1.Study on Mechanism of HL-60 Cell Apoptosis Induced by DMDAI-L

Objective: To explore the effect and mechanism of DMDAI-L in inducing the HL-60 cells apoptosis. Methods:Caspase-3 activity in HL-60 cells was measured with the enzymatic visible substrate DEVD-pNA. The fluorescence changes of mito-chondrial membrane potential (△Ψm) in HL-60 cells were investigated with the fluorescent probe JC-1. Results:The caspase-3 activ-ity was significantly increased in HL-60 cells after the DMDAI-L treatment at the concentration of 1. 25, 2. 5, 5, 10 and 20μg·ml-1 for 24h(P<0. 05). DMDAI-L could significantly reduce the mitochondrial membrane potential in HL-60 cells. Conclusion: The mechanism of DMDAI-L in inducing HL-60 cells apoptosis may involve the activation or regulation of caspase-3 activity as well as the reduction of mitochondrial membrane potential in HL-60 cells within certain concentration and time range.

Objective To solve the encountered problem of lack of space and dyskinesia of detector in dual energy X-ray absorptiometry system, Methods The method used PartitionMagic8.0 software to partition space in hard disk and added the load to keep the detector balance for moving, Results The original software can run in hard disk, and the pulleys are not gotten force from side. Conclusion The reason of problem is analyzed to find the method that can reduce the percentage for system failure and improve precision and accuracy of data.
3.Mechanisms of inhibition on human pancreatic cancer PC-3 cell line by a diterpenoid compound 5 F isolated from Pteris semi pinnata L treatment

Objective:To investigate the effect of 5F on proliferation and apoptosis of human pancreatic cancer.Methods:The pcDNA3.1-PUMAAS and pcDNA3.1 were transfected into AsPC-1 cells by Lipofectamine 2000 methods,stable transfected clony was chosen through G418.AsPC-1 cells were divided into three groups:transfected pcDNA3.1-PUMAAS,transfected pcDNA3.1 and control group without transfection AsPC-1 cells.Three groups was treated with serial concentrations(8.875,37.5,142?mol/L,respectively)of 5F 10?L.MTT assay was used to observe the inhibitory actions of 5F on AsPC-1 cells.The apoptotic rate of the cells was detected by flow cytometry.And the apoptosis was assessed by Hoechst 33258 dye staining and TUNEL staining.The expression of PUMA protein was detected by Western blotting and RT-PCR.Results:The inhibitory action on cell growth was seen in AsPC-1 control cells and pcDNA3.1-cells dealing with 5F.It could also promote the occurrence of apoptosis.5F inhibited the proliferation of AsPC-1 cells in a concentration-dependent manner.The apoptosis induced by 5F was accompanied with the up-regulation of PUMA.In cells treated with 5F,the apoptosis rate decreased greatly and proliferation rate increased compared with the AsPC-1 control cells and pcDNA3.1-cells dealing with 5F.The expression of PUMA was decreased greatly comparing with pcDNA3.1-cells treated with 5F,Conclusion:5F can depress the proliferation of AsPC-1 cell in vitro,mainly through the induction of apoptosis,and it was a potential agent for pancreatic cancer chemotherapy,the mechanism was probably related to its effect on the regulation of PUMA expression.

The objective of this paper is to find out a way that calculate the consumed peak value of oxygen in medical central oxygen supply system,which is the basis for discussion of purchasing hospital oxygen generating system.The author established a math model by poisson distribution to describe the consumed peak value of oxygen in medical central oxygen supply system to find out a convenience way to calculate the consumed peak value of oxygen.
5.Pressure-based Monitoring of Medical Central Compressing Air Supply System

Objective To determine the indexes for evaluating medical central compressing air sypply system.Methods According to the gas equation,pressure calculation was perfomed with the pressure variation as the monitoring subject.Results Such parameters could be used for monitoring medical central compressing air supply system as as average velocity & instanteous velocity of gas consumption,air compressor's average exhaust velocity & exhaust velocity at start-stop point,and pressure-exhaust velocity curve of air compressor.Conclusion The method can be applied to enhancement of quality management & maintenance of the equipment.

7.Effect of the area of paries medialis defect on the stability of fracture ends after percutaneous compression plating for femoral intertrochanteric fracture

Objective To analyze the effect of different areas of paries medialis defect of femoral trochanter on the stability of fracture ends after percutaneous compression plating for femoral intertrochanteric fracture.Methods Thirteen preserved cadaveric specimens of adult femur were used in this experiment.The age of death ranged from 30 to 50 years.Oblique osteotomy was conducted under the same condition using a wire saw to create experimental models of fracture of Evans-Jensen type Ⅳ.The ratios of the area of paries medialis defect to the projected area of lesser trochanter in the specimens 1 to 13 were set respectively as 0,10.0%,14.5%,20.2%,23.4%,30.6%,45.8%,56.8%,76.8%,88.7%,99.8%,121.3% and 149.4%.All the fractures were fixated by standard percutaneous compression plating.Cyclic load was applied on each specimen using Instron 2000 universal material tester.A vertical load of 600 N was applied for respectively 1,10,100 and 1,000 times to measure the displacements of proximal and distal fracture ends.Results When the ratio of the area of paries medialis defect to the projected area of lesser trochanter was 30.6%,the average displacements of the proximal and distal ends were 0.14 cm and 0.10 cm.When the ratio was 45.8%,the average displacement of the proximal end was 0.53 cm,278.6% greater than that for 30.6%;the average displacement of the distal end was 0.41 cm,310.0% greater than that for 30.6%.When the ratio was 149.4%,the average displacement of the proximal end was 0.93 cm,564.3% greater than that for 30.6%;the average displacement of the distal end was 0.65 cm,550.0% greater than that for 30.6%.Conclusions When the ratio of the area of paries medialis defect to the projected area of lesser trochanter reaches > 45.8%,the area of paries medialis defect will have a significant effect on the stability of fracture ends after percutaneous compression plating for femoral intertrochanteric fracture.The displacement will increase linearly under the same compression with the increase in defect area.
8.Changes of visual acuity and visual evoked potentials before and after gene therapy for Leber hereditary optic neuropathy

