (P =0.029 and P =0.014,respectively).There were no significant differences in adverse events among the two groups (P > 0.05).Conclusions Chemotherapeutic agent CPT-11 in the form of TACE significantly improved overall survival when compared with doxorubicin for the treatment of unresectable HCC.
5.Chemoradiotherapy for unresectable pancreatic cancer patients after percutaneous transhepatic cholangiodrainage

Objective To evaluate the efficacy of chemoradiotherapy in patients with unresectable pancreatic cancer who were previously treated with PTCD.Methods From September 2005 to December 2012,47 unresectable pancreatic cancer patients with obstructive jaundice were enrolled in this study.They were divided into two groups.21 patients received after PTCD chemotherapy or radiation,or chemoradiotherapy.26 patients in support care group received only nutrition,analgesia and other related support treatment.Survival analysis was performed with Kaplan-Meier statistics.Differences in survival between the groups were assessed for statistical significance with the log-rank test.Results The median overall survival time of patients after PTCD was 7.19 months.The median overall survival time of chemoradiation group was 9.07 months,which was higher than that of support care group (5.52 months),P=0.017.12 patients received single therapy (either chemo or radiation),and 9 patients received chemoradiotherapy.The median overall survival times were 8.31 months and 11.15 months,respectively (P =0.325).Conclusions Post PTCD chemoradiotherapy helps prolong the survival time in unresectable pancreatic cancer patients.
6.Application value of the electrocautery in uvalopalatopharyngoplasty

Objective To evaluate the effects of electrocautery assisted uvalopalatopharyngoplasty (UPPP) for obstructive sleep apnea and hyponea syndrome(OSAHS) .Methods Patients with OSAHS were randomly divided into two groups ,with 14 cases in each group .Group A was operated on with electrocautery ,while group B was operated on with the traditional method .The operative blood loss ,the operation time ,the tunica albuginea off time ,post operative pain ,surgical outcomes and complications were compared between two groups .Results The operative blood loss and the operation time of group A were much less than in group B (all P<0 .05) ,while no difference in the tunica albuginea off time ,post operative pain and surgical outcomes was found (P>0 .05) .Two groups of patients both had no serious complications .Conclusion The advantages of electrocautery assisted UPPP consists of less operative blood loss and less operation time .It deserves to generalize and apply in the future clinical treatments .

Objective To discuss the significance of platelet rich plasma in promoting bFGF and VEGF expression in would healing of rabbit buns .Methods The 24 rabbits were randomly divided into control group and experimental group .The sulfadiazine silver paint to the wound for control group and the platelet rich plasma gel evenly spread to the wound for experimental group .At 7th ,10th ,14th day ,4 rabbits of each group were randomly selected to sacrificed after anesthesiaed ,wound healing rate was compare in 2 groups ,HE staining and testimmunohistochemistry were conducted in each group .Results At 7th ,10th ,14th day ,the wound healing rate of experimental group were higher than those of control group .At 7th d ,the focal granulation ,cells and vessels inten-sive were more significant in experimental group than in control group ,at 10th day ,the wound fibroblast cells and capillary number are more significant in experimental group ,and at 14th day ,the most of fibroblasts translate into fiber cell and the capillary number decreased ,the fibroblasts proliferation was still active and fiber cell was less in the control group .At 7th ,10th day ,the expression of bFGF and VEGF were both higher ,but experimental group is obvious higher than control group(P<0 .05) ,but at 14th day ,the ex-pression of bFGF and VEGF were gradually declined and was not obvious statistics differences in the two group (P>0 .05) .Conclu-sion The platelet rich plasma could promote the big white rabbit scald wound healing ,while the main mechanism maybe the ex-pression of bFGF and VEGF increased in the early .

Objective:To investigate the effect of Ku70 silencing on the expression of apoptosis-related genes in human NSLC cell line,A549 and drug-resistant A549DDP.Methods:Expression of Ku70mRNA in human A549 cells was examined by RT-PCR.Expression of apoptosis-related genes was examined by PCR-Array assay.Results: The Ku70mRNA was higher in A549DDP cells than in A549 cells.The expression of BAX,TP53 and TP73 was significantly increased whereas the expression of TNFRSF1A and BFAR decreased in siRNA-Ku70 A549DDP cells when compared to those of A549DDP cells.Conclsion:The amount of Ku70 mRNA is higher in A549DDP cells,suggesting that Ku70 is important for drug-resistance; Silencing Ku70 might reverse the drug-resistance in A549DDP cells through regulation of expression of apoptofic genes.
9.The value of non-invasive examinations in the diagnosis of arterosclerosis obliterans of lower limbs

