1.Color Doppler ultrasound in the diagnosis and treatment of arterial injury of extremities

Objective To explore the clinical significance of the color Doppler ultrasound in diagnosis and treatment of artery injury of extremities. Methods Color Doppler ultrasound was used in 69 patients with limb trauma. Results Ultrasonography showed completely or partially vascular rupture in 13 cases (surgery confirmed 11 cases), artery injury and thrombosis in 18 cases (surgery confirmed 14 cases), highly suspicious vascular injuries in 19 cases (surgery confirmed 12 cases), vascular pressure, cramps or bone room fascia caused by high blood pressure obstacles in 19 cases (all confirmed afterclinical treatment). Conclusion Color Doppler ultrasound is the best approach for diagnosis and outcome evaluation of of artery injury of extremities.
2.Drug-Resistance of Staphylococcus in Nosocomial Infection

OBJECTIVE To study drug-resistance status in nosocomial infection with Staphylococcus in the local region,and take effective measures to control the nosocomial infection.METHODS Totally 129 strains of Staphylococcus were clinically isolated from various specimens from Jan 2006 to Dec 2008.The strain identification,detection of meticillin-resistant Staphylococcus(MRS) and K-B disk diffusion test were performed.The data were analyzed statistically.RESULTS Among all 129 strains consisting of 6 species of Staphylococcus,the isolating rate of S.aureus predominated with 39.5%.Among them,meticillin-resistant S.aureus(MRSA) accounted for 29.4%.Among all coagulase-negative Staphyloccocus(CNS),meticillin-resistant coagulase-negative Staphylococcus(MRCNS) accounted for 32.1%.Meticillin-susceptible S.aureus(MSSA) showed significantly lower drug-resistance than MRS.CONCLUSIONS Nosocomial infection status caused by MRS is serious in the grass-roots hospitals.Effective measures should be taken to control it.
3.Infection Status and Drug-susceptibility Analysis of Mycoplasma Strains Isolated from Genitourinary Tract

OBJECTIVE To investigate the infection status and drug resistance of Mycoplasma strains isolated from genitourinary tract. METHODS The IST kit produced by Bio-Merieux was used to culture the Mycoplasma strains and to do drug susceptibility test. RESULTS The Mycoplasma infection ratio was 41.0%,with Ureaplasma urealyticum(Uu) 71.9%,Mycoplasma hominis(Mh) 2.3%,and mixed infection 25.8%,respectively.The drug susceptibility test showed that drug resistance ratios were as follows: to erythromycin 58.7%,ciprofloxacin 50.0%,ofloxacin 41.3%,pristinamycin 3.3%,josamycin 5.4% and doxycycline 7.6%. CONCLUSIONS Mycoplasma infection in genitourinary tract is mainly due to Uu infection.We should rationally choose and use antibiotics according to the drug susceptibility,and josamycin is very effective.
4.A composite of massive frozen allograft and prosthesis replacement used in limb salvage surgery

Objective To analyze the effect of reconstruction for the proximal or distal femoral defect by a composite of massive frozen allograft and prosthesis replacement. Methods According to Enneking's classification, malignant bone tumors of the proximal or distal femur were widely resected in 28 cases. Reconstruction of the affected hip or knee joint after major joint removal was done by a composite of allograft and prosthesis. A conventional femoral component with a stem long enough to provide good fixation in the femoral shaft was threaded over a customized allograft. After the assembly had been reduced and checked for size and position, the allograft was cemented to the femoral component, and the composite was in turn cemented into the femoral shaft with PMMA. Results 26 patients were followed up from 6 to 78 months with an average of 41 months. 3 cases died within 18 months following operation; 4 case sustained recurrences within 8 months postoperatively, and underwent re-resection; and the others were alive without diseases. All patients had no dislocation, loosening or breakage of prosthesis. X-rays showed the time of bone healing between allograft and host femur was an average of 7.4 months (range, 5 to 11 months), and the rate of bone healing was 92% (24/26). Absorption was seen around allograft greater trochanter of femur in 9 cases and at femoral condyle in 6 cases. 13 cases with total hip composite replacement had good active extension-flexion degrees of an average of 85?( range, 71? to 124?), and 13 with total knee composite replacement had a rang of motion of 92?(range, 68? to 135?). The postoperative average ISOLS scale score of 26 cases by Enneking's system was 26.2 point (range, 22 to 28). Conclusion The clinical result demonstrated the composite of massive frozen allograft and prosthesis replacement has the advantages of both allograft and prosthesis. Its best indications are that the patients with a major femoral defect from benign or low grade malignant tumor have longer survival expectancy.
5.Research progress of p70 ribosomal protein S6 kinase inhibitors.

