1. The association between a genetic variant in MKK4 gene promoter and lung cancer risk in Southern Chinese population
Tumor 2011;31(12):1082-1086
Objective: To investigate the association between the single nucleotide polymorphism (SNP) of mitogen-activated protein kinase kinase 4 (MKK 4) promoter and the risk of lung cancer development in Southern Chinese population. Methods: A hospital-based case-control study including 800 cases of lung cancer and 900 healthy controls was conducted. TaqMan assay was used to test the SNP of rs3826392 (-1304T>G) in MKK 4 promoter, and the software SAS 9.13 was used to analyze the association of MKK4 polymorphism and the susceptibility of lung cancer. Results: The observed genotype frequency of -1304T>G in MKK4 promoter was appropriate for Hardy-Weinberg equilibrium in the healthy controls (P = 0.149). The distribution of genotypes was significantly different between the cases of lung cancer and the healthy controls (P=0.001). Compared with homozygous genotype (TT), the risk for lung cancer was decreased by 25% in the carriers of heterozygous genotype (TG) [adjusted odd ratio (OR ) = 0.75, 95% confidence interval (CI) = 0.58-0.97)], and the risk for lung cancer in carriers of GG homozygote was decreased by 45% (adjusted OR = 0.55, 95% CI = 0.33-0.94). There was a significantly decreased trend in the risk for lung cancer along with the increased number of mutation-typic G allele (Ptrend<0. 001). Conclusion: These findings demonstrate that -1304T>G genotype in promoter region of MKK4 gene may contribute to the decreased risk of lung cancer. Copyright© 2011 by TUMOR.

Acta Nutrimenta Sinica 1956;0(02):-
The dynamic variation of folate content in the whole blood from ?-irra-diated rats have been observed. The level of folate, which did not alter significantly 24 hours after whole-body exposure of 800 rads, decreased dramatically to about 50 percent at the 5th day and to about 67 percent at the 10th day. After irradiation the haemoglobin concentration in rats also decreased, but there was no coincidence between the dynamic variation of folate level and haemoglobin concentration. It is probable that ionizing radiation induces the folic acid malnutrition of rats.

Aim The changes of the levels of blood sugar and plasma insulin in obese woman pa-tients with insulin resistance and the effect of insulin intravenous infusion during oper-ation were observed. Methods 10 patients of normal weight(contrast group, group A)and 20 patients of obesity women(ASA grade Ⅰ- Ⅱ) undergoing selective surgerywere investigated. The patients of obesity were randomly divided into two groups(group B and C, respectively). Groups A and B liquids without glucose, and in group Ctransfuse liquids with glucose and insulin(4g glucose add 1U insulin) were transfusedduring operation. The levels of blood sugar and plasma insulin were measured beforeand after anesthesia, during operation(30min) and after operation respectively. ResultsThere was an insulin resistance phenomenon in patients with obesity. The levels ofblood sugar and plasma insulin in patients with obesity were remarkably higher thanthose in patients of normal weight. The levels of blood sugar and plasma insulin wereextremely raised after anesthesia and during operation. The levles of blood sugar wasinhibited as insulin was infused during operation, but no hypoglycemic reaction wasfound. Conclusion There are insulin resistance phenomenon in patients with obesity.Insulin infused during operation can prevent blood sugar rise and minimize glucosemetabolism turbulence.

Chinese Journal of Analytical Chemistry 2009;37(11):1651-1656
The derivatization towards the hydroxyl groups of steroid environmental endocrine disrupting chemicals, such as estrone(E1),17β-estradiol(E2),estriol(E3) and 17α-ethynylestradoil( EE2),with A,O-bis (trimethylsilyl) trifluoroacetamide ( BSTFA ) as derivatization reagent followed by gas chromatography-mass spectrometry(GC-MS) was studied. The effects of the amount of BSTFA,derivatization temperature and time on the derivatization of target compounds,as well as the stability of steroid derivatives, calibration curves, instrumental quantification limits were investigated. The cleavage mechanism of steroid derivatives characteristic fragment ions was also interpreted. The results showed that better derivatization results were obtained when the amount of BSTFA was 25 μL for 100 μL steroids standard mix solution at 0.01 g/L;it is unnecessary to heating in the derivatization, better results were obtained when the steroids were derivatized with BSTFA at room temperature (20℃) for 10 min. The derivatives of target compounds were stable because their relative response factors(RRFs) had no observable change when deposited at -20℃ for 48 h. Under the optimal conditions, good linear correlation was obtained and the instrumental quantification limit was 0. 3μg/L for E1 and E2,while it was 5μg/L for EE2 and E3.
