1.Study of relationship between T helper cell type-1 and type-2 cytokines and intrahepatic cholestasis of pregnancy

Objective To investigate the effect of T helper cell type-1(TH 1) cytokines [interferon-? (IFN-?), tumor necrosis factor-a (TNF-?)] and T helper cell type-2(TH 2) cytokines [interleukin-4 (IL-4)] on pathogenesis of intrahepatic cholestasis of pregnancy (ICP). Methods The peripheral blood mononuclear cells (PBMC) from 20 cases of ICP patients (study group) and 20 cases of normal pregnant women (control group) were cultured with photohemglutinin stimulation. TNF-?, IFN-? and IL-4 were determined in the supernatant by enzyme- linked-immunoassay. Results (1) In supernatants of cultured PHA induced PBMC, the levels of IFN-? and TNF-? of study group were (639?156) ng/L and (1 021?231) ng/L, which were significantly higher than those of control group, (464?147)ng/L and (728?175) ng/L( P
2.The clinical application of plug mesh in the repairment of abdominal hernia

Objective To assess the value of plug mesh tension free hernia repair in the treatment of abdominal external hernia. Methods 123 tension free repairs of hernia in 115 patients performed in our hospital from April 1988 to May 2000 were retrospectively reviewed. 50 4% (58/115) of cases were complicated by diseases such as cardiopulmonary dysfunction, hypertension, diabetes mellitus, prostate hypertrophy, cirrhotic ascites and thrombocytopenia. Results All patients had healing by first intention with no incisional infection or death , and they were discharged 3~7 days after operation(90% of patients were discharged 3 days after operation).90 4% of patients were followed up with an average of 20 months (1~25 months). There was no recurrence during the follow up.Conclusion Plug mesh tension free repair has the advantages of less trauma, little postoperative discomfort, rapid recovery and low recurrent rate.
3.Effect of silodosin, an alpha1a-adrenoceptor antagonist, on benign prostatic hyperplasia in rat

Objective To investigate the effect of silodosin,a selective alpha1 a-adrenoceptor antagonist on a rat model of testosterone-induced benign prostatic hyperplasia (BPH) and its mechanisms.Methods The rats were divided into three groups:control,testosterone-induced BPH,and silodosin +BPH groups.BPH was induced by subcutaneous injection of testosterone [20 mg/(kg · d)] for 4 weeks.Meanwhile silodosin + BPH groups rats were administered silodosin 4 weeks [100 μg/(kg · d)].After 4 weeks,all animals were sacrificed to examine the blood biochemical profiles,prostate volume,weight,histopathological changes,and epidermal growth factor receptor (EGFR) and B-cell chronic lymphocytic leukemia (CLL)/lymphoma 2 (BCL-2) protein expressions.Results Each group showed an increase compared to their initial body weight;however,differences in weight change between groups were not significant (P > 0.05).The BPH group displayed lower glucose levels than the control group.The serum levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were not significantly different among groups (P > 0.05).The group treated with silodosin showed significantly lesser prostate size and weight than the testosterone-induced BPH group [volume:(0.93 ± 0.14) cm3 vs (1.75 ± 0.15)cm3,P <0.01;weight:(0.97 ±0.06)g vs (1.30±0.05)g,P <0.01].In addition,silodosin decreased the expressions of EGFR and BCL-2 in prostate tissues (P < 0.05).Conclusions These results suggest that silodosin suppress the development of BPH by inhibiting the expressions of EGFR and BCL-2.

Objective: To observe the effects of Thermafil obturat ion technique.Methods: 60 premolars with pulp pathema or periapical dese ase were divided into two groups with 30 in each at random,The teeth were treate d and the root canals were obturated by Thermafil obturation technique and later al condensation technique respectively. Preoperative and postoperative radiograp hs of each tooth were taken. Objective To compare the expression of renal transforming growth factor-?_1(TGF-?_1),its receptors type Ⅰ and Ⅱ(T?RⅠ and T?R Ⅱ) in two rat models of chronic nephropathy induced by CsA and FK506 respectively.Methods Rat models of chronic CsA-/FK506-induced nephropathy were established by administering sandimun Neoral and Prograf separately.Then their kidneys were dissected and the protein expression of TGF-?_1,T?RⅠand T?RⅡ,the mRNA levels of T?RⅠ and T?RⅡ was detected by using immunohistochemistry(IHC) and in situ hybridization(ISH) respectively.Results The IHC revealed that the integrated optical densities(IODs) of renal TGF-?_1,T?RⅠ and T?RⅡ were(605.24)?(140.24),(876.28)?(208.73) and(981.59)?(258.65) respectively in CsA-treated rats,(488.37)?(101.76),(586.63)?(131.21) and(543.34)?(105.26) respectively in FK506-treated ones.The ISH indicated that the IODs of renal T?RⅠ and T?RⅡ mRNA were(905.08)?(158.75) and(1090.92)?(242.73) respectively in CsA-treated rats,(661.37)?(205.65) and(716.27)?(195.55) respectively in FK506-treated ones.The difference of the above-mentioned five factors between the two groups was significant(P
10.A study on porcine acellular cardiac scaffold

Objective To study the change of the structural and biological properties of the cardiovascular tissue after cell extracting. Methods The native and treated pulmonary cusp and pulmonary artery wall were evaluated by HE stain, ETVG stain, immunochemistry stain and scan electron microscopy. The contents of the main macro-molecules were measured. The content and position of calcium was investigated by subcutaneous implantation in a rat model. Results The acellularization procedure resulted in an almost complete removal of the original cells while the construction of the matrix remained. The contents of colleagen and elastin increased while soluble GAG reduced. The content of calcium in acellular tissue by xenogenic implantation was less obviously than that in fresh one. Conclusion A suitable acellularization procedure can remove all cells and most soluble proteins, and matrix remains nearly intact, so as to alleviate obviously calcification via xenogenic implantation in vivo.
