1. Study of dynamic expression of NF-κB and ICAM-1 after cerebral ischemic preconditioning

Objective: To study the roles of nuclear factor (NF-kB) and intercellular adhesion molecule 1 (ICAM-1) in cerebral ischemic preconditioning induced brain ischemic tolerance. Methods: A total of 100 clean rats were randomly allocated into 4 groups: control, ischemic, preconditioning, and ischemic preconditioning groups. Both focal and ischemic preconditioning models were induced. The neuroethological score, infarct volume ratio and expression of NF-kB and ICAM-1 in the ischemic region at the corresponding time points were observed. Results: Circled digit one The neurological deficit score in the ischemic group was higher than that in the ischemic preconditioning group. The cerebral infarction volume ratio was higher than that in the ischemic preconditioning group (28.6 ± 3.2% vs. 16.2 ± 3.8%, t = 2.668 [P < 0.05]), and there were significant differences (P < 0.05). Circled digit two The number of NF-KB positive cells in the ischemic preconditioning group were lower than those in the ischemic group at the same time points, but they were higher than those in the preconditioning group, and there were significant differences (P < 0.05). The peak time of the number of NF-KB positive cells in the ischemic preconditioning group was delayed for 48 hours. Circled digit three The numbers of ICAM-1 positive cells in the ischemic preconditioning group were less than those in the ischemic group at the same time points, but they were higher than those in the preconditioning group, and there were significant differences (P < 0.05). Conclusion: Ischemic preconditioning decreases the expression of NF-KB and ICAM-1 after ischemia. The inhibition of inflammatory reaction may be one of the mechanisms of the ischemic tolerance induced by ischemic preconditioning.
2.Bone regenerationvia a novel honeycomb-like polycaprolactone-calcium silicate crystal compound scaffold in extreme-sized cranial defects

BACKGROUND:Polycaprolactone as a polymer material has poor biocompatibility, and needs to be combined with other natural biological materials to increase biocompatibility, thereby promoting tissue regeneration.OBJECTIVE:To develop a novel honeycomb-like polycaprolactone-calcium silicate crystal compound scaffold, and observe its osteogenic effects in Sprague-Dawley rats with skull defects.METHODS: Eighteen Sprague-Dawley rats were used to make skull defect models and randomized into three groups: blank control group with no implantation, control group with implantation of normal polycaprolactone-calcium silicate crystal compound scaffold, and experimental group with implantation of the novel honeycomb-like polycaprolactone- calcium silicate crystal compound scaffold. Six weeks after implantation, bone regeneration effect in the defect region measuredvia X-ray scanning, Micro-CT three-dimensional reconstruction, and histological analysis.RESULTS AND CONCLUSION: (1) X-ray scan: in all the rats, the size of bone defect was reduced, the fracture line became vague, and the marginal bone density was increased. The percentage of new bone area was highest in the experimental group, successively followed by the control group and blank control group. (2) Micro-CT three-dimensional reconstruction: new bones in the blank control group were mainly distributed on the both sides of the defect, but those in the control and experimental groups distributed in the defect region. The bone regeneration capacity was ranked as follows: experimental group > control group > blank control group (P < 0.05). (3) Histological analysis: new bone ingrowth was visible in all the three groups to different extents. Compared with the other two groups, new bone formation and microvessel density were significantly higher in the experimental group (P < 0.05). To conclude, this novel honeycomb-like polycaprolactone-calcium silicate crystal compound scaffold can obviously promote bone formation in the skull defect region.
3.Effect of forchlorfenuron on fruit morphology and lignans content of Schisandra chinensis.

The effect of plant growth regulator forchlorfenuron (CPPU) 1 x 10(-6), 0.67 x 10(-6), 0.5 x 10(-6) on fruit morphology and effective components lignans was studied. Those morphologies were the combination of four basic morphological changes. The result showed, diametre were increased and longitudinal diametre of fruits were inhibited by foliage fertilizers including CPPU. At the same time, 1 000-grain weight and yield showed the varying degrees increase under CPPU. The order of the degree was 0.5 x 10(-6) > 1 x 10(-6) > 0.67 x 10(-6). Six lignans content of Schisandra chinensis of different harvest time and different CPPU processing groups were determined, the results showed that lignans accumulation occurred mainly in periods of premature the half mature fruiting stages. Under the 0.67 x 10(-6) CPPU treatment, schisandrol B, schisandrin B, schisandrin C content of S. chinensis showed different increase.

