Chinese Journal of Immunology 2016;32(3):345-349
Objective:To investigate the protective effect of N-acetylcysteine(NAC)on right ventricular remodeling in a rat model of monocrotaline-induced pulmonary arterial hypertension ( PAH ) .Methods: PAH rats were induced by a single injection of monocrotaline(60 mg/kg,sc)and were ad ministered with NAC[500 mg/(kg? d)]for 6 weeks. At the end of 4 weeks, the right ventricular systolic pressure ( RVSP ) and mean pulmonary artery pressure ( mPAP ) were monitored via the right jugular vein catheterization into the right ventricle .Right ventricle ( RV ) to left ventricle ( LV )+septum ( S ) were calculated.Right ventricular morphological change was observed by HE staining .Sirius red was used to demonstrate collagen deposition .The expressions of collagenⅠ,collagen Ⅲ, NADPH oxidase 4 ( NOX4 ) and nuclear factor-kappa B ( NF-κB ) were analyzed by RT-PCR and ( or ) Western blot. Results:NAC attenuated RVSP ,mPAP and right ventricular remodeling index ( RV/LV+S) of PAH rats induced by monocrotaline after treatment for 4 weeks.Furthermore ,monocrotaline-induced right ventricular collagen accumulation and collagen Ⅰand collagenⅢexpression were both significantly suppressed by NAC .The expressions of NOX 4 and NF-κB were obviously decreased in right ventricule from PAH rats with NAC treatment. Conclusion:NAC ameliorates right ventricular remodeling of PAH induced by monocrotaline in rats through down regulating the expression of NOX 4 and antioxidant activity ,and inhibiting activation of NF-κB and collagen accumulation .

Osteoarthritis (OA) is the main reason of joint pain and dysfunction in the elderly in China, and its incidence is increasing year by year. In addition to the joint peripheral osteophyte formation and degeneration of articular cartilage, in?flammation, as one of the dominant pathological changes in OA, is causing more and more attention. Pro-inflammatory cyto?kines (PIC) are important mediators of inflammation. The increased level of PIC in OA can lead to systemic and local inflam?mation, results in further destruction of many kinds of tissues in joint (such as cartilage), and accelerates the development of OA. Besides, the severity of inflammation is closely related to the clinical symptoms of OA. Therefore, it is important to un?derstand the role of PIC in the pathogenesis of OA. From the perspective of the relationship between pro-inflammatory fac?tors and OA and the molecular mechanism, this article reviews the research progress in this field, which provides new con?cepts for diagnose and treatment of OA.

Aim To establish a new simple, easy and economical PCR method for determining cyp2d6~*10 allele and studying its distributive frequencies in Chinese population. Methods The new PCR are established and compared with the classical PCR-RFLP. The 224 samples have been determined with the new PCR.Results the results of the two methods go on the way. The allele distribution frequencies of cyp2d6~*10 resembles the reports. Conclusion the new method is proved to be accurate and convenient. It provides a genetics basis for the drug therapy with individuals in clinics.

Objective To explore the influence of the number and orientation of the beams on the optimization of IMRT plan. Methods Four IMRT plans were designed for 9 patients with cervical cancer, and 7 and 15 fields were applied. The 15-field plans had 30 segments and the 7-field plans had 55 segments. The initial beam angle degrees were 0° and 180°,respectively.Dose delivery time,MUs of plans, the dose distributions of the targets, organs at risk and normal tissues were analyzed and compared in the plans. Results Compared the plans with different beam directions under the same amount,no difference of the irradiation dosimetry in the target and organs at risk was found, except for irradiation dosimetry received by the 7-field 180° small intestine was about 4% higher than the other three plans(F=6.164,P<0.05).The terms of the volume of organs at risk got high dose irradiation(V40 and V30 of the rectum and bladder,V40 of the small intestine),which was similar in the 7-and 15-field plans.V20 and Dmean of organs at risk were significantly smaller(F=3.665-10.503,P<0.05)in the 15-field plans. The 15-field plans needed a little longer treatment time(F=0.312,P<0.05)and HI was slightly worse (F=12.933,P<0.05),but the number of MUs was significantly reduced(F=4.650,P<0.05).Conclusions Increasing the beam number will offset the negative impact of sub field reduction and get the similar dose distribution result.

Rheumatoid arthritis (RA) is a chronic inflammatory disease. Joint deformity and dysfunction can occur in the late stage of the disease, which is seriously harmful to human health. Anti-inflammatory factors (AIC), as a protective factor, together with pro-inflammatory factors (PIC) play important roles in the pathogenesis and progression of RA. It is widely accepted by the majority of scholars that the decrease of AIC and the increase of PIC in RA can aggravate the systemic and local inflammatory reactions and accelerate the articular cartilage and subchondral bone destruction, resulting in further progress of RA. A new generation of biological therapy for RA targeting at AIC is in the ascendant. Therefore ,it is important to understand the role of AIC in the pathogenesis of RA. From the perspective of the relationship between AIC and RA and the mechanism, this article reviews the research progress in this field, which provides new concepts for the diagnosis and treatment of RA.

Objective To investigate the inhibitory effects of a recombinant adenovirus vector ex-pressing human IFN-λ1 ( r-Ad-hIFN-λ1) on the growth of orthotopic gastric cancer and to analyze its possi-ble mechanism in a nude mice model of orthotopic human gastric carcinoma .Methods Orthotopic trans-plantation was performed to establish the nude mice model of orthotopic gastric cancer .Thirty mice were ran-domly divided into three groups including PBS control group , Ad-Lac Z empty vector group and r-Ad-hIFN-λ1 experiment group .The mice in each group were given the corresponding interventions once a week for three times. The sizes of tumor were detected by using B-mode ultrasound at different time points to draw growth curves .The expression of IFN-λ1 at mRNA level in skeletal muscle tissues was analyzed by RT-PCR.Western blot assay and immunohistochemical staining were used to detect IFN-λ1 protein in gastric tumor samples .The apoptosis of cells in paraffin-embedded tumor tissues was detected by TUNEL .The per-centages of natural killer ( NK) cells in spleen tissues were analyzed by flow cytometry .Results Compared with control group and empty vector group , the mice in r-Ad-hIFN-λ1 experiment group showed the smallest tumor size [(331.25±6.00) mm3, (322.92±5.92) mm3 vs (248.39±7.60) mm3; P<0.05].hIFN-λ1 was transfected into skeletal muscle successfully and expressed in gastric cancer tissue .Highly expressed hIFN-λ1 was observed in cytoplasm of tumor cells from experiment group by immunohistochemical staining . The apoptotic index of tumor tissues for experiment group was 0.700±0.059 which was significant different from that of control group (0.271±0.026, P<0.05) and empty vector group (0.333±0.028, P<0.05). The percentage of NK cells in spleens from mice in experiment group [(26.49±1.89)%] was significantly higher than that of control group [(13.94 ±1.31)%, P<0.05)] and empty vector group [(19.12 ±1.69)%, P<0.05)].Conclusion Transfection of r-Ad-hIFN-λ1 and expression of hIFN-λin skeletal muscle could significantly inhibit the growth of gastric cancer by inducing tumor cells ′apoptosis and enhan-cing NK cells.
