1.Analysis of cardiovascular events and risk of tumor in aged patients with diabetes mellitus

Objective:To observe cardiovascular events and risk of tumor in aged patients with diabetes mellitus (DM) .Methods :A total of 2743 DM patients ,who visited to our department of endocrinology from 2004 to 2010 , were enrolled as DM group ,another 2642 people without glucose metabolic disorder ,who visited to our hospital dur‐ing the same period with the same age ,were regarded as normal control group .Incidence rates of cardiovascular dis‐eases and morbidity of malignant tumor ,levels of tumor markers were compared between two groups (both groups were followed up for 2~7 years) .Results:Follow‐up found that cardiovascular disease rate in DM group was signif‐icantly higher than that of normal control group (21.47% vs .6.43% , P<0.05 );tumor marker CA19‐9 level in DM group was significantly higher than that of normal control group [ (17.89 ± 3.61) kU/L vs .(9.78 ± 2.64) kU/L ,P=0.001] ,and incidence rate of malignant tumor was significantly higher than that of normal control group (18.34% vs .8.18% , P=0.001) .Conclusion:Incidence rate of cardiovascular events and tumor in aged patients with diabetes mellitus is significantly higher than that of people without glucose metabolic disorder of same age .
2.Clinical study of pelvic autonomic nerve preservation in curative excision of male patients with rectal cancer

Objective To evaluate the effects of pelvic autonomic nerve preservation (PANP) during the radical resection of rectal carcinoma on the sexual and urinary function of male patients.Methods The sexual and urinary dysfunction rates from 45 male patients who undergone PANP and 45 control patients who did not undergo PANP during radical resection of rectal carcinoma were analyzed.Results In the PANP group,the incidences of erectile dysfunction,ejaculatory dysfunction and urinary dysfunction were 26.67%,24.44% and 28.89%,respectively.But in control group,the above three rates were 57.78%,60.00% and 62.22% in order.The rates of these three dysfunctions significantly were different between the two groups ( x2 =8.92,11.66,10.08,P < 0.01 ).Conclusion PANP during radical resection of rectal carcinoma could reduce the post-operative sexual dysfunction and urinary dysfunction.
3.Effect of methazolamide in patients with refractory uveitic macular edema

AIM:To evaluate the efficacy and safety of methazolamide in treating refractory uveitic macular edema.METHODS: Retrospective self-controlled study was designed.A total of 15 patients (20 eyes) with refractory uveitic macular edema which used methazolamide as adjuvant therapy were enrolled in Shanghai First People`s Hospital from January 2015 to June 2016.The changes of central macular thickness (CMT) and best corrected visual acuity (BCVA) were observed at baseline and 2, 4, 8wk after treatment.We also focused on the incidence of complications and relapse.RESULTS: The CMT was 445.95±154.10μm, 338.83±138.34μm, 251.50±40.20μm, 244.90±35.68μm at baseline, 2, 4 and 8wk after treatment, respectively.The differences among them were statistically significant (F=15.467, P<0.05).The BCVA (log MAR) were 0.40±0.17, 0.28±0.21, 0.19±0.20, 0.18±0.21 at baseline, 2, 4 and 8wk respectively, with a significant difference among them (F=5.208, P<0.05).When the cumulative dose reached to 700mg and 1400mg, no one had methazolamide-related complications;and when it came to 2800mg, 5 patients (33%) had methazolamide-related complication.After the withdrawal of methazolamide 1wk, 1 and 3mo, 3 patients (20%), 5 patients (33%) and 8 patients (53%) relapsed, respectively.CONCLUSION: Methazolamide is beneficial in improving macular edema and vision in 4wk.When the cumulative dose is more than 1400mg, we need pay attention to the complications.After discontinuing methazolamide for 1wk, macular edema relapsed in some patients, and more than half of patients recurred after 3mo.So the patients should be followed closely in 3mo after withdrawal of methazolamide.
5.Executive control for methadone maintenance patients:an event-related potential study

Objective To discuss the neuropsychological mechanism of executive control for methadone maintenance patients(MMP).Methods Using the event-related potentials (ERPs) technique to reveal the time course of information processes with high temporal resolution,and exam the function of attention monitoring and response inhibition in heroin-dependent MMP.Results (l) The behavioral results: there were significant withinsubjects Stroop interference effects for the MMP and control groups (reaction time:control group' s congruent condition(766.57±75.64) ms,incongruent condition(879.52±62.31) ms,MMP group' s congruent condition (821.89±64.44) ms,incongruent condition (906.29 ± 69.46) ms,P< 0.001 ; error rate: control group' s congruent condition (4.15± 2.92) %,incongruent condition (8.70 ± 6.12) %,P< 0.001,MMP group' s congruent condition (12.07 ±10.80) %,incongruent condition (16.60±12.43)%,P<0.01).(2)ERPs data showed that MMP demonstrated significantly smaller incongruent-N2/N450/SP amplitudes than controls,and all disappearing incongruent effects in N2,N450 and SP,comparing statistically significant incongruent effects for controls in N2,N450 and SP.In MMP group,the amplitudes of N2 and N450 incongruent condition (N2 (1.40± 2.91) μV、N450 (1.29 ± 0.55) μV) were bigger than congruent condition amplitudes(N2(0.77±3.61) μV 、N450(0.83± 1.07) μV,P<0.05),but the amplitude of SP (0.37±3.58)μV was smaller than congruent condition(1.53±3.05) μV,P<0.001 ;in control group,the amplitudes of N2 and N450 incongruent condition((-0.30±3.45) μV,N450(1.77± 1.55)tμV) were smaller than congruent condition (N2(1.10±3.64) μV,P<0.001; N450(2.37±2.12) μV,P<0.05),the amplitude of SP ((1.93±1.65) μV) was bigger than congruent condition((0.98±2.02)μV,P<0.01).Conclusion There are impaired executive control functions during the conflict monitoring process and the conflict resolution process for MMP.These results provide the neural electrophysiological evidence to explain relapse behaviors in methadone maintenance patients.
6.Molecular biology of 5-lipoxygenase

