1.Establishment of atherosclerosis models induced by feeding high-fat diet plus arterial intimal injury of the ventral aorta with balloon in rabbits

BACKGROUND:Studies regarding atherosclerosis models preparation commonly established by feeding high-fat diet or arterial intimal injury to healthy or hyperlipoidemia animals.OBJECTIVE:To establish atherosclerosis models induced by feeding high-fat diet combined with arterial intimal injury of ventral aorta with balloon.DESIGN,TIME AND SETTING:A comparative observation experiment was performed at the Animal Experimental Center of Chinese PLA General Hospital from January to March 2007.MATERIALS:Twenty New Zealand rabbits were randomly divided into high-fat diet,and high-fat diet combined with arterial intimal injury groups,with 10 animals in each group.High-fat diet was composed of common forage,4% cholesterol,10% pork fat and 10% egg yolk powder.METHODS:Rabbits in the high-fat diet group were fed with high-fat diet.Animals in the high-fat diet combined with arterial intimal injury group were underwent operation of sacculus injury after fed with high-fat diet for 4 weeks.MAIN OUTCOME MEASURES:Arteriae aorta was harvested after 12 weeks of operation and received pathological examination,thickness ratio of fatty streak and plaque,intima,and tunica media was calculated.Serum total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL) and low density lipoprotein cholesterol(LDL) was measured.RESULTS:After 12 weeks,the levels of TC,TG,HDL and LDL were increased,which was more obviously in the high-fat diet combined with arterial intimal injury group(P

Objective To explore the relations between UA and serum hs-CRP, the effects of early intervention with atovastatin on serum hs-CRP in patients with UA and its clinical significance in the early management of UA. Methods T 60 patients with UA (UA group),53 patients with stable angina (SA group) and 50 healthy controls (control group) were enrolled to the study. The serum hs-CRP levels were measured by particle enhanced immunoturbidimetric assay. UA group were randomly assigned to the atovastatin group and the routine group for a 4-week treatment immediately after admission. Selected coronary artery angiography was performed in 78 patients and 29 healthy controls with Judkin's technique. 4.Clinical study of anti-neurohormone drugs on heart function and the plasma concentrations of neurohormone of patients with different grade heart function after myocardial infarction

ObjectiveTo observe the effects of anti-neurohormone drugs on ventricular remodelling and the plasma concentrations of neurohormone of patients with different grade heart function after myocardial infarction.MethodsAccording to ejection fraction of patients,103 consective patients with myocardial infarction were divided into two groups:normal heart function group:ejection fraction ranged from 50% to 70%.Abnormal heart function group:ejection fraction was less than 50%.All patients were given the same beta-receptor blocking agent and angiotensin-converting enzyme inhibitor and then were followed up 6 months.Heart function and the plasma concentrations of neurohormone were compared before and after anti-neurohormone drugs were given;at the same time,changes of heart function and the plasma concentrations of neurohormone were compared in statistical aspect between two groups.ResultsThe improvement of heart function and change of the plasma concentrations of neurohormone were significantly different in statistics before and after drugs were used.Besides,the patients with different grade heart function had significantly different changes of heart function and plasma concentrations of neurohormone.ConclusionAnti-neurohormone drugs can significantly reduce the level of the plasma neurohormone,improve heart function and inhibit heart remodeling.What’s more,there are better treatment effects in patients with heart failure.
5.Color Doppler ultrasound in diagnosis and localization of scrofula in operation

Objective To explore the two-dimensional images features and color flow characteristics of scrofula, as well as their application in the localization of scrofula. Methods A total of 110 cases of the scrofula confirmed by operation were retrospectively analyzed. Two-dimensional images and color flow characteristics, the relationship with vascular traveling and the invasion site were recorded. Results Lymph nodes of the lateral neck region along with large vessels were most commonly involved, then followed the submaxillary lymph nodes and the lymph nodes anterior neck longitudinally. There might be multiple lesions behaving differently on two-dimensional images and with different color flow characteristics. Conclusion Most of scrofula can be diagnosed clearly according to the two-dimensional images and color flow characteristics. The description of relationship between involved lymph nodes with vascular, the depth and position of scrofula make the operation safety.
6.Allogeneic bone marrow mesenchymal stem cell transplantation for acute myocardial infarction in rats

BACKGROUND:A number of studies have shown that bone marrow mesenchymal stem cells can survive in the infarcted myocardium and improve cardiac function. OBJECTIVE:To investigate the effects of al ogeneic rat bone marrow mesenchymal stem cells on heart failure in acute myocardial infarction models of rats and possible mechanisms. METHODS:Rat bone marrow mesenchymal stem cells were isolated from the bone marrow of 39 male Wistar rats by density gradient centrifugation with Percol . After ligating anterior descending coronary artery, 39 female Wistar rats were randomly divided into three groups:control group (Dulbecco’s modified Eagle’s medium, n=12), mesenchyma stem cells group (n=15) and mononuclear cells group (n=12). Eight weeks later, hemodynamics and left ventricular function were measured. Histopathological and immunohistochemical examinations were performed. RESULTS AND CONCLUSION:Compared with the control group, left ventricular end diastolic pressure, left ventricular relative weight, the col agen volume fraction of type I and type III in the infarction zone of the left ventricle were al significantly decreased, in contrast to ±dp/dtmax,-dp/dtmax/left ventricular systolic pressure, body weight and vascular density in infarction zone were al significantly increased both in mesenchymal stem cells group and mononuclear cells group. There were no significant differences between two treatment groups except for interventricular septal thickness and vascular density in non-infarction zone. 5-Bromo-2'-deoxyuridine positive cells were observed in the infarction area of mesenchyma stem cells group but no positive cells in mononuclear cells group. Some bal-like cellmasses were found positively stained with desmin and cardiac troponin T. Results have suggested that embedded bone marrow mesenchymal stem cells survived in exogenous host hearts. The therapy of mononuclear cells and mesenchymal stem cells could limit the left ventricular remodeling after acute myocardial infarction and improve left ventricular function through angiogenesis inducing and col agen deposition decreasing.
7.DCE-MRI assessment of adjacent vertebral blood perfusion of degenerated lumbar discs

