1.Thinking on Several Ethical Problems in the Clinical Research

This study was conducted in the First Affiliated Hospital of Zhejiang Chinese Medical University for the project of Establishment and Assessment of Ethics Review Platform for TCM Clinical Research. The Ethics Review Platform for Clinical Research is a complete system, which is a Participant Protection System. It includes four as-pects, which are the medical and health institution, ethics committee, ethics committee office, research departments and researchers. In this system, the establishment of research departments and researchers platform is the most cru-cial part. All treatment, protection, risk control, adverse event supervision, which are closely related to the Partici-pant Protection System are completed by researchers. However, during the assessment process, a series of problems were found, such as the current weak ethical consciousness of researchers, less familiar with ethical review process and do not pay equal attention to the participant protection as clinical research. This kind of problem was also found in other hospital assessments in China, which has become a short board of the Participant Protection System.Therefore, from the aspect of the ethics committee, this article presented several ethics problems which should be paid more attention to by researchers such as participant protection, plan design and conduction during the clinical research and also gave some useful advices to change the situation, according to the Assessment Standard of Ethics Review Platform for TCM Clinical Research. In this way, researchers will develop their ethical consciousness and be more familiar with the ethics review process in order to complete key task of participant protection.
2.Design of an microwave applicator using for tumor in superficial layer.

OBJECTIVEA 2.45 GHz microstrip applicator using single rectangle sheet structure is presented.
METHODSBased on the radiant principle of microstrip antenna, the applicator's parameter is designed and the simulating model is set and optimized in HFSS.
RESULTSMeasured by network analyzer, the technical target of this applicator is complied with design demand. During irradiation experiment, based on 30 W power, 30 mm radiation distance and 15 min duration experiment condition, the thermal field distribution map of phantom is obtained from the far-infrared image instrument. The 3D map shows that the region of thermal field centre has small radius and deep heat penetration.
CONCLUSIONThe microwave energy from this applicator can reach the tumor in superficial layer without heat injuring normal tissue around it.

3.Non-thoracoscopic Nuss procedure for correction of pectus excavatum

Objective To review our experience in correction of pectus excavatum.by Nuss procedure without thoracoscopic assistance.Methods 412 patients with pectus excavatum were surgically corrected using Nuss technique without thoracoscopic assistance.There were 329 boys and 83 girls with average age of 7 years and 8 months ( range from 2 years and 8 months to 28 yeats and 5 months).The bars were removed after surgery in 102 patients.Results The operation in all patients had been performed successfully without any severe complications The average time of the operation was 40 minutes.The average blood lose during operation was ( 10 ±2) ml.Conclusion Nuss procedure without thoracoscopic assistance is safety and effective.It could be minimize the trauma and shorter operating time.
4.Survey of vitamin A nutritional status of primary and secondary school students in Qiongzhong,2014-2017

Objective:
To understand vitamin A level of middle and primary school students in Qiongzhong Area in Hainan Province implementing the "Nutritional Compulsory Education Student Nutrition Improvement Plan" (hereinafter referred to as "Student Nutrition Plan"), and to provide suggestions for the followup implementation of student nutrition improvement work.
Methods:
The multi-stage cluster sampling method was used to select three junior high schools and elementary schools in Qiongzhong County, Hainan Province, and students from 1-2 classes from each grade were randomly selected as survey objects. In March 2014, November 2015, December 2016 and December 2017, fasting venous blood was collected from students and serum vitamin A (Retinol) levels were detected by high-performance liquid chromatography.
Results:
From 2014 to 2017, vitamin A levels were (358.77±88.44) (333.54±81.91) (345.84±86.08) (370.70±87.94)μg/L respectively, the subclinical deficiency rate of vitamin A were 22.3%, 31.6%, 27.9%, 18.0%, the vitamin A level of elementary school students were (332.92±71.80) (315.34±73.41) (327.44±77.02) (356.84±80.88)μg/L, the vitamin A level of junior high school students were (412.20±95.56) (383.20±83.53) (396.63±89.48) (411.60±95.14)μg/L, the difference were statistically significant by year (F=26.43, 4.01, P<0.05); vitamin A level and vitamin A subclinical rate of primary school students showed differences by year in both gender. The deficiency rate was statistically significant regardless of the annual difference between men and women(P<0.05); there was no statistically significant difference in the annual rate of vitamin A deficiency and sub-clinical vitamin A deficiency among junior high school students of different grades and genders (χ2=0.85, 2.08, 1.40, 2.36, P>0.05).
Conclusion
The implementation of "Student Nutrition Plan" in Qiongzhong area of Hainan Province shows pasitive effect on the vitamin A nutritional status of students. It’s suggested that prevention should be further strengthened, and nutrition knowledge publicity should be promoted.
5.Design of broadband power divider in microwave hyperthermia system.

