Objective To compare the expression of renal transforming growth factor-?_1(TGF-?_1),its receptors type Ⅰ and Ⅱ(T?RⅠ and T?R Ⅱ) in two rat models of chronic nephropathy induced by CsA and FK506 respectively.Methods Rat models of chronic CsA-/FK506-induced nephropathy were established by administering sandimun Neoral and Prograf separately.Then their kidneys were dissected and the protein expression of TGF-?_1,T?RⅠand T?RⅡ,the mRNA levels of T?RⅠ and T?RⅡ was detected by using immunohistochemistry(IHC) and in situ hybridization(ISH) respectively.Results The IHC revealed that the integrated optical densities(IODs) of renal TGF-?_1,T?RⅠ and T?RⅡ were(605.24)?(140.24),(876.28)?(208.73) and(981.59)?(258.65) respectively in CsA-treated rats,(488.37)?(101.76),(586.63)?(131.21) and(543.34)?(105.26) respectively in FK506-treated ones.The ISH indicated that the IODs of renal T?RⅠ and T?RⅡ mRNA were(905.08)?(158.75) and(1090.92)?(242.73) respectively in CsA-treated rats,(661.37)?(205.65) and(716.27)?(195.55) respectively in FK506-treated ones.The difference of the above-mentioned five factors between the two groups was significant(P

Corneal endothelial cell(CEC)is the most critical part for the cornea, of which activity can influence the postoperative vision.It is very important for the clinical cornea preservation considering the function and its self-purification of donor cornea.There are a variety of classical methods, which can significantly prolong the saving time of donor cornea with its good quality of CEC.We reviewed the published papers about present preservation methods of cornea, which can give us many suggestions for the clinical cornea preservation.

OBJECTIVE: To establish a rapid and sensitive liquid chromatography-tandem mass spectrometry(LC-MS/MS) method for quantification of vincristine sulfate (VCR) in dog plasma and to study the pharmacokinetics of vincristine sulfate thermosensitive liposomes (VSTL) in dogs. METHODS: The plasma was extracted with terl-butyl methyl ether (TBME). VCR and IS(vinblastine sulfate) were separated on an Agela Venusil XBP C18(2.1 mm × 30 mm, 3.0 μm) with a mobile phase gradient at a flow rate of 0.4 mL · min^-1. The injection volume was 10 μL. An Agilent 6460A QQQ triple-quadrople mass spectrometer equipped with an electros-pray ionization(ESI) source was used as detector and was operated in positive ion mode. Multiple-reaction monitoring(MRM) was performed and the m/z of ions selected for quantitation were m/z 825.4→807. 2(VCR) and m/z 811.3→223.9(IS, vinblastine sulfate). Dogs were injected VSTL and vincristine sulfate injection (VSI) via vein at the dose of 0.07 mg^-1, respectively. VCR and internal standard were quantified in dog plasma using a high-performance liquid chromatography-tandem mass spectrometry, Pharmacokinetic parameters were calculated, and a comparative study of VSTL and VSI with six dogs was conducted. RESULTS: The chromatograms showed no endogenous interfering peaks in blank dog plasma. The linear range of VCR in plasma was 0.25-500 ng · mL^-1 (r=0.994 3). The lower limit of quantification was 0.25 ng · mL^-1. The intra-run and inter-run relative standard deviations(RSD) were less than 15%. The pharmacokinetic parameters of VSTL and VSI were as following: ρmax (121.00 ± 42.31) and (61 ±23.36) ng · mL^-1; t1/2λz(23.95±9.03) and (37.91±8.02) h; CLz(0.37±0.07) and (0.35 ±0.09) L · h · kg^-1; Vz(12.15 ±2.14) and (18.95 ±3.27) L · kg^-1; AUC0-t(144.87 ± 1.10) and (127.7 ±2.45) ng · h · mL^-1; AUC0-∞ (152.97 ±12.56) ng · h · mL^-1; and (131.61 ±13.22) ng · h · mL^-1. CONCLUSION: The method is shown to be sensitive, accurate, and convenient for assaying the concentration of vincristine sulfate in preclinical pharmacokinetic studies. The ρmax and AUC of VSTL are significantly higher than VSI after intravenous infusion, the other pharmacokinetic parameters are no significant difference.

The endocytosis/phagocytosis of calcium alginate nanocapsules by peripheral blood derived dendritic cells was confirmed with the use of Quantum dots labeling.Results further demonstrated these nanoparticles could cause the upregulation of HLA-DR expression to induce the maturation of dendritic cells in vitro.Dendritic cells stimulated by nanocapsules covalently loaded with BSA were shown to stimulate the proliferation of self T cells with blank capsules and free BSA as controls,suggesting their potential applications in cancer cell therapy as a new antigen delivery vehicle with a strong adjuvant effect.

Objective To clone the human soluble interleukin-6 receptor(hsIL-6R) gene and expression in insect cell line. Methods The hsIL-6R gene was cloned into plasmid pAcGP67B. After co-transfection of recombinant plasmid and wild type AcNPV DNA, the rAcNPV was confirmed by the end point dilution assay and dot blot. Then it was purified by plaque assay. Results SDS-PAGE showed molecular weight of the expressed product was about 47 000. The expressed recombinant protein was confirmed to be specific and capable of binding its ligand IL-6 by Western blot and ligand-receptor binding assays. Conclusions Secretory expression of hsIL-6R gene in baculovirus expression system was indicated. The expressed product had immunological and biological activities.