Objective To analyze the correlation of intervertebral disc degeneration with the blood perfusion of its adjacent border of vertebral body by dynamic contrast‐enhanced MRI (DCE‐MRI) .Methods 3 .0T DCE‐MRI examinations were performed in 15 patients (50 discs) .Intervertebral discs from L1 to L5 were classified to normal group(17 discs) and degeneration group(33 discs) based on the Pfirrmann grading system .The DCE‐MRI pharmaeokinetic parameters of the region of interest(ROI):volume transfer constant (Ktrans ) ,blood volume(BV) and blood flow (BF) were obtained .Independent sample t test was used to compare normal group and de‐generation group .Results In normal group (n=17) ,the mean Ktrans ,BV and BF were (0 .036 2 ± 0 .011 2)min-1 ,(4 .81 ± 1 .17) mL/100 mL ,(5 .68 ± 1 .55) mL · min-1 · 100 mL -1 for the superior border of vertebral body ,and (0 .042 ± 0 .012 9)min-1 ,(5 .03 ± 0 .797) mL/100 mL ,(6 .02 ± 1 .28) mL · min-1 · 100 mL -1 for the inferior border of vertebral body .In degeneration group (n=33) ,the mean Ktrans ,BVand BF were (0 .030 4 ± 0 .011 2)min-1 ,(4 .22 ± 1 .03) mL/100 mL ,(4 .73 ± 1 .38) mL · min-1 · 100 mL-1 for the superior border of vertebral body ,and (0 .033 8 ± 0 .012 5) min-1 ,(4 .07 ± 0 .940) mL/100 mL ,(4 .72 ± 1 .43) mL · min-1 · 100 mL -1 for the inferior border of vertebral body .By statistical analysis :(1)There were no significant differences between superior border and inferior border of vertebral body in Ktrans ,BV and BF in normal group(t= -0 .98 ,-0 .479 ,0 .533 respectively ,P>0 .05 ) .(2)There were no significant differences between superior border and inferior border of vertebral body in K trans ,BV and BF in degeneration group(t= -0 .906 ,0 .497 ,0 .013 respectively ,P>0 .05) .(3)There were significant differences between normal group and degeneration group in Ktrans ,BV and BF(t= -2 .26 ,-3 .01 ,-2 .99 respectively ,P<0 .05) .Conclusion The bone marrow per‐fusion in degeneration group is decreased when compared to normal group with dynamic contrast‐enhanced MRI .Intervertebral disc degeneration correlates with the loss of its blood supply .
8.Establishment of chemiluminescence immunoassay for AngiotensinΙ

Objective:The chemiluminescence immunoassay for AngiotensinⅠ ( AngⅠ ) was developed by the competition method. The renin activity was calculated by the determination of Ang Ⅰ. Methods: AngⅠ antigen was labeled with biotin and fluorescein labeled AngⅠantibody,and anti fluorescein antibody was used to coated with the microporous plate . Results:The standard range of the method was 100-7 800 pg/ml. The assay sensitivity was 24. 3 pg/ml. The intra- and inter-assay coefficients of variance were 5. 6%-8. 7% and 6. 8%-10. 4% respectively. Analytical recovery was 95. 4%-105. 3%. The correlation coefficients between measured and expected values were 0. 999 after serial diluted. Compared with radioimmunoassay kit,the correlative equation was y=0. 95x+235. 70,correlation coefficient was 0. 973. Coated microporous plate and biotin labeled Ang Ⅰ antigen,and fluorescein labeled Ang Ⅰantibody at 37℃ for a week with good stability. Conclusion: The results of the method were in accord with the basic requirements of immunoassay.
9.Effect of Yigan Granules on Bile Secretion in Rats

Objective To study the effect of Chinese Herb of Yigan Granules in different dosages on bile secretion in rats.Methods The rats were randomly divided into 5 groups: blank control group,positive control group and high,mod erate,low dosages of Yigan Granules groups.The medicines were administered thr ough duodenal intubation for the rats.Then the volume of bile were gathered by common bile duct intubations in different time,and the volume of bile and the contents of bilirubin in bile were tested before and after given drugs.Results Yigan Granules increased the bile secretion in a dose-dependent manner and promote the secretion of bilirubin.Conclusion Chinese Herb of Yigan granules has a choleretic effect.
10.Determination of Astragaloside in Bushen Wenfei Mixtures by HPLC-ELSD

Objective:To establish the content determination method for astragaloside in Bushen Wenfei mixtures by HPLC-ELSD. Methods:HPLCwithanAgilentZORBAXC18(150mm×4.6mm,5μm)columnwasused. Methanol-water(75∶25)wasusedasthe mobile phase and the flow rate was 0. 8 ml·min-1 . The drift tube temperature was 80℃ and the gas flow rate was 2. 5 ml·min-1 . Results: The linear range of astragaloside was 0.297-4.944 μg (r =0.999 7). The average recovery was 98.13% with RSD of 1. 27%(n=9). Conclusion:The method is with sufficient accuracy, stability and repeatability, and can be used in determination of Bushen Wenfei mixtures.
