1.Evaluation of the effects of methylene blue photochemical viral inactivation of red blood cells by using animal models

Objective To evaluate the safety and effectis of methylene blue photochemical inactivation of virus in red blood cell (RBC) by using animal models. Methods ①The effects of improving anemia was assessed by transfusing MB P RBC to Beagle’s dog. ②Duck hepatitis B virus(DHBV)was serially diluted and added into human RBC, and then one day old ducklings were inoculated with that treated RBC by intravenous injection. Sera from ducklings were collected and tested by dot hybridization. Infectivity of DHBV (ID 50 ) in RBC was calculated before and after the treatment. Results Dog RBC treated by MB P were not hemolysed after autologous transfusion. There were no changes in serum ALT, AST, BUN, and creatinine after transfusion. The ID 50 contained in RBC changed from 10 3.35 to 10 8.35 after treatment. Conclusion MB P viral inactivation of RBC is safe and effective.
2.Research on performance evaluation index system for the innovative medical devices transforming project in Zhejiang Province

Objective Establish an overall performance evaluation index system for innovative medical devices transforming project,a major science and technology project in Zhejiang province.Methods The weight of indexes was calculated using Delphi method,analytic hierarchy process and weight of percentile method.Results A three-level performance evaluation index system was established.First level included 3 indexes,as condition index,process index and result index,with weight of 0.3401、0.4042 and 0.2557 respectively.Meanwhile,second level and third level included 13 and 38 indexes.Conclusions The index system provide a performance evaluation tool for the innovative medical devices transforming project.
3.The long-term results of radical nephrectomy for small renal carcinoma

Objective To investigate the long-term follow-up results of patients treated with radical nephrectomy for small renal carcinoma.Methods Between January 1992 and June 2004,a total of 56 pa- tients(41 men and 15 women;mean age,54 years;age range,19-71 years)underwent radical nephrectomy for small renal carcinoma.The clinical data and long-term follow-up results of the 56 cases were analyzed ret- rospectively.All the patients were followed by questionnaire;and the 5-and 10-year survival rates were calcu- lated by Kaplan-Meier method.Results None of the patients was found to have metastasis preoperatively. Postoperative pathology showed renal clear cell carcinoma in 44 cases,granular cell carcinoma in 7,and mixed cell carcinoma in 5.Among these cases,ipsilateral adrenal metastasis was found in 1 case,local lymph- aden metastasis in 2,and perirenal fat metastasis in 4.Postoperatively,49 of the 56 patients(87.5%)were followed for 11-155 months with a mean of 71 months.The 5-and 10-year survival rates were 81.7% and 66.9%,respectively.Five patients died of metastasis from renal carcinoma.Conclusions Radical ne- phrectomy for small renal carcinoma has favorable long-term effects,therefore it is an optimal surgical proce- dure for small renal carcinoma.
4.Effect of type 2 diabetes mellitus on the platelet derived endothelial cell growth factor and vascular endothelial growth factor in patients with cerebral infarction

Objective To observe the effect of type 2 diabetes mellitus on serum platelet-derived endothelial cell growth factor (PD-ECGF) and vascular endothelial growth factor (YEGF) in patients with acute cerebral infarction.Methods Seventy-three patients with acute cerebral infarction (32 patients with non-concomitant diabetes were assigned to group A,and 41 patients with diabetes were assigned to group B),30 cases of type 2 diabetes mellitus patients with non-cerebrovascular disease hospitalized in the same period (group C),and 30 patients without cerebrovascular disease and diabetes mellitus (group D) were the subjects.The serum levels of PD-ECGF and VEGF in group A and group B were measured at 1,3,7,10 to 14 days after onset by double antibody sandwich enzyme-linked immunosorbent assay,which in group C and group D were measured at 1 st day..NIHSS scores were performed on group A and group B using the NIHSS Score (NIHSS) scale.Results The levels of PD-ECGF in group A were (5.93 ± 1.25),(5.93 ± 1.25),(4.19 ± 1.23),(3.67 ± 1.06) μg · mL-1,at 1,3,7,10 to 14 days after onset,The levels of PD-ECGF in group B were (2.88 ±0.54),(2.84 ±0.53),(2.81 ±0.41) and (2.86 ± 0.49) μg · mL-1.The VEGF levels in group A were (172.32 ± 31.91),(158.91 ± 31.84),(158.69 ± 29.27),(156.92±38.16),(159.64 ±27.21) and (159.91 ±40.25) pg· mL-1,The levels of VEGF in group B were (154.91 ±31.84),(158.69 ±29.27),(156.92 ±38.16),(159.64 ±27.21) pg · mL-1,the differences were statistically significant (all P < 0.05).On the first day of admission,the PD-ECGF of group C and D were (2.25±0.49),(2.79±0.51) μg·mL-1,the VEGF were (94.90±19.85),(151.11±130.33) pg· mL-1,the differences were statistically significant (all P < 0.05).The scores of NIHSS in group A and group B were (12.52 ±3.25) and (12.89 ±2.56).The difference was not statistically significant (P > 0.05).On the 10 th to 14th day after onset,the NIHSS scores of group A and group B were (6.48 ± 2.15) and (4.24 ± 1.87) respectively,the difference was statistically significant (P < 0.05).Conclusion Type 2 diabetes can reduce the expression of PD-ECGF and VEGF in patients with cerebral infarction and affect the prognosis of patients with type 2 diabetes mellitus.
5.Research progress of exosomes in treating spinal cord injury

