2.Determination of Chloramphenicol and Dapsone in Compound Chloramphenicol Cream by HPLC

OBJECTIVE: To establish the HPLC method for the determination of chloramphenicol and dapsone in compound chloramphenicol cream. METHODS: The analytical column was Hypersil ODS and the mobile phase consisted of 0.1%sodium heptanesulfonate solution (a mixture of 0.1% sodium heptanesulfonate 500 mL and dimethylformamide 5 mL plus acetic acid glacial 0.5 mL)-acetonitrile (75∶25) at a flow rate of 1.0 mL?min-1. The detection wavelength was 272 nm and the sample size was 20 ?L. RESULTS: The linear ranges of chloramphenicol and dapsone were 25.59~153.54 ?g?mL-1 (r=0.999 9) and 12.57~75.42 ?g?mL-1 (r=0.999 9), respectively. The average recovery rates were 100.62% (RSD=0.92%) and 100.93% (RSD=1.22%), respectively. CONCLUSION: The method is simple and accurate and can be used for the quality control of compound chloramphenicol cream.

Objective: To explore the myocardial protection of induction and reperfusion with warm blood cardioplegia for hypoxic immature myocardium. Methods: A hypoxic model of neonatal rabbits was established in this study. Some indexes,such as myocardial energy metabolism, water content, SOD, MDA and myocardial ultrastucture, were observed after induction and reperfusion with warm blood cardioplegia, comparing to cold crystalloid cardioplegia. Results: Hypoxic neonatal rabbits in this study presented cyanotis, hypoxemia and higher ratio of RV/(LV+S) simulating the pathophysiological changes in cyanotic congenital heart disease. It could be built and repeated easily in experimental researches of immature myocardial protection. Objective To investigate the serum level of vascular endothelial growth factor (VEGF) in patients with coronary artery disease (CAD) and its clinical significance. Methods The study included 36 patients with CAD and 10 non-coronary heart disease patients as the control. The concentrations of serum VEGF in the coronary veins and the aorta were measured by an enzyme-linked immunsorbent assay (ELISA). Total mononuclear cells were isolated from peripheral blood by Ficoll density gradient centrifugation, and were cultured in M199 medium supplemented with 20% fetal bovine serum. After 7 days culture, attached cells were collected. The effect of VEGF on the migration of endothelial progenitor cells (EPCs) were assayed with modified Boyden chamber assay. ve To evaluate the protective effect of low-dose aprotinin on lungs from acute injury induced by cardiopulmonary bypass(CPB).Methods Twenty-eight ASA II-III patients (13 male, 15 female) aged (40.39?8.51) years, weighing (56.79?7.61) kg scheduled for elective valve replacement, were divided into two groups of 14 each: control group and aprotinin group. Patients who had infectious disease or received glucocorticoid before operation were excluded. The mean duration of CPB was (127.93?52.96) min and the mean aortic cross-clamping time was (68.54?30.71) min.The patients were premedicated with intramuscular phenobarbital 0.1 and scopolamine 0.3 mg. Anesthesia was induced with midazolam 0. 1-0.2mg?kg, fentanyl 10-20?g,kg-1 and intubation was facilitated with vecuronium 0.1-0.12 mg?kg-1. Radial artery and right internal jugular vein were cannulated for direct BP and CVP monitoring. Anesthesia was maintained with intermittent boluses of fentanyl and vecuronium supplemented with enflurane inhalation. The patients were mechanically ventilated. Tidal volume was maintained at 10-12 ml?kg , respiratory rate 12 bpm and I:E ratio 1:2.In aprotinin group 1 ? 10~6KIU of aprotinin was added to the priming solution, 1 ? 10~6 KIU aprotinin was infused after induction of anesthesia until thoracotomy and 2.5 ? 105KIU.h-1 was infused from thoracotomy until the end of surgery. In control group aprotinin was replaced by same volume of lactated Ringer solution. Peripheral blood samples were taken before induction of anesthesia, before CPB and 1,24 h after termination of CPB, for determination of IL-10 and TNF-?. Respiratory index [P( A-a)DO2/PaO2] was calculated before CPB, 10 min and 60 min after CPB. Results There was no significant difference in age, weight, duration of operation CPB time and aortic cross-clamping time between the two groups. The TNF-? concentration was not significantly different before induction of anesthesia and CPB between the two groups. But at 1 and 24 h after CPB TNF-? level was significantly higher than the baseline value before anesthesia in control group and that in aprotinin group. The IL-10 concentration greatly increased at 1 h after CPB in both groups as compared with that before anesthesia, but was much higher in the aprotinin group than that in the control group. Objective To explore the role of sCD44v6 in the diagnosis of colorectum cancer. Methods ELISA was used to detect the concentration of sCD44v6 of 81 patients after or before surgery.Results The level of sCD44v6 in the group of colorectum cancer was 200.5?31.1 ng/ml, which was higher than that of normal controls (45.6?9.5ng/ml) and that of patients with coloritis (48.3?7.6ng/ml). The concentration of sCD44v6 in the group of colorectum cancer with metastasis(280.1?26.9ng/ml) was higher than that of the group without metastasis(160.9?34.4ng/ml). The level of sCD44v6 was related to tumor burden.Conclusions The level of sCD44v6 is closely related to colorectal cancer and its metastasis. It would be useful in the diagnosis, prognosis, and forcasting of metastasis.
10.Post-operative prevention of early complications of closed spinal dysraphisms in children

ObjectiveTo investigate the preventive strategy of early post-operative complications of closed spinal dysraphisms in children.MethodsOne hundred and nine children with closed spinal dysraphisms underwent surgery from January 2004 to December 2008,were enrolled in this study.After dural closure,the wound was washed completely with saline to clear the tissue debris to prevent postoperative infection. We routinely made a horizontal incision to prevent postoperative infection. Results After operation,all patients were recovered and were discharged from hospitalNo post-operative complications,including cerebrospinal fluid leakage, wound infection and flap necrosis, were encountered. ConclusionThe sophisticated microsurgical technique and the delicate surgical manipulation is the prerequisite for the prevention of early operational complication of closed spinal dysraphisms in children.
