Objective:To observe slit expression on the model of DMBA induced cheek pouch caricnoma in hamsters in various stages, so as to elucidate the relationship between slit expression and angiogenesis. Methods:Carcinogenesis model (HBPCa) was established in 60 hamsters by topical application of 5 g/L DMBA in left buccal pouch three times per week for 14 weeks.Slit expression, vascular endothelial growth factor (VEGF) and von Willebrand factor (v-WF) were examined by immunohistochemical staining, microvascular dense(MVD) was measured 6-16 weeks respectively after application of DMBA.Results:Carcinoma was developed in all the hamsters.Slit,VEGF and v-WF expression and MVD were all increased with the development of carcinoma(P

Objective To investigate the risk factors related with the recurrence of multifocal papillary thyroid microcarcinoma(PTMC).Methods The data of 126 patients undergoing surgery from Jan.2001 to Jun.2011 proved to be multifocal PTMC by pathology were rctrospcctively analyzed.Logistic multifactor regression analysis was used to analyze the clinicopathological factors that may influence the recurrence of PTMC.Results The median age of the 126 patients was 46 years(ranging from 15 to 78 years old). The recurrence rate was 14.3% (18/126) and the overall survival rate was 98.4% with the medium follow-up of 75 months (ranging from 24 to 150 months).Multivariate analysis showed that extra-thyoidal extension, neck lymph node metastasis and non-total(or near total)thyroidectomy were the independent risk factors for the recurrence of multifocal PTMC (P =0.008,0.027,0.022 respectively). Conclusions Extension beyond the thyroid and metastatic neck lymph node predict a high risk of recurrence.Total or near total thyroidectomy can reduce the risk of recurrence.

Objective To observe the effect of acupuncture on cerebral palsy children. Methods90 children with cerebral palsy were divided into the observation group (acupuncture and comprehensive rehabilitation) and control group (comprehensive rehabilitation). They were assessed with Gross Motor Function Measure and the Gesell Development Schedules before and 3 months after treatment. ResultsThe total effective rate was 95% in observation group, and 80% in the control group (P<0.01). The score of gross motor function and Gesell adaptive development quotient improved more in the observation group than in the control group (P<0.01). ConclusionAcupuncture is efficacious on functional recovery in children with cerebral palsy

Objectives To study the effect of preoperative platelet transfusion for splenectomy and devascularization in the prevention of intraoperative and postoperative bleeding. Methods The 230 patients with cirrhosis and portal hypertension who received splenectomy and periesophagogastric davascularization were divided into strata A,B and C according to the platelet counts.Stratum A patients had a platelet count of less than 30× 10/L,B between 30× 10/L and 50× 109/L,and C more than 50 × 109/L.The patients in each stratum were then randomly divided into a preoperative transfusion group (T group) and a non-transfusion group (NT group). The amounts of intraoperative bleeding, postoperative drainage in 48 hours after operation, rates of postoperative bleeding, and general medical conditions were compared. Results A comparison in stratum A showed lower amounts of intraoperative bleeding and 48 hour postoperative drainage, and a lower rate of bleeding in the T group (P<0.05).There were no significant differences between the T and the NT groups in strata B and C (P>0.05).Conclusions For patients with a platelet count lower than 30 × 109/L,preoperative platelet transfusion significantly reduced bleeding suggesting that preoperative platelet transfusion for splenectomy and periesophagogastric devascularization should be a routine. For those patients whose platelet count was above 30 × 109/L,platelet transfusion is not recommended.

Objective To investigate the construction of compound membrane with corneal stromal cells and collagen-chitosan by tissue engineering technique and its biocompatibility. Methods Rabbit and human corneal stromal cells were separated and seeded into collagen-chitosan membrane. The compound membrane was transplanted into rabbit corneal stroma. Then the growth condition of keratocytes, the effect on normal keratocytes and degradation of compound membrane were detected by corneal confocal microscope, anterior OCT and histological and immunohistochemical methods ex vivo 1,2,4 weeks after grafting. Results The rabbit and human corneal stromal cells grown well in collagen-chitosan scaffold. The compound membrane degradated gradually after grafting. There was no necrosis and dissolvation.Corneal epithelium, stroma and endothelial cells were all normal. Conclusion Collagen-chitosan can be used as a biological scaffold for construction of corneal stroma.Corneal confocal microscopy and anterior OCT are new methods to observe the biological activity of constructed corneal stroma.

Background and purpose:The genesis and progression of cervical cancer are closly related to E6 and E7 oncogenes of HPV.Special ribozyme and antisense oligonucleotide could inhibit the expression of E6 or E7.but both of them were not the best methods. The aim of this study was to investigate the inhibitive effect of chemically synthetic siRNA on E6 gene in the cervical cancer cell line SiHa and the following biological changes of SiHa cells. Methods:Three specific siRNAs targeting to HPV16 E6 were synthesized and transfected into SiHa cells by lipofectamine. The level of E6 mRNA was tested by RT-PCR,proliferation activities, the cell cycle distribution, expression of p53 protein and the ultramicrostructure changes of SiHa cells were detected by MTT,FCM,immunochemistry and TEM respectively. Results:All the 3 siRNAs could inhibit the level of E6 mRNA singificantly, among which siRNA1 was most effective.Decreased cellular proliferation activity was observed by MTT,especially when the cells were transfected with siRNA1 at 50 nmol/L concentration.Flow cytometry revealed obvious G1 arrest in cell cycle. The expression of p53 protein in transfected cells(0.75?0.06)was increased compared to the control group(0.43?0.03),the difference was singificant.Cell-substance concentration and vacuolus were found in endochylema by TEM.Conclusions:Chemically synthetic siRNA can interfere in the expression of E6 mRNA in SiHa cells specifically and induce the biological changes of the target cells.

Objective: To investigate the protective effect and mechanism of JXHG Decoction on alcoholic liver injury. Methods: Each group of mice were fed for 30 days. Then 50% alcohol used to establish hepatic injury in mice by intragastric administration.16 hours later, the levels of TNF-α, AST, ALT in serum and MDA, GSH, TG, Caspase-3, Caspase-8 in liver were measured accordingly. Results: JXHG Decoction could reduce the levels of TNF-α, AST, ALT, MDA, TG, Caspase-3, Caspase-8, NF-κB and increase GSH, while alleviated tissue necrosis and aelipose degeration of hepar. Conclusion: JXHG Decoction has favorable anti-effects on alcoholic liver injury. Its possible mechanisms are reducing the inflammatory reaction by depressing the expression of TNF-alpha and NF-kappa B in liver cells, and alleviating the hepatic cell apoptosis induced by Caspase-8 and Caspase-3.