p70 ribosomal protein S6 kinase (p70S6K), an important member of AGC family, is a kind of multifunctional Ser/Thr kinases, which plays an important role in mTOR signaling cascade. The p70 ribosomal protein S6 kinase is closely associated with diverse cellular processes such as protein synthesis, mRNA processing, glucose homeostasis, cell growth and apoptosis. Recent studies have highlighted the important role of S6K in cancer, which arose interests of scientific researchers for the design and discovery of anti-cancer agents. Herein, the mechanisms of S6K and available inhibitors are reviewed.

6.Analysis on prevalence and influencing factors of eye diseases in Chongqing area

Objective To understand the prevalence situation of common eye diseases and frequently‐occurring eye diseases in Chongqing area to provide the epidemic basis for the prevention and treatment of eye diseases .Methods Totally 80 384 people examined in the First Affiliated Hospital of Chongqing Medical University during 2013 were selected and performed the examination of ocular adnexa ,anterior eye segment ,vision ,color vision and fundus .Results Among 80 384 cases of physical examination ,16 897 cases of eye diseases were detected with the prevalence rate of 21 .03% ,males were more than females ,the difference showed the statistical significance(P<0 .05) .Top three identified diseases were refractive error (14 .21% ) ,cataract (7 .87% ) and eye ground arteriosclerosis (1 .70% ) .Age ,sex ,diabetes ,serum total cholesterol and triglyceride significantly affected the eye diseases (P<0 .05) .Conclusion Age ,sex ,diabetes ,serum total cholesterol and triglyceride are the risk factors influencing eye diseases .Control‐ling blood glucose and lipid has an important significance in preventing eye diseases .
7.Effects of Leukocyte-and Platelet-rich Fibrin on Tendon-bone Healing of Rabbits' Extra-articular Bone Tunnel in the Early Period

Objective To evaluate the early effect of leukocyte-and platelet-rich fibrin (L-PRF)on tendon-bone healing in rabbits' extra-articular bone tunnel.Methods Thirty healthy New Zealand rabbits were randomly divided into an experimental group and a control group,each of 15.The semitendinosus tendon harvested from the hind leg of each rabbit was prepared as free autografts.Each autograft was implanted into the bone tunnel created at ipsilateral proximal tibial metaphysis.The experimental group was added with autologous peripheral blood-derived L-PRF,while the control group did not add anything.Speciments from each group were harvested at 4,8 and 12 weeks after operation.The morphological changes were observed and evaluated at each time point.The fibroblasts on the tendon-bone interface were counted and analyzed under the high magnification microscope.Results The number of fibroblasts between the tendon and bone in the experimental group was significantly higher than that of the control group at each time point (P<0.05).And the connection between tendon and bone was closer and the collagen fibers were more regular in the experimental group than the control group at each time point.Sharpey's-like fibers which marked the early healing of the tendon to bone,were observed at 4th week in the experimental group,while they were observed at 8th week in the control group.As the healing time extended,Sharpey's like fibers continue to increase in both groups and significant differences were observed in the histological morphology (Buark grades)between the two groups (P<0.05).Conclusion Autologous peripheral blood-derived L-PRF can promote early healing of autologous tendon to bone in the extra-articular bone tunnel of model rabbit.
8.The characteristics of epithelial-mesenchymal transition in human prostate cancer stem progenitor cells