OBJECTIVETo investigate the factors influencing the pregnancy outcomes of artificial insemination with donor sperm (AID), improve the pregnancy rate, and evaluate the safety of the offspring.

METHODSWe retrospectively analyzed 7,761 cycles of AID for 5,109 infertile couples performed between July 1, 2005 and June 30, 2013 in the Center of Reproductive Medicine of Shenyang No 204 Hospital, the outcomes of pregnancy, and the incidence of birth defects.

RESULTSTotally, 2 252 clinical pregnancies were achieved by AID, in which the pregnancy rate per cycle was 29. 02% and the cumulative pregnancy rate was 44. 08%. The clinical pregnancy rate was remarkably higher in the females of ≤ 35 years than in those of > 35 years old (30.31% vs 20.18%, P < 0.01), in the women with < 5-year infertility than in those with > 5-year infertility (30.83% vs 28.16%, P < 0.01), and in the patients of the ovarian stimulation group than in those of the natural cycle group (33.22% vs 28.68%, P < 0.01) The clinical pregnancy rate was the highest in the first treatment cycle (29.87%), with statistically significant difference from the fourth cycle (23.61%) (P < 0.05), but not between the other cycles (P > 0.05). There were 28 cases of birth defects in the offspring (1.40%), including 6 cases (21.43%) involving the cardiovascular system, 4 (14.29%) involving the musculoskeletal system, 3 (10.71%) involving the urogenital system, 3 (10.71%) involving the central nervous system, 2 cases (7.14%) of cleft lip and palate, 2 (7.14%) involving the respiratory system, 2 (7.14%) involving the gastrointestinal digestive system, and other anomalies.

CONCLUSIONFemale age, infertility duration, and ovarian stimulation treatment are important factors influencing the clinical pregnancy rate of AID. Artificial insemination with cryopreserved donor sperm does not increase the incidence of birth defects, which is considered as a relatively safe technique of assisted reproduction.

A total of 1 100 patients underwent renal transplantation in the Organ Transplantation Center,Second Affiliated Department of General Hospital of Chinese PLA between 1988 and 2008 were collected,and retrospective analysis was performed in five female patients with malignant tumor,which appeared at 68 months (20-132 months) following renal transplantation,including 2 with renipelvic and uretal cancer and 3 with bladder cancer. Two of the 3 patients with bladder cancer presented homolateral renipelvic and uretal metastasis. Three cases and 1 case of upper uretal cacer were observed at the homolateral or heterolateral of kidney grafts respectively. The main characteristic of sign was iterative and painless gross hematuria. The 5 patients underwent renal transplantation with intravesical instillation therapy and nephrectomy. All patients were survived without rejection in the 1-62 months follow-up. The incidence of malignancy in renal allograft recipients is much higher than that in normal ones,which is related to the long term use of immunosuppressants. Urinary epithelial cancer is the main complicating carcinoma and the first choice of treatment is surgical operation. Based on the normal renal grafts function,the dose of immunosuppressants should be as low as possible,Moreover,radiotherapy or chemiotherapy should be adopted according to the types and stages of tumor complicating renal transplantation.

Objective:To observe the clinical effect of warm needling moxibustion plus acupoint sticking therapy for cervical radiculopathy.Methods:A total of 120 cases were allocated into an observation group,a warm needling group and an acupoint sticking group according to the random number table,with 40 cases in each group.Cases in the observation group received warm needling moxibustion plus acupoint sticking therapy;cases in the warm needling group received the same warm needling moxibustion in the observation group;cases in the acupoint sticking group received the same acupoint sticking therapy in the observation group.The scores of Japanese Orthopaedic Association (JOA) and visual analog scale (VAS) were recorded before and after treatment.Results:The total effective rate was 95.0％ in the observation group,versus 77.5％ in the warm needling group and 75.0％ in the acupoint sticking group (both P＜0.05).Inter-group differences in JOA and VAS between the observation group and the other two groups were statistically significant (all P＜0.05).Conclusion:Warm needling moxibustion plus acupoint sticking therapy is effective in treating cervical radiculopathy,and it can significantly alleviate pain and enhance clinical efficacy,and thus is worth clinical popularization.

Objective To preliminarily evaluate the value of color power doppler ultrasonography in the diagnosis of sacroiliitis in ankylosing spondylitis(AS). Methods Fifty-seven sacroiliac joints in 31 patients with active AS and 40 sacroiliac joints in 20 volunteers were detected by color power doppler ultrasonography.The color flow signs inside the sacroiliac joints were observed,and the resistance index(RI) was measured. Results In active AS,color flow signs were seen in 55 joints,and the mean RI value was 0.53?0.08 in 45 joints,while the other 10 color flow signs represented reversed phase in diastolic phase on pulse doppler ultrasonography.In the volunteers,color flow signs were seen in 16 joints,and the mean RI value was 0.97?0.01 in 6 joints,while the other 10 color flow signs represented reversed phase in diastolic phase on pulse doppler ultrasonography. Conclusion The abnormal flow signs at the sacroiliac joints can be detected by color power doppler ultrasonography.Low RI values provide diagnosis evidence for active AS.
