Objective To observe the effects of sodium arsenite (NaAsO2) on the expression of transcription factor nrf2 (Nrf2) and heme oxygenase 1 (HO-1) in Chang liver cells, and to explore the possible mechanism by using buthionine sulfoximine ( BSO), a GSH synthesis inhibitor. Methods Chang liver cells were treated with NaAsO2 at the doses of 5, 10 and 20 ?mol/L, alone, for 24 h, or pretreated with BSO (3 mmol/L, 12 h). Western blot assays were used to detect the protein expression of Nrf2 and HO-1. Objective To apply the virtual reality training in clinical education of rehabilitation therapy. Methods 26 undergraduate students majored in rehabilitation therapy were randomly divided into 2 groups during clinical education of physiotherapy. The experimental group (n=14) received virtual reality training first, and then with routine teaching, while the control group (n=12) learnt with routine teaching only. Self-assessment and physiotherapy test were conducted after training. Results The experimental group reported more interested, initiative and positive in training than those of the control group. The experimental group achieved higher scores in practical tests than the control group (P<0.05), while no significant difference in the theoretical examination between the groups. Conclusion Application of virtual reality training may improve the teaching of physiotherapy.

Objective To evaluate the effects of electrocautery assisted uvalopalatopharyngoplasty (UPPP) for obstructive sleep apnea and hyponea syndrome(OSAHS) .Methods Patients with OSAHS were randomly divided into two groups ,with 14 cases in each group .Group A was operated on with electrocautery ,while group B was operated on with the traditional method .The operative blood loss ,the operation time ,the tunica albuginea off time ,post operative pain ,surgical outcomes and complications were compared between two groups .Results The operative blood loss and the operation time of group A were much less than in group B (all P<0 .05) ,while no difference in the tunica albuginea off time ,post operative pain and surgical outcomes was found (P>0 .05) .Two groups of patients both had no serious complications .Conclusion The advantages of electrocautery assisted UPPP consists of less operative blood loss and less operation time .It deserves to generalize and apply in the future clinical treatments .

Corneal endothelial cell(CEC)is the most critical part for the cornea, of which activity can influence the postoperative vision.It is very important for the clinical cornea preservation considering the function and its self-purification of donor cornea.There are a variety of classical methods, which can significantly prolong the saving time of donor cornea with its good quality of CEC.We reviewed the published papers about present preservation methods of cornea, which can give us many suggestions for the clinical cornea preservation.

· AIM: To evaluate the combined effect of rapamycin ophthalmic solution and cyclosporin A ophthalmic solution on allogeneic transplantation in a rat model.rats were used as donors. The animals were randomly assigned to 4 groups. Group A: control (ophthalmic solution solvent 100μ L); group B: 2g/L rapamycin ophthalmic solution 100μ L; group C: 10g/L cyclosporin A ophthalmic solution 100μ L; group D: 2g/L rapamycin ophthalmic solution 50μ L and 10g/L cyclosporin A ophthalmic solution 50μ L, 4 times every day. The treatment was started on 2d after operation, and the animals were administered until rejection. The grafts were inspected by slit-lamp microscope and the corneal survival time was recorded. The pathologic changes were measured by light microscope.significantly prolonged compared with the control group (P ＜ 0.01). However, the combined therapy (group D) was significantly superior compared with group B and C (P＜0.05, P＜ 0.01). The histopathological findings showed that the inflammation cells, neovascularity in each treated group were significantly fewer than that in control group at 14d after operation.a double drug regimen with rapamycin ophthalmic solution and cyclosporin A ophthalmic solution for the control of acute corneal allograft rejection. It indicats that the combined therapy produced synergistic effect.

Objective To evaluate the value of non invasive examinations in the diagnosis of vascular disease of lower extremity. Methods Segmental systemic pressure (SEG), pulse volume recorder (PVR) and continuous wave (CW) were adopted to examination of 36 patients with arterosclerosis obliterans of lower limbs, and the foundings were compared with that of MRA and operation . Results In all 36 cases, non invasive examinations detected abdominal aorta and common iliac artery with femoral artery occlusion in 12 cases ,femoropoplitea artery occlusion in 28 cases, popliteal artery and the distal artery occlusion in 16 case; MRA together with operation confirmed abdominal aorta and common iliac artery with femoral artery occlusion in 11 cases, femoropopliteal artery occlusion in 32 cases and popliteal artery and the distal artery occlusion in 13 cases. The accurent diagnosis rate of non invasive examination was 85.7%. Conclusions Although PVR has not yet been used to replace the arteriography to directly observe the local situation of arterial lesions , it can preliminarily determine the obstruction sites and severe degree of the occluded arteries. PVR has convenient and reliability in the diagnosis,evaluation of the therapeutic effect and the postoperative follow up of blood vessel diseases,it is worthy to be widely applied in the clinical practice.

