2.Kidney theory of traditional Chinese medicine and stem cell research

Kidney theory is an important figure in traditional Chinese medicine (TCM), it proposes that the kidney is the root of life, in which stores congenital essence and energy of human body that promotes reproduction, growth and development. The facts that body development begins from oosperm have long been elucidated in modern medicine and life science. Furthermore, oosperm is a totipotent stem cell. The totipotent stem cell with its derived embryonic stem cell (ESC) and stem cell constitute the source of growth, development and tissue regeneration. It is rational to comprehend ESC by means of the Kidney theory of TCM, investigate the effect of Kidney-nourishing on the activities of ESC, such as proliferation, aging and apoptosis, and on its related genes regarding transcription, expression and functional regulation. Our works have suggested that classical Kidney-yin-nourishing herb formula Zuo-gui-wan could inhibit the ESC differentiation and apoptosis, stimulate proliferation, and promote the process of cell cycle. These observations coincide with up-regulate expression of Wnt and Oct3/4, and down-regulate expression of P16INK4a. At the same time, classical Kidney-yang-nourishing herb formula You-gui-wan could inhibit ESC apoptosis induced by H2O2, up-regulate Notch expression and down-regulate P16INK4a expression. In addition, there have been some evidences indicated Kidney-nourishing could promote mesenchymal stem cells proliferate. These observations strongly suggest that, Kidney theory of TCM, as the root of the life, is associated with ESC and stem cell; it may provide insight into Kidney theory of TCM and stem cell research.

Objective To investigate the relationship between the different deep venous drainage patterns in the brain and the perimesencephalic subarachnoid hemorrhage (PMSAH).Methods From January 2014 to January 2017,the clinical data of 90 patients with subarachnoid hemorrhage (SAH) diagnosed and treated in the Second Affiliated Hospital of Anhui Medical University were analyzed retrospectively.Thirty patients with PMSAH were in a PMSAH group and 60 patients with aneurismal SAH were in a control group.Unilateral cerebral hemisphere venous drainage was divided into type A (normal continuous):the basilar vein had deep middle cerebral vein drainage and was drained into the great cerebral vein of Galen;type B (normal discontinuous):there was discontinuous venous drainage between the basal vein and the anterior uncal vein and the posterior Galen vein;type C (primitive variant):did not drained into great cerebral vein of Galen,perimesencephalic vein was drained into the superior petrosal sinus or basal vein was directly drained into the transverse sinus or straight sinus.The different combinations of bilateral cerebral hemisphere venous drainage were divided into normal type drainage (typeⅠ:AA),discontinuous type drainage (types Ⅱ:AB or BB),and primitive type drainage (types Ⅲ:AC,BC,or CC).The differences of venous drainage between the two groups were compared.Results In the PMSAH group,both types Ⅰ and Ⅱ drainages accounted for 26.7% (n=8 in each type) and type Ⅲ accounted for 46.7% (n=14).In the control group,typeⅠaccounted for 48.3% (n=29),type Ⅱ accounted for 28.3% (n=17),and type Ⅲ accounted for 23.3% (n=14).There were no significant differences in the distribution of three venous drainage patterns between the two groups (χ2=5.804,P=0.055).However,there was significant difference in the types Ⅲ venous drainage between the two groups (χ2=5.081,P=0.024).Conclusion Most of the deep cerebral venous drainage in patients with PMSAH showed basilar venous drainage into the venous sinuses of dura mater,but not to the large cerebral vein drainage,suggesting the way of primitive drainage into the dural sinus was more prone to rupture compared with that of drainage into large cerebral veins.
5.Design and optimization of pulse sequence for quantitative measurement of ?-aminobutyric acid concentration using MR spectroscopy

Objective To design and optimize the MRS pulse sequence which used to measure gamma-aminobutyric acid(GABA) in vivo. Methods In-depth research for the MRS sequence was carried out and a theoretical analysis and simulation by software using product-operator algebra for the design and optimization of pulse sequence was discussed. Results Through the simulation of MRUI software, Cr signal at ?3.0 was seen to be suppressed and the weak GABA signal was displayed clearly. Conclusion The sequence was designed and optimized. Method to increase the efficiency and the stability of the measurement was studied. Our research gives theoretical preparation for the coming programming work.
6.Observations on short-term curative effect of resynchronization therapy for patients with chronic heart failure

Objective To observe the short-term curative effect of biventricular cardiac resynchronization therapy in patients with chronic heart failure.Methods Twenty-six patients with chronic heart failure underwent biventricular cardiac resynchronization therapy,and a left ventricle lead was implanted from the coronary sinus to vein.During the 3-34(13.8?10.4)months of follow-up period,the following parameters were recorded:the cardiac function,patients' activity,heart rate at night,QRS wave width,left ventricular end-systolic dimension(LVESD),left ventricular end-diastolic dimension(LVEDD),end-systolic volume(ESV),end-diastolic volume(EDV),left ventricular ejection fraction(LVEF),left ventricular filling time,and standard deviation of time to regional peak systolic velocity(Ts-Sd-12).Results One out of the overall 26 patients died from sudden cardiac death.The cardiac function was improved in the 25 survivals in the 3-months follow-up,showing that the effective rate of CRT was 96.2%.For the 25 patients,the cardiac function was improved according to New York Heart Association(NYHA)functional class from Ⅲ-Ⅳ grade to Ⅱ-Ⅲ grade.The average total activity time was increased from 0.31?0.40 hours per day to 2.35?1.20 hours per day.Average heart rate during night-time was decreased from 84.8?15.7 beats per minute to 63.4?4.5 beats per minute,variability of cardiac rhythm was raised from 47.3?7.1ms to 96.4?15.1ms;QRS duration was shortened from 158.6?31.8ms to 129.5?30.2ms(P
7.Permeability increase of prostate cancer cells membrane under low intensity ultrasound in vitro

