Objective To evaluate the role of δ receptor in reduction of hypoxia-reoxygenation (H/R) injury to cardiomyocytes by morphine preconditioning in rats with chronic heart failure. Methods Adult male Sprauge-Dawley rats weighing 220-250 g were used in the study.Chronic heart failure was induced by injection of adriamycin 2 mg/kg via the tail vein once a week for 6 weeks.Their hearts were removed 2 weeks after the last injection and the cardiomyocytes were isolated and cultured. The cells were randomly divided into 5 groups ( n =8 each):control group (group C); H/R group; morphine preconditioning group (group MPC); morphine preconditioning + naloxone (opioid receptor antagonist ) group (group MPC + Naloxone) ; morphine preconditioning + naltrindole ( δ receptor antagonist) group ( MPC + Naltrindole group). The cells were cultured in normal culture atmosphere in group C and were exposed in hypoxic air for 3 h followed by 1 h reoxygenation in the other groups.Morphine preconditioning was performed immediately before hypoxia in group MPC.Naloxone and naltrindole were added before morphine preconditioning in groups MPC + Naloxone and MPC + Natrindole respectively. At 1 h of reoxygenation, the cell viability ( by MTT assay),activities of lactate dehydrogenase ( LDH ) and creatine kinase (CK),and cell apoptosis were detected. The apoptotic rate was calculated. Results The cell viability was significantly lower, and the activities of LDH and CK and apoptotic rate were significantly higher in groups H/R,MPC + Naloxone and MPC + Natrindole than in group C (P < 0.05).The cell viability was significantly higher, and the activities of LDH and CK and apoptotic rate were significantly lower in group MPC than in group H/R ( P < 0.05).Conclusion Morphine preconditioning reduces H/R injury to cardiomyocytes through activating δ receptor in rats with chronic heart failure.

The gene nhaA encodes functional protein that may play an important role in salt tolerance of Escherichia coli In order to study bacteria salt tolerance, a pair of primers were designed according to public nhaA sequence and was used to amplify 1 1kb DNA fragment with PCR The nhaA gene from E coli DH5?was cloned into a T vector and sequence analyses reveal that the cloned fragment contains entire nhaA gene coding region To apply the method of homology analysis, the result shows that many kinds of bacterium have nhaA gene, such as E coli K12, E coli O157:H7, Salmonella typhimurium and Salmonella enterica , et al This analysis suggests that nhaA gene lie generally in bacterial; and it has intimate relation with salt tolerance of E coli that may be of great importance in improvement of the salt tolerance of plant

Chinese Pharmaceutical Journal 2018;53(4):249-257
OBJECTIVE: To study the synthesis and activities of AHPN derivatives. METHODS: Starting from p-bromophenol and 1-adamantanol, a series of AHPN derivatives were synthesized by substitution reaction, condensation reaction, oxidation reaction and reduction reaction. These new compounds were characterized by 1H-NMR, 13CNMR and HR-MS. Biacore technique was used to test the derivatives' combining activities with RARγ. RESULTS: Four compounds, 7c, 6c, 6e, and 6h, exhibited significant combining activities with RARγ compared with AHPN. The introduction of phosphoric acid groups and nitrogen heterocyclic ring increased the activities of these compounds. CONCLUSION: Compounds 7c, 6c, 6e, and 6h show significant combining activities with RARγ, which are worthy of further study.

Objective To investigate the physicochemica l properties and immunobiological activity of a polysaccharide (RAP-B-1) from stems of Rubus amabilis. Methods The crude polysaccharide (RAP) was obtained successively by boiling, ethanol precipitating and dialyzing. RAP was isolated with DEAE-cellulose and Sephadex G-100 to obtain a polysaccharide RAP-B-1. The physicochemical properties of RAP-B-1 were studied by hydrolysis, periodate oxidation, Smith degradation and methylation, CE, IR, NMR and GC-MS. The immunobiological activities were estimated by the proliferative activity of spleen lymphocytes and phagocytic activity of peritoneal macrophages in mice. Results The molecular weight of RAP-B-1 was 4.80×104 with specific optical rotation value [α] 20D+68.3 (c=1,H2O), and was composed of eight monosaccharides. The molar ratios were as Xyl: Ara: Glc: Rha:Gal: Man: GlcA: GalA = 1.0:6.9:0.8:1.1:6.9:0.3:0.5:3.3. RAP-B-1 was an arabinogalactan. The linkages of arabinose were →1) Ara (2,3→,→1) Ara(5→and→1) Ara, and the linkages of galactose were→1) Gal(4→,→1) Gal(6→and→1) Gal. RAP-B-1 could improve the proliferative activity of spleen T cells(P<0.05) and booste phagocytic activity of peritoneal macrophages at 50μg/ml concentration(P<0.01). Conclusion RAP-B-1 is an arabinogalactan and has immunobiological activity.

Objective To observe the efficacy and safety ofYiqi Huatan Granules combined with carbocisteine to reduce the acute exacerbation of chronic obstructive pulmonary disease (COPD). Methods Totally 108 patients in the stationary phase of COPD with qi deficiency or merging phlegm blockage/blood stasis syndrome were randomly divided into two groups. Patients in the control group were treated with carbocisteine and inhaled bronchodilators on-demand, while patients in the treatment group were treated withYiqi Huatan Granules combined with carbocisteine and bronchodilators for 24 weeks. Times and severity of acute exacerbations of COPD, TCM syndrome scores, lung function, and 6-minute walking distances of the two groups during the treatment period were observed. Results The average times of acute exacerbations of COPD in the treatment group (0.33 ± 0.55) were fewer than the control group (0.62 ± 0.71);the average times of severe acute exacerbations of COPD in the treatment group (0.12 ± 0.33) were fewer than the control group (0.28 ± 0.45), with statistical significance (P<0.05). Comparison between the TCM syndrome scores before and after treatment, TCM total syndrome scores, breathless/dyspnea score, breath shortness and fatigue score, and 6-minute walking distance in the treatment group were better than the control group (P<0.05), with better safety. Conclusion The treatment ofYiqi Huatan Granules combined with carbocisteine for COPD patients can effectively decrease times and severity of acute exacerbations of COPD, and improve TCM syndrome scores and 6-minute walking distance.

Objective To assess apoptosis mediated by the cell death receptor Fas in peripheral T lymphocytes of patients with abdominal aortic aneurysm. Methods The apoptotic pathway was triggered by anti-Fas monoclonal antibody in cultured and activated peripheral T-cells from 20 AAA patients. Control groups consisted of 15 patients with aortic atherosclerotic occlusive disease(AOD)and 25 healthy individuals. Cell survival and death rate were assessed. Results Cross-linkage of Fas receptor exerted a strong apoptotic response on T cells from AOD patients and healthy controls, while the effect on T cells was very limited from that of AAA patients. The evaluation of cell Survival rate showed a significantly higher percentage in AAA group(98.9%±10.3%)than in the AOD subjects(58.9%±15.2%)or the healthy group(59.4%±12.9%;P<0.001=.Apoptosis assessment by annexin V and propidium iodide staining and flow cytometry showed similar results. The defect in AAA group was not due to decreased fas expressed at normal levels. Moreover,it specifically involved the Fas system because cell death was induced in the normal way by methylprednisolone. Conclusions Fas-induced apoptosis in activated T cell from AAA patients is impaired. This may disturb the normal down-regulation of the immune response and thus provide a new insight into possible mechanisms and routes in the pathogenesis of AAA.
