Chinese Journal of Laboratory Medicine 2017;40(1):7-10
Integron was a novel bacterial resistance gene horizontal transmission element. In recent years, many researchers made a lot of research on the resistance mechanism of pathogen. In this paper, the detection and the novel discovery of the integron gene cassette were summarized , including Enterobacteria, Acinetobacter baumanii, Pseudomonas aeruginosa and other bacteria , on the basis of the multidrug resistance mechanism mediated by integron. The prospect the research was descixbed and more attention should be paid to the prevention and control of multidrug resistant bacteria.

Objective:To investigate the distribution of TNF microsatellite polymorphisms in Chinese Han population of Hubei province Methods:DNA samples were extracted from 164 unrelated healthy individuals’EDTA blood TNF microsatellite alleles were typed using PCR technique, followed by High Voltage denaturing PAGE,with silver staining At the same time, the PCR products were cloned and sequenced Results:Detected seven alleles and eleven kinds of genotypes at the TNFb locus;four alleles and seven kinds of genotypes at the TNFe locus The polymorphism information contents (PIC) were 0 67 and 0 33 respectively No deviation from Hardy Weinberg equilibrium were observed Statistical analysis showed, the distribution of TNFb and TNFe alleles in Chinese Han population were significantly different from that in European or in American Caucasian(P

Chinese Journal of Digestion 2001;0(07):-
Objective To investigate whether tumor necrosis factor (TNF)a, TNFb and TNFc microsatellite polymorphisms correlate with chronic atrophic gastritis and gastric adenocarcinoma in Chinese Han population. Methods TNFa, TNFb and TNFc microsatellite alleles in 164 healthy subjects, 53 patients with chronic atrophic gastritis and 56 patients with gastric adenocarcinoma were typed using PCR technique combined with High Voltage denaturing PAGE and silver staining. At the same time, the PCR products were cloned and sequenced. Results The frequency of TNFa10 allele was significantly higher in patients with chronic atrophic gastritis than in healthy individuals ( 19.81% vs. 11.89% , P = 0.04 ). However it was not related to age, gender, degree of atrophy or intestinal metaplasia in patients with chronic atrophic gastritis. The frequency of TNFa6b5c1 haplotype homozygote was significantly lower in patients with gastric adenocarcinoma than in healthy individuals ( 1.79% vs. 15.85% , P = 0.006 ). The sequence result revealed that the copy number of dinucleiotide repeating within the same TNFa allele was not consistent with that in the reports from Western countries. Conclusions It should be more accurate and clear to define TNFa alleles. TNFa10 allele is associated with the susceptibility to chronic atrophic gastritis. TNFa6b5c1 haplotype homozygote is negatively associated with gastric adenocarcinoma and thus may play a resistant role in the shifting process from chronic atrophic gastritis to gastric adenocarcinoma.

Objective To investigate the features and prognosis of noncompaction of ventricular myocardium (NVM) in Chinese. Methods The clinical features of 16 NVM patients were analyzed and 15 patients were followed prospectively. Results Familial occurrence were found in seven patients.Six came to hospital for heart failure and two for chest pain.Arrhythmias were found in six patients.Six patients were asymptomatic.Fifteen patients were followed prospectively for 52.06?8.46 months. In the follow-up,three patients died and heart failure occurred in one asymptomatic patients. One patients accepted permanent pacemaker.Five patients were still asymptomatic. Conclusion In general, the prognosis of NVM is poor, but the outcome is better in patients without symptom. The early diagnosis is important.

Objective To evaluate the prognostic value of dynamic delta model of end stage liver disease(MELD)in patients with decompensated liver cirrhosis. Methods Ninty-seven patients with decompensated liver cirrhosis were enrolled in the study and followed for 1 year followed up.Child-Turcotte- Pugh(CTP)score and MELD score were calculated twice for each patient on the first day of admission and one month later. The difference between two MELD scores represented the delta MELD.The predictive value related with delta MELD,MELD and CTP scores was determined by the area under receiver operating characteristic(ROC)curve. Results Ten patients died within 3 months, whose delta MELD(3.23?2.77) were higher than those of survivors(0.15?0.39)(P

Objectives To explore the calcium signaling mechanism of STIM1 in breast cancer cells. Meth-ods After SiRNA interruption, Western blot and Transwell were used to measure protein expression of STIM1 and cell migration in MDA-MB-231 cells respectively. The relationship between STIM1 and SOCE calcium signaling were analysed by Laser confocal microscopy. Western blots were used to measure protein expression of FAK after si-lence STIM1. Results The numbers of cells without STIM1 were significantly lower than those cells with STIM1 by Transwell assay. STIM1 mediated SOCE in MDA-MB-231. Blocking SOCE might inhibite cells migration. Si-lence STIM1 did not affect the expression or activation of FAK in MDA-MB-231 cells. Conclusion STIM1 influ-ences cell migration through SOCE pathway in breast cancer cells, which is independent on the expression or activa-tion of FAK.

Objective To analyze the morphological matching rate with a digital image recognition system in pre-and post-orthodontic treatments, and to provide forensic dentistry evidence for individual identification. Methods High-resolution digital images of the palatal rugae were taken under particular conditions from each patient in pre-and post-orthodontic treatments.Features of each digital image were extracted using a digital image recognition system and collected in the palatal rugae database.Data of the palatal rugae werematched by MATLAB software system. Results From the matching results, the accuracy rate of individual identification of orthodontic patients was 100%.The accuracy rate in matching palatal rugae patterns in pre-and post-orthodontic treatments was 95.67%.The results were statistically significant with P<0.05.Conclusion Palatal rugae pattern is highly individual-specific.The morphologyofpalatal rugaecould bechangedby theorthodontic treatment, however, these changes may not greatly influence individual identification in forensic dentistry, providing a new potential to individual identification.

Objective To estimate reliability of magnetic resonance elastography (MRE) in measuring liver stiffness of volunteers and patients with chronic liver disease and to assess inter-rater consistency. Methods MRE was performed on a 3.0 T scanner in all subjects, including 24 volunteers (control group) and 64 patients with liver disease (chronic liver disease group).Liver stiffness was measured blindly by two raters. The pathological fibrosis score was applied as a standard reference for liver fibrosis in 22 patients. The intraclass correlation coefficient (ICC) was used to evaluate inter-rater reliability. The differences of liver stiffness between two groups were evaluated using non-parametric MannWhitney U test.Spearman analysis was used to analyze the correlation between fibrosis stages and liver stiffness. Results The intraclass correlation coefficient of liver stiffness was perfect (ICC =0.99,P < 0.01)between two raters. There was significant difference of mean stiffness between control group and patient group (U =90.5,P <0.01) with(2.35 ±0.34) kPa and(4.17 ± 0.47) kPa,respectively. The correlation between fibrosis stage (3,3,5,5 and 6 patients in fibrosis stage S0,S1,S2,S3 and S4) and stiffness (2.13,3.25,3.82,5.45 and 7.35 kPa) was very strong (r =0.96,P <0.01).Conclusion MRE is a reliable and promising tool to measure liver stiffness and to assess liver fibrosis.