In clinical application of microwave hyperthermia, multi-applicators are often simultaneously required to irradiate the tumor because of its large volume or its deep location. Power divider separates the input microwave energy into equal, or unequal, energy to each applicator. In this paper, the design procedure for the three-section transmission-line transformer based one-to-two equal-split Wilkinson power divider is introduced. By impedance analysis on equivalent scheme, the design parameter of power divider is provided, and by simulation and optimization on Ansoft HFSS, a microstrip structure Wilkinson power divider operating frequency 2. 45 GHz is given. Measurement test results from network analyzer show that it has 25% bandwidth and good isolation in output with this structure. Besides, it is characterized by small size and easy processing. This power divider suits microwave hyperthermia.

6.The relationship of methylenetetrahydrofolate reductase G1793A gene polymorphism, hyperhomocysteinaemia and ulcerative colitis

Objectives The present study aimed to investigate the associations between genetic polymorphism of methylenetetrahydrofolate reductase ( MTHFR) G1793A, plasma homocysteine (Hcy) levels, vitamin status and ulcerative colitis ( UC) in a cohort of patients in Hubei Han nationality. Methods Two hundred and ninty-nine UC patients and 764 age- and sex-matched healthy controls were recruited in this study. Polymorphism of MTHFR G1793A was examined using a PCR-RELP method.Plasma levels of Hcy, folate and vitamin B12 were determined by enzymatic cycling assay and corpuscle immune chemiluminescence assay, respectively.Results Both variant allele and genotype frequencies in MTHFR G1793A gene were significantly higher in the UC patients compared to the controls (22.24% vs 14.20% , P<0.001 ;42.81% vs 26.97%, P < 0.001, respectively).Plasma Hcy levels were increased in UC patients compared to the controls [(20.67 ±6.42)mmol/L vs (13.21 ±5.11)mmol/L, P <0.001] while folate and vitamin B12 concentrations were significantly decreased [(11.37±6.34) nmol/L vs (14.89±7.21) nmol/L, P < 0.001; (324.15±127.53 ) pmol/L vs (421.54±128.45 ) pmol/L, P < 0.001, respectively].Furthermore, hyperhomocysteinaemia (HHcy) and folate deficiency were also more prevalent in the UC patients (32.44% vs 25.78% , P = 0.029; 23.41% vs 17.01%, P =0.016, respectively).Conclusions Genetic polymorphism of MTHFR G1793A Wag strongly associated with UC.HHcy,folate deficiency and low vitamin B12 concentration were common phenomena in the UC patients of Hubei Han nationality.Our findings demonstrate that the genes relmed to Hey metabolism may play an important role in the pathogenesis of UC.
7.Establishment and application of an in vitro neruoinflammation model in astrocytes after ischemic injury

Objective To establish a in vitro cell model of inflammation following ischemic injury in rat astrocytes, and study the effects of mild hypothermia on the response of astrocytes to ischemia and inflammation. Methods Primary cultured neonatal SD rat astrocytes identified by GFAP immunofluorescence staining were divided into normal control group (C), hypoxia group (H), and hypoxia and leukocyte treatment group (H+W). Hypoxic injury of the cells was induced by exposure to glucose-flee DMEM medium in 5% CO2 and 95% N2 for 4 h, and for leukocyte treatment, the astrocytes were co-cultured with leukocytes isolated from rat blood. The 3 groups of cells were subjected to hypothermia treatments at 37, 34, 32 and 30 ℃, and the cell viability was estimated by lactic dehydrogenase (LDH) release assay and live/dead assay. Results Ischemic injury occurred in the astrocytes after 4 h of hypoxic exposure. Hypoxic treatment of the cells at 37 ℃ resulted in increased LDH release in all the 3 groups, and the increment intensified in the order of groups C, H and H+W. Mild hypothermia at 34 ℃ and 32 ℃ significantly reduced the LDH release in groups H and H+W (P<0.05), but hypothermia at 30 ℃ significantly increased LDH release in comparison with that at 32 ℃ (P<0.05). Conclusion Mild hypothermia can significantly reduce the severity of ischemic and inflammatory injuries in rat astrocytes possibly mediated also by other mechanisms in addition to inhibited astrocyte metabolism due to low temperatures.
8.Radiation protection and possible mechanisms for low intensity microwave.