Exosomes are membrane vesicles secreted by cells into the extracellular environment. Exosomes are a new cell component in the treatment of spinal cord injury. Exosomes are secreted by mesenchymal stem cells have anti-apoptotic, anti-inflammatory and pro-angiogenic effects in the treatment of spinal cord injury, which play a key role in nerve cell-cell communication and can delivery exogenous genetic material. Exosomes can penetrate blood-brain barrier and are more stable than their parent cells, reducing the safety risks in the administration of viable cells.
6.Bortezomib combined with autologous peripheral blood hematopoietic stem cell transplantation for therapy of patients with multiple myeloma.

This study was aimed to evaluate the therapeutic efficacy of bortezomib combined with autologous peripheral blood hematopoietic stem cell transplantation (autoPBSCT) for patients with multiple myeloma (MM). 5 patients underwent autologous hematopoietic stem cell transplantation. Bortezomib treatment was supplied for patients before autoPBSCT and in the conditioning of transplantation, it was also used in maintaining treatment. Patients with transplantation adopted bortezomib plus melphalan conditioning regimen. The number of infused MNC and number of CD34(+) cells were 4.06×10(8) (4.09×10(8) - 4.37×10(8))/kg and 3.98×10(6) (2.49×10(6) - 8.2×10(6))/kg respectively. The results showed that hematopoiesis was reconstituted in 5 patients, with a neutrophil cell count more than 0.5×10(9)/L at day 14 (13 - 25 days) after transplantation and platelet count more than 50×10(9)/L at day 28 (21 - 41 days) after transplantation. Transplantation-associated death was not observed. 5 patients were disease-free survival. In conclusion, treatment of bortezomib combined with autologous peripheral hematopoietic stem cell transplantation is an effective method for patients with multiple myeloma. Use of bortezomib after transplantation might still be favourable to MM patients, for survival prolongation and life quality improvement.

7.Effect of Nocardia rubra cell wall skeleton (Nr-CWS) on oncogenicity of TC-1 cells and anti-human papillomavirus effect of Nr-CWS in lower genital tract of women.

OBJECTIVETo detect the effect of Nocardia rubra cell wall skeleton (Nr-CWS) on tumorigenicity induced by TC-1 cells and to clinically study anti-human papillomavirus effect of Nr-CWS in lower genital tract of women.
METHODSTumor model was established by injecting TC-1 cells subcutaneously in SCID mice, then divided them into 3 groups randomly and injected with isovolumetric physiological saline, 60 micrograms/ml Nr-CWS and 120 micrograms/ml Nr-CWS respectively, the growth of tumors was measured one week later. Nr-CWS was applied on 45 HPV positive women whose TCT test was normal and without cervical erosion 2-3 days after menstruation. HPV was detected again 3 months later to explore the effect of Nr-CWS on HPV infection in female lower genital tract.
RESULTSThe animal experiment showed the weight of transplanted tumors in treated group was less than that of control group (chi2=12.5, P= 0.002). The tumor inhibition rate was 59.1 percent and 84.2 percent in the groups treated with Nr-CWS 60 and 120 micrograms/ml Nr-CWS; the results of HPV detection in 23 out of the 45 cases (51.1 percent) became negative after the 3-month treatment; the viral load was reduced in 9, and there was no change in viral load in 13 cases. Significant difference was found between the rates of undetectable viral load and the natural viral disappearance rate (P less than 0.05).
CONCLUSIONNr-CWS has an inhibitory effect to TC-1 cell tumorigenesis and clinical application of Nr-CWS may eliminate the HPV infection in lower genital tract of a considerable proportion of women with HPV infection.