Objective To evaluate the value of non invasive examinations in the diagnosis of vascular disease of lower extremity. Methods Segmental systemic pressure (SEG), pulse volume recorder (PVR) and continuous wave (CW) were adopted to examination of 36 patients with arterosclerosis obliterans of lower limbs, and the foundings were compared with that of MRA and operation . Results In all 36 cases, non invasive examinations detected abdominal aorta and common iliac artery with femoral artery occlusion in 12 cases ,femoropoplitea artery occlusion in 28 cases, popliteal artery and the distal artery occlusion in 16 case; MRA together with operation confirmed abdominal aorta and common iliac artery with femoral artery occlusion in 11 cases, femoropopliteal artery occlusion in 32 cases and popliteal artery and the distal artery occlusion in 13 cases. The accurent diagnosis rate of non invasive examination was 85.7%. Conclusions Although PVR has not yet been used to replace the arteriography to directly observe the local situation of arterial lesions , it can preliminarily determine the obstruction sites and severe degree of the occluded arteries. PVR has convenient and reliability in the diagnosis,evaluation of the therapeutic effect and the postoperative follow up of blood vessel diseases,it is worthy to be widely applied in the clinical practice.
10.Regional blood flow fluorescence visualization in robotic partial nephrectomy: preliminary clinical experience

Objective To explore the clinical utilization value of regional blood flow fluorescence visualization in selective arterial clamping robotic partial nephrectomy.Methods 12 cases of robotic partial nephrectomy with indocyanine green-based regional blood flow fluorescence visualization selective arterial clamping between October 2016 and June 2017 by our team were retrospectively analyzed.There were 9 males and 3 females with age between 36-78 years,mean age(51.2 ± 11.0) years,BMI 20.1-36.2 kg/m2,mean of (25.6 ± 4.8) kg/m2,tumor diameter 2.0-5.1 cm,mean of(3.3 ± 0.9) cm,and R.E.N.A.L.score 4-10,mean(7.3 ± 2.0).Preoperative renal function status of eGFR (estimated glomerular filtration rate) was 82-133 ml/(min · 1.73 m2),mean (101.9 ± 13.7)ml/(min · 1.73 m2) and split ECT-GFR of 44.5-70.6 ml/min,mean of(53.8 ± 8.5) ml/min in operated kidney;48.2-71.1 ml/min,mean of(56.8 ±6.8) ml/min in contralateral kidney;in total,92.7-139.1 ml/min,mean of(109.8 ± 14.6)ml/min.Perioperative information including operative time,blood loss,warm ischemia time,surgical complications,pathologic outcomes and follow-up data of included patients were analyzed.Results All procedures were done smoothly without open or radical conversion,with operative time of 95-203 min,mean of (170.6 ±38.6)min,and estimated blood loss of 60-1 000 ml,mean of(178.3 ± 206.9)ml.According to the visualize uptake of fluorescence imaging perfusion area visualization after selective arterial clamping,1 case underwent unclamping robotic partial nephrectomy,1 case converted to main renal artery clamping with warm ischemia time of 18 min,10 cases performed via renal arterial branch clamping with an average ischemia time of (25.5 ± 10.5) (range 17-46)min,1 of which due to parenchymal bleeding obscuring visualization necessitated clamping of the main renal artery with 1000 ml blood loss,46 min of arterial branch occlusion and 16 min of main artery clamping.The average postoperative hospital stay was (5.8 ± 0.9) (range 5-8)days,and the duration of drainage was (3.5 ± 0.5) (range 3-4) days.No postoperative complications occurred.Postoperative pathology:all margins were all negative,11 cases of clear cell carcinoma,eosinophilic adenoma in 1 case.The average eGFR of 9 cases of renal arterial branch clamping was (94.5 ±22.5)(range 56-140)ml/(min · 1.73 m2) at discharge with a 5% percentage decrease versus preoperative level.Among them,4 cases obtained with ECT-GFR data 1 month postoperatively had a mean of(37.6 ±13.2)(range 20.8-55.8) ml/min with 29% percentage decrease on the surgical side,an average of (58.5 ± 6.9) (range 51.2-68.4) ml/min with 2% compensatory increase of the contralateral side,and (98.7 ± 16.2) (range 79.3-124.3) ml/min in total with a 10.4% overall decrease.Conclusions The visualization of blood flow imaging based on indocyanine green fluorescence can clearly and intuitively show the effect of branching arterial occlusion in robotic partial nephrectomy,guide the optimization of surgical resection strategy,and improve safety and clinical outcome.