4.Imageology and significances of rotational axes of distal femur on Chinese adults with total knee arthroplasty.

OBJECTIVETo explore clinical significance of rotational axis of distal femur on Chinese adults in total knee arthroplasty (TKA).
METHODSThere were 86 Chinese adults (106 normal knees) including 47 males (53 knees) and 39 females (53 knees), 54 knees were on left and 52 on right. The CT scan was employed in the distal femur. The scan direction was aligned to be on the plane perpendicular to the mechanical axis of the femoral. The CT images of cross sections across lateral and medial femoral epicondyle were moved to personal computer,lateral angle between anterior posterior line (APL) and surgical transepicondylar axis (STEA) (ATA),lateral angle between posterior condylar line (PCL) and APL (APA), angle between perpendicularity of APL and PCL (A-PA), posterior condylar angle (PCA), condylar twist angle (CTA), angle between clinical transepicondylar axis (CTEA) and STEA (CSA) were measured. These values were divided into different groups according to gender and side, the values of CTA, PCA, A-PA, angle PT (varus of tibia plateau), constant 3, ATA, APA and constant 90° were compared by statistically. A-PA and PCA, and CTA were analysed statistically with the liner regression, the relationship among CTEA, STEA ,PCL, APL and PLP were performed to assess by liner regression.
RESULTSATA was (89.79 ± 1.22)°, APA was (84.84 ± 1.83)°, A-PA was (5.16 ± 1.83)°, PCA was (4.80 ± 1.23)°, CTA was (8.23 ± 1.40), CSA was (3.45 ± 0.68)°. All the parameters had no differences on sex and side,but CSA had difference on male and female. There was no difference among angle PT, PCA, A-PA. There was significant difference in CAT, constant 30 and angle PT, PCA,A-PA. There was no difference between ATA and constant 90°, but there was difference between APA and constant 90°. There was relativity between PCA and CTA, and also PCA and A-PA, CTA and A-PA. There was significant relativity between STEA and CTEA, between STEA and APL, between STEA and PCL, and also between APLP, APL and PCL, but there was no significant relativity between PCL and CTEA.
CONCLUSIONTKA for Chinese, the section of femoral posterior condyle should be external 5° to obtain the optimum rotational orientation. The property is different entirely between STEA and CTEA, the rotational alignment is not performed according to parallel to the CTEA in distal femur. Among STEA, APL, PCL, the STEA is the most reliable mark ofrotational alignment of the distal femur, and the PCL is the less reliable mark.

5.Effects of lipopolysaccharide on actin reorganization and actin pools in endothelial cells.

OBJECTIVETo investigate the dose and time-dependent effects of lipopolysaccharide (LPS) on cytoskeletal F-acitn and G-actin reorganizations by visualizing their distribution and measuring their contents in human umbilical vein endothelial cell line ECV-304.
METHODSF-actin was labeled with rhodamine-phalloidin and G-actin with deoxyribonuclease I (DNase I)conjugated with fluorescein isothiocyanate (FITC). Contents of cytoskeletal proteins were obtained by flow cytometry.
RESULTSF-actin was mainly distributed peripherally in endothelial cells under normal conditions. LPS stimulation caused the formation of stress fibers and filopodia. G-actin was normally seen in perinuclear and nuclear areas in control ECV-304 cells. Under LPS stimulation, G-actin dots appeared in the cytoplasmic region. The actin disorganization was accompanied by the time- and dose- dependent decrease in F-actin pool and increase in G-actin pool.
CONCLUSIONSLPS can induce characteristic morphological alterations of actin cytoskeleton and formation of intercellular gap in endothelial cells, accompanied by changes in F-actin and G-actin pools.