Objective To investigate the permeability changes of prostate cancer cells membrane under low intensity ultrasound in vitro.Methods The culture of monolayer adherent LNCaP prostate cancer cells in six-well plate was exposed to continuous ultrasound at frequency of 1 MHz.The cells membrane permeability (stained with Calcein)and cells viability(stained with PI)were evaluated by fluorescent microscope (FM) and cells morphological changes were evaluated by scanning electron microscope (SEM) under ultrasound with acoustic intensity of 160 mW/cm2 for 5 s.The rate of cells with increased cells membrane permeability as function of acoustic intensity (80 mW/cm2,120 mW/cm2 and 160 mW/cm2,for 5 s) and exposure duration (5 s,10 s and 15 s,acoustic intensity of 120 mW/cm2) was evaluated by flow cytometry.ResultsAfter low intensity of ultrasound,the cells with increased cell membrane permeability could be clearly shown with Calcein uptake under FM while no cell showed Calcein uptake in the control group.The SEM showed less microvilli on the cells after low intensity of ultrasound exposure and few cells showed holes on the cell membrane.The rate of cells with increased membrane permeability increased with acoustic intensity and exposure duration.Conclusion Low intensity ultrasound alone could increase membrane permeability of prostate cancer cells and cells with increased membrane permeability showed surface plane,uncommon holes on the cells membrane.The rate of cells with increased membrane permeability positively correlated with acoustic intensity and exposure duration.
8.Application of comprehensive experiment evaluation indicators to the fundamental nursing assessment

Objective To evaluate the comprehensive experiment in fundamental nursing by using the indicator system and analyze the teaching effect.Methods 450 nursing graduates of grade 2011 were divided into 56 groups and comprehensive experiments were graded by using the indicator system in each group.Results The four indexes in level one scored (18.08±1.50),(19.99±2.31),(28.18±3.35),(18.16±1.73) points and accounted for 90% 、80% 、81%、91% respectively.The total points' scorings and proportions were (84.41±5.84) points and 84% respectively.Some deficiencies were found to be improved during evaluation.Conclusions Nursing comprehensive experiment evaluation indicator system can assess comprehensive experiments well,and the related competencies of nursing graduates need to be improved.
9.Framework-building and Promotion Tactics of Patient Electronic Engagement Under E-Health Environment

Patient engagement is considered as one pillar of new stage of health care reform and new vitality of health care quality improvement.In recent years,the rapid development of E-Health technology provides richer opportunities for patients to actively participate in health care.The article concludes the basic connotation of patient electronic engagement and proposes the framework on the basis of foreign actions as well as literature reviews,expounds the type,media and methods of patient electronic engagement as well.From the views of projects support,patient education and information rights,feasible policy measures are proposed for patient electronic engagement advancement under E-Health environment.
10.The Mechanism of laboratory research for xiaoaiping treating SGC 7901 gastric carcinoma cellular strains

Objective On the purpose of assessing inhibition efficacy and mechanism of Xiaoaiping (XAP, a kind of Chinese medicine) for SGC 7901 gastric carcinoma cellular strains.Methods 1. Extracorporal inhibition experiments were undertaken for SGC 7901 gastric carcinoma cellular strains using XAP 50,40,30,20,10mg/ml concentrations respectively. 2. SGC 7901 gastric carcinoma cellular strains on the logarithmic growth periods(4?10 7 cells) were concentrated to 1ml in volume. It was coagulated after mixing with thrombin and fibrin and then the coagulated clot was implanted in renal capsules of Kunming water mouse(certifications: No.005, CSAC) for intracorporal experiments. After continual injecting Xiaoaiping intra abdominal cavity was seven days, with 0.4,0.6,0.8ml/kg concentrations respectively. On the 8th day all the water mice were sacrificed using eyepicec ruler for calculating tumor size under microscope including inhibition hate. 3. In order to pursue mechanism of Xiaoaiping on gastric carcinoma, analyses were carried out on FACSTar with cellular analysis of cellfit DNA analysis software, after Xiaoaiping(30mg/ml) mixed with SGC 7901 cellular strains(1?10 6 cells) seven days. Simultaneously Giemsa decoration method as used for observing tumor cell morphologic changes. Results 1. Extracorporal inhibition rates were 100%,59%,29%,19% and 15% to SGC 7901 gastric carcinoma cells using XAP 50,40,30,20,10mg/ml concentrations respectively. Obvious differences were found in comparison with control groups, inhibition rate of XAP in high concentration is similar with HCPT(98%), IC 50 concentration was 21mg/ml after used XAP for seven days. 2. Tumor inhibition rates were 50%,53% and 71% respectively with continual injecting Xiaoaiping into abdominal cavity seven days by 0.4,0.6,0.8ml/kg concentrations. ED 50 was 0.44/kg for Kunming Water Mouse by logit method. Obvious differences were found in comparison with control groups. Inhibition rate(71%) of XAP in high concentration reached favorabler standard with 5 Fu (76%). 3. DNA analyses showed that cellular period were 65.70% in G 0 G 1 period, 16.22% in G 2 M period, 18.09% S period respectively. G 2/G 1 ratio was 1.86 after XAP(30mg/ml) mixed with SGC 7901 cellular strains seven days, the ratio of cellular nucleus to syrup was reduced as tumor cell morphology. Conclusion Xiaoaiping appears to have good inhibition effect on gastric carcinoma cellular strains and should be as an alternative choice for treatmenting gastric carcinoma and metastases.
