1.Experience on Management of Hospital Preparation by Analyzing the Cause for a Set of ADR Events

OBJECTIVE:To provide reference for the quality management of hospital preparation in medical institution.METHODS:The relationship between the cause for some typical ADR events such as "Armillarisin A Injury","Xinfu Injury","Methotrexate Injury","Aesculus Wilsonii Rehd Injury" and drug production were discussed and analyzed by combining with status quo of preparation management in hospital.RESULTS & CONCLUSIONS:Above ADR events were connected with quality management in drug production closely.The quality management system of hospital preparation should be established to strengthen management of whole preparation process and guarantee the safety of hospital preparation.
2. Killer peptide-loaded nanoparticles: Characterization and evaluation of anticryptcoccal efficacy

Objective: To prepare killer peptide-loaded PLGA nanoparticles which have better effect in treating systemic crypotococcosis. Methods: We adopted a modified double-emulsion method to prepare the killer peptide-loaded PLGA noparticles, and the nanoparticles were characterized by using TEM, SEM, Dynamic Laser Scattering, drug loading analysis, ecapsulation efficacy assessment and in vitro release test. We also evaluated the antifungal activities of killer peptide and killer peptide-loaded PLGA nanoparticles against Cry ptOcoccus neoformans ATCC32609 by plate count method. In the in vivo study, the anticryptococcal efficacies of killer peptide and killer peptide-loaded PLGA nanoparticles were compared by observing the survival and colony-forming in vital organs in a systemic murine cryptococcosis model. Results1 We successfully synthesized killer peptide-loaded PLGA nanoparticles. In vitro anticryptococcal test showed that the killer peptide and killer peptide-loaded PLGA nanoparticles had significantly stronger anticryptococcal effect compared with the normal saline group (P<0.05), and there was no significant difference in the efficacies between free killer peptide and killer peptide-loaded PLGA nanoparticles (P>0.05). In vits test demonstrated that mice treated with killer peptide-loaded PLGA nanoparticles (3 mg/kg or 5 mg/kg) had longer survival period and less fungal burden in vital organs than mice treated with free killer peptide (P<0.05); there was no significant difference in fungal burden and survival period between empty PLGA nanoparticle treated group and normal saline group (P>0.05). Conclusion: The killer peptide-loaded PLGA nanoparticles are more effective in treating systemic murine cryptococcosis.
3. The development of quorum sensing phenomenon in pathogenic fungi

The quorum sensing commonly exists in procaryote kingdom, regulating various biologic functions. Recently similar phenomenon was also found in fungi world, which affecting both biofilm formation and dimorphism. In this article, we focus on the recent progress on quorum sensing of pathogenic fungi and discuss the possibility of taking quorum sensing molecule as a potential target for antifungl therapy.
4.Influence of fluoxertin on level of IL-2,IL-6 and TNF-αin brain tissue and serum of rats with post-stroke depression

Objective To explore the influence of fluoxertin on level of IL-2 ,IL-6 and TNF-α in brain tissue and serum of rats with post-stroke depression(PSD) .Methods 60 rats were divided into sham operation group ,depression group ,stroke group ,PSD group and fluoxertin group ,12 cases in each group .ELISA was used to detect the level of IL-2 ,IL-6 and TNF-αin brain tissue and serum .Results The level of IL-2 ,IL-6 and TNF-αin depression group ,stroke group and PSD group were significantly increased and higher than that in sham operation group and fluoxertin group(P<0 .05) .There was no significant difference between sham op-eration group and fluoxertin group(P>0 .05) .Conclusion Fluoxertin could significantly decrease the level of IL-2 ,IL-6 and TNF-αand play an important role in therapy of PSD .
5.Game Theory Analysis on Quality and Safety Control of Chinese Medicine Decoction Pieces from the Perspective of Supply Chain

Game relationship of mutual restriction and mutual connection exist among various segments of Chinese medicine decoction pieces supply chain. Any problems in any segments will affect quality and safety of the whole supply chain, and ultimately affect the safety of consumers. Based on the theory of supply chain, this article established a linear supply chain and static game model, including planting farmers, production and marketing society, processors of Chinese medicine decoction pieces, terminal consumers and middle sales section whichis made up of whole sale and retail markets and manufacturing enterprises of Chinese patent medicine. This article also proposed the suggestions about quality safety of Chinese medicine decoction pieces through quantitative methods in game theory.
6.Polyvinyl alcohol and its composite materials for tissue engineering scaffolds

