2.The promoter methylation of death-associated protein kinase influences expressions of its mRNA and protein in tissue of gastric cancer

Objective To investigate the regulation effect of promoter methylation of deathassociated protein kinase (DAPK) on mRNA and protein expression of DAPK in tissue of primary gastric cancer (GC). Methods The cancerous and noncancerous samples from 62 patients with GC were determined by RT-PCR for mRNA expression of DAPK. The DAPK promoter methylation was detected by methylation-specific PCR. The protein expression of DAPK in 34 patients with methylation was determined by Western blot. Results mRNA and protein expre.ssions of DAPK in cancerous tissues were reduced significantly compared to noncancerous tissues (0. 2863d±0. 2027 vs 0. 57364±0. 1968,0. 2616±0. 0913 vs 0. 65294±0. 1808, P<0.01). Methylation frequency of DAPK in cancerous tissues was higher than that in noncancerous tissues (54.8% vs 17.7%, P<0.01). Furthermore, DAPK mRNA expression was decreased in methylation group compared to unmethylation group (0.1399±0. 0835 vs 0. 46404±0. 1569, P<0. 01). Moreover, a significant correlation was demonstrated between the TNM stage and DAPK promoter methylation (P = 0. 04). Conclusion Expression of DAPK is down-regulated in cancerous tissues at mRNA and protein levels. Low expression of DAPK is associated with hypermethylation of the promoter of DAPK gene.
3.A PRELIMINARY STUDY ON SURVIVIN AND BCL-2 EXPRESSION IN CERVICAL CARCINOMAS

Objective To study the expression of a novel inhibitor of apptosis and survivin in cervical carcinoma and its relationship to the expression of Bcl-2.Methods Using SP immunohistochemical technique, we examined the expression of survivin and Bcl-2 in 59 cervical invasive squamous cell carcinomas.Results Survivin was expressed in 41 of 59 cases(69.5%) of cervical carcinomas. In contrast, no expression of survivin in normal cervical tissues was observed. Overexpression of survivin was related to the tumor grade and clinical stage. Survivin positive cases were strongly associated with Bcl-2 expression(80% versus 35.7%;P<0.005).Conclusion Apoptosis inhibition by survivin abnormal expression, alone or in cooperation with Bcl-2, may participate in the onset and progression of cervical carcinoma. Survivin is a new diagnostic/therapeutic target in cervical cancer.
4.Neuroprotective effect of sodium oxybate against focal cerebral ischemia-reperfusion injury and the relation to GABA in rats

AIMTo study the protective effect of sodium oxybate (SO) against focal cerebral ischemia-reperfusion injury in rats, and the relationship between the effects of SO and ?-aminobutyric acid (GABA). METHODSThe reversible middle cerebral artery occlusion (MCAO)model in rats was established to investigate the role of SO. The scores of neurological deficits was detected by Longa EZ method in MCAO rats. The extracellular levels of glutamate (Glu) and GABA in CSF were measured by high performance liquid chromatography-fluorometer (HPLC-FR) method, and the weight of cerebral infraction was detected. RESULTSThe scores of neurological deficits and the weight of cerebral infraction markedly decreased by SO while the ratio of GABA/Glu obviously increased administered SO in MCAO rats. CONCLUSIONSSO could prevent MCAO rats from ischemia-reperfusion injury, the protective effect is related to SO keeping dynamic balance of excition-inhibition, and persisting inhibition-depended effect.

Objective To reveal and forecast the incidence trend of Brucellosis, in order to provide acientific basis for future intervention and policy-making. Methods Descriptive epidemiological method was used to analyze and statistically describe the distribution of the disease in different times, different locations and different (7.0783/10 million to 13.1257/10 million) and Qingxu ( 1.4811/10 million to 8.5241/10 million) were higher,followed by Yangqu county(0 to 5.8232/10 million), Xiaodian(0.8108/l0 million to 2.4229/10 million) and Jinyuan district ( 0.5329/ 10 million to 1.5896/10 million), and the remaining counties(districts) in the annual There were 223 cases of Brucellosis patients from 2006 to 2009 in Taiyuan. Vocational high risk population was farmers, with a total of 140 cases, accounting for 62.78% of the total number of incidence, followed by students and workers, respectively, 13, 14 cases, accounting for 5.83% and 6.28%, other occupational groups, 56 cases,77.58%;28 cases aged above 60 years, accounting for 12.56%;22 cases aged younger than 19 years, accounting identical in the four years, most cases occurred in spring and summer and showing a clear seasonal high.Conclusions The incidence trend of Brucellosis is on the rise from 2006 to 2009. High risk population is farmer,and the number of younger patients is on the rise, we propose strengthen protection for high risk groups.
