OBJECTIVE To study the effect of chronic intermittent hypoxia and removal of hypoxia on learning and memory ability in Sprague-Dawley rats and the synaptophysin expression in hippocampus CA3 region.METHODS Experiments were performed on 24 three-month-old male Sprague-Dawley rats,They were randomly assigned to three experimental groups(n=8/group):unhandled control group(UC),chronic intermittent hypoxia group(CIH),and Removal of hypoxia group(RH).Rats in UC group were raised normally,those in CIH group suffered 8H intermittent hypoxia everyday,the duration of experiment was 4 weeks,and those in RH group was treated as same as those in CIH group in the first 4 weeks and were raised normally in the next 4 weeks.The learning and memory ability of rats in each group were assessed with the Morris water maze test at the end of the experiments.The expression of synaptophysin in hippocampus CA3 region was investigated by immunohistochemistry staining and image analysis.RESULTS ①Morris water maze test learning scores(place navigation):From the beginning,the escape latency in CIH rats was significantly longer than that in UC rats and RH rats(P

Objective To explore the feasibility of the application of autoregressive integrated moving average(ARIMA) model to forcast the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan ,and to provide scientific references for prevention and control .Methods SAS 9 .2 software was used to fit the ARIMA model for the monthly incidence rate of bacil‐lary dysentery from 2005 to 2013 .Thereafter ,the fitted model was used to forecast the data from January to June in 2014 ,and was evaluated by comparing the actual incidence .Results The ARIMA(0 ,1 ,1)X(0 ,1 ,1)12 model had a better fitted effect ,it was pre‐dicted that the incidence of bacillary dysentery of Jianghan District of Wuhan City from January to June 2014 would increase slight‐ly ,and the forecast data of bacillary dysentery from January 2005 to December 2013 consisted with the actual situation .Conclusion ARIM A model can be used as short‐term forecasting model to predict the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan .

Objective To study the clinical characteristics of obstructive sleep apnea-hypopnea syndrome in the aged. Methods Forty-two over 60 patients with obstruction sleep apnea-hypopnea syndrome (OSAHS) diagnosed by polysomnography (PSG) and 42 matched but less than 60 years OSAHS patients were selected, who were admitted to our hospital from February 2003 to December 2005. The clinical presentations and the results of PSG monitoring were analyzed between the two groups. Objective To investigate the effects of the serum from chronic renal failure (CRF) rats on endoplasmic reticulum stress (ERS) and activation of activating transcription factor 4 (ATF4) of rat arterial smooth muscle cells (ASMCs) cultured in vitro and explore the possible mechanism. Methods The rat model of CRF was established by 5/6 nephrectomy. ASMCs were incubated in the media with 10% or 20% serum of CRF rats cultured in vitro. ATF4 nuclear translocation were analyzed by immunofluorescence. GRP78 (glucose regulated protein 78), p-PERK (pancreatic ER kinase) and ATF4 proteins in response to the CRF serum in ASMCs were determined by Western blot. The ATF4-DNA binding activity was analyzed by electrophoretic mobility shift assay (EMSA). Results The CRF serum could significantly promote the expression of GRP78 of ASMCs in dose-dependent manner. Under the stimulation of 20% CRF serum, nuclear translocation of ATF4 in ASMCs was found. Compared with control serum group, the expression of ATF4 and p-PERK of CRF serum group was increased significantly (P<0.01). The CRF serum could increase the ATF4-DNA binding activity. Conclusion The CRF rat serum can effectively induce ERS of ASMCs and activate the pathway of PERK-ATF4.
7.Biological characteristics and hematopoietic support of bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus

Objective To explore the biological characteristics and karyotype of bone marrow-derived mesenchymal stem cells(MSCs)in patients with systemic lupus erythematosus(SLE)and hematopoietic sup- port of MSCs.Methods MSCs were isolated from bone marrow of 11 SLE patients and 6 healthy controls by density centrifugation and adhesive culture in vitro.The surface markers were detected by flow cytometry (FCM).The morphological changes of MSCs were observed in primary and passage cultures.The growth curves were assayed.The karyotype of MSCs was detected by blocking cellular mitosis with colchicines.The MSCs from SLE patients and healthy controls were infused to ICR mice after high-dose chemotherapy.The changes of peripheral blood counts of the mice were recorded.Results Approximately(6~9)?10~9 MSCs from SLE were obtained after 5 passages and their growth was slower than normal controls(P<0.01).Both groups were positive for CD29,CD44 and CD105,and negative for CD14,CD34,CD45 and HLA-DR.MSCs from SLE had a normal karyotype.MSCs infusions of the two groups were accompanied by no adverse event and the recovery of white blood cell,hemoglobin and platelet count was quicker when compared with the controls(P<0.05).Conclusion MSCs from SLE have demonstrated abnormalities in expansion in vitro.MSCs from SLE have a normal karyotype.Ex vivo MSCs infusion from SLE patients can support hematopoiesis as normal MSCs.
8.Diagnosis and scr eening for antimalarial retinopathy:two cases report and literature review

Objective To analyze antimalarial assosiated retinopathy detected by different methods with of modern ocular fundus examination, and explore screening strategy and the diagnosis of hydroxych-loroquine assosiatedretinopathy according to clinical experience and international guidelines. Methods Full fundus examination was performed in two patients with antimalarial retinopathy. The related literature were reviewed. Results Two patients had bull's eyes maculopathy and abnormal visual fields. Conclusion Clin-icians' alertness to hydroxychloroquine related retinopathy may improve early diagnosis of hydroxychloroquine toxicity. New objective tests are more sensitive than visual fields examination. Visible bull's-eye maculopathy is a late change, and the goal of screening is to recognize toxicity at early stage.
9.Effects of p27mt gene transfection mediated by replication deficient adenovirus on the proliferation and apoptosis of human hepatocellular carcinoma cells

Objective To assess the effects of p27mt gene transfection on the proliferation and apoptosis of human hepatocellular carcinoma cell (HCC) lines SMMC-7721. Methods A replication deficient adenovirus vector encoding p27mt (Ad-p27mt) was used and p27mt cDNA was transfected into human SMMC-7721 cell lines in vitro. The synthesis of DNA in SMMC-7721 cells was determined by using 3H-thymidine incorporation; the cell apoptosis was determined by flow cytometry, TUNEL method and DNA fragmentation analysis. Results The virus titer was 7.95?1012 cfu/ml, the transduction efficiency was 100 % when multiplicity of infection ≥50, FCM analysis revealed a sub-G1 cell peak in Ad-p27mt transduced hepatocellular carcinoma cell lines. 10.Application of Transnasal Gastroscope in Children With Upper Gastrointestinal Tract Disorders

Objective To explore the property and diagnostic significancec of transnasal gastroscope(EG-470N5)in children with upper gastrointestinal tract disorders.Methods Sixty children with upper gastrointestinal tract disorders were examined by using EG-470N5.The scope was inserted either through nose(n=28)or via mouth(n=32).Beside the routine preparations,nasal spray of lidocaine or nasal drib of ephedrine was added to those who received nasal intubation.Results Examination of upper gastrointestinal tract subjects in all succed with no obvious side response in 28 cases(46.7%)who received the examination via nose,27 cases(45%)had mild nausea and 5 cases(8.3%)had serious nausea and dysphoria.Children with esophagostenosis and pyloristenosis and 2-day-old neonate were examined without adverse effect.Conclusions Transnasal gastroscope is preponderant to children due to its slim diameter and supple body,especially to those having swallow difficulty and confined tract.It is more favorable practical prospect than routine gastroscope.