BACKGROUND:Polyvinyl alcohol is a biocompatible and biodegradable polymer. It is widely used in clinical areas because of its water-soluble, film forming, emulsification, adhesiveness, tasteless, and nontoxic.
OBJECTIVE:To review the applications of polyvinyl alcohol and its composite materials in bone, cartilage, skin, vessels and other tissue engineering scaffolds.
METHODS:A computer-based online search of CNKI database from January 2000 to December 2011, PubMed database and Elsevier (ScienceDirect) database from January 1980 to December 2012, was performed by the first author with key words of“poly(vinyl alcohol), composite material, tissue engineering scaffold”both in Chinese and English. Literatures concerning polyvinyl alcohol and its composite materials in bone, cartilage, skin, vessels and other tissue engineering scaffolds were included, and repetitive research was excluded.
RESULTS AND CONCLUSION:Although there are not enough strength, complications and other shortcomings in vivo, due to its good biocompatibility and biodegradable properties, polyvinyl alcohol and its composite
materials have made great progress in tissue engineering applications from the laboratory to the pre-clinical
research. But its long-term effects need further research. It wil be a main research aim of scaffold materials in the future to improve the interaction of cel s with the scaffold materials by surface modification, to prepare biomimetic materials by cel microenvironment simulation, to improve the hydrophilicity, the adhesion of cel s, and cel
differentiation and proliferation, to bionic the structure and function of the natural extracel ular matrix by building three-dimensional porous structure and control ing the release of cel growth factors, to meet the need of tissue regeneration by congruity or harmony of degradation and mechanical strength.
7.Investigate the efficacy of autologous serum combined with recombinant human epidermal growth factor in repairing of corneal defect

AIM: To investigate the efficacy of autologous serum combined with recombinant human epidermal growth factor in repairing of corneal defect after corneal foreign body eliminating operation.
METHODS: Totally 92 patients with corneal foreign body(110 affected eyes)as the research object in our hospital from October 2015 to March 2016. All the patients were divided into the treatment group and the control group according to the random number table. There was no significant difference between the two groups in age, sex and disease course. Both groups received corneal foreign body elimination by slit lamp. The treatment group was given eye drops containing epidermal growth factor combined autologous serum and tobramycin after surgery, while the control group was only administrated with epidermal growth factor and tobramycin. Treatment effects were compared 1wk after treatment.
RESULTS: One week after the treatment, the cure rate in the treatment group(96.4%)was much higher than that in the control group(78.2%). Two days after treatment, the fluorescein staining in the treatment group was 98.2%, it was higher than that in the control group(83.6%). The concentration of TNF-α and IL-6 of the treatment group was lower than that in the control group. One week after the treatment, AS-OCT images showed that the repair of corneal matrix in the treatment group was closer to the preoperative normal level.
CONCLUSION: The treatment effect that AS combined rhEGF is much better than that only usde rhEGF, shortened healing time of corneal epithelial wound. It is more effective and security to treat traumatic corneal epithelial defect, better repair function to corneal matrix.

9.Expression of Pleurocidin from winter flounder in Escherichia coli and optimization of culture conditions.

To express Pleurocidin in Escherichia coli and to enhance the secretory efficiency of the fusion protein, the gene encoding Pleurocidin was ligated with Cherry DNA sequence via blunt-end ligation. Then this fusion gene was cloned into pET22b (+) vector and the recombinant plasmid was transformed into E. coli BL21 (DE3). Lactose was used to induce expression of fusion protein. The recombinant plasmid pET22b (+) -CP was successfully constructed and high-level expression of fusion protein was induced with lactose. Statistics showed that addition of glycine after 16 h of induction significantly enhanced the secretory efficiency of the fusion protein. After hydrolysis of the fusion protein by diluted hydrochloric acid and some further purification steps, r-Pleurocidin was obtained with antibacterial activity against E. coli DH5α and Bacillus subtilis BS168. In conclusion, the fusion protein was expressed in E. coli and biologically active r-Pleurocidin was obtained after hydrochloric acid cleavage and purification.

10.Effects of non-saccharomyces albicans metabolic products on the proliferation of human umbilical vein endothelial cell ECV304.

OBJECTIVETo evaluate the effects of non-Saccharomyces albicans metabolic products on the cell cycle distribution and proliferation of human umbilical vein endothelial cell ECV304 cells in vitro.
METHODSThe parallel dilution supernatant of Saccharomyces tropicalis, Saccharomyces krusei and Saccharomyces glabrata were prepared, and 1, 4, 16-fold(s) diluted concentration and control group were set up. The line of human umbilical vein endothelial cell ECV304 was cultured in vitro and treated by non-Saccharomyces albicans supernatant. The proliferous effect of ECV304 induced by non-Saccharomyces albicans supernatant after 24, 48, 72 h was detected by the methods of MTT, and the changes of cell density and cycle after 48 h were investigated by inverted microscope and flow cytometry.
RESULTSAt the 24th hour, all of the higher concentration (1-fold) of non-Saccharomyces albicans supernatant and the 4-folds diluted Saccharomyces krusei could promote ECV304 proliferation(P < 0.05). After adding various non-Saccharomyces albicans supernatant at 48h and 72th hour, Saccharomyces krusei supernatant and Saccharomyces glabrata supernatant significantly increased proliferation rate of ECV304, while Saccharomyces tropicalis supernatant group showed no significant change no matter which concentration was tested. At 48th hour after adding the non-Saccharomyces albicans supernatant, the ECV304 cells density treated by Saccharomyces krusei supernatant and Saccharomyces glabrata supernatant were significantly higher under the inverted microscope. The G0/G1 population of ECV304 cells decreased while cell proliferation index (PI) increased after incubated with Saccharomyces krusei supernatant and Saccharomyces glabrata supernatant for 48 hours (P < 0.05). Saccharomyces tropicalis group showed no significant change (P > 0.05).
CONCLUSIONThe metabolic products of Sacharoymces krusei and Saccharomyces glabrata could induce proliferation of ECV304 cell, which suggests non-Saccharomyces albicans should be undergone more attention clinically in detection and treatment.
