Objective To investigate the changes of bone mineral density (BMD) and its causing factors in menopausal female patients with type Ⅱ diabetes mellitus (DM2). Methods Dual energy X ray absorptimetry was used to measure the BMD of lumbar spines (L 2-4 ) and proximal femur in 82 female patients with type Ⅱ diabetes and 46 normal controls. According to BMD, DM2 patients were divided into two groups (DM A and DM B). Fasting plasma glucose (FPG), fasting plasma insulin (FINS), fasting plasma C peptide (FCP), glycated hemoglobin (HbAc1), urine albumin excretion rate (UAER), and urine ? 2 microglobin (? 2 MG) were measured and compared between the two groups. OBJECTIVETo study the effect of Weifuchun on inflammation of Helicobacter pylori (Hp)-infected gastric epithelial cells (GES-1) and its correlation with NF-kappaB signaling pathway.
METHODSHp standard home-made strain (CagA +, VacA +) NCTCI 1637 infected GES-1 cells were used. Weifuchun was used as intervention. Weifuchun of different concentrations (5,10, and 20 microg/mL) were screened by MTT assay. A blank group and the model group were set up. Then the growth inhibition rate of drugs on gastric epithelial GES-1 cells was detected with MTT assay. Cell cycle was detected using flow cytometry. The supernatant liquid was separated to detect the contents of IL-8 and IL-4 by ELISA.The protein expression level of NF-kappaB was detected by Western blot analysis.
RESULTSMTT assay indicated significantly inhibitory effect of Weifuchun on GES-1 cells [5% inhibiting concentration (IC5)] was 10 microg/ml in the Weifuchun group. After GES-1 and Hp were cultured together,the contents of IL-8 in the supernatant were more obviously higher in the model group than in the blank group (P < 0.05), and then gradually decreased. After treatment with different concentrations of Weifuchun, the levels of IL-8 in the supernatant were less when compared with the model group at 12, 24, 48, and 72 h (P < 0.05). The decrement was the most significant in the high dose Weifuchun group. The IL-4 level in the supernatant was obviously lower in the model group than in the blank group. It obviously increased in the high concentration Weifuchun group (P < 0.05). There was no statistical difference in the IL-4 level between middle, low concentration Weifuchun group and the blank group (P > 0.05). The protein expression of intranuclear P65 increased and that of IkBalpha decreased 60 min after Hp infection. But the protein expression of intranuclear P65 decreased and the protein expression of IkBalpha increased after intervention of Weifuchun.
CONCLUSIONSWeifuchun adjusted H. pylori induced IL-8 and IL-4 production by gastric epithelial cells through blocking NF-kappaB pathways. Its mechanisms might possibly lie in inhibiting p65 from entry into nucleus and the degradation of IkBalpha. Weifuchun was an effective drug for treatment of Hp correlated chronic gastritis.

5.Clinical curative effect observation of PPH in treatment of complexity hemorrhoids and circular hemorrhoid

Objietive Observe the clinical curative effect of PPH in treatment of complexity hemorrhoids and circular hemorrhoid. Methods From January 2010 to January 2014, the clinical data of 278 patients who underwent PPH to treat complexity hemorrhoids, mixed hemorrhoid and circular hemorrhoid were retrospectively analyzed. The postoperative recent and forward curative effect was observed. Results The average operativetime was 28 minutes, and the average hospital stay were 6. 1 days. There were postoperative complications such as pain, anal skin edema, bleeding and urinary retention. 186 cases were cured (66. 9%),35 cases were markedly improved (12. 6%),20 ca-ses were of poor effect (7. 2%),and recurrence occured in 37 cases (13. 3%). Conclusion PPH has the advantages of simple in operation and quick in postoperative recovery. But there were to many postoperative complications and the recurrence rate is high. So PPH needs more further observation and study.

Objective To explore the relationship of D-Dimer and high sensitive C-reactive protein in serum in acute cerebral infarction. Methods 64 patients with acute cerebral infarction were measured with the D-Dimer and Hs-CRP and compared with that of 50 healthy subjects as the controls. Their scores of National Institutes of Health Stroke Scale (NIHSS) were recorded and the correlation with the D-Dimer and Hs-CRP level was analyzed. Results Compared with those in controls, the levels of D-Dimer and Hs-CRP in patients with acute cerebral infarction were significantly higher (P<0.05), and those in patients with large infarction area were significantly higher than those in patients with small infarction area (P<0.01). The levels of D-Dimer and Hs-CRP uncorrelated with the scores of NIHSS.Conclusion The D-Dimer and Hs-CRP level is helpful to assess the acute cerebral infarction.
8.Expression of p38 mitogen-activated protein kinase in hippocampus CA1 and CA3 area of rats with conditioned fear