Objective:To observe the changes of vision and visual evoked potentials (VEP) in patients with Leber hereditary optic neuropathy (LHON) before and after gene therapy.Methods:A retrospective cohort study. From December 2017 to October 2018, 35 cases of 70 eyes of m11778G.A/MT-ND4 mutant LHON patients who were diagnosed in the Tongji Hospital of Huazhong University of Science and Technology and received gene therapy were included in the study. There were 30 males (87.71%) and 5 females (12.29%), with the mean age of 23.31±6.72 years. The gene therapy method was intravitreal injection of rAAV2-ND4 (recombinant adeno-associated virus carrying NADH-ubiquinone oxidoreductase subunit 4 gene) into one eye. The eye with poor visual acuity was chosen as the injection eye. If both eyes had the same visual acuity, the right eye was designated as the injection eye. Seventy eyes were divided into the injected eye group and the non-injected eye group, in which were both 35 eyes. The best corrected visual acuity (BCVA) and pattern VEP (PVEP) examinations were performed in the injected eye group and the non-injected eye group before treatment (baseline), 1, 3, and 6 months after injection. Compare the changes of BCVA and PVEP between the injected eye group and the non-injected eye group at baseline, 1 month, 3 months, and 6 months after injection. Independent sample t test, paired sample t test or two independent sample nonparametric test were performed to compare the two groups. Results:Compared with baseline, 1, 3, and 6 months after treatment, the BCVA of the injected eye group ( t=3.530, 4.962, 5.281; P=0.001, 0.000, 0.000) and the non-injected eye group ( t=3.288, 2.620, 2.252; P=0.002, 0.013, 0.031) increased, and the difference was statistically significant; there was no statistically significant difference between VEP IT ( tinjected eye group=-0.158, 1.046, -1.134; Pinjected eye group = 0.875, 0.303, 0.190; tnon-injected eye group=0.773, -0.607, -0.944; Pnon-injected eye group = 0.445, 0.548, 0.352) and VEP A ( Zinjected eye group=-0.504, -0.934, -1.065; Pinjected eye group = 0.614, 0.351, 0.287; Znon-injected eye group=-0.521, -0.115, -0.491; Pnon-injected eye group = 0.602, 0.909, 0.623). Conclusion:After gene therapy, the visual acuity of the injected and non-injected eyes of LHON patients improved; PVEP did not change significantly, and remained stable compared with baseline.
9.Effects of salmeterol and fluticasone propionate combined montelukast in the treatment of patients with bronchial asthma

Objective To compare the effects of salmeterol and fluticasone propionate combined montelukastin the treatment of patients with bronchial asthma.Methods Ninety-eight bronchial asthma patients from April 2014 to March 2016 in the Fourth People's Hospital Sichuan Province were selected anddivided into observation group and control group with 49 patients in each group according to the above principles by prospective study.The control group was given salmeterol treatment,and the observation group was added given montelukast treatment based on the control group.The treatment days were 15 d recorded the prognosis of the two groups.Results The total effective rates of the observation group and the control group were 95.9% and 81.6% respectively,the total effective rate of the observation group was significantly higher than that of the control group (P < 0.05).The FEV1 and FVC values after treatment in the observation group and control group were significantly higher than before treatment (P < 0.05),while the FEV1 and FVC values in the observation group after treatment were significantly higher than those in the control group (P < 0.05).The scores of ACT scale after treatment of two groups were obviously higher than before treatment,and the score of observation group was statistically higher than control group after treatment (P < 0.05).During treatment,there was no statistical significance on adverse reaction between two groups.Conclusion Salmeterol and fluticasone propionate combined montelukast in the treatment of patients with bronchial asthma can promote the improvement of lung function,so as to improve the quality of life and clinical treatment.
10.Research of the mechanism of pre-B-cell colony-enhancing factor in pulmonary vascular endothelial permeability increasing after CPB

Objective To investigate the mechanism of PBEF in pulmonary vascular endothelial permeability increasing after car-diopulmonory bypass ,in order to provide the basis for a better lung protective measures during cardiopulmonary bypass .Methods Animal models were established ,group A :the rats were transfected with the lentiviral AD-PBEFshRNA ;group B :the rats were took 30 min deep hypothermic circulatory arrest ;group C :the rats were took 30 min deep hypothermic circulatory arrest ,then trans-fected with the lentiviral AD-PBEFshRNA ;control group :the rats were anesthetized and established CPB tube ,without CPB by-pass .Lung tissue was detected with Western blotting and ELISA .Results PBEF ,phosphorylation of P38MAPK ,ERK ,MLC ,VE-cadherin ,FAK in group C had significant difference with group A ,B and the control group(P<0 .05) .VEGF ,MMP2 ,MMP9 ,W/D in group C had significant difference with group A ,B and the control group(P<0 .05) .The pathological results showed that rat lung tissue of the control group was normal ,while in group C ,it had severe pathological damage ,the pathological damage degree in group A ,B reduced compared to group C .Conclusion PBEF can through MAPK/PI3K-Akt/VEGF signaling pathway ,increase pulmonary vascular endothelial permeability .