OBJECTIVETo investigate radiation protection and possible mechanisms of low intensity microwave on gamma-ray exposed mice.
METHODS96 healthy Kunming mice were randomly divided into the following four groups: normal control, microwave (120 microW/cm(2), 900 MHz), gamma-ray irradiation (5 Gy), combined exposure of microwave and gamma-ray (120 microW/cm(2) + 5 Gy). The microwave group and combined group were exposed to 120 microW/cm(2) microwave firstly, 1 h/d, for 14 days. Then the ionization and combined group were exposed to 5 Gy (60)Co gamma-ray irradiation on the 15th day. Animals were sacrificed on the third, 6th, 9th and 12th day after irradiation. The sternum and spleen paraffin section were produced, and the histological changes were observed. Apoptosis rate of mice splenic cells in each group was examined by flow cytometry, and serum concentration of antioxidant and lipid peroxide was detected at the same time.
RESULTSBone marrow was obviously injured either by radiation or microwave exposure, characterized by undergoing four-phase lesions, namely apoptosis-necrosis, void, regeneration and recovery phase. Compared with the gamma-ray group, the pathological changes in combined group were slighter and the recovery was quicker. The pathological injuries of spleen were similar to that of bone marrow. Injuries in the combined group were slighter than gamma-ray group. It showed that apoptosis rate of splenic cells in combined group was significantly lower on the 6th and 9th day after gamma-ray radiation (23.02% +/- 15.18%, 25.37% +/- 11.62% respectively) from FCM results. Assays of oxidative damages suggested that serum superoxide dismutase (SOD) level in combined group increased while lipid peroxide level decreased significantly (P < 0.05).
CONCLUSIONLow intensity microwave may exert protection effects on injuries induced by ionizing radiation. The underlying mechanisms might be related with suppression on the hematopoietic cells apoptosis induced by gamma-ray radiation, inhibition of oxidative damages, and thus enhanced reconstruction of the hematopoietic system.

9.Effect of quercetin on neural stem cell proliferation in the subventricular zone of rats after focal cerebral ischemia-reperfusion injury.

OBJECTIVETo study the effect of quercetin on the proliferation of neural stem cells in the subventricular zone (SVZ) of rats after focal cerebral ischemia.
METHODSAn adult rat model of middle cerebral artery occlusion (MCAO) model was established by placement of an intraluminal filament at the origin of the MCA. Quercetin was administered intraperitoneally in the rats at a dose of 50 mg/kg every 3 days starting at 6 h after MCAO, and BrdU (50 mg/kg daily) was also injected intraperitoneally starting at 4 h after MCAO. BrdU-positive cells in the SVZ were counted at 7, 14 and 21 days after MCAO.
RESULTSCompared with the sham-operated group, the rats in the ischemic model group showed significantly increased BrdU-positive cells in the ipsilateral SVZ 7 days after MCAO, reaching the peak level on day 14 and beginning to decrease on day 21 (P<0.05). The number of ipsilateral BrdU-positive cells in quercetin group was significantly greater than that in the model group on days 7, 14 and 21 (P<0.05), and maintained the high level on day 21.
CONCLUSIONQuercetin can maintain a high level of neural stem cell proliferation in the SVZ after focal cerebral ischemia in adult rats.

10.Construction of delta-pIRES2-EGFP plasmid and its expression in HEK293 cells.

OBJECTIVETo construct the delta-pIRES2-EGFP plasmid and investigate its expression in HEK293 cells.
METHODSFull length cDNA of rat delta opioid receptor gene amplified from rat brain tissues using reverse transcription and nested PCR was cloned into pMD20 T vector. The delta cDNA was inserted into pIRES2-EGFP plasmid to construct the recombinant eukaryotic plasmid delta-pIRES2-EGFP, which was transfected into HEK293 cells via Lipofectamine2000. The expression of delta was examined under fluorescence microscope.
RESULTSThe recombinant delta-pIRES2-EGFP plasmid was successfully constructed, and high expression of delta was detected in HEK293 cells transfected by the plasmid.
CONCLUSIONdelta-pIRES2-EGFP has been successfully cloned, which shows high expression of delta in HEK293 cells.