Objective To observe attenuated store-operated Ca2+ entry in superior mesenteric artery endothelial cells of aged rat.Methods The pressure myograph was applied to measure the vasodilation of superior mesenteric artery induced by bradykinin (BK) in aged versus non-elder rats.Primarily cultured MAECs were treated by thapsigargin (TG) and BK to deplete intracellular Ca2+ stores for comparing SOCE.SOCE of MAECs was detected by calcium imaging technology and was compared between aged and non-elder rats.The expression levels of Orai 1 and stromal interaction factor 1 (STIM 1) were observed by immunofluorescence method.Results Compared with the nonelder rats (70.0 ± 4.1 ％),BK-induced vasodilation in aged rats (27.0 ± 4.9)％ was declined by (60.7±4.3) ％.SOCE induced by BK and TG in primarily cultured MAECs was attenuated by 37.6％ and 39.2％ in aged rats (1.71±0.18 and 1.06±0.03) respectively as compared with non-elder rats (2.72±0.39 and 1.75±0.06).The expressions of SOCE's components Orai 1 and STIM 1 in MAECs were decreased obviously in aged rats than in non elder rats.Conclusions SOCE of MAECs and SOCE-induced vasodilation are significantly attenuated in aged rats.The decreased expression level of Orai 1 and STIM 1 may contribute to this alteration.

Objective To explore the effect of RNA interference targeting hypoxia-inducible factor-1α(HIF-1α) gene on the proliferation of human tongue cancer cells (Tca8113 and Tscca). Methods HIF-1α-specific short hairpin RNA (shRNA) expression vector and empty shRNA vector were respectively transferred into two kinds of cultured human tongue cancer cells. The expression of HIF-1αgene was detected by RT-PCR and Western blot method. The clone forma-tion assay was used to observe the cell proliferation. The cell counting was used to monitor the number of living cells. Flow cytometry was applied to detect cell cycle distribution. Additionally, the cells were injected into nude mice, and tumor form-ing condition was observed. Results After the shRNA HIF-1αplasmid was transfected into tongue cancer cells, the expres-sions of both HIF-1αmRNA and protein were significantly decreased in tongue cancer cells under hypoxia condition. The number of colony formation was significantly lower in HIF-1αsmall interfering RNA (siRNA) group than that in empty and negative control group. The cell counting revealed that the number of living cells was remarkably lower in HIF-1αsiRNA group than that in negative control group. Also, the cell cycle in HIF-1α siRNA group was arrested at G0/G1 phase after transfection and 48 hours hypoxia culture. Besides, the tumor volume was remarkably smaller in HIF-1αsiRNA group than that in negative control group after the tongue cancer cells transplanted into nude mice for several days. The statistical analy-sis indicated that the difference was significant (P＜0.05). Conclusion RNA interference targeting HIF-1α gene could suppress the proliferation of human tongue cancer cells effectively. HIF-1αgene could be researched as a new therapeutic target of tongue cancer in the future.

OBJECTIVE: To observe the effects of different concentrations of arsenic trioxide (As(2)O(3)) on apoptosis and proliferation of human lung cancer cell line A549 in vitro under hypoxia and normoxia. METHODS: A549 cells were treated with 0, 1, 2, 4 micromol/L As2O3 for 12, 24 and 48 h under hypoxia (5% O(2)) and normoxia (21% O(2)). The proliferative inhibition rate of A549 cells was measured with methyl thiazolyl tetrazolium assay, and the apoptotic rate of A549 cells was detected by Annexin V/propidium iodide (PI) double staining. RESULTS: Under normoxia and hypoxia, 1, 2, 4 micromol/L As(2)O(3) could significantly inhibit the proliferation of A549 cells and induce the apoptosis of A549 cells. The results depended on the drug concentration and action time. And the hypoxia couldn't influence the effects of As(2)O(3). CONCLUSION: As(2)O(3) can inhibit the proliferation and induce the apoptosis of A549 cells under hypoxia and normoxia.

7% (OR=4.640,95%CI=1.064 to 20.239,P=0.041) were the high risk factors for myocardial ischemia in patients with type 2 diabetes.Conclusion Poor control of the 4 factors is the high risk factor for myocardial ischemia in patients with type 2 diabetes.Overall and effective control of such risk factors can decrease the incidence of myocardial ischemia and improve its treatment.