5-lipoxygenase (5LO) is the key enzyme in the synthesis of leukotrienes, which are potent biological mediators of inflammation and anaphylaxis.After lock out of the 5LO gene by homologous recombination in embryonic stem cells, 5LO deficient-mice can develop normally and stay healthy, with a selective opposition to certain inflammatory insults. The cellular distribution of 5LO in unstimulated cells among different cell types is different; it can present in the cytosol, the nucleus, or both. 5LO was also found capable of shuttling in and out of the nucleus under certain conditions. It is suggested that 5LO might have other functions besides its traditional catalytic role.
8.Influence of radiosensitivity by mild hyperthermia in a pulmonary adenocarcinoma cell line

Objective:To investigate the radiosensitization and the cell-cycle of mild hyperthermia(≤42℃)on human pulmonary adenocarcinoma cell line SPC-A-1 in vitro. Methods: The human pulmonary adenocarcinoma cell line SPC-A were treated with radiation and the combination of radiation with mild hyperthermia. Radiosensitivity was determined by clonogenic assay and quantified by calculating the thermal enhancement ratio (TER). Flow cytometry was used to observe the cell-cycle. Results: Do, Dq calculated from the dose-response curve for radiation combined with 41.5℃ were 1.390 Gy, 1.426 Gy, whereas 1.693 Gy, 2.453 Gy for radiation alone, respectively. TER was 1.218. The proportion of cells in S phase was found to be 14.81% in the radiation group. The values, after 48 hours and 72 hours, with 6Gy radiation combined immediate 41.5℃ one hour mild hyperthermia, were 5.89% and 9.08%, respectively, versus 18.8% and 31.91% with 6 Gy radiation alone. Conclusion:Radiosensitization of mild hyperthermia in SPC-A-1 cells associated with the hyper-radiosensitization of the cells in S phase.
9.Effect of continuous adenosine infusion on mesentery microcirculation during cardiopulmonary resuscitation in rats with asphyxia

Objective To evaluate the effect of continuous adenosine infusion on mesentery microcirculation during cardiopulmonary resuscitation in rats with asphyxia. Methods Rat model of asphyxia was established in 22 healthy Wistar rats. The animals were then randomly divided into normal saline group (group A, n=10), epinephrine group (group B, n=6) and epinephrine plus adenosine group (group C, n=6). After a 3-min asphyxia without intervention, cardiopulmonary resuscitation (CPR) was started. The cardiac compression was carried out with an electric cardio-pulmonary resuscitation machine (200 times/min). Respiration was restored and maintained with a ventilator with tidal volume of 4ml, breathing rate 50times/min and FiO2 100%. After a 4-min CPR, rats in group A and group B were given normal saline and epinephrine (bolus of 90?g/kg) respectively, and in group C epinephrine (bolus of 90?g/kg) plus 70?g/kg adenosine were given. Electric defibrillation would be initiated if there was ventricular fibrillation. The reperfusion rate of mesentery arterioles and venules, diameter of blood vessels and relative blood velocity were observed. 10.Inducing Mesenchymal Stem Cells from Adult Human Adipose Tissue to Differentiate into Cardiomyocytes In Vitro

ObjectiveTo study the differentiation of adipose tissue-derived mesenchymal stem cells (ADMSCs) into cardiomyocytes in vitro. MethodsADMSCs were isolated and purified by the method of digesting and adhering to the culture plastis. The third generation of cells were determined with immunocytochemistry method and induced by 5-aza. On the 7th, 14th, 21st, 28th day after being induced, the surface antigen of myoblast cell and cardiac myocyte were determined, the expression of gene of GATA4 and Nkx2.5 were tested, and the content of atrial natriuretic polypeptide (ANP) assayed. ResultsImmunocytochemical staining showed CD44, CD13, CD105 positive, CD45, CD34, HLA-DR, factor Ⅷ negative. After being induced by 5-aza, the direction of cell arraying was gradually similar. The cells were stained positively for Desmin, α-Sarcomeric Actin, myosin heavy chain and Troponin T. The cells express GATA4 and Nkx2.5 and secrete ANP. ConclusionThere was mesenchymal stem cells in human adipose tissue. 5-aza may induce ADMSCs to differentiate into cardiomyocytes in vitro.