8.Study of mandibular anterior alveolar bone thickness in subjects with different facial skeletal types.

OBJECTIVETo study the association of vertical facial skeletal types and sagittal facial skeletal types with anterior alveolar bone thickness.
METHODSAmong 168 cases with malocclusion in early permanent dentition stage, 93 patients were male and 75 patients were female. All patients (aged 10-14 years) were divided into 9 groups by different facial skeletal types, mandibular anterior alveolar bone thickness in patients' lateral cephalometric films were measured. ANOVA were performed to measurement results with the SPSS 13.0 statistical software.
RESULTSThough sagittal facial skeletal types were the same, there were significant differences between different vertical facial types groups. The order was low-angle group, average-angle group and high-angle group according to the size. A high-angle individual often had a thin anterior alveolar bone while a low-angle individual often had the opposite morphology character. There was no statistical significance between skeletal type I, II and III. But group of skeletal type III also had a thin alveolar bone thickness which had no significant difference with high-angle group. Low-angle group III had no significant difference with average-angle group I and II in alveolar bone thickness.
CONCLUSIONSagittal facial skeletal types have little influence on anterior alveolar morphology, but the vertical facial skeletal types have strong connection with anterior alveolar bone thickness.

Toxicity of different processed was evaluated Polygoni Multiflori Radix by determining the hepatotoxic potency for selecting processing technology. Process Polygoni Multiflori Radix using high pressure steamed, Black Bean high pressure steamed, atmospheric steamed for different time. Using normal human hepatocytes (L02) as evaluation model, hepatotoxic potency as index to evaluate hepatotoxic potency of different processed Polygoni Multiflori Radix. Analysis chemical composition of some processed products by UPLC-MS. Hepatotoxic bioassay method cloud evaluate the toxicity of different Polygoni Multiflori Radix samples. Different processing methods can reduce the toxicity of Polygoni Multiflori Radix, high pressure steamed three hours attenuated was better. Different processing methods have different effects on chemical constituents of Polygoni Multiflori Radix. Comparing with crude sample, the contents of gallic acid, 2,3,5,4-tetrahydroxyl diphenylethylene-2-O-glucoside, emodin-8-O-beta glucoside and emodin were decreased in processed products with 3 kinds of different methods. The change trend of 2,3,5,4-tetrahydroxyl diphenylethylene-2-O-glucoside content was similar with hepatotoxic potency. Different processing methods can reduce the toxicity of Polygoni Multiflori Radix. Processing methods and time attenuated obvious impact on toxicity. Recommended further research on the attehuated standard control of Polygoni Multiflori Radix concocted.

10.Clinical Effect of Laparoscopic Radical Prostatectomy Bladder Cancer and the Influence on Serum Levels of SF, SIL-2R and TSGF

Objective:To analysis clinical curative effect of laparoscopic radical prostatectomy bladder cancer and influence on serum levels of ferritin (SF),soluble interleukin-2 receptor (SIL-2 R) and rumor specific growth factor (TSGF).Methods:98 cases of bladder cancer who were treated in our hospital from August 2012 to February 2016 were selected and randomly divided into the control group 0=49) and the research group (n=49).The patients in the control group were treated with open radical radical cystectomy,while the patients in the research group were treated with laparoscopic radical cystectomy.Then the operation time,intraoperative blood loss,anal exhaust time,hospitalization,the lymph node cleaning,the serum levels of SF,SIL-2R,TSGF,white blood cells and cortisol,the complications and recurrence rate in the two groups were observed and compared.Results:The operation time of research group was longer than that of the control group,while the intraoperative blood loss,the hospitalization and the anal exhaust time were less than those of the control group,and the differences were statistically significant (P<0.05).There was no statistically significant difference about the numbers of the lymph node and the recurrence rate between two groups (P<0.05).After treatment,the serum levels of SF,SIL-2R and TSGF in the two groups decreased,while there was no statistically significant difference between the two groups (P>0.05);After treatment,the white blood cell count and cortisol rise in the two groups increased,while the research group was lower than that of the control group (P<0.05).Conclusion:LRC and ORC clinical efficacy similar,both of which can reduce the serum levels of SF,SIL-2R and TSGF of patients with laparoscopic radical prostatectomy bladder cancer.