6.Clinical application of hybrid operating room in cardiovascular disease

Hybrid operating room refers to clean operating rooms equipped with advanced MRI(magnetic resonance ima-ging), DSA(digital subtraction angiography) and CT(computer tomography), radiation therapy C-arm and navigation equip-ment.Currently, hospitals attach great importance to install and use DSA, so that the doctor can not only perform the conven-tional operation, but also directly explore the angiograpic and interventional therapy, which greatly improve the efficiency and the success rate of the surgery.After check out database, choosen about 40 papers of high quality.This paper introduces the concept of hybrid operating room, the development process, clinical application status and advantages in cardiovascular dis-ease, and prospects for its development in the future , and provides some suggestions to more easily build hybrid operating room.
7.Effect of pulchinenoside on FZD8 expression of adjuvant arthritis rats.

To study the effect of pulchinenoside (PULC) on the Frizzled (FZD) expression of adjuvant arthritis ( AA) rats. AA rats were prepared through the toe injection with complete Freund's adjuvant to culture fibroblast-like synoviocytes (FLS). The effect of the oral administration with PULC on the FZD8 expression was detected by the real time qPCR. The effect of FZD8 knockout on the expressions of IL-1, IL-6, IL-8 were detected by MTT and ELISA. The role of miR-375 in the abnomal expression of FZD8 was detected by the real time qPCR. The results showed signfiicant decrease in the FZD8 expression among AA rats, FLS proliferation ater FZD8 knockout and IL-1, IL-6, IL-8 expressions and notable increase in miR-375 expression after the oral administration with PULC. The up-regulated miR-375 expression can inhibit the FZD8 expression. PULC may inhibit the FZD8 expression by up-regulating the miR-375 expression.

Objective To investigate the effect of Panax notoginsenosides monomers ginsenoside Rg 1 in inhibiting hepatic fibro-sis .Methods The rat model of hepatic fibrosis was established by using 50% Ccl4 ,total 35 d .The different doses of Rg1was ad-ministered by hypodermical injection .At the end of the treatment ,the pathological changes of hepatic tissue were observed by light and transmission electron microscope .The stereological method was adopted to measure the volume density (Vvm) ,area density (Svm) ,specific surface(Qm) and surface number density (Nam) of liver cell mitochondria in various groups .Results The stereo-logical data of liver cell mitochondria showed that the statistical differences existed among various groups .Vvm in the Panax Notog-insenosides ,low dose Rg1 and isotonic saline groups were significantly increased compared with the normal control group with sta-tistical difference(P<0 .01);Vvm in the high dose Rg1 ,middle dose Rg1 and colchicine groups showed the increasing trend com-pared with the normal control group without statistical difference (P>0 .05);Vvm in the high ,middle and low dose Rg1 ,Panax No-toginsenosides and colchicine groups showed the decreasing trend compared with the isotonic saline group without statistical differ-ence(P>0 .05) .Svm in the low dose Rg1 ,Panax Notoginsenosides ,colchicine and isotonic saline groups were significantly increased compared with the normal group with statistical difference (P<0 .01);Svm in the high dose Rg1 ,middle dose Rg1 ,Panax Notogin-senosides and colchicine groups was significantly induced compared with the isotonic saline group with statistical difference (P<0 .01);Svm in the high dose Rg1 was reduced compared with the middle dose Rg1 group(P<0 .05) .Nam in the low dose Rg1 ,col-chicine and isotonic saline group was significantly increased compared with the normal group (P<0 .01);Nam in the high dose Rg1 , middle dose Rg1 and Panax Notoginsenosides groups were significantly reduced compared with the isotonic saline group with statis-tical difference(P<0 .01);Nam in the high dose Rg1 group was reduced compared with the middle dose Rg 1 group with statistical difference (P<0 .05) .Qm in all groups was reduced compared with normal group without statistical difference (P>0 .05) .Conclu-sion Rg1 has antifibrosis effects of Panax notoginsenosides ,even exceeds Panax notoginsenosides in some aspects ,and the above-mentioned effect is positively correlated with dose .Rg1 is an ideal drug for preventing and treating liver fibrosis .