Objective To implore the characteristics of epithelial-mesenchymal transition(EMT) in human prostate cancer stem progenitor (S/P) cells isolated from LNCaP cell lines.Methods The S/P cells were obtained through florescence-activated cell sorting (FACS).Western blot and immunofluorescence assay were used to detect the S/P cells' EMT markers expression,such as E Cadherin,N Cadherin,Vimentin and Snail.Soft agar assay was used to detect the tumorigenesis ability of S/P cells.Cell migration assay was used to detect the migration ability of S/P cells.Results Compared with non S/P cells,the expressions of EMT markers,such as N Cadherin,Vimentin and Snail,were increased in S/P cells,while the expressions of epithelial marker and E Cadherin were decreased in S/P cells.After cultured for three weeks,S/P cells and non S/P cells both clonally grew.The colony numbers were (18.34±1.21) and (82.27±7.54),respectively (t =8.617,P =0.001).After cultured for 48 hours,the migration cells number was (25.33±5.13) in non S/P cells and (74.33±7.64) in S/P cells (t =7.953,P =O.001).Conclusions Human prostate cancer S/P cells isolated from LNCaP cell line have some characteristics of EMT,such as stronger tumorigenesis and migration ability,which could promote tumor invasion and metastasis.
9.Tightropeversus Endobutton in the reconstruction of the cruciate ligament

BACKGROUND:Because of its advantages, Endobutton has been widely used in clinic. Currently, its shortcomings are increasingly recognized. Tightrope that overcomes the shortcomings of Endobutton has been gradualy accepted by a doctor skiled in sports injuries.
OBJECTIVE: To analyze and compare the differences in the effects of Tightrope and Endobutton in the reconstruction of the cruciate ligament.
METHODS:Totaly 60 cases of anterior cruciate ligament rupture were selected and subjected to anterior cruciate ligament reconstruction under arthroscopy, of which 30 cases were randomly assigned to reconstruction by Endobutton device and 30 cases underwent reconstruction by Tightrope device. Al operations were performed by the same surgeon. Al patients were subjected to regular functional exercise and were folowed up regularly after operation. Effects of Tightrope and Endobutton in the cruciate ligament reconstruction were evaluated by comparing various indexes in the two groups.
RESULTS AND CONCLUSION:Compared to the Endobutton fixation system, the Tightrope fixation system could shorten the operation time, reduce the length of tendon incision, and decrease the loss of bone mass in the femoral bone tunnel. There were no significant differences in the maximum knee flexion, knee joint score and Tegner movement level score between the two groups at 3 and 6 months after operation. These findings indicate that the Tightrope fixation system is superior to the Endobutton fixation system, because it is more simple and convenient to operate and has less bone loss. However, their clinical efficacy has no difference after 6 months.
10.Detection of Th17 cells and interleukin-17 expression in peripheral blood from patients with atopic dermatitis

ObjectiveTo determine the protein and mRNA expression levels of interleukin-17 (IL-17) and proportions of Th17 cells in peripheral blood of patients with atopic dermatitis (AD),and to analyze their clinical significance.MethodsVenous blood samples were obtained from 63 patients with AD and 30 normal human controls.Enzyme linked immunosorbent assay was performed to measure the plasma level of IL-17 protein,real time fluorescence reverse transcription (RT)-PCR to detect the mRNA expression level of IL-17 in peripheral blood,flow cytometry to determine the percentage of Th17 cells in peripheral blood mononuclear cells(PBMCs).Spearman's rank correlation analysis was performed to assess the relationship of peripheral blood Th17 cell percentage and IL-17 levels with disease severity in patients with AD.Results The percentage of Th17 cells in PBMCs was 1.83% ± 0.47% in patients with acute AD,significantly higher than that in normal human controls (0.85% ± 0.45%,t =4.128,P < 0.01 ) and in patients with chronic AD ( 1.12% ± 0.69%,t =2.439,P < 0.05).There was a significant increase in the plasma level (98.37 vs.63.75 ng/L,U =168,P < 0.05) and mRNA expression level of IL-17 in peripheral blood in patients with AD compared with normal controls.Both the Th17 cell percentage and plasma IL-17 level were positively correlated with eczema area and severity index(EASI) score in patients with AD (r =0.681,P < 0.01;r =0.427,P < 0.05).ConclusionsThere is an elevated proportion of Th17 cells and an abnormal expression of IL-17 protein and mRNA in peripheral blood from patients with acute AD,which are positively correlated with disease severity,implicating the possible roles of IL-17 and Th17 cells in the pathogenesis of AD.
