2.The analysis of ABR about newborns in the neonatal intensive care unit after failing hearing screening

Objective To investigate the auditory brainstem response (ABR) of newborns in the neonatal intensive care unit (NICU) after hear screening failure.Methods To test ABR of 833 healthy newborns with no perinatal diseases at the clinic,and 425 newborns born in NICU who failed the hearing screening of monaural or both-ear from March 2014 to May 2015.Results 49.6% of children′s wave V response thresholds were ≥30 dB in the NICU group and 36.1% children′s wave V response thresholds were ≥30 dB in healthy group.The hearing abnormality rate of the NICU group was significantly higher than that of the healthy group(P<0.05);The average auditory thresholds healthy of the group[(19.76±6.59)dB] was significantly lower than those of the NICU group[(41.52±20.35)dB](P<0.05);both ear latency for waves Ⅰ,Ⅲ and Ⅴ and interpeak latency for waves Ⅰ-Ⅴ in healthy group were significantly shorter than those in NICU group (P<0.05).Conclusion The hearing loss extent of the NICU group is greater than that of the healthy group.The ABR abnormality reflects auditory pathway is abnormal.
3.To evaluate the therapeutic effects of preoperative platelet transfusion for splenectomy and devascularization

Objectives To study the effect of preoperative platelet transfusion for splenectomy and devascularization in the prevention of intraoperative and postoperative bleeding.Methods The 230 patients with cirrhosis and portal hypertension who received splenectomy and periesophagogastric davascularization were divided into strata A,B and C according to the platelet counts.Stratum A patients had a platelet count of less than 30× 10/L,B between 30× 10/L and 50× 109/L,and C more than 50 × 109/L.The patients in each stratum were then randomly divided into a preoperative transfusion group (T group) and a non-transfusion group (NT group).The amounts of intraoperative bleeding,postoperative drainage in 48 hours after operation,rates of postoperative bleeding,and general medical conditions were compared.Results A comparison in stratum A showed lower amounts of intraoperative bleeding and 48 hour postoperative drainage,and a lower rate of bleeding in the T group (P<0.05).There were no significant differences between the T and the NT groups in strata B and C (P>0.05).Conclusions For patients with a platelet count lower than 30 × 109/L,preoperative platelet transfusion significantly reduced bleeding suggesting that preoperative platelet transfusion for splenectomy and periesophagogastric devascularization should be a routine.For those patients whose platelet count was above 30 × 109/L,platelet transfusion is not recommended.
4.The effects of medium pH and purification method on product rate of expression rhGM-CSF in recombinant E.coli

Purpose The aim is to study the influence factors of recombinant E.coli cell growth,product expression and purification,in order to raise the product rate of rhGM-CSF.Methods Three pH media were planed to observe the effects of pH on E.coli cell growth and expression with the indexes of the cell density and expression level.The influences of various treatments on product rate were observed with the indexes of the volume and purity of inclusion bodies and the yield,purity,activity of product.Using SDS-PAGE method for analysis of purity.The product bioactivity was determined by using TF1 cell and MTT colorimetric method.Results The cell density in the medium pH 7.0 was 2.22 g/L;the product expression rate was 23%.It was two times higher than other pH media.The inclusion body prepared by bacteria without freezing store was 2.5 times volume than freezing store.During renatura-tion PEG 4000 could raise 0.8 times active product.The optimum conditions used rhGM-CSF product rate could raise 8 times.Concluston The product rate of rhGM-CSF expressed in E.coli was affected by many factors.The optimum condition for culture,expression and purification could raise quite a number of product rate.
5.Effect of occlusal contacts on stress distribution in the dentin and peridentium after post-core crown restoration

BACKGROUND:The post-core crown system is an effective method for dental defect repair, but its repair outcomes are influenced by various factors;thereby, a further study is needed to optimize this method.OBJECTIVE:To investigate the stress distribution in different degrees of dental defect after post-core crown restoration under different occlusal contacts. METHODS:Three-dimensional models of the mandibular first molar were established by CT images, and models of complete natural teeth, six different defected teeth (total maxil ofacial slight and severe defects, distal middle maxil ofacial slight and severe defects, proximal middle maxil ofacial slight and severe defects) and two different post-core crowns (fiber post and zirconia post) were created by software. These models were imported into ANSYS software to analyze the stress distribution in dentin and periodontium under stable occlusal contact (ABC) and unstable occlusal contact (AC), respectively. RESULTS AND CONCLUSION:The stress in dentin and the periodontium after post-core crown restoration under AC was obviously higher than that under ABC, and AC affected the restored teeth more markedly. The post-core crown material with higher elastic modulus could induce higher stresses, and severe defected teeth showed higher stress distribution. Different defected types made no overt effects on the stress distribution in the periodontium. These results suggest that the B area contact is critical in regulating occlusal contacts of restoration.
6.Assay and Analysis on Three Virus Pathogen of Pneumonia in Children

