Objective To construct two ribozyme genes (gⅠand gⅢ) by using PCR according to the requirement of ribozyme's domains and the nucleotide sequences of ?_1Ⅰand Ⅲ procollagen genes. Methods First we constructed two ribozyme genes (g, gⅢ) by using PCR, and then separately cloned into the downstream of T7 promoter of pGEM-T vector. Results The resulting recombinant plasmids (pT-gⅠ, pT-gⅢ) were verified by restriction enzymes, PCR and sequencing. Conclusion The expected two ribozyme genes are constructed successfully and this work affords the basis for the preparation of large amounts of ribozyme molecules and for the study on interaction between mRNA and ribozyme in vivo.

Objective: To evaluate the diagnostic value of the special sequences and methods of MRI in liver cirrhosis and portal hypertension. Methods:100 cases of liver cirrhosis were examined by 3DT1W/FFE and SENSE-BABLANCE sequences and compared with normal livers. Results: ① 3DT1W/FFE imaging has clear advantage over conventional scan in detecting cirrhotic nodules. ② The peak of signal intensity ASD (average standard deviation) in normal livers and the minimum of signal intensity ASD in liver cirrhosis cases were found . If signal intensity standard deviation in liver was over 30, liver cirrhosis should be considered. ③ The degree of hypertension and the size of portal vein and its branches were evaluated by SENSE-BABLANCE sequences. Conclusion: MRI special sequences and methods play a significant role in the early diagnosis of liver cirrhosis and portal hypertension.

Objective: To discuss the application of BALANCE-FFE sequence in abdominal MRCP.Methods:100 patients of abdominal diseases were examined by BALANCE technique. Results: The signal of BALANCE-FFE sequence was high in abdominal MRCP examination, and BALANCE image of the coronal scan was excellent. Conclusion:The biliary tract had obviously high signal intensity acquired by BALANCE sequence. The gallstone and the stenosis could be seen. The tissue out of the bile duct, the head of pancrease and the lymph could also be seen. The BALANCE sequence could be used as a supplement of conventional MRCP.

BACKGROUND: Construction of recombinant adenovirus plasmid plays a central role in preparation of recombinant adenovirus. However, conventionally intracellular homologous recombination method is limited by complex procedures, low successful ratio, and long experimental cycle.OBJECTIVE: To construct the recombinant replication-defective adenovirus vector containing interleukin-10 (Ad.dL-10) in a SD rat, and to provide experimental evidences for eukaryotic expression and animat model studying.DESIGN, TIME AND SEI-FING: Opening study was conducted in the First Affiliated Hospital of Sun Yat-sen University from July 2005 to April 2006.MATERIALS: A SD rat, AdEasy system (USA), ThermoscdptTMRT kit & Tdzol (USA), and HEK-293 (Guangzhou, China) were used in this study. The primer of rlL-10 gene of clone rat was synthesized and sequenced in Shanghai, China.METHODS: rlL-10 gene was cloned from total RNA of healthy SD rat spleen tissue with RT-PCR, and then recombinant adenovirus plasmid named pAd.rlL-10 was obtained by homologous recombination within E.ColiBJ5183 carded with AdEasy-1 system. Last, the recombinant adenovirus was packaged, proliferated by HEK-293 cells and purified.MAIN OUTCOME MEASURES: The Ad.rlL-10 was identified using Western blot and RT-PCR methods.RESULTS: rlL-10 gene was successfully cloned from fresh spleen tissue of a SD rat and incorporated into recombinant adenovirus plasmid pad in order to obtain the Ad.rlL-10. Western blot and RT-PCR showed the rlL-10 gene and protein expressions in the cells. Finally, the rlL-10 recombinant adenovirus was obtained with the titers of 1.0x1014 pfu/mL after amplification and purification.CONCLUSION: AdEasy-1 system characterizing by simple operation and reliable results is commonly used to obtain enough quantity and quality adenovirus after homologous recombination, and HEK-293-induced package, amplification, and purification.

Objective The solution for the cross-platform of visualizing human meridian acupoints by three-dimensional (3D) virtual technology was explored based on existing researches to improve interactive method. Methods 3D virtual technology was utilized to construct a 3D visualization model of human meridians and acupoints. The cross-platform 3D visualization system was realized by integrating the existing relevant standards and clinical knowledge. Results The system can visually and dynamically show the meridians and acupoints of human body. Moreover, it can reveal the details including part meridian circulation, the spatial structure of acupoints, etc which fail to be reflected by the real acupuncture model and two-dimensional (2D) planar chart.Meanwhile, the system can be employed to realize the visualization interaction between meridians model and human beings, and the multi-screen cross-platform user scenarios in the whole network. Conclusion It is expected to be widely applied in fields, such as teaching of traditional Chinese medicine (TCM), scientific research, science popularization, etc.

OBJECTIVETo analyze the relationship between the pulse pressure (PP) and atherosclerotic renal artery stenosis (ARAS) in middle-age and eldery paients with hyperensio, so as to provide the basis for the diagnosis and treatment of ARAS.
METHODSWe tetro spectively analyzed the data of 257 patients with hypertension undergoing renal arteriography at General Hospital of PLA between 2009 and 2014. Their mean age was (64.09 +/- 7.81) years. According the results of arteriography, they were divided into two groups: ARAS group (n = 145), and non-ARAS group (n = 112). Their PP and multinomial clinical indexes with ARAS were statistically analyzed.
RESULTSSingle factor correlation and Logisitic analysis showed that age, PP, serum creatinine (SCr) levels, and new onset of hypertension after 50 years of age were strongly positively related to ARAS. The age and PP and SCr was a team of continuous variable.
CONCLUSIONPP was the first new risk factor of ARAS in middile-aged and elderly patients with hypertension. It is suggested that PP may be a early predictive indicator and a new therapeutic target for ARAS.

Objective To investigate the metabolic changes and the metabolites distribution of different hippocampal regions (head, body and tail)in patients with Alzheimer’s disease (AD)using 1 H-MRS for early diagnosis. Methods The hippocampal multivoxel 1H-MRS at 3.0 T was scanned in 30 patients with AD and other 30 normal cognitive elders (NC)as contrast. The data obtained were processed at a workstation. The hippocampus was divided into 3 parts (head, body and tail),and the ratios of N-acetylaspartate (NAA)/creatine (Cr),myoinositol (MI)/Cr and MI/NAA were calculated respectively. The metabolite ratios and distribution changes were compared between group AD and NC.Results Compared with the group NC,the NAA/Cr in group AD in bilateral hippocampal body and tail was decreased, whereas the MI/Cr and MI/NAA in bilateral body and tail,MI/NAA in left head were opposite (P<0.01).In group NC,the NAA/Cr was gradually decreased from the bilateral hippocampal heads to tails (P<0.01),however,the MI/NAA was opposite (P<0.01).No distribution differences in every metabolic ratios of bilateral hippocam-pus were found in AD group (P>0.05).Conclusion Metabolic changes and disappearance of the normal distribution in different hippocampal regions detected by 1 H-MRS provide helpful clues for early diagnosis of the AD.
