1.Constructing an artificial skin model of melanoma through seeding MV3 melanoma cells and HaCaT cells onto the de-epidermized dermis*

BACKGROUND:We have built the three-dimensional human skin melanoma model with human epidermal keratinocytes and MV3 melanoma cel s co-cultured on the de-epidermized dermis in vitro.
OBJECTIVE:To establish a skin model of melanoma by mixed culture of MV3 melanoma cel s and HaCaT cel s on the de-epidermized dermis in vitro.
METHODS:MV3 melanoma cel s and HaCaT cel s were mixed with different percentages and inoculated on the surface of de-epidermized dermis fol owed by a liquid culture and air-liquid culture, and then the tissue-engineered skin model was established in vitro. Routine biopsy immunohistochemical observation was performed on the constructed skin melanoma model.
RESULTS AND CONCLUSION:Hematoxylin-eosin staining showed that MV3 melanoma cel s were distributed on the surface layer of de-epidermized dermis and formed tumor masses, while the HaCaT cel s were mixed growth with tumor cel s and formed a typical epidermoid structure. Some tumor cel s infiltrated into the surface or deep of de-epidermized dermis and showed a tumor foci distribution. The CK10, CK-pan and S-100 proteins were positive for immunohistochemical staining. With the increasing of MV3:HaCaT cel percentage, CK10 and CK-pan gradual y down-moved from the surface, and changed from layer distribution to lumpy distribution, while the staining of S-100 protein was gradual y distributed layer-by-layer, and some area showed tumor-like distribution. The results show that the skin model of melanoma can be in vitro constructed successful y by mixed culture of MV3 melanoma cel s and HaCaT cel s on de-epidermized dermis.
2.Hemi or total arthroplasty replacement for severe damages of proximal part of humerus

[Objective]To appraise the indications,essential technical points and selection of prostheses of hemi or total arthroplasty of the shoulder for severe damages of proximal part of humerus.[Method]Twelve cases had an arthroplasty of the shoulder;4 total-arthroplasties and 8 hemi-arthroplasties of the shoulder;6 cases of four parts fractures,2 cases of humeral head osteonecrosis,1 case of osteonecrosis of humeral head and glenoid cavity,2 cases of proliferative arthritis with the shoulder rigidity,1 case of bone humor.The function of the shoulder was evaluated postoperatively according to the methods of Amercian Society of Surgeons for Shoulder and Elbow Joints.All cases were followed up with a mean duration of 14 months.[Result]Twelve cases revealed obvious relief of pain with absence of pain in 8 and mild in 4.Twelve cases had no prosthetic stems loosening.The motion range of the shoulder was improved with a mean abduction of 100.Ten cases had the satisfactory results for treatment.[Conclusion]Hemi or total arthroplastyof shoulder is effective for complicated proximal humeral damages.The key of success is related with scientific operative method,appropriate selection of prostheses,effective rehabilitation plan.
3.The evaluation of multislice spiral computed tomography angiography in the diagnosis of moyamoya disease

Objective To evaluate the diagnostic value of the multislice spiral computed tomography angiography (MSCTA) in the diagnosis of moyamoya disease and explore its future application. Method The image data of 10 patients with moyamoya disease undertaken MSCTA and digital subtraction angiography (DSA) were reviewed analysis. Results MSCTA could clearly show stenosis, multiple occlusion or abnormalities of the cerebral vessels. Volume-rendering helped to show the relationship between the abnormal vessels and the surrounding tissues. Combined maximum intensity projection (MIP) and multiplsnar reconstruction (MPB) images could clearly show abnormally increased vessels (moyamoya disease vessels). The rate of occlusion and stenosis showed by MSCTA were 66.2%(53/80)and 67.5%(54/80)by DSA. There was no significant difference between the two methods (P>0.05). The images of MSCTA were basically same as those of DSA. Conclusions MSCTA is sensitive in diagnosing moyamoya disease, which is an important basis for early diagnosis. Early diagnosis and treatment is effective in improving prognosis of moyamoya disease.

7.ISOLATION OF ROSMARINIC ACID FROM CALLUS OF PERILLA FRUTESCENS AND STUDIES OF ITS INHIBITION ON THE GROWTHS OF BACTERIA AND FUNGAL

Perilla frutescens callus were induced from leave explants on MS medium suplemented with NAA and 2,4-D. The Rosmarinic acid (RosA) content of dried callus was 0.85%. The RosA was extracted from the callus with 80% alcohol and purified through extraction with ethyl acetate and a Sephadex LH-20 column chromatography. The parity of the final product was 95% as analyzed by HPLC RosA could inhibit the growths of Escherichia colt, Staphylococcus aueris and Rhizotonia with MICs of 300, 400, 800?g/mL, respectively.

10.One-stage foci debridement for cervical tuberculosis

Objective To evaluate the effectiveness of one-stage debridement for cervical tuberculosis at different segments.Methods Clinical data of 54 patients (male 20,female 34) with cervical tuberculosis treated by one-stage debridement from Jan 1998 to Dec 2011 were reviewed retrospectively.The average age of these patients was 45.4 years (range,26-75 years).Among them,12 cases were involved in single level (C2 1 case,C4 2 cases,C5 3 cases,C6 4 cases,C7 2 cases); 36 cases in the adjacent two levels (C1,2 3 cases,C2,3 2 cases,C3,4 5 cases,C4,5 6 cases,C5,6 14 cases,C6,7 5 cases,C7T1 1 case); 5 cases in three levels(C4 6 2 cases,C5-7 2 cases,C3,5,6 1 case)and 1 case in four levels (C4-7).Five cases were involved in other spinal levels (T6 1 case,T 3 cases,L3,4 1 case).Comorbidity of cervical ossification of the posterior longitudinal ligament was found in 1 case.Before surgery,the mean VAS and JOA scores were 5.9 (range,4-9) and 10.5 (range,7-12) respectively and the mean Cobb angle of lesion segment was 26.7°± 9.1°.All cases underwent regular anti-TB treatment preoperatively,and surgical treatment were performed when blood sedimentation (ESR) was lower than 50 mm/1 h.According to the segmental involvement,different surgical approaches were performed including anterior debridement with anterior or anterior-posterior internal fixation and fusion,submandibular approach debridement with posterior occipital cervical fusion or atlantoaxial fusion.Results Mean follow-up duration was 27.3 months (range,13-52 months).Symptoms were improved significantly in all cases.Mean time of union was 3.2 months (range,2-4 months).At the last follow-up,the mean VAS and JOA scores were 5.9 (range,4-9)and 10.5 (range,7-12) respectively,and the mean Cobb angle of lesion segment was 6.8°.Regular anti-TB treatment was performed postoperatively for 18 to 20 months.Intraoperative esophageal injury was found in one case of upper cervical tuberculosis which was cured uneventfully after gastrointestinal tubation for 1 week.No cervical tuberculosis recurrence,graft loosening or pseudarthrosis was found in follow-up.Conclusion With preoperative anti-TB treatment,cervical tuberculosis can be treated by one-stage foci debridement according to the segmental involvement and deformity.Postoperative regular anti-TB treatment is a crucial factor for the final recovery of cervical tuberculosis.