Objective To explore the child pneumonia in infection circumstances of Coxsackie B virus(CVB), Cytomegalovirus(CMV),Epstein-Barr virus (EBV).Methods Indirect ELISA were used to detect the specific IgM antibody of CVB, CMV, EBV. Results The total positive rate,which suffer from the three kinds of viruses in 140 children wich pneumonias was 38.6%.The positive rates of the CVB- IgM, CMV- IgM, EBV- IgM were 24.3%,8.6%,6.5%.Conclusions CVB has the higher infection rate in the pneumonia, is one of the main causes in the infection of child pneumonia.CMV,EBV also has the certain infection rate, although the infection rate is not high, they can cause damage of many organs and systems .They should be watched out for and valued.
7.The mechanism research of STIM1 in breast cancer cells

Objectives To explore the calcium signaling mechanism of STIM1 in breast cancer cells. Meth-ods After SiRNA interruption, Western blot and Transwell were used to measure protein expression of STIM1 and cell migration in MDA-MB-231 cells respectively. The relationship between STIM1 and SOCE calcium signaling were analysed by Laser confocal microscopy. Western blots were used to measure protein expression of FAK after si-lence STIM1. Results The numbers of cells without STIM1 were significantly lower than those cells with STIM1 by Transwell assay. STIM1 mediated SOCE in MDA-MB-231. Blocking SOCE might inhibite cells migration. Si-lence STIM1 did not affect the expression or activation of FAK in MDA-MB-231 cells. Conclusion STIM1 influ-ences cell migration through SOCE pathway in breast cancer cells, which is independent on the expression or activa-tion of FAK.
8.The high-risk factors and audiology characteristics analysis of 952 cases of neonatal failed hearing screening

Objective To analyze the related factors in new born failed in hearing screening and its audiological characteristics . Methods From May 2009 to Febrary 2012 ,952 infants within the 3 to 6-month-old with detailed birth records and hearing screen-ing records were reviewed in the study .They were born in Chongqing and surrounding areas ,but failed in the first and second hear-ing screening .They were tested by auditory brainstem response (ABR) ,distortion product otoacoustic emissions (DPOAE) ,tympan-ometry and stapedius reflex .Results The normal ratio of tympanogram ,normal group(74 .25% ) was higher than the high-risk group(69 .61% )(P<0 .05);The pass proportion of DPOAE in normal group(34 .57% ) was higher than high-risk group(27 .94% ) (P<0 .05);In normal group ,ABR response threshold >30 dBnHL was 55 .80% ,60 .36% in high-risk group .High-risk group′s hearing abnormality rate was significantly higher than the normal group (P<0 .05);In normal group ,mean threshold of ABR was (47 .64 ± 22 .86)dBnHL .While it was(50 .58 ± 25 .02)dBnHL in high-risk group .The threshold was higher in normal group than high-risk group(P<0 .05) .Conclusion premature birth ,low birth weight(<1 500 g) ,hyperbilirubinemia ,neonatal asphyxia ,pal-ace the virus infection ,mechanical ventilation ≥ 5 days ,≥ 2 high-risk factors are risk factors for hearing loss .Newborn hearing screening is a long and arduous task ,We should enhance screening efforts in infant with high risk factors ,and actively prevent and treat neonatal perinatal risk factors and reduce the incidence of hearing loss .
9.Comparison of two different workflows of crossmaching by full-automatic blood grouping instrument

Objective To provide an optimal working process for crossmatching by full-automatic blood typing instrument.Methods Two workflows were applied to crossmatch test by full-automatic blood typing instrument.One was diluting red blood cells of donor samples to concentrate of 1%,the other was detecting directly the donor′s packed red blood cells.Compared consistency and processing time of the two different workflows.Results Cross match results of two workflows were consistent well(U=0,P>0.05).The average processing times before testing and undergoing testing were not significantly different(t=0.692,t=0.562,P>0.05),whereas the average processing times after testing and throughout testing were significantly different(t=146.485,t=67.053,P<0.05).Conclusion The workflow of diluting donor′s sample before testing saved processing time and better suits hospital having a large quantity of specimens.
10.Application of conscious sedation analgesia in superficial bladder tumor resection for elderly patients

Objective To evaluate the effects of conscious sedation analgesia during superficial bladder tumor resection for elderly patients. Methods Forty elderly patients who needed 2 μm laser resection for bladder tumors were treated with target controlled infusion (TCI) remifentanil and propofol. The initial target concentration of remifentanil was 2.5 ng/mL and that of propofol was 1 μg/mL. The target concentration of those two agents was adjusted until loss of painfulness to the operative stimulus. The MAP, HR, SpO2, RR and PetCO2, BIS and scores by OAA/S were compared between pre-and post-surgery at different time points (time before anesthesia, T0; time before analgesia, T1; time when the resection began, T2; 5 minutes after the resection began, T3;10 minutes after the resection began, T4; 15 minutes after the resection began, T5; the end of operation, T6;5 minutes after operation). Results All patients lived through the surgery under a conscious sedation. After the anesthesia, blood pressure, heart rate and breathing rate were declined, while oxygen saturation was unchanged. Conclusion TCI with remifentanil and propofol provides better hemodynamic stability, satisfactory sedation analgesia and faster recovery of surgery.