ObjectiveTo investigate the expression and activity of p38 mitogen-activated protein kinase (p38 MAPK)in hippocampus CA1 and CA3 area of rats with conditioned fear.Methods 24 h after rat model of conditioned fear was established,rats were sacrificed,then expression of p38 MAPK and phosphorylated p38 MAPK (Thr180/Tyr182) in hippocampus CA1 and CA3 area of rats were detected by western blot.ResultsCompared with control group ( ratios of value of gray scale were 1.0 ± 0.1 and 1.0 ± 0.2,respectively),expression of p38 MAPK and phosphorylated p38 MAPK (Thr180/Tyr182) in hippocarpus CA1 area of rats with conditioned fear (9.4 ± 2.6 and 7.8 ± 2.1,respectively) were significantly increased ( n =9,P < 0.05 ).Compared with control group ( 2.1 ± 0.5 and 1.4 ± 0.5,respectively),expression of p38 MAPK and phosphorylated p38 MAPK (Thr180/Tyr182 ) in hippocampus CA3 area of rats with conditioned fear (6.2 ± 3.3 and 2.6 ± 0.6,respectively)were also significantly increased ( n =9,P < 0.05 ).Conclusionp38 MAPK may play important role in the formation of long term memory of conditioned fear.
9.Evaluation on capability of parasitic disease control and prevention among professional and technical personnel in medical and health institutions in Wuxi City

Objective To understand the capability of parasitic disease control and prevention among professional and tech?nical personnel in medical and health institutions in Wuxi City,so as to provide the evidence for promoting relative capability building. Methods Forty?one professional and technical persons from 22 medical and health institutions received the evalua?tion through the theoretical knowledge exam and laboratory operation skill assessment. Results The average score of theoretical knowledge exam was(76.5±15.6)with the pass rate of 80.5%and excellent rate of 48.9%. The average score,pass rate and ex?cellent rate for Plasmodium blood slide making were(7.3 ± 1.5),87.8%and 41.5%respectively,the average score,pass rate and excellent rate for Plasmodium blood slide reading were(14.0 ± 7.2),31.7% and 12.2% respectively;the average score, pass rate and excellent rate for helminthes microscope examination were(19.4 ± 10.4),24.4%and 0 respectively;the average score,pass rate and excellent rate for Oncomelania hupensis snail identification were(8.6±1.1),95.1%and 73.2%respectively. The average scores of helminthes microscope examination and Oncomelania hupensis snail identification were higher in the par?ticipants with middle?level professional title or above than in the participants with primary level professional title (both P <0.05). The average scores of theoretical knowledge exam,Plasmodium blood slide reading and helminthes microscope examina?tion were higher in the participants from disease control and prevention institutions than in the staff who came from medical insti?tutions(all P<0.05). Conclusions The professional and technical personnel in medical and health institutions in Wuxi do bet?ter in theoretical knowledge,Plasmodium blood slide making and Oncomelania hupensis snail identification. However,the capa?bility of parasite microscope examination is urgently needed to be improved in the future.
10.Effect of APP 5-mer peptide on production and differentiation of cultured embryo hippocampal neural stem cells originated from SD rats

Objective To look for a small molecular neurotrophin which could promote production and/or differentiation of neural stem cells (NSCs). Methods 1.Embryo hippocampal NSCs were cultured in vitro . 2. The neurospheres were identified by antibodies of bromodeoxyuridine (BrdU), glial fibrillary acid protein (GFAP), microtubule associated protein 2 (MAP2) and Galactocerebroside (GalC). 3. The cells were divided into four groups, which were control group, FBS treated group, trans-sequence of APP 5-mer peptide treated group, APP 5-mer peptide treated group. The morphology of NSCs was observed in above four groups. 4. Cell counts, detection of clone information rate and diameter of clone were done to study the effect of APP 5-mer peptide on production of NSCs. 5. In addition, we also detected the MTT metablism rates in all groups. Results 1. The NSCs formed neurospheres and grew in floating. They were BrdU-positive. GFAP-positive, MAP2-positive and GalC-positive cells appeared after FBS were added into the medium. 2. The morphology of NSCs was not changed in APP 5-mer peptide treated group and trans-sequence group compared with the control group. 3. The cell number increased in APP 5-mer peptide treated group as compared with the control group. There were no apparent differences between the control group and the trans-sequence treated group. 4. The clone formation rate and diameters of neurospheres increased in APP 5-mer peptide treated group. 5. The MTT metabolism rate increased in APP 5-mer peptide treated group. Conclusion APP 5-mer peptide could promote the production of embryo hippocamal NSCs in vitro. APP 5-mer peptide doesn't promote the differentiation of embryo hippocamal NSCs in vitro.
